Abstract

L'invention concerne un procédé d'utilisation d'un bioéacteur à double compartiment, qui comporte une poche souple (1) ayant deux compartiments (2, 3) séparés par un filtre (4), ainsi qu'au moins une ouverture d'entrée (5) en communication avec un des compartiments (2) et au moins une ouverture de sortie (6) en communication avec l'autre compartiment (3) comportant sa mise en place dans un appareillage de traitement biologique notamment de lavage ou de transformation d'un produit tissulaire ou cellulaire, caractérisé en ce qu'il met en œuvre, pour une dissociation tissulaire, l'utilisation dudit bioréacteur en tant que chambre de digestion et un malaxage manuel de la poche souple pour dissocier les tissus au cours d'une étape de digestion au cours de laquelle une solution de digestion contenant de la collagénase circule entre des ouvertures d'entrée (5) et de sortie de la poche (6).

Description

BIOREACTEUR A DOUBLE COMPARTIMENT
La présente invention a pour objet un bioréacteur à double compartiment apte à permettre une transformation d'un produit tissulaire ou cellulaire, et un procédé pour sa mise en œuvre.
Les dispositifs actuellement utilisés dans une application à l'extraction des îlots de Langerhans sont constitués de chambres à parois rigides.
Dans l'article de Camillo RICORDI et Collaborateurs, intitulé "Automated Method for Isolation of Human Pancreatic Islets", paru dans DIABETES volume 37, année 1988, pages 413 à 420, la dissociation du tissu pancréatique est réalisée par agitation de la chambre de séparation en acier inoxydable qui contient des billes. Il en résulte plusieurs inconvénients, à savoir :
1) le caractère traumatisant de la dissociation mécanique par billes ;
2) un risque d'hyperpression par obturation du filtre de la chambre à parois rigides ;
3) l'impossibilité de visualiser l'état d'avancement de la digestion tissulaire.
Une variante de ce procédé de dissociation est décrite dans la Demande de Brevet FR 2 696 754 (HOSPAL INDUSTRIE - INSERM).
L'article de Denis DUFRANE et Collaborateurs, intitulé "A simple Method Using a Polymethylpenten Chamber for Isolation of Human Pancreatic Islets" paru dans Pancréas, volume 30 n° 3, année 2005, pages e51 à e59 met également en oeuvre une technique d'agitation, à partir d'une chambre conique en polyméthylpentène (PMPC) qui contient des billes et qui présente deux compartiments séparés par un filtre en treillage métallique (acier inoxydable) ayant un diamètre de passage de 500 μm.
Cette technique présente les inconvénients 1 et 2, l'inconvénient 3 étant supprimé parce que la chambre conique est transparente.
La présente invention propose de remédier aux deux premiers inconvénients présentés en proposant une technique dont l'idée de base est d'utiliser une chambre de séparation en matériau souple qui permet à un opérateur d'effectuer une dissociation tissulaire par des manipulations manuelles externes dans des conditions d'asepsie et éventuellement de rendre possible une thermorégulation du produit par immersion dans un liquide compte tenu de l'étanchéité du dispositif.
L'invention concerne ainsi un bioréacteur à double compartiment, caractérisé en ce qu'il comporte une poche souple séparée en deux compartiments séparés par un filtre, ainsi qu'au moins une ouverture d'entrée en communication avec un des compartiments et au moins une ouverture de sortie en communication avec l'autre compartiment.
Au moins un des compartiments peut comporter à la fois une ouverture d'entrée et une ouverture de sortie.
L'invention concerne également un procédé d'utilisation d'un tel bioréacteur, caractérisé en ce qu'il met en œuvre la mise en place dans un appareillage de traitement biologique notamment de lavage ou de transformation d'un produit tissulaire ou cellulaire, la circulation d'une solution entre une ouverture d'entrée et une ouverture de sortie de la poche. Plus particulièrement l'invention concerne un procédé qui met en œuvre, pour une dissociation tissulaire, notamment le tri d'îlots de Langerhans, la mise en place dudit bioréacteur en tant que chambre de digestion et un malaxage manuel de la poche souple, réaliser ladite dissociation, notamment pour séparer des îlots de Langerhans, au cours d'une étape de digestion pendant laquelle une solution de digestion contenant de la collagénase circule entre des ouvertures d'entrée et de sortie de la poche.
D'autres caractéristiques et avantages de l'invention apparaîtront mieux à la lecture de la description ci-après en liaison avec la figure unique.
La figure 1a représente une poche souple utilisable en tant que bioréacteur stérile à usage unique pour un procédé de transformation d'un produit tissulaire ou cellulaire, dont la figure 1b est une coupe selon AA.
La poche souple 1 en matière plastique souple présente deux compartiments 2 et 3 séparés par un filtre 4 fixé en position sagittale. La poche souple 1 peut être similaire à une poche de sang, et le filtre 4 présente un pourtour 7 qui est soudé pour positionner le filtre 4 en position sagittale dans la poche 1.
La taille des mailles du filtre 4 est fonction de l'application dévolue au dispositif. L'introduction, le renouvellement et la circulation des fluides ou du matériel biologique à l'intérieur du dispositif, sont réalisés au travers d'embouts d'accès d'entrée 5 et de sortie 6 ouvrant sur chacun des compartiments 2 et 3 de la poche. Le système est ainsi intégrable à un équipement ou à un circuit conventionnel. L'étanchéité du dispositif autorise son immersion complète dans un milieu liquide (régulation thermique), ainsi qu'une aseptie.
L'invention s'applique à la dissociation tissulaire, par exemple extraction d'îlot de Langerhans avec dans ce cas un maillage du filtre compris entre 50 μm et 600 μm ou bien à une utilisation pour des réactions biologiques par exemple culture cellulaire, co-culture, ou bien encore ingénierie tissulaire ou bien en tant que dispositif de lavage de tissus ou de suspensions cellulaires, sans nécessiter de centrifuger la suspension cellulaire, avec notamment dans le cas de cultures cellulaires ou de lavage de suspensions cellulaires, une taille de filtre n'autorisant que le passage d'un liquide acellulaire (sans cellules).

