Claims (6)
1. Användning av pSG5-replikonen för framställning av en temperaturkänslig hybridplasmid.
2. Användning av pSG5-replikonen för detektion av IS-element och transposoner, kännetecknad därav, att en marker insätts i en plasmid omfattande pSG5-replikonen, Streptomyces-organismer transformeras med den sä erhällna hybridplasmiden, transformanterna inkuberas 10 vid en temperatur över 36 °C, screenas pä basis av markern och DNA-omrädet, i vilket rekombination skett, analyseras.
3. Förfarande för framställning av Streptomyces-mutanter, kännetecknat därav, att hela DNA:t isoleras frän en Streptomyces-utg&ngsstam, det omvandlas 15 till korta fragment, dessa integreras i en hybridplasmid som omfattar markern samt pSG5-replikonen, den erhällna hybridplasmidpopulationen transformeras i utgängsstammen, transformanterna screenas under användning av pä markern ·' baserad screening, hybridplasmiderna elimineras genom att 20 höja temperaturen över tröskelvärdet och mutanterna screenas under användning av upprepad pä markern baserad screening.
4. Användning av Streptomyces-mutanter framställda medelst förfarandet enligt patentkrav 3 för isolering av ·*·'' 25 muterade gener, kännetecknad därav, att mute- rade gener isoleras ur de screenade mutanterna.
5. Användning av Streptomyces-mutanter framställda medelst förfarandet enligt patentkrav 3 för isolering av gener av motsvarande vildtyp, kännetecknad 30 därav, att motsvarande vildtypsgener isoleras med hjälp av *; de muterade gener som anges i patentkrav 4.
6. Användning enligt patentkrav 5, kännetecknad därav, att hybridplasmiden som omfattar pSG5-replikonen innehäller en marker som kan screenas i E. 3. coli och att man vid isoleringen av de muterade generna använder en kosmidbank.
