inyección intramuscular o subcutánea. En algunas realizaciones, la composición de la invención puede administrarse por vía oral. Como se usa en el presente documento, una "composición" se refiere a cualquier composición que contiene una cantidad farmacéuticamente eficaz de una apelina pegilada.
El tratamiento descrito en el presente documento puede usarse para tratar cualquier mamífero adecuado, que incluye primates, tales como monos y seres humanos, caballos, vacas, gatos, perros, conejos, y roedores tales como ratas y ratones. En una realización, el mamífero que va a tratarse es el ser humano.

EJEMPLOS
EJEMPLO 1
Apelina 36 conjugada con PEG
Se produjo satisfactoriamente una apelina-36 conjugada con PEG de 40 kDa (PEG-apelina-36) con conjugación del extremo N, alta pureza (>98 %) y mínima reducción de la afinidad de unión del receptor APJ Usando un ensayo de inhibición de adenilato ciclasas, se observó bioactividad in vitro comparable entre PEG-apelina-36 y apelina-36 no modificada. La evaluación in vivo de PEG-apelina-36 se realizó en ratas normales y ratas con infarto de miocardio (IM). Se evaluó la función cardíaca mediante ecocardiografía antes, durante una infusión IV de 20 minutos y hasta 100 minutos después de la infusión del péptido. Se observaron aumentos similares en la fracción de eyección (FE) cardíaca durante la infusión de PEG-apelina-36 y apelina-36 en ratas normales. Sin embargo, los animales que recibieron PEG-apelina-36 mantuvieron la FE significativamente elevada durante el periodo de monitorización después de la infusión de 100 minutos en comparación con los animales que recibieron apelina-36 no modificada. De forma interesante, los aumentos de la FE observados con PEG-apelina-36 y apelina-36 fueron mayores en ratas con IM. PEG-apelina-36 tuvo una vida en circulación prolongada en comparación con la apelina-36 en ratas. No hubo cambios en la tensión arterial aórtica cuando se administró PEG-apelina-36 o apelina-36.
EJEMPLO 2
Materiales y métodos:
PEGilación de apelina
Conjugación de apelina-36 (AnaSpec, Fremont, CA): La conjugación con aldehído-PEG ramificados de 10 kDa, 30 kDa y 40 kDa (NOF America Corp., White Plains, NY) se llevó a cabo a péptido 0,2 uM y PEG 0,4 uM en presencia de cianoborohidruro 20 mM (Sigma-Aldrich, St. Louis, MO) en tampón acetato sódico 0,1 M a pH 5. La conjugación se realizó a 25 ºC durante la noche y se extinguió con Tris 2 mM a pH 7,5. Los conjugados se dializaron contra agua para eliminar el péptido no conjugado. El conjugado de apelina-36 PEGilado de 40 kDa (PEG-apelina-36) se purificó adicionalmente por cromatografía de intercambio catiónico débil en una columna HiTrap SP FF (G.E. Healthcare, Piscataway, NJ) para eliminar el PEG libre. El conjugado se eluyó a concentración 1,5 M de sales de la serie de columna de 15 minutos. Se usó Nanodrop A280 para determinar la concentración del conjugado de apelina-36.
Determinación del sitio de PEGilación: Se secuenciaron en el extremo N 200 pmoles de apelina-36 y PEG-apelina36 en el secuenciador Procise (Applied Biosystems, Foster City, CA) usando el método de líquido pulsado preprogramado durante 18 ciclos. Se analizaron apelina-36 y PEG-apelina-36 también por espectrómetro de masas de MALDI-TOF (Voyager-DE PRO) (Applied Biosystems, Foster City, CA) en modo lineal usando DHB (ácido 2,5dihidroxibenzoico) como matriz. El voltaje de aceleración fue 25.000 V.
Ensayo de unión al receptor APJ: La unión se realizó usando competición con sonda de [Glp 65, Nle 75, Tyr 77]apelina 13 marcada con 125I para unirse al receptor APJ en una fracción de membrana de las células CHO transfectadas (Perkin Elmer, Waltham, MA). El péptido diluido en serie se incubó con membrana y sonda durante 1 hora a temperatura ambiente. La mezcla se filtró entonces sobre una placa de filtración de fibra de vidrio tipo C de 1,2 um (Millipore Corp., Billerica, MA), bloqueada previamente con 0,5 % de polietilenimina. La placa se lavó 4 veces con 200 ul de tampón de unión helado y luego otras 4 veces con Tris-HCl helado 50 mM, pH 7,4, se secó y se contó con fluido de centelleo. Los datos se analizaron con Graphpad Prism 4 (GraphPad Software, Inc, La Jolla, CA) usando regresión no lineal.
Ensayo de bioactividad in vitro
Generación de un clon celular con expresión del receptor APJ de alto nivel: Se mantuvieron células HEK293 (ATCC, Manassas, VA) a 37 ºC / 5 % de CO2 en medio Eagle modificado por Dulbecco (DMEM) complementado con Lglutamina, penicilina/estreptomicina y 10 % de suero bovino fetal (FBS). El plásmido pCMV6-XL4 (Origene Technologies, Inc., Rockville, MD) que contenía un ADNc que codifica APJ humano se subclonó en pCMV6-Neo (Origene Technologies, Inc.) para generar pCMV6-APJ. Se transfectaron transitoriamente células HEK293 con pCMV6-APJ usando Superfect (Qiagen Technologies Inc., Valencia, CA), y se seleccionaron en medio de selección
