Abstract
Nuevos p-terfenilos hexakis-sustituidos con grupos bilaterales para el tratamiento de la infección por el virus de la inmunodeficiencia humana tipo 1 (VIH-1) y otras enfermedades.#La invención consiste en nuevos compuestos p-terfenilos hexakis-sustituidos con grupos bilaterales y composiciones farmacéuticas que los contengan con utilidad como agentes farmacéuticos.#Debido a su capacidad de interaccionar con un bucle interno de ARN y de mimetizar una {al}-hélice proteica, estos compuestos son efectivos especialmente en el tratamiento de la infección por el virus de la inmunodeficiencia humana tipo 1 (VIH-1) así como otras enfermedades, tales como enfermedades causadas por otros virus ARN, enfermedades infecciosas o crónicas donde estos compuestos puedan ser usados para modular la función de bucles internos de ARN, o enfermedades infecciosas o crónicas donde estos compuestos puedan ser utilizados como agonistas o inhibidores de dominios proteicos de {al}-hélice en interacción con otras biomoléculas.

Por impedimento estérico, las cadenas bilaterales 2aminoetílicas en posiciones 2’ y 6’ (orto) de JB398 inducen la adopción por este bifenilo de una conformación escalonada, similar a la adoptada por los pterfenilos 3,2’,2’’trissustituídos que se comportan como miméticos de αhélice13. Este impedimento estérico está maximizado en SC30, donde las cuatro cadenas

5 Este ensayo fue validado midiendo los valores IC50 y Ki del péptido revp y el antibiótico de referencia neomicina B. La constante de inhibición Ki obtenida para revp (7.0 ± 1.4 nM) concordó muy bien con la constante de disociación Kd determinada independientemente mediante SPR (4.2 ± 3.4 nM). El valor Ki obtenido para el antibiótico de referencia neomicina B (3.5 ± 0.4 �M) también coincidió con la Kd calculada por SPR (2.2 ± 1.4 �M) y con valores
10 Ki previamente publicados en la literatura22 .
Cuando los ligandos bifenílicos y terfenílicos se evaluaron mediante este ensayo, los resultados indicaron que algunos de ellos eran capaces de inhibir la interacción RRERev3450 con valores IC50 de hasta 7.0 µM (Fig. 5 y Tabla 2) y valores Ki de hasta 3.8 �M. De 15 manera similar a lo observado con los experimentos SPR y RMN para la interacción RREligando, la composición de los sustituyentes del anillo de benceno central tuvo un impacto importante en la actividad inhibitoria de la serie tetrakis (2aminoetilo) terfenílica. Los compuestos más potentes resultaron ser nuevamente aquellos conteniendo grupos hidrofóbicos en el anillo de benceno central. Los más activos fueron IIS420 e IIS375, con 20 dos grupos metilo en las posiciones bilaterales 2’ y 6’ de este anillo, que exhibieron concentraciones IC50 de 7.0 y 32.2 �M, respectivamente (valores Ki de 3.8 y 17.4 �M) (Fig. 5), seguidos por JB399, IIS806, IIS771 e IIS758 (con valores IC50 de 23.4, 56.4, 77.2 y
78.1 �M, respectivamente). Las moléculas terfenílicas conteniendo un sustituyente más polar (metoxi) en el anillo central, exhibieron valores IC50 más altos. Los bifenilos JB391 y
25 JB398, con valores IC50 de 5.6 y 22.1 �M, respectivamente, resultaron ser sorprendentemente activos en este ensayo. Sin embargo, estas moléculas interaccionan pobremente con el bucle RRE (e.g. Fig. 3, abajo), por lo que es probable que ambas inhiban la interacción RREfrevp a través de un mecanismo diferente.

Los terfenilos IIS420, IIS758, IIS375, IIS771, IIS792, IIS806 y, en menor medida, el bifenilo JB391 mostraron actividad antiviral sin citotoxicidad tras ser evaluados con un ensayo celular, como se muestra en la Tabla 3a y en la Figura 6.

