Abstract

Ethambutol (I), and antitubular agent, is prepd. by condensing 2 moles of a cpd. (II): (+)-ROCH2-CHEt-NHOH (where R = H, alkyl, lower acyl or benzyl) with 1 mole of glyoxal in a suitable solvent, pref. EtOH, at between 0 degree C and the b.pt. of the solvent, pref. room temp. for between a few mins. and several hrs., to give a cpd. (III): and reducing (III) in the presence of a suitable catalyst, pref. Pd, to give (I), which is isolated and purified or converted into a non-toxic salt. Cpd. (II; R = H) is pref. prepd. by electrolytic redn. of 2-nitro-1-butanol, and sepn. of the obtd. racemate using a di-O-benzoyl-tartaric acid (opt. ring-substd. by NO2, halogen, alkyl or alkoxy), e.g. di-O-isopropylidene-2-ketogulonic acid.
