Description
Concentrados de isoflavonas y métodos para su
preparación.
La presente invención se refiere a concentrados
de isoflavona y a un método para su preparación.
Las hormonas son compuestos químicos que se
sintetizan en órganos o células especiales y luego son transportadas
a un lugar diferente, para coordinar allí el crecimiento, el
desarrollo y las actividades fisiológicas y metabólicas a muy baja
concentración.
Las plantas contienen compuestos que no tienen
carácter hormonal para ellas y que sin embargo producen en los
mamíferos efectos similares a los hormonales. Los mejor conocidos
son las llamadas isoflavonas, procedentes de las leguminosas. La
soja, p. ej., contiene genisteína, isogenisteína y formononetina.
Estos agentes poseen efectos estrógenos y por tanto tienen una
influencia positiva en síntomas postmenopáusicos tales como
problemas de circulación sanguínea, diabetes de tipo II y
osteoporosis, afortunadamente sin efectos secundarios. De este modo
influyen indirectamente en la tasa de mortalidad. Los fenómenos
generales de envejecimiento, como los de la piel, también forman
parte de este grupo de cualidades protectoras. Durante décadas, la
absorción media de isoflavonas en Asia es de 20 a 100 mg/día; en
los países occidentales es muy inferior a esta cantidad.
Las isoflavonas del tipo de la genisteína están
contenidas en la soja y en muchas otras plantas. La nutrición a
base de soja, habitual sobre todo en Asia, se relaciona con el hecho
de que allí se dan muchos menos casos de carcinomas (de mama, de
próstata, de piel) y que las mujeres raramente tienen síntomas
postmenopáusicos. Las isoflavonas se conocen como fitoestrógenos y
se fijan al receptor de estradiol. En ensayos con animales y
modelos de piel se halló que estas isoflavonas aumentan el potencial
antioxidante intracelular de tipo enzimático, lo cual se debe en
parte a su efecto.
El documento WO 99/38509 describe la elaboración
de una formulación de genisteína, en cuyo proceso de preparación se
utiliza preferentemente el emulsionante Polisorbato 80, a una
temperatura de reacción de 80ºC, con el fin de obtener una
microemulsión.
Por consiguiente, la presente invención se
plantea por una parte el objetivo de incrementar la
biodisponibilidad y la resorción de dichos principios activos y por
otra parte facilitar su incorporación en los productos acabados de
la industria alimentaria, cosmética y farmacéutica.
La presente invención prevé para ello un
concentrado de isoflavona que lleva un polisorbato y también una
mezcla de genisteína, daidceína, gliciteína y sus glicósidos. El
concentrado de la presente invención es soluble en agua y forma una
solución transparente. Cuando se administra esta solución las
isoflavonas del tipo genisteína se resorben fácilmente en el tracto
gastrointestinal. Por lo tanto cabe la posibilidad p. ej. de añadir
una dosis del concentrado según la presente invención a una bebida
sin alcohol, con lo cual la absorción global de isoflavonas por el
organismo se aumenta de manera casi casual y simultánea con el
consumo de bebidas. Las isoflavonas del tipo genisteína en forma de
concentrado soluble en agua se pueden emplear en el sector
alimentario clásico y ventajosamente en los sectores de cosmética o
de la higiene corporal, ya que la solubilidad en agua asegura una
absorción mucho mayor de las isoflavonas a través de la piel.
Se prefieren los principios activos de origen
puramente vegetal. Para un concentrado de principio activo que
contenga isoflavonas se recomienda usar un extracto de habas de
soja, generalmente preparado en forma de polvo, que lleva 400 g por
kilogramo de una mezcla de genistín, daidcín y glicitín, así como
sus agliconas. Entonces, de modo ventajoso, La relación
cuantitativa de genistín, daidcín y glicitín es aproximadamente de
1,3:1,0:0,3.
Para preparar el concentrado de la presente
invención se usa preferentemente Polisorbato 80. No obstante, para
la finalidad de la presente invención también resultan adecuados el
Polisorbato 20, el Polisorbato 40, el Polisorbato 60 y el
Polisorbato 65, que se obtienen como ésteres parciales de los ácidos
láurico, palmítico o esteárico con sorbitol.
Por tanto, además de bebidas no alcohólicas, son
objeto de la presente invención licores, diversos productos
alimenticios, y también productos para el cuidado de la piel,
cosméticos y otros similares, que lleven un concentrado de
isoflavona formado por polisorbato y una mezcla de genisteína,
daidceína, gliciteína y sus glicósidos. De modo conveniente, una
bebida, p. ej., lleva aproximadamente 0,5 g hasta 3 g de concentrado
de isoflavona por litro. Este concentrado exento de agua puede
empaquetarse sin diluir en cápsulas de gelatina y/o libres de
gelatina, como un antioxidante o un suplemento dietético.
