SEGUN ESTE METODO SE AISLA Y CARACTERIZA EL GEN HIS4, DESCUBRIENDOSE, AISLANDOSE Y CARACTERIZANDOSE UNA REGION REGULADORA RESPONSABLE DE LA PRESENCIA O AUSENCIA DE AMINOACIDOS UTIL PARA LA EXPRESION CONTROLADA DE PRODUCTOS POLIPEPTIDICOS EN LEVADURAS. TRANSFORMACION INTEGRADORA DE LAS CEPAS DE LEVADURA DEL GENERO PICHIA, QUE PROPORCIONA UN MEDIO PARA RECOMBINACION DE SECUENCIAS DE VECTORES EN EL ADN CROMOSOMICO DEL HOSPEDANTE. ESTA RECOMBINACION DA COMO RESULTADO Y QUE SE MANTENGAN ESTABLES SECUENCIAS DE ADN INSERTADAS COMO ADN EXTRACROMOSOMICO.
