Abstract
Un método para producir una planta de pimiento dulce Capsicum annuum transgénica fértil, dicho método comprende: (a) transformar una célula vegetal de pimiento dulce Capsicum annuum con un vector de expresión que codifica una proteína babyboom (BBM), en donde dicho vector de expresión produce una actividad de transcripción nuclear inducible de dicha proteína babyboom (BBM); (b) regenerar dicha célula vegetal de pimiento dulce Capsicum annuum transformada en estructuras tipo brote (SLS) en condiciones inductoras que dan como resultado una actividad de transcripción nuclear de dicha proteína babyboom (BBM), dichas condiciones inductoras comprenden poner en contacto dicha célula vegetal de pimiento dulce Capsicum annuum transformada con un medio que comprende un agente que produce dicha actividad de transcripción; (c) cultivar dichas estructuras tipo brote (SLS) en una planta de pimiento dulce Capsicum annuum transgénico fértil en condiciones no inductoras que dan como resultado la ausencia substancial de actividad de transcripción nuclear de dicha proteína babyboom (BBM); en donde dicha proteína babyboom (BMM) tiene al menos 70%, preferiblemente al menos 80%, más preferiblemente al menos 90%, incluso más preferiblemente al menos 95%, y lo más preferiblemente al menos 99% de identidad de secuencia a nivel de aminoácido con la SEQ ID NO: 1 en condiciones en que dicha proteína babyboom (BBM) tiene actividad de transcripción.

Se describe la secuencia de nucleótidos que codifica una proteína BBM que está operativamente ligada a un elemento genético seleccionado del grupo que consiste en un activador de transcripción, un activador de 5 transcripción o un sistema de localización nuclear.
Tales elementos genéticos permiten el control de la actividad de la proteína BBM para permitir una actividad espacial y/o temporal de la proteína BB6M. En esta memoria la actividad espacial quiere decir que se puede lograr actividad de BBM específica de un órgano o tejido. Una ventaja de usar tales elementos genéticos es que se puede prevenir la aparición de problemas relacionados con la expresión o actividad inapropiadas de la proteína
En una realización preferida la proteína BBM está operativamente ligada a un sistema de localización nuclear. En esta realización es posible controlar la actividad de BBM originando la proteína BBM::GR fusionada translacionalmente para migrar o translocarse al núcleo.
Cuando el elemento genético según la invención es un activador de transcripción, es posible regular la actividad de
15 BBM a un nivel transcripcional. Tal sistema de inducción de la transcripción puede ser un sistema que comprende un promotor inducible por etanol o un promotor inducible por choque térmico. Se puede usar un activador de traducción, que se puede localizar en la secuencia 5’UTR o 3’UTR, para regular la actividad BBM mediante control de la traducción. Quedará claro para el experto qué sistemas se pueden usar para inducir actividad BBM según los elementos genéticos mencionados anteriormente. Otros sistemas apropiados incluyen un sistema inducible por
20 estrógeno, un sistema inducible por PRP, un sistema inducible por UAS, cualquier sistema que comprenda VP16.
En una realización preferida de la presente invención la secuencia de nucleótidos que codifica una proteína BBM está operativamente ligada a una secuencia de nucleótidos caracterizada por la SEQ ID NO: 2. El péptido codificado por esta secuencia de nucleótidos permite que se induzca la actividad de la proteína BBM cuando el material vegetal se pone en contacto con un medio que comprende dexametasona (DEX).
25 Según la presente invención, la célula vegetal o material vegetal transformado regenerador se pone en contacto con un medio que comprende un agente adecuado para la inducción de actividad de la proteína BBM.
Se describe el etanol como el agente que es adecuado para la inducción de la actividad de la proteína BBM. El etanol se puede usar para inducir la transcripción del gen exógeno BBM. También son adecuados otros sistemas

5 Discusión
El resultado muestra que usando un gen BBM sobre-expresado inducible, se pueden generar brotes de buena calidad de plantas de pimiento dulce transgénicas directamente o a través de embriogénesis somática y que el transgén se hereda a la siguiente generación.
Se ha estudiado intensivamente la inducción de la formación de embrión somático en especies y variedades de
10 pimiento picante que conduce a la regeneración en plantas normales. En el pimiento dulce, sin embargo, fue posible la inducción de embriones somáticos pero los embriones carecen del meristemo apical y no se desarrollan

en plantas normales. Mediante activación del gen BBM durante el proceso de regeneración fue posible regenerar plantas de pimiento dulce fértiles.
Ejemplo 2
Transformación genética de Petunia recalcitrante W138 con BBM
La línea de Petunia W138 ha demostrado ser recalcitrante a la regeneración y la posterior producción de plantas transgénicas.
En un intento de obtener plantas de Petunia transgénicas maduras, se transformaron explantes de W138 usando un protocolo estándar para la transformación de petunia.
Se transformaron dos líneas de origen W138 (NC2676-10, NC2676-11) y una de origen W5 (293) con el constructo BBM inducible (35S::BBM:GR), o un constructo T-DNA control, en este caso que contiene el gen reportero GUS. Se obtuvieron callos resistentes a Kanamicina de todas las tres líneas con ambos constructos, con la mayor eficiencia para NC2676-11, seguido de 293. Los callos del constructo control presentaron una fuerte expresión GUS.
Aparecieron primero estructuras tipo brote (SLS) y primordios en la línea NC2676-11 seguido de las otras en el mismo orden, aunque se observó desarrollo y crecimiento solamente en callos transformados con 35S::BBM:GR (Fig. 14 y 15).
Al igual que en el pimiento dulce, la sobre-expresión de BBM lleva a la formación de brotes transgénicos en una planta recalcitrante.
Ejemplo 3
Análisis de la progenie de líneas 35S::BBM:GR
Se creció la progenie homocigota T1 de múltiples líneas 35S::BBM:GR in vitro en medio de co-cultivo que comprende además TDZ, DEX y/o TDZ y DEX o sin TDZ/DEX como un control.
El cultivo de explantes de cotiledón de líneas 35S::BBM:GR homocigotas reveló diferencias de desarrollo causadas por la proteína BBM inducida nuclear y no nuclear. En el borde del cotiledón donde ha sido transversalmente cortado se forman embriones somáticos (Fig. 19). Estos efectos no se observan en explantes de cotiledón crecidos en medio sin DEX.
Los resultados en esta memoria muestran que se puede usar la expresión de BBM exógena y controlada para producir brotes morfológicamente normales y de alta calidad de pimiento dulce transgénico. Tales embriones somáticos de hecho tenían una semejanza sorprendente con embriones cigóticos. De hecho, se pueden inducir un alto número (>100) de embriones somáticos en cotiledones de pimiento. La alta cantidad de tales embriones que se pueden identificar en explantes de pimiento es suficiente para permitir un factor de multiplicación de al menos 100 veces a alcanzar en la generación de plantas de pimiento dulce maduras en cultivo de tejidos. Este hallazgo también permite multiplicar plantas de pimiento dulce estériles masculinas sin la necesidad de usar una línea de mantenimiento.
