Abstract
Un polipéptido con una longitud menor de 44 restos de aminoácidos, en donde dicho polipéptido comprende, en un orden desde el extremo amino al extremo carboxi: (a) la secuencia mostrada en SEQ ID NO:1 o la secuencia mostrada en SEQ ID NO:1 con no más de tres adiciones o sustituciones de aminoácidos, (b) la secuencia mostrada en SEQ ID NO:2 o la secuencia mostrada en SEQ ID NO:2 con no más de cinco adiciones, sustracciones o sustituciones de aminoácidos, y (c) la secuencia mostrada en SEQ ID NO:3 o la secuencia mostrada en SEQ ID NO:3 con no más de tres adiciones, sustracciones o sustituciones de aminoácidos.

Ejemplo 3-Evaluación de CU-NP en células endoteliales de aorta humana in vivo
Se ensayó CU-NP en células endoteliales de aorta humana (HAEC), y se definió el trascurso del tiempo de 5 activación de GMPc in vivo. Se incubó CU-NP con HAEC (pasos 2 a 5, a 80 a 90 % de confluencia) a 10-10, 10-8 o 10-6
M durante 10 minutos en un incubador de CO2. CU-NP (n=6) o CNP (n=3) se administraron por infusión intravenosamente a perros anestesiados normales a 14 pmol/kg/min durante 75 minutos. Se recogió sangre preinfusión, a 25, 30, 45, 60, 75 minutos durante la infusión (I) y a 1, 2, 4, 6, 10, 20, 30, 45, 60 minutos post-infusión (post-I). Se cuantificó el GMP cíclico por radioinmunoensayo.
10 Tal como se muestra en la Tabla 3, CU-NP estimuló la producción de GMPc en HAEC (P<0,01 para 10-6 M frente a no tratamiento). Además, CU-NP incrementó GMPc en plasma in vivo frente a pre-I (todos los momentos durante I y post-I, P<0,01, excepto a 45 minutos post-I, P<0,05). CU-NP activó GMPc a un mayor grado en comparación con CNP (P<0,001, 25 minutos I a 30 minutos post-I). Por tanto, CU-NP activó significativamente GMPc en HAEC, y
15 también estimuló significativamente GMPc a mayor grado que CNP in vivo.

20 Ejemplo 4 –acciones estimulantes de GMPc de CU-NP en glomérulos caninos aislados
Se condujeron experimentos para determinar si CU-NP estimula directamente GMPc en glomérulos aislados, y para comparar las acciones activadoras de GMPc de CU-NP con CNP, URO y CNP-C, que consistían en la longitud completa de 22 aminoácidos de CNP con un duplicado del extremo N fusionado en la posición C terminal. Los 25 glomérulos se aislaron tras la extracción de riñones caninos normales. CU-NP, CNP, URO, y CNP-C (10-5 M) se incubaron con glomérulos frente a control. Se midió GMP cíclico por RIA, con corrección para niveles de proteína. CU-NP y URO suscitaron mayores respuestas de GMPc que los respectivos controles (P<0,01), pero sin diferencia entre grupos. CU-NP y URO estimularon mayores respuestas de GMPc frente a CNP y frente a CNP-C (P<0,001). Por tanto, CU-NP estimuló GMPc en glomérulos en mayor grado que CNP, pero en similar grado que URO. CNP-C

35 Ejemplo 5 – Efectos de hemoconcentración de CU-NP
Se estudiaron tres NP humanos (BNP, CNP, URO) y CU-NP para evaluar sus efectos sobre la permeabilidad vascular manifestada por un incremento en hematocrito. Perros anestesiados normales recibieron intravenosamente infusiones de BNP (n=7), CNP (n=6), URO (n=5), o CU-NP (n=6) a 14 pmol/kg/minuto. Se recogió sangre en tubos
40 EDTA sobre hielo y se centrifugaron. Los datos se informan para la línea base y a 60 minutos de infusión. Además de incrementar el hematocrito (Tabla 5), CU-NP redujo la presión de ECP in vivo (línea base 6 ± 0.9 mmHg, 60 minutes 3 ± 0.9 mmHg; P<0,05). Por tanto, los efectos de la hemoconcentración de CU-NP pueden contribuir a sus acciones de descarga cardiaca in vivo.

Ejemplo 6 – Acciones cardiorrenales y neurohumurales de CU-NP en insuficiencia cardiaca experimental canina
5 Se evaluó CU-NP en insuficiencia cardiaca (IC) canina para determinar si CU-NP activaría el segundo mensajero GMPc y ejercería acciones favorables sin excesiva hipotensión. Se indujo IC suave marcando el ritmo (180 lpm durante 10 días). CU-NP se administró por infusión intravenosamente en 6 perros anestesiados a 75 ng/kg/minuto durante 75 minutos. TFG se midió por la depuración de inulina. Se valoró la reabsorción fraccional de Na+ (RFNa)
10 por depuración de Li+. Los datos se recogieron a pre-infusión (pre-I), 30 y 60 minutos I, y post-I. La actividad de la renina en plasma, la angiotensina II, la aldosterona y el GMPc también se midieron. Los resultados (media ± EEM) se presenta en la Tabla 6.
CU-NP incrementó GMPc en plasma, la excreción urinaria de GMPc, la generación de GMPc renal neta, el flujo de
15 orina, y la excreción urinaria de Na+. Se redujeron RFNa proximal y distal. Se aumentaron el flujo sanguíneo renal y la TFG, con un descenso suave en PAM. Tanto GC como RVS eran iguales. Se redujeron PECP y PAP. Se suprimieron también la actividad de renina en plasma, la angiotensina II y la aldosterona. Por tanto, CU-NP activó GMPc en IC canina y ejerció las acciones de aumento renal, descarga cardiaca y de supresión del sistema reninaangiotensina-aldosterona sin excesiva hipotensión.
