Abstract

1468849 Determining glutamate and glutamic transaminases DOW CHEMICAL CO 10 July 1974 [19 July 1973] 30597/74 Heading G1B A method for the determination of glutamate or glutamic transaminases comprises mixing a specimen to be analyzed with a solution of oxidized nicotinamide adenine dinucleotide, glutomate dehydrogenase, a tetrazolium dye and diaphorase or N-methyl phenazonium metho-sulphate, the solution additionally containing a transaminose substrate when glutamic transaminases are to be determined, incubating the mixture under conditions of pH and temperature conducive to the formation of a measurable colour and measuring the colour. The solution used is essentially free of ammonium ions. A buffer may be incorporated in the solution to provide a pH of 7À9 to 8À3. The tetrazolium dye is preferably 2-p-iodophenyl-3-nitrophenyl-5- phenyltetrazolium chloride. The method is particularly applicable to the analyses of biological fluids.
