Abstract

Procedimiento para el cultivo de células CHO para la producción de inmunoglobulinas o fragmentos de inmunoglobulinas, caracterizado por que se cultiva una célula CHO que produce sustancias con limitación de glucosa DGL, siendo el grado de limitación de glucosa DGL >= qGlc/qGlcmáx con qGlc >= tasa de consumo de glucosa específica observada en el momento y qGlcmáx >= máxima tasa de consumo de glucosa específica conocida para estas células CHO, mayor que el DGL que conduce al mantenimiento exclusivo, DGLmantenimiento, de la célula CHO y es >= 0,5, siendo el DGLmantenimiento >= qGlcmantenimiento/qGlcmáx, con qGlcmantenimiento >= tasa de consumo de glucosa específica observada durante el metabolismo de mantenimiento puro y qGlcmáx >= máxima tasa de consumo de glucosa específica conocida para estas células CHO, siendo la cantidad de glucosa suministrada no superior al 50 % de la que puede consumir como máximo el número de células esperado máximo sin limitación de glucosa, en el que se usa un medio que contiene glucosa que con respecto a otros componentes de sustancias nutritivas no está limitado antes del inicio de la limitación de glucosa, en el que el procedimiento se inicia como procedimiento discontinuo y se continúa como procedimiento continuo, en el que antes del inicio del procedimiento continuo se hace funcionar el cultivo como procedimiento discontinuo.