Para preparar un concentrado hidrosoluble que
contenga isoflavonas se parte, por ejemplo, de 100 g de un extracto
de habas de soja en polvo, vendido por la firma
Archer-Daniels-Midland Company, USA,
bajo la marca NOVASOY. Este extracto de habas de soja contiene como
mínimo 40% de genistín, daidcín y glicitín y sus agliconas, según
la proporción cuantitativa 1,3:1,0:0,3. 100 g de dicho extracto
contienen por lo tanto 20,0 g de genistín, 15,4 g de daidcín y 4,6
g de glicitín, es decir 40,0 g de isoflavonas en total.
Para preparar un concentrado de isoflavona exento
de agua que lleve isoflavonas, sirve, por ejemplo, el citado
extracto de habas de soja en polvo, que puede adquirirse con la
marca NOVASOY de la
Archer-Daniels-Midland Company,
Decatur, Illinois, USA, el cual contiene principalmente los
glicósidos de genisteína, daidceína y gliciteína.
Unos 166 g de este extracto de soja en polvo se
vierten en aproximadamente 834 g de Polisorbato 80 calentado a unos
75ºC y la mezcla (1,0 Kg) se agita uniformemente durante una media
hora a esta temperatura. Se obtiene un concentrado transparente de
color marrón oscuro sin sedimento. Añadiendo 1-2 ml
de este concentrado a una cantidad diez veces mayor de agua
destilada se obtiene una solución transparente después de completar
el proceso de mezcla.
1,0 Kg de este concentrado lleva aproximadamente
66 g de isoflavona. Si se añaden 1,5 g del concentrado a 1,0 l de
una bebida terminada, esta bebida contendrá unos 100 mg de dicha
isoflavona. Por lo tanto un litro de una bebida terminada y
preparada de este modo cubre suficientemente la necesidad diaria de
isoflavona.
En lugar de NOVASOY también pueden emplearse
extractos de soja de otras procedencias, que se caracterizan por
una mayor carencia de azúcar. Por ejemplo, la firma K.-W.
Pfannenschmidt GmbH, de Hamburgo, suministra un extracto de soja
con un contenido total de isoflavona de al menos 40%. Este 40%
aproximado consta de 7,58% de genistín, 25,43% de genisteína, 5,48%
de daidcín y 1,67% de daidceína. Empleando este extracto de soja
tal como se ha descrito para el NOVASOY, se obtienen aproximadamente
los mismos resultados.
Arriba se describe la producción de un
concentrado de principio activo exento de agua. No obstante se puede
obtener un concentrado acuoso de principio activo, según los
ejemplos siguientes:
La obtención del concentrado acuoso de isoflavona
según la presente invención consiste en verter dichos compuestos en
agua precalentada aproximadamente a 60ºC y agitar la mezcla a dicha
temperatura durante un periodo de tiempo prefijado de unos diez
minutos. Luego se agrega el polisorbato a la mezcla y la temperatura
se incrementa hasta unos 100ºC. Se continúa agitando a esta
temperatura hasta que la mezcla se vuelve clara y transparente.
Como alternativa puede añadirse primero el polisorbato a los
compuestos e incorporar luego el agua.
Se agitan uniformemente, por ejemplo, 100 g del
citado extracto de soja NOVASOY en 400 g de agua destilada
precalentada aproximadamente a 60ºC. La mezcla (cantidad total 500
g) se agita continuamente mediante un agitador magnético,
manteniendo la temperatura de 60ºC durante unos diez minutos. Sin
dejar de agitar se añaden 500 g de Polisorbato 80 y luego la
temperatura se incrementa hasta aproximadamente 100ºC.
Se continúa agitando a esta temperatura, hasta
que la mezcla (cantidad total 1 Kg) se vuelve clara y transparente.
Esta mezcla transparente contiene los 40 g de isoflavona
incorporados.
Si se desea que un concentrado hidrosoluble de
isoflavona contenga menos agua específicamente como producto final,
la cantidad de agua que se debe agregar según el siguiente ejemplo 3
puede compensarse o reducirse parcialmente incorporando
triglicéridos o un aceite vegetal ligero (como el de cártamo). La
relación ponderal de NOVASOY a Polisorbato 80 debería elegirse
aproximadamente 1:6 en este caso, mientras que en los ejemplos
anteriores era aproximadamente de 1:5.
Se agitan uniformemente 100 g del mencionado
extracto de soja NOVASOY en 230 g de agua destilada precalentada
aproximadamente a 60ºC. La mezcla (cantidad total 330 g) se agita
continuamente mediante un agitador magnético, manteniendo una
temperatura aproximada de 60ºC durante unos cinco minutos. Sin dejar
de agitar y mantener la temperatura se añaden unos 70 g de aceite
vegetal (aceite de cártamo) y se agita durante unos cinco minutos.
Luego se añaden 600 g de Polisorbato 80 a la mezcla (400 g) y la
temperatura se incrementa hasta aproximadamente 100ºC. Se sigue
agitando a esta temperatura, hasta que la mezcla (cantidad total 1
Kg) se ha vuelto transparente o se obtiene una solución acuosa
clara y marronosa. Esta mezcla transparente contiene los 40 g de
isoflavona incorporados.
