Abstract
1318568 Determination of dehydrogenases MILES LABORATORIES Inc 17 Sept 1970 44498/70 Heading B1X A composition for the assay of a dehydrogenase enzyme comprises nicotinamide adenine dinucleotide or nicotinamide adenine dinucleotide phosphate, a substrate for the enzyme, a terminal acceptor dye and meldola's blue as intermediary hydrogen carrier. The terminal acceptor dye may be 2-(P iodophenyl)-3 (pnitrophenyl)-5-phenyl tetrazolium chloride, bromide or iodide or 2,6-dichloroindophenol. The substrate-enzyme pair may be lactic acid, lactic dehydrogenase; glucose 6-phosphate, glucose-6-phosphate dehydrogenase; 6-phosphogluconate, 6-phosphogluconate dehydrogenase. The composition may be used in aqueous buffered solution or in or on a carrier which may be filter paper thickness 0.01 to 0.2", wood strip, synthetic plastics or woven or non-woven fabric. In use, the composition in solution and the sample are mixed and the rate of change of optical density at 500 mu observed.