Claims

REVENDICATIONS
1) Procédé d'utilisation d'un bioéacteur à double compartiment, qui comporte une poche souple (1) ayant deux compartiments (2, 3) séparés par un filtre (4), ainsi qu'au moins une ouverture d'entrée (5) en communication avec un des compartiments (2) et au moins une ouverture de sortie (6) en communication avec l'autre compartiment (3) comportant sa mise en place dans un appareillage de traitement biologique notamment de lavage ou de transformation d'un produit tissulaire ou cellulaire, caractérisé en ce qu'il met en œuvre, pour une dissociation tissulaire, l'utilisation dudit bioréacteur en tant que chambre de digestion et un malaxage manuel de la poche souple pour dissocier les tissus au cours d'une étape de digestion au cours de laquelle une solution de digestion contenant de la collagénase circule entre des ouvertures d'entrée (5) et de sortie de la poche (6).
2) Procédé selon la revendication 1 , caractérisé en ce que la dissociation tissulaire est le tri d'îlots de Langerhans et en ce que le malaxage sépare les îlots de Langerhans.
3) Procédé selon une des revendications 1 ou 2, caractérisé en ce que le filtre (4) a un maiilage compris entre 50μm et 600μm adapté au tri d'îlots de Langerhans
4) Procédé selon la revendication 1 , caractérisé en ce que le filtre a une taille de pores n'autorisant que le passage d'un liquide sans cellules, adapté notamment à une culture cellulaire ou à un procédé de lavage de suspensions cellulaires.
5) Procédé selon la revendication 1 , caractérisé en ce que la poche est utilisée pour des réactions biologiques telles que culture cellulaire, co-culture, ou bien ingénierie cellulaire.
6) Procédé selon la revendication 1 , caractérisé en ce que la poche est utilisée en tant que dispositif de lavage en biologie, sans nécessiter de centrifuger la suspension cellulaire.
7) Procédé selon une des revendications précédentes, caractérisé en ce qu'au moins un des compartiments présente à la fois une ouverture d'entrée et une ouverture de sortie.