2,5 g de este concentrado acuoso transparente y
estable contienen 100 mg de isoflavona. Esta cantidad, por ejemplo,
se puede agregar a un litro de una bebida terminada, que de este
modo cubre el doble de la demanda diaria de isoflavona,
correspondiente a una cantidad de 50 g.
En lugar de aceite de cártamo también puede
utilizarse aceite de girasol, aceite de linaza, aceite de soja o
aceite de oliva. Todos estos aceites se caracterizan por un elevado
contenido de triglicéridos, tales como ácido
\alpha-linolénico, ácido
\gamma-linolénico, ácido linoleico o ácido oleico.
Por ejemplo, el aceite de cártamo contiene hasta 83% de ácido
linoleico y hasta 24% de ácido oleico; el aceite de linaza contiene
hasta 71% de ácido linolénico, hasta 31% de ácido linoleico y hasta
23% de ácido oleico. Así pues, se pueden usar uno o más aceites
ricos en triglicéridos, preferiblemente de origen vegetal, en vez
del aceite de cártamo.
Para acelerar y optimizar el proceso de mezcla y
la solubilidad, se recomienda calentar los concentrados de
isoflavona según los ejemplos 1 a 3 a unos 70ºC, antes de efectuar
la mezcla y el agua donde deben disolverse, a unos 40ºC, también
antes de efectuar la mezcla.
El concentrado de la presente invención también
es estable en cuanto a su transparencia y solubilidad en agua si se
añade ácido gástrico al concentrado de la presente invención o a un
preparado a partir del mismo (p. ej. una bebida). Esto también es
cierto si el concentrado se calienta p. ej. a unos 100ºC. Por tanto
el concentrado se caracteriza por una excelente estabilidad.
En los concentrados de la presente invención hay
micelas, en cuyo interior se hallan las isoflavonas rodeadas por
moléculas de polisorbato. El radio medio de las micelas es inferior
a 20 nm. Para verificarlo, el concentrado de 4% de isoflavona
resultante del ejemplo 2 se diluye en agua en una proporción de
1:200 y la dilución del concentrado resultante, con 0,02% de
isoflavona, se somete al fraccionamiento campo/lujo con detector
DAWN-EOS conectado a continuación, según la
tecnología Wyatt. El resultado está reflejado en el diagrama
adjunto, según el cual la curva 1 indica un radio R.M.S. de la
mezcla de aproximadamente 16 nm. La curva 2 es una curva de
referencia.
Claims (15)
1. Concentrado de isoflavonoide que contiene un
polisorbato y una mezcla de genisteína, daidceína, gliciteína y sus
glicósidos.
2. Concentrado según la reivindicación 1, que
lleva aproximadamente un 6,64% en peso de mezcla y aproximadamente
83,4% en peso de Polisorbato 80.
3. Concentrado según la reivindicación 1, que
contiene 4% en peso de mezcla, 40% en peso de agua y 50% en peso de
Polisorbato 80.
4. Concentrado según la reivindicación 1, que
contiene 4% en peso de mezcla, 23% en peso de agua, 7% en peso de
aceite vegetal con triglicéridos y 60% en peso de Polisorbato
80.
5. Concentrado según una de las reivindicaciones
anteriores, en que el polisorbato es el Polisorbato 80.
6. Método para preparar un concentrado de
isoflavonoide, según una de las reivindicaciones anteriores, que
contiene una mezcla de genisteína, daidceína, gliciteína y sus
glicósidos, caracterizado porque el polisorbato calentado
desde aproximadamente 60ºC hasta aproximadamente 100ºC se incorpora
a un extracto, con preferencia de origen vegetal, que lleva la
mezcla y la mezcla caliente se agita uniformemente, hasta obtener un
concentrado hidrosoluble transparente y libre de sedimentos.
7. Método según la reivindicación 6,
caracterizado porque el extracto se agita primero en agua
caliente, se agrega el polisorbato y la temperatura se incrementa
hasta aproximadamente 100ºC.
8. Método según la reivindicación 7, en el cual
el extracto que contiene la mezcla se agita en agua caliente a
aproximadamente 60ºC y la agitación se prosigue durante un periodo
de tiempo prefijado, por ejemplo de diez minutos, a continuación se
añade el polisorbato y la temperatura se incrementa hasta
aproximadamente 100ºC, manteniendo la agitación.
9. Método según una de las reivindicaciones 6 a
8, caracterizado porque se emplea el Polisorbato 80.
10. Método según una de las reivindicaciones 6 a
9, caracterizado porque la relación ponderal entre extracto y
Polisorbato 80 es aproximadamente de 1:5 o 1:6.
11. Método según una de las reivindicaciones 6 a
10, en que a la mezcla se le añade un aceite rico en triglicéridos,
como por ejemplo aceite de cártamo o aceite de girasol,
principalmente de origen vegetal.
12. Método según la reivindicación 11, en que se
añade hasta aproximadamente 7% en peso de aceite de cártamo.
