Description
Dérivés de cyanine, conjugués fluorescents les contenant et leur utilisation
Domaine de l'invention
La présente invention a pour objet des dérivés de cyanine qui absorbent la lumière et émettent de la fluorescence dans le rouge et le proche inf ra-rouge Ces dérivés de cyanine sont capables de traverser les membranes cellulaires et sont fonctionnalisés de telle sorte qu'ils puissent être aisément utilisés comme marqueurs fluorescents
L'invention concerne également des conjugués fluorescents comprenant un dérivé de cyanine de l'invention, lié de manière covalente à un agent de couplage permettant le marquage de molécules dans la cellule. Les dérivés de cyanine selon l'invention sont particulièrement adaptés au marquage de protéines dans les cellules.
Enfin l'invention concerne également des conjugués fluorescents comprenant un dérivé de cyanine de l'invention, lié de manière covalente à une substance que l'on souhaite faire rentrer dans la cellule, ladite substance étant rendue fluorescente par le couplage avec le dérivé de cyanine.
L'invention a également pour objet l'utilisation des dérivés de cyanine de l'invention pour marquer des produits présents dans les cellules vivantes.
Les composés selon l'invention sont particulièrement utiles pour marquer des biomolécules se trouvant dans le milieu intracellulaire sans modifier l'intégrité de la membrane plasmique, en particulier sans utiliser de produits destinés à perméabiliser cette membrane.
Les composés selon l'invention ont en effet un caractère lipophile, dans la mesure où ils ne possèdent pas de groupes sulfates, sulfonates, phosphates, phosphonates ou carboxylates classiquement utilisés dans l'art antérieur pour résoudre les problèmes d'agrégation. Ces groupes ont été remplacés par des esters (de préférence diester) de phosphonate ou phosphate qui n'affectent pas le caractère lipophile de ces composés et permettent, après hydrolyse par les enzymes intracellulaires, de prévenir les phénomènes d'agrégation dans la cellule
L'invention concerne donc également l'utilisation de dérivés de cyanine selon l'invention comportant un agent de couplage, en tant que marqueurs fluorescents
L'invention concerne en outre une méthode de marquage d'une biomolécule présente dans une cellule intacte, qui consiste a introduire dans le milieu extracellulaire un dérivé selon l'invention comportant un agent de couplage, ladite biomolécule comportant un domaine de couplage
Cette méthode est très avantageuse puisqu'elle permet de marquer des composés à l'intérieur des cellules sans perméabiliser leur membrane
Domaine technique
Les colorants fluorescents qui absorbent et émettent de la lumière dans la région spectrale du visible et du proche infra-rouge ont été couramment utilisés pour diverses applications d'analyse dans le domaine de la biochimie, de la biologie et du diagnostic, notamment en raison de leur absorption molaire et/ou de leur rendement quantique de fluorescence élevé.
En particulier, seuls les dérivés de cyanme absorbent et émettent dans le proche infra-rouge, région du spectre électromagnétique particulièrement intéressante pour étudier les cellules vivantes car cette gamme de longueurs d'onde permet d'éviter le blanchiment («bleaching») dû à la décomposition photochimique des colorants utilisés comme marqueurs fluorescents (ce phénomène augmente en fonction de l'énergie des photons et est donc peu prononcé dans le proche infra-rouge). De plus, les molécules biologiques ne présentent pas d'autofluorescence dans le proche infra-rouge, ce qui limite les problèmes de bruit de fond non spécifique.
Malgré ces avantages, le caractère lipophile des cyanines, dû à la présence de groupes polycycliques et d'une chaîne polyméthine hydrophobe, n'est pas toujours compatible avec de bonnes performances en tant que marqueur fluorescent. Par exemple les cyanines ont tendance à former des agrégats en solution. Ainsi le vert d'indocyanine voit son rendement quantique chuter suite à une extinction de fluorescence due à la formation d'agrégats lorsqu'il est utilisé à des concentrations supérieures à 125 μM. Ce phénomène, assez général (il existe aussi dans le cas de la fluorescéine), est particulièrement prononcé dans la série des cyanines.
Par ailleurs, et malgré ce caractère lipophile, les cyanines traversent très mal, voire pas du tout, les membranes biologiques, notamment en raison de leur masse moléculaire élevée (de l'ordre de 700-800 daltons), alors que des molécules plus petites comme la fluorescéine (337 daltons) ne présentent pas cet inconvénient.
