Abstract

Procedimiento para la obtención de substratos enzimáticos de fácil desdoblamiento para proteasas, según la fórmula general. R1-p-Glu- A1-A2-NH-R2 en la que R1=h o un grupo protector preferentemente t-butiloxicarbonilo, benzyyloxicarbonil); A1=Gly, Ala, Val, Leu, Ile, Ser, Thr, Pro, Pip, Phe o Tyr A2= Arg O Lys R2= un grupo hidrocarburo aromático posiblemente sustituido en el que -NH-R2 es un grupo determinable por medios físicos-químicos, preferentemente un grupo cromogénico o fluorogénico que se desdobla con una enzima presente y forma a continuación un producto de excisión de fórmula H2N-R2, cuya cantidad se puede cuantificar. Caracterizado porque la síntesis se realiza gradualmente, acoplando a A2 que lleva R2 o un grupo protector removible de carboxilo que a continuación se sustituye por R2 o partiendo de R1-p-Glu-A en donde R1 es un grupo protector, preferentemente t-butiloxicarbonilo o benciloxicarbonilo y acoplando a A2-NH-R2 en donde R2 se puede introducir separadamente en sustitución de un grupo protector removible de carboxilo.
