Abstract
Preparation of placental lactogen by (a) treating frozen human placenta in order to extract the proteins and the placental hormone; (b) precipitating the hormone from the solution eluted from an ion-exchanger by increasing the ionic strength with (NH4)2SO4; (c) suspending the precipitate in NH4HCO3 and passing through a column of 'Sephadex' C25 (RTM) and a column of 'Sephadex' G100 buffered with NH4HCO3 in order to eliminate (NH4)2SO4 and to equilibrate the protein; (d) eliminating higher molecular impurities by recycling; and (e) precipitating the resulting placental lactogen with (NH4)2SO4, dialysing in the presence of H2O and lyophilizing. Placental lactogen is useful inter alia for diagnostic purposes.