Les solutions techniques pour parer au problème de solubilité et d'agrégation des cyanines ne sont pas très satisfaisantes. Des conjugués de cyanines avec des peptides hydrophiles, des polyéthylèneglycols ou des oligosacchandes ont été utilisés et plusieurs études ont été consacrées à l'utilisation de colorants pour le proche infrarouge ainsi que sur leurs conjugués avec des biomolécules. Cette approche a l'inconvénient de nécessiter des synthèses multi- étapes, ce qui implique des rendements modestes D'autre part dans le cas de l'utilisation d'oligosacchaπdes ou d'oligopeptides hydrophiles le coût de revient peut être élevé et la
purification de ces conjugués nécessite des techniques de purification autres que celles utilisées dans la synthèse organique classique
L approche la plus utilisée pour améliorer les propriétés photophysiques et donc les performances analytiques en tant que traceur fluorescent a été de limiter le phénomène d'agrégation en augmentant le caractère hydrophile par l'introduction de groupes chimiques portant une charge négative sur les noyaux indoles, tels que des sulfonates et/ou des groupes alkylesulfonates (sulfoalkyle) ainsi que des groupes carboxyliques Ainsi, les dérivés non- sulfonés d'indocyanine (comme le produit connu sous la dénomination commerciale IR-786 de Sigma) ont des solubilités inférieures à 10 μM, et leur utilisation en milieu aqueux nécessite l'utilisation d'adjuvants. La tétra-sulfonation de ces dérivés permet d'augmenter la solubilité en milieu aqueux jusqu'à plus de 10 mM. En revanche, la sulfonation n'a qu'un faible impact sur le rendement quantique de fluorescence qui en moyenne est inférieur à 15% pour un composé tétra-sulfoné.

La demande internationale WO 2005/056689 A2 concerne des composés colorants à pH physiologique possédant un squelette cyanine et comportant un substituant chargé négativement.
La demande internationale WO 2005/061456 A1 concerne un milieu de contraste fluorescent proche infrarouge contenant un composé de cyanine qui présente un groupe soluble dans l'eau.
La demande internationale WO 2005/089813 A2 concerne des colorants de cyanine, ainsi que des conjugués biologiques de ceux-ci, utilisés dans l'imagerie diagnostic et en thérapie. Les conjugués consistent notamment en plusieurs colorants de cyanine avec une variété d'homologues d'acide bis- et tétrakis carboxylique.
La demande internationale WO 2005/056687 A2 a pour objet des colorants de cyanine dont le reste méthine est substitué, qui conviennent au marquage des acides nucléiques et qui consistent en des dérivés de cyanine substitués notamment par des chaînes sulfoalkyle ou carboxyalkyle
La demande internationale WO 2004/039894 A2 porte sur des colorants de cyanine fonctionnalisés et sur leurs dérivés utiles comme intermédiaires dans la préparation de colorants de cyanine, ainsi que sur des procédés de préparation de ces colorants et sur les colorants ainsi obtenus Les dérivés de cyanine comportent tous des fonctions sulfonates
La demande internationale WO 03/082988 A1 a trait a des fluorochromes hydrosolubles de proche infrarouge, utiles par exemple pour l'imagerie biomédicale
La demande internationale WO 02/068537 A2 concerne des colorants fluorescents notamment des colorants de cyanine fluorescents et solubles dans l'eau qui contiennent des sites supplémentaires de fixation à des biomolécules La solubilité dans l'eau est assurée par la présence de groupes sulfonates.
La demande internationale WO 02/24815 A1 décrit des dérivés de cyanine comportant des groupes sulfonates,
La demande internationale WO 01/77229 A2 concerne des colorants de cyanine qui présentent des substituants alkyle en position méso de la chaîne méthine, des groupes sulfoaryle ainsi qu'au moins un groupe réactif permettant la liaison à une substance cible.
La demande internationale WO 01/52746 A1 concerne des conjugués colorant-peptide qui conviennent pour l'imagerie médicale et la thérapie. Ces conjugués colorant-peptide comprennent plusieurs colorants à base de cyanine avec une variété d'homologues bis- et tétrakis (acide carboxylique).
La demande internationale WO 00/66664 A1 concerne des colorants cyanines asymétriques contenant un fragment cyclique aza-benzolium, notamment des colorants cyanines substitués par une chaîne latérale cationique, des colorants cyanines monomères et dimères, des colorants cyanines réactifs au plan chimique et des conjugués de colorants cyanines.