Se describe la invención adicionalmente mediante los siguientes ejemplos de realización, que son realizaciones preferidas de la invención. Estos ejemplos son ilustrativos y no limitativos. Debe entenderse por tanto que puede haber otras realizaciones que se encuentran dentro del espíritu y alcance de la invención tal como se define por las reivindicaciones adjuntas al presente documento.
Ejemplo 1: Diseño basado en estructura de inhibidores RRERev.
Modelado molecular. El análisis conformacional de una molécula terfenílica hexakissustituida con grupos bilaterales se llevó a cabo utilizando el campo de fuerzas Merck MMFF9424 integrado en el paquete de programas MOE (CGC Inc.). Se hicieron variaciones sistemáticas de las torsiones bencenobenceno en intervalos de 30º y se minimizó la energía potencial de los confórmeros. Los cálculos de docking de moléculas biy terfenílicas con diferentes patrones de sustitución se realizaron utilizando la estructura de RRE 1ETF6 y el algoritmo Dock 5.025 .
Para construir un modelo mejorado de un complejo RREterfenilo, se llevaron a cabo cálculos de docking con los programas Autodock 3.0526 y Gold 5.027 apoyados en los resultados de RMN. En base a los datos de RMN (Fig. 3), el sitio de unión del terfenilo IIS311 se definió alrededor de la base C20 y en el surco mayor del bucle interno. Los cálculos permitieron una total flexibilidad del ligando, que fue previamente minimizado con el campo de fuerzas MMFF94s de MOE (CGC Inc.). A pesar de que tanto Autodock como Gold están parametrizados para proteínas, ambos programas permiten emplear ARN como receptor y un trabajo previo había mostrado que Autodock 3.05 se comporta satisfactoriamente con este tipo de dianas28 . Los cálculos con Gold emplearon la función de ajuste GoldScore, optimizada para predecir conformaciones de unión de ligandos27 . Para los cálculos con Autodock, se asignaron cargas Amber 199429 y PEOE30 a los átomos de ARN y terfenilo, respectivamente, y la carga negativa de los grupos fosfato de ARN fue neutralizada28. Con objeto de tener en cuenta la posible flexibilidad del ARN, se utilizaron como dianas varias estructuras de RRE depositadas en el PDB: aislado (estructuras 1CSL31 y 1DUQ32) y unido al péptido Rev3450 (1ETF y 1ETG6) y a péptidos alternativos (1G7033 y 1I9F34). Los mejores resultados (Fig. 4) se obtuvieron con estructuras de RRE extraídas de complejos con Rev34

50 (1ETF y 1ETG) y con estructuras de RRE aislado (1CSL y 1DUQ). Ambos algoritmos dieron lugar a energías de unión considerablemente peores con estructuras de RRE extraídas de complejos con péptidos diferentes a Rev3450 (1G70 y 1I9F). Estos resultados refuerzan la idea de que las moléculas pterfenílicas bilateralmente sustituidas mimetizan la estructura de Rev3450 en su complejo con el bucle interno de RRE. Estos resultados se muestran en la Figura 4.
Ejemplo 2: Muestras de ARN y péptido.
El ARN RRE (SEQ ID No. 1) utilizado en los experimentos de espectroscopia de RMN y polarización de fluorescencia (FP) se adquirió de Dharmacon (Thermo Fisher Scientific Inc.) y, tras eliminar los grupos protectores 2’ACE, se purificó siguiendo un protocolo basado en electroforesis en gel y diálisis. Para los experimentos de SPR, RRE biotinilado en 5’ y dos ARN control 5’biotinaRREc (SEQ ID No. 2) y 5’biotinaTARc (SEQ ID No. 3) se adquirieron de Microsynth AG ya purificados por HPLC, y se microdializaron en un tampón HBSEP (10 mM HEPES pH 7,40, 150 mM NaCl, 3 mM EDTA y 0,005% (v/v) de surfactante P20) antes de su inmovilización en chips de estreptavidina. En comparación con RRE, la horquilla RREc contiene una oposición G:G en lugar del bucle interno GGCG:ACGGUA que forma el sitio de alta afinidad de Rev, mientras que en la horquilla TARc el bucle interno de RRE es sustituido por el pequeño bucle interno UCU, reconocido por la proteína Tat del virus VIH118 .
Para los experimentos de polarización de fluorescencia (FP), se empleó el péptido FITCAhxGTRQARRNRRRRWRERQRAAAARamida (SEQ ID No. 5; denominado frevp), conteniendo el fluoróforo FITC unido a la glicina Nterminal (Genscript Inc.). Este péptido contiene el tracto Rev3450 rico en argininas (TRQARRNRRRRWRERQR) que forma la αhélice esencial para la interacción con el bucle interno del RRE20 . Con el fin de poner a punto los experimentos de FP y SPR, se utilizó un péptido similar pero sin etiquetar, succinilTRQARRNRRRRWRERQRAAAARamida (SEQ ID No. 4; identificado como revp), que también se adquirió de Genscript Inc. Ambos péptidos contienen aminoácidos AAAAR adicionales en su extremo Cterminal que favorecen la conformación αhélice21. El antibiótico de referencia neomicina B fue suministrado por Sigma. Este antibiótico, los péptidos revp y frevp, y los compuestos bifenílicos y terfenílicos, fueron disueltos en agua y congelados hasta su utilización en los ensayos de RMN, SPR, FP y celulares.

