Abstract

PROCEDIMIENTO PARA LA OBTENCION DE UN ADN QUE CODIFICA U POLIPEPTIDO CON ACTIVIDAD DE INTERFERON, SIGUIENDO TECNOLOGIA DE ADN RECOMBINANTE. SE OBTIENEN FIBROPLASTOS HUMANOS A PARTIR DE FORESKINA DERIVADA DE FETO O SIMILARES. SE AÑADEN UNA PEQUEÑA CANTIDAD DE INTERFERON A UN CALDO DE CULTIVO DE FIBROPLASTOS QUE CONTIENEN POLI (I): POLI (C) PARA INDUCIR LA SINTESIS DE ARNM DE INTERFERON. SE AÑADE CICLOHEXIMIDA PAR AUMENTAR EL NIVEL DE ARNM DE INTERFERON, SE RECOGEN LAS CELULAS Y SE DESTRUYEN, SEPARANDO LOS NUCLEOS. EL ARN TOTAL CITOPLASMICO SE EXTRAE CON FENOL, SE DISUELVE EN UNA DISOLUCION SALINA Y SE PURIFICA POR CROMATOGRAFIA EN COLUMNA. UTILIZANDO ESTE ARN SE SINTETIZA EL ADN. EL INTERFERON TIENE APLICACIONES TERAPEUTICAS EN ENFERMEDADES VIRALES, OSTEOSARCOMA Y CARCINOMA MAMARIO.
