Abstract
A composition for the determination of glutamic oxalacetic transaminase and glutamic pyruvic transaminase e.g. in the body fluids of animals or man comprises an a -keto-glutarate together with an aspartate (for glutamic oxalacetic transaminase) or with alanine (for glutamic pyruvic transaminase), glutamate dehydrogenase, nicotinamide adenine dinucleotide, 2:6-dichloroindophenol, phenazine methosulphate and a triethanolamine buffer of pH 7-8.5. In the presence of the transaminase a glutamate is formed from the first two components, and this is then oxidised to a -ketoglutarate by the glutamate dehydrogenase in the presence of, and with consequent reduction of, the dinucleotide: the dyestuff system (dichloroindophenol, phenazine methosulphate) is then reduced by the reduced dinucleotide. The materials may be separately packed in the form of the following aqueous solutions: (1) and (2) triethanolamine buffer, nicotinamide adenine dinucleotide, dichloroindophenol, aspartate or alanine (3) phenazine methosulphate, (4) glutamate dehydrogenase stabilised with glycerol and (5) a -ketoglutarate. Solutions 1, 3, 4 and 5 comprise the test reagent, solutions 2 (which has different proportions to solution 1) and 3 are used as a control. Alternatively the compositions may be impregnated into a suitable carrier, e.g. filter paper strips, and the product lyophilised.
