Abstract

1278036 Determination of niacin WARNERLAMBERT CO 21 May 1970 [18 June 1969] 24702/70 Heading B1X [Also in Division C6] A diagnostic composition for the positive detection of niacin produced by mycobacteria comprises a bibulous material containing at least four reagent-impregnated zones, separated from one another by an untreated area of the bibulous material the zones containing respectively:- A, 0.2 to 6 mg alkali metal salt of para aminobenzoic acid, B, 0.6 to 20 mg sodium or potassium thiocyanate; C, 0.2 to 10 mg crystalline acid selected from citric acid, oxalic acid and malonic acid; D, 1 to 20 mg chloramine T. The material may also have zones containing E a chemically inert hydrophobic barrier composition and F, a dyestuff, zone F optionally also being coated with the barrier composition. The order A, B, C, D is preferred, but not essential. The bibulous material may be paper, felt, porous ceramic woven or matted glass fibre. The hydrophobic material may be polymethyl methacrylate and the dyestuff Sudan IV or Sudan Red GGA. In use, the strip may be placed with the non-hydrophobed end in a saline extract of the mycobacteria and the container immediately stoppered. A colour develops in the solution and/or on the strip on standing.
