Vector vírico que comprende un ácido nucleico codificante de la colesterol 24-hidroxilasa para su utilización en el tratamiento de la enfermedad de Alzheimer en un sujeto, en el que dicho vector vírico debe administrarse directamente en el cerebro del sujeto o mediante inyección intravascular o intratecal.

Análisis de oligómeros Aβ solubles
En primer lugar se homogeneizó el córtex e hipocampo diseccionados en un homogeneizador Dounce en Tris-HCl
5 10 mM, pH 6,8, complementado con cóctel inhibidor de proteasas (Complete, Roche). Se ajustó la concentración de proteínas a 6 mg/ml para todas las muestras. Se llevó a cabo una primera etapa de ultracentrifugación (100.000g a 4ºC) para recoger el sobrenadante Tris y el sedimento se disolvió en un tampón de Tris-HCl 10 mM, Triton al 2% (pH 6,8). Otra etapa de ultracentrifugación (100.000g, 4ºC) permitió eliminar el sobrenadante de Triton y se resuspendió el sedimento en un tampón de Tris-HCl 10 mM, SDS al 0,5% (pH 6,8). Tras una última etapa de ultracentrifugación
10 (100.000g a temperatura ambiente), se recogió el sobrenadante de SDS y el sedimento se disolvió directamente en solución Blue Laemmli.
Aislamiento de membranas resistentes a detergentes (MRD)
15 Se aislaron balsas lipídicas tal como se ha descrito previamente (Vetrivel et al., 2004). En primer lugar se lisó un sedimento de diez millones de células en tampón MBS (MES 25 mM, NaCl 150 mM, EDTA 1 mM, pH 6,5) que contenía Triton X-100 al 1% y cóctel inhibidor de proteasas (Complete, Roche).
Todas las etapas se llevaron a cabo a 4ºC. Se ajustó la concentración de proteínas a mg/ml y se introdujeron los
20 lisados celulares en yodixanol al 40% (gradiente de densidad OptiPrep, Sigma) diluido en un tampón apropiado (sacarosa 0,25 M, EDTA 6 mM, tricina 120 mM, pH 7,6). Se superpusieron dos capas de tampón de yodixanol al 30% seguido de al 20% en la parte superior del tubo de ultracentrífuga. Tras la ultracentrifugación a 39.000 rpm durante 20 h a 4ºC, se recogieron fracciones (de 1 ml) y se analizaron mediante transferencia western siguiendo el protocolo estándar. SE utilizó la detección de flotilina-2 para identificar las fracciones de MRD.
25 RT-PCR cuantitativa
Se llevó a cabo la extracción del ARN mensajero de células o tejidos utilizando el kit RNAble (Eurobio Laboratories). Se llevó a cabo la RT-PCR cuantitativa en tiempo real en el sistema de detección de secuencias ABI Prism 7700
30 (Perkin-Elmer Applied Biosystems) tal como se ha descrito (Bieche et al., 2004).
Como control de ARN endógeno, los presentes inventores cuantificaron los transcritos del gen de proteína de unión a caja TATA (PUT). Se normalizó la cantidad de transcrito diana (Ndiana) basándose en el contenido de PUT en cada muestra y posteriormente se normalizó respecto a un nivel basal de ARNm utilizando la ecuación: Ndiana=2∆Ctmuestra,
35 enlaque ∆Ct es el valor Ct del gen diana menos el valor Ct del gen PUT. Se listan los cebadores en la Tabla 2.

24S-hidroxicolesterol se incrementó en dos veces en el córtex cerebral e hipocampo diseccionados de ratones en los que se había inyectado el vector VAA5-CYP46A1wt, mientras que el colesterol total no experimentó cambios (figura 6C).
La expresión del gen CYP46A1 estaba asociada a una reducción marcada del número de placas amiloides (63% a 68%) y del porcentaje de área ocupada por depósitos amiloides (71%) en el hipocampo y córtex cerebral (figura 7A, B). La mayoría de los depósitos amiloides remanentes se concentraron en regiones en las que muy pocas o ninguna célula expresaba la proteína CYP46A1wt. Los péptidos Aβ40 y Aβ42 evaluados mediante ELISA se redujeron en 50±3% y 57±4%, respectivamente, en muestras agrupadas de córtex cerebral e hipocampo (figura 7C). La cantidad de oligómeros Aβ hexaméricos y dodecaméricos se encontraba ligeramente reducida sin alcanzar la significancia estadística. En contraste, los oligómeros Aβ triméricos se redujeron en 30% a 50% (P<0,05, figura 7D). El análisis de transferencia western mostró que la cantidad de producto C83 de corte de la α-secretasa permanecía sin cambios, mientras que el producto C99 de corte de la β-secretasa se incrementaba en 40% en ratones en los que se había inyectado VAA5-CYP46A1wt (figura 7E, F). Los fragmentos DICP generados por corte γ eran prácticamente indetectables (figura 7E), sugiriendo que la expresión de CYP46A1 inducía una reducción del corte de FTC por la γsecretasa in vivo.