donde [L] es la concentración total de frevp y Kd es la constante de disociación RREfrevp. Estos valores únicamente se calcularon para inhibidores competitivos capaces de unirse específicamente al bucle interno de RRE con bajas estequiometrías, según los experimentos de SPR y/o RMN. Los resultados se muestran en la Figura 5 y en la Tabla 2.
Ejemplo 6: Plásmidos, virus y células.
El vector pNL4.3Luc fue generado clonando el gen luciferasa en la región nef del clon proviral pNL4.340. El plásmido pNL4.3Ren fue generado clonando el gen renilla en la región nef del clon proviral pNL4.341 .
Células MT2 (American Type Culture Collection, Rockville, MD)42 fueron cultivadas en medio RPMI1640 completo suplementado con suero bovino fetal 10% (v/v), Lglutamina 2 mM, penicilina (50 UI/mL) y estreptomicina (50 �g/mL) (todos Whittaker M.A. BioProducts). Las células HeLa TetOn Luc fueron cultivadas en medio DMEM suplementado con suero bovino fetal al 10% (v/v), Lglutamina 2 mM, penicilina (50 IU/ml), Estreptomicina (50 µg/ml) (todos Whittaker M.A. BioProducts), higromicina (100 µg/ml) (Invitrogen) y geneticina (100 µg/ml) (Sigma). Las dos líneas celulares fueron cultivadas a 37ºC con atmósfera humidificada con CO2 al 5% y pasadas dos veces a la semana.
Ejemplo 7: Ensayos celulares muestran que los inhibidores de RRERev3450 bloquean la replicación del VIH1 in vivo y producen este efecto a nivel posttranscripcional.
Evaluación de la actividad antiVIH1. Los sobrenadantes infecciosos fueron obtenidos mediante transfección de los plásmidos en células 293T mediante fosfato cálcico. Estos sobrenadantes fueron utilizados para infectar células MT2 en placas de 96 pocillos de fondo en U en presencia de diferentes concentraciones de los compuestos. La cuantificación de la actividad antiviral se realizó 48 horas después de la infección. Para ello, las células fueron lisadas con 100 ÉL/pocillo del tampón apropiado (Luciferase Assay System Kit with Reporter Lysis Buffer o Renilla Lysis Buffer, Promega, Madison, WI). Las unidades relativas de luminiscencia (RLUs) se obtuvieron en un luminómetro (Berthold Detection Systems, Pforzheim, Germany) después de añadir el sustrato a los extractos celulares. La viabilidad celular fue evaluada en células tratadas de igual manera que en los ensayos de infección pero sin infectar. Después de 48 horas, la viabilidad se cuantificó

mediante el sistema CellTiter Glo (Promega) siguiendo las instrucciones del fabricante. La concentración inhibidora 50% (EC50) y la concentración citotóxica 50 % (CC50) se calcularon mediante el programa GraphPad Prism. Los resultados se muestran en la Tabla 3a y Figura 6a. Para el ensayo HeLa TetOn Luc, se sembraron células HeLa TetOn Luc en placas de 5 24 pocillos el día antes del experimento y se mantuvieron en cultivo durante un día. A continuación el medio se cambió por medio fresco con diferentes concentraciones de los compuestos y las células se trataron con 1 �g/mL de doxiciclina. 6 horas después, los sobrenadantes se eliminaron, las células se lisaron con el tampón de lisis Luciferasa (Promega), y se midió la actividad luciferasa en un luminómetro. Los resultados se
10 muestran en la Tabla 3a y Figura 6a.
Ensayos de Transfección. Células MT2 se mantuvieron en cultivo sin estímulos y antes del ensayo se resuspendieron en 350 ÉL de RPMI sin suero y antibióticos y se transfectaron a 320 V, 1500 �F y resistencia máxima con los plásmidos a una concentración de 0.5 �g/106 15 células con un Easyject plus Electroporator (Equibio, Middlesex, UK). Después de la transfección, las células fueron inmediatamente cultivadas en RPMI completo suplementado con suero fetal bovino, glutamina y antibióticos, y tratadas o no con diferentes concentraciones de compuestos. Las células fueron recogidas 48 horas después y lisadas con el buffer de lisis Luciferasa (Promega). Las RLUs fueron cuantificadas en un
20 luminómetro. Los resultados se muestran en la Tabla 3b y Figura 6b.
Ejemplo 8: Síntesis de compuestos bifenílicos y terfenílicos bilateralmente sustituidos.
Los fragmentos de partida 1 y 2 fueron piezas clave para la introducción de la agrupación
25 amínica en la molécula final. El fragmento 1 se preparó a partir del reactivo comercial 4bromo3,5dimetilfenol (6). El grupo fenol de 6 se alquiló con yodometano y, a continuación, se llevó a cabo la oxidación del éster metílico resultante. La esterificación del ácido carboxílico 8 dio lugar al compuesto 9, cuya posterior reducción con LiBH4 (borohidruro de litio), seguida del tratamiento con NBS (Nbromosuccinimida) y NaCN (cianuro sódico)
30 originó el fragmento 1 con buen rendimiento (Esquema 2).
