Abstract
Una molécula de ácido nucleico aislada que comprende una secuencia de nucleótidos que tiene la capacidad de iniciar, en una planta, la transcripción de un gen de una manera preferida de tejido de xilema y cámbium, en la que dicha molécula de ácido nucleico aislada comprende una secuencia de nucleótidos como se expone en la SEQ ID NO: 6.
Description
grupos, se expresaban preferentemente en los tejidos del cámbium y/o xilema de Populus. Estas doce regiones promotoras corresponden a las secuencias desveladas en el presente documento como SEQ ID NOS: 1-12.
Para el aislamiento de regiones promotoras específicas, se diseñaron pares de promotores específicos de genes
5 (normalmente de una longitud de 30 nt) de las secuencias de los cóntigos promotores descritos anteriormente, para ampliar por PCR un fragmento de 700 a 3.500 nucleótidos desde la región promotora de cada uno de los doce genes cuyas secuencias promotoras se desvelan en el presente documento. La primera ronda de PCR se realizó sobre una muestra de ADN genómico de Populus deltoides o P. trichocarpa, que se preparó de hojas usando el método de extracción del bromuro de cetil-trimetil amonio (CTAB) (Aldrich y Cullis (1993) Plant Mol. Biol. Report.
10 11:128-141). Los cebadores se diseñaron para amplificar la región aguas arriba de la región codificante, es decir, la región no traducida 5’ y la región promotora del gen seleccionado. A continuación se ofrecen las secuencias de los cebadores usados para cada promotor:

La PCR se realizó usando reactivos disponibles en el comercio y los parámetros de ciclo de 5 minutos a 94 ºC seguido de 35 ciclos de 94 ºC durante 1 minuto, después una temperatura de hibridación modificada, como se describe más adelante durante un minuto, después 72 ºC durante 3 minutos. La temperatura (T) de hibridación se 20 ajustó para cada par de cebadores y varió de 50 ºC a 59 ºC. Finalmente, las muestras se mantuvieron a 72 ºC durante 7 minutos, después a 4 ºC hasta análisis posterior. Diez µl de cada uno de los fragmentos de ADN amplificados resultantes se procesaron en un gel de agarosa al 0,8 %, se purificaron usando el kit de Purificación de gel GFX (Amersham), se subclonaron en el vector pGEM-T-Easy (Promega) y después en sitios EcoRI y BgIII del vector pAPROM-ATG. Las secuencias finales se determinaron en los plásmidos resultantes. La Figura 1 ilustra
25 esquemáticamente el casete de expresión pAPROM-ATG que comprende el gen GUS unido operativamente a un promotor desvelado en el presente documento. La Figura 4 ilustra esquemáticamente el vector plasmídico que comprende un gen de interés unido operativamente a un promotor de la invención.
EJEMPLO 3
30 Transformación de plantas de Arabidopsis
Usando un protocolo de transformación mediado por Agrobacterium tumefaciens (Bechtold et al., (1993) C. R. Acad. Sci. Paris 316:1194-1199; Bent et al., (1986) Mol. Gen. Genet. 204:383-396) se transformaron plantas de 35 Arabidopsis thaliana Columbia con construcciones individuales que contenían uno cualquiera de los promotores de la invención unido operativamente a un gen de interés. Las construcciones también contenían el gen marcador de selección Bar que confiere resistencia a análogos de fosfinotricina herbicida como glufosinato de amonio (Thompson et al. (1987) EMBO J. 9:2519-2523). En este ejemplo, el gen de interés unido operativamente a los promotores preferidos de cámbium/xilema de la invención es el gen indicador GUS que codifica la enzima beta-glucuronidasa
40 (GUS) (Jefferson (1987) Plant Mol. Biol. Rep. 5: 387-405) que posibilita la inspección visual del fenotipo deseable, es decir, la expresión de GUS de una manera preferida de cámbium/xilema.
Se sembraron semillas de Arabidopsis thaliana ecotipo Columbia en macetas que contenían vermiculita. Las plantas se cultivaron en un régimen de oscuridad/luz de 16/8 horas a 22 ºC. Después de 4-5 semanas, las plantas se
45 transformaron con la cepa GV3101 de Agrobacterium tumefaciens de acuerdo con Bent et al., (1986) Mol. Gen. Genet. 204:383-396; que lleva el vector plasmídico que comprende el gen de interés unido operativamente a cada uno de los promotores descritos en el presente documento.
Para la transformación de la planta, 1 litro de medio LB que contenía rifampicina, gentamicina y kanamicina se
50 inoculó con una alícuota de cultivo iniciador de Agrobacterium durante una noche. Después, el cultivo se dejó crecer durante una noche a 28 ºC en un agitador rotativo, hasta que la DO600 fue ≥ 8,0. La Agrobacterium se precipitó por centrifugación y el sedimento bacteriano se resuspendió en 300 ml de sacarosa al 5 % y Silwet L-77 al 0,03 %. Esta suspensión de Agrobacterium se pulverizó sobre las plantas. Las macetas se colocaron en una bandeja que se cubrió con una envoltura de plástico para mantener la humedad y las plantas se dejaron crecer bajo el régimen
55 anterior para madurar y asentar las semillas.
Las semillas se recogieron y la superficie se esterilizó con una solución que contenía lejía 50 % y Tritón X-100 al 0,02 % durante 7 minutos. Después, las semillas se lavaron 3 veces con agua destilada estéril y se sembraron en placas en medio MS que contenía 6 mg/l de Finale como agente de selección. Después de 5 a 7 días, los