La expresión de Adam9, 10, 17, Bace1 y Psen1 y, más inesperadamente, de los genes diana LXR (Abca1, Abca2, Abcg1, Abcg4, Abcg5 y ApoE) no resultó modificada. La expresión de los genes de la 3-hidroxi-3-metilglutarilcoenzima A reductasa (Hmgcr) y de la proteína 2 de unión a esterol (Srebp2) se incrementó en 1,6 veces (figura 7G) sin cambios en la expresión de los genes de la acil-coenzima A:colesterol aciltransferasa 1 (Acat1), del receptor de lipoproteína de baja densidad (Ldlr), de la proteína 1 relacionada con lipoproteínas de baja densidad (Lrp1), de lecitina:colesterol aciltransferasa (Lcat) y de C1 de la enfermedad de Niemann-Pick (Npc1).
La expresión de CYP46A1 reduce la microgliosis y mejora los déficits cognitivos antes de la aparición de depósitos amiloides
Los depósitos amiloides inducen una respuesta microglial y la astrocitosis en ratones PPA23 (Bornemann et al., 2001). Se redujo el número de células microgliales positivas para Iba-1 en 41±7% y 46±4%, respectivamente, en el córtex cerebral y el hipocampo de ratones en los que se había inyectado vector VAA5-CYP46A1wt (figura 8A, B). De manera similar, se redujo el número de células positivas para GFAP en prácticamente el 50% en ratones PPA23 en los que se había inyectado el vector VAA5-CYP46A1wt.
Los ratones PPA23 desarrollan déficits cognitivos a los 3 meses de edad, mientras que los depósitos amiloides sólo se detectan en el cerebro de los mismos a los 6 meses (Van Dam, 2003). Con el fin de evaluar los efectos de la inyección del vector VAA5-CYP46A1wt sobre las funciones cognitivos, los ratones fueron sometidos a ensayo a los 6 meses utilizando el procedimiento del laberinto acuático de Morris (LAM). Todos los ratones que habían recibido inyecciones de vector VAA5-CYP46A1wt o VAA5-CYP46A1mt mostraron rendimientos mejorados durante la etapa de adquisición (P<0,001) (figura 8C, D). Sin embargo, los inventores observaron un efecto significativo de la inyección de VAA5-CYP46A1wt sobre el parámetro longitud de trayectoria (P=0,054, RM-ANOVA de dos factores) sin ninguna interacción significativa entre la inyección del vector y el bloque de ensayo (P=0,583) (figura 8C). La ausencia de efectos de tratamiento sobre la velocidad de nado (figura 8E) indica que la mejora de la longitud de trayectoria no se debe a un mejor rendimiento motor. Los inventores no observaron ningún efecto significativo de la inyección de vector sobre la latencia de escape (figura 8D). Durante el ensayo de prueba, los ratones en los que se había inyectado vector VAA5-CYP46A1wt no permanecieron más tiempo en el cuadrante diana (figura 8F) pero cruzaron la posición anterior de la plataforma más frecuentemente (1,4±0,4) que los ratones PPA23 con tratamiento simulado (0±0, P=0,018), indicando que su memoria espacial había mejorado.
La expresión de CYP46A1 reduce la cantidad de colesterol y presenilina-1 en membranas resistentes a detergentes aisladas a partir de células neuroblastoides murinas que expresan PPA humano mutado
Con el fin de obtener más información sobre los mecanismos por los que la sobreexpresión de CYP46A1 reduce los fragmentos DICP generados por el corte γ in vivo, los inventores construyeron líneas celulares N2a neuroblastoides murinas para sobreexpresar constitutivamente el gen PPA humano que incluye las mutaciones sueca y London (PPAsl). Los inventores seleccionaron el clon 17 (en lo sucesivo denominado en la presente memoria N2a-PPA17), que se encontró que secretaba los niveles más altos de péptidos Aβ40/42 (figura 9A) y demostraron que dicho clon presentaba una expresión reducida del gen CYP46A1 murino y una reducción de 43% del contenido de 24Shidroxicolesterol (figura 9B).
