Abstract

The invention comprises compounds of formula <FORM:1115954/C2/1> wherein RO- is a free, etherified or esterified hydroxy group, R1 is a C2- 4 alkyl group, Y is a carbonyl, ketalized carbonyl, hydroxymethylene or acyloxymethylene group, Z is a carbonyl, hydroxymethylene or acyloxymethylene group and the hydrogen atoms H at the junctions in ring C have trans-anti-trans configuration with respect to the group R1, and their preparation (1) by reaction of a 17-acyloxy-gona-1,3,5(10), 16 tetraene with (a) a peracid to give a 16a ,17a -epoxide followed by hydrolysis or hydrogenolysis; (b) a hydroborating agent, followed by mild oxidative hydrolysis; or (c) a hydroxylating or acyloxylating agent, (11) reaction of a 16-unsubstituted gonatrien-17-one with a nitrite to give a 16-oximino-compound and subjecting this to reductive hydrolysis, or (111) interconversion of the groups RO-, Y and Z. (\sB) - 3 - Methoxy and acetoxy - 17 - acetoxy-13b - ethyl - gona - 1,3,5(10),16 - tetraenes are prepared by enol esterification with isopropenyl acetate. The compounds of the invention have estrogenic, anti-lipaemic and pituitary gonadotrophin inhibiting activity and may be used in pharmaceutical compositions in combinations with a carrier, for example in forms suitable for oral, parenteral or rectal administration.