A continuación, los inventores restauraron la expresión de la colesterol-24-hidroxilasa mediante la construcción de células N2a-PPA17 para sobreexpresar el gen CYP46A1 humano. Obtuvieron dos líneas celulares, denominadas N2a-PPA-CYP-A y -B, que mostraron un incremento del 24S-hidroxicolesterol sin cambios significativos en el nivel de colesterol total (figura 9C). En estas células, se observó una reducción de la secreción de los péptidos Aβ40 y Aβ42, que era inversamente proporcional al contenido incrementado de 24S-hidroxicolesterol (r2=0,91) (figura 9D). Los péptidos Aβ42 eran prácticamente indetectables en las células que expresaban el nivel más alto de gen CYP46A1. Este efecto no se debía a modificaciones de la expresión del transgén PPAsl ni de Adam9, Adam10,

Adam17, Bace1 y Presenilin1 (Psen1) murinos que participan en el procesamiento de PPA, ni del gen de la 3hidroxi-3-metilglutaril-Coenzima A reductasa (Hmgcr), el enzima limitante de la velocidad de la síntesis del colesterol, y tampoco de los genes del casete transportador A1 de unión a ATP (Abca1) y de la apolipoproteína E (ApoE).
Con el fin de caracterizar el procesamiento de PPA por las secretasas en las células N2a-PPA-CYP, los inventores cuantificaron los fragmentos C-terminales (FTC) de la PPA mediante transferencia western en extractos celulares brutos. El contenido del producto C83 de corte de la α-secretasa no resultó modificado en las células N2a-PPA-CYP-A y N2a-PPA-CYP-B. El péptido C99 producto del corte de la β-secretasa se incrementó en 37% en las células N2a-PPA-CYP-A y se redujo en 34% en las células N2a-PPA-CYP-B. El producto DICP de corte de la γ-secretasa se redujo en 50% a 65% tanto en células N2a-PPA-CYP-A como células N2a-PPA-CYP-B (figura 10A). La reducción concomitante de DICP y el incremento de β-FTC en las células N2a-PPA-CYP-A son consistentes con el hecho de que la sobreexpresión de CYP46A1 actúa principalmente al nivel del corte de FTC por la γ-secretasa. Al nivel más alto de expresión, CYP46A1 aparentemente también reduce el β-FTC sin modificar la cantidad de BACE1 en las MRD, lo que sugiere un efecto adicional sobre la actividad de la β-secretasa.
Existen cada vez más pruebas de que las balsas lipídicas, definidas bioquímicamente como membranas resistentes a detergentes (MRD), son los microdominios de membrana principales en los que se produce el procesamiento amiloidogénico de la PPA (Cordy et al., 2006). Las balsas lipídicas se encuentran enriquecidas en colesterol y el complejo de γ-secretasa co-reside con la PPA y la β-secretasa en estos microdominios (Li et al., 2000; Wahrle et al., 2002; Vetrivel et al., 2004). Los cambios en el contenido de colesterol en las balsas lipídicas puede influir marcadamente sobre la producción de Aβ (Ehehalt et al., 2003; Won et al., 2008).
En las células N2a-PPA17 que sobreexpresan el gen CYP46A1, la cantidad de colesterol se redujo en 28% y 45% en las dos fracciones de MRD positivas para flotilina-2 de células N2a-PPA-CYP-A y N2a-PPA-CYP-B, respectivamente (figura 10B). La PPA reside tanto en dominios de balsa como en dominios no de balsa y el porcentaje de PPA localizado en las dos fracciones de MRD positivas para flotilina-2 de las células N2a-PPA17 representa el 40% de la cantidad total de PPA (figura 10C). En las mismas fracciones enriquecidas en colesterol de las células N2a-PPA-CYP-A y N2a-PPA-CYP-B, la cantidad de BACE1 no resultó modificada pero la PPA se redujo en 32% y 45%, mientras que PSEN1, un componente de la γ-secretasa, se redujo en 25% y 67%, respectivamente (figura 10C). Conjuntamente, estos resultados sugieren que la reducción del corte de FTC por la γ-secretasa en células N2a-PPA17 que sobreexpresan CYP46A1 resulta de una reducción del colesterol en las MRD que se asociaba a un menor reclutamiento o a una estabilización de la PPA y de PSEN1 en los mismos microdominios.