La demande internationale WO 01/57237 A2 décrit des cyanines non fluorescentes, comportant un groupe NO2 et des esters d'acides carboxyliques.
Une tentative a été faite d'utiliser une fonction phosphonate (acide phosphonique) dans le but de réduire le phénomène d'agrégation des cyanines. [Oswald B., et al Novel diode laser- compatible fluorophores and their application to single molécule détection, protein labelmg and fluorescence résonance energy transfer immunoassay. Photochem Photobiol. (2001 )74(2):237- 45].
La demande internationale WO 01/36973 A2 concerne un procédé d'amélioration de la solubilité dans l'eau de marqueurs optiques consistant à introduire des restes d'acide phosphoπque ou des restes d'acide phosphonique, ou leurs sels ou monoesters, dans les marqueurs. Ces marqueurs sont utiles pour le marquage de biomolécules, de polymères et d'agents pharmaceutiques Cette demande décrit, en particulier, une cyanine pentaméthine portant un alkyl-phosphonate-monoester sur l'azote du cycle indole
Gruber et al (Journal of Fluorescence, Vol. 15, No 3, May 2005, 207-214), décrivent deux composés dérivés de cyanines (dénommés chroméon 546 et chroméon 642), dans lesquels un des atomes d'azote est substitué par un éthyl ester de phosphonate porté par un bras diméthylène, et l'autre par un groupe réactionnel N-hydroxysuccinimide porté par un bras polyméthylène. On notera que le groupe phosphonate ne permet pas, à priori, à ces composés de traverser les membranes biologiques
Mazières et al (Dyes and Pigments, 74 (2007) pp 404-409) décrit des cyanines symétriques substituées au niveau des azotes des cycles indoles par des groupes diéthoxy-phosphoryl- propyle Ces composés, après hydrolyse, conduisent à des cyanines portant des groupes acides phosphoniques solubles dans l'eau.
La demande de brevet US 2006/0199955 divulgue des cyanines comportant un groupe phosphonate zwittenonnique, destiné à augmenter la polarité de la cyanine et donc sa solubilité ; les composés décrits sont également porteurs d'un groupe sulfonate et ne peuvent donc pas traverser les membranes cellulaires de cellules intactes.
Les colorants fluorescents dans le proche infra-rouge et possédant une structure carbocyanine ainsi que des fonctions chargées, tels que ceux décrits ci-dessus (sulfate, sulfonate, carboxyle, phosphate ou phosphonate), sont rapportés comme présentant une faible perméabilité membranaire à cause de leur masse moléculaire élevée (comparé par exemple à celle de la rhodamine = 334 Da) et de la présence de charges multiples. On a également observé qu'avec l'augmentation du nombre de charge les colorants fluorescents restent dans les compartiments extra-cellulaires. [Frangioni JV, Curr Opin Chem Biol. 2003, 7(5):626-34. In vivo near-infrared fluorescence imaging]. Enfin, lorsque plusieurs composés fluorescents de ce type sont fixés à une même biomolécule une extinction de fluorescence a été observée [Lm Y. et al. II existe donc un besoin pour des dérivés de cyanines capables de traverser les membranes cellulaires, ne présentant pas de propension à l'agrégation, et pouvant être utilisés comme marqueurs fluorescents. Il est important de noter que les solutions de l'art antérieur pour pallier l'agrégation (ajout de groupes chargés négativement), s'opposent à une bonne perméabilité membranaire A l'inverse, les cyanines lipophiles susceptibles de traverser les membranes ont tendance à s'agréger.
Les paramètres gouvernant la propension pour un colorant à traverser les membranes des cellules sont connus (caractère lipophile, charge et caractère polaire qui pris ensemble définissent un caractère amphiphile, taille et forme de la molécule) mais leur influence relative et leur utilisation comme outil prédictif de la faculté de traverser les membranes reste limité
Maigre ces obstacles, on a maintenant mis au point de nouveaux dérivés de cyanmes capables de traverser les membranes cellulaires sans pour autant présenter de problème d'agrégation et de solubilité
Les cyanmes selon l'invention ont la particularité de comporter
• au moins un ester (de préférence diester) de phosphate ou un ester (de préférence diester) de phosphonate, greffé sur la cyanine via un bras de liaison, et
• un groupe fonctionnel, ou bien un agent de couplage, ou encore une substance conjuguée que l'on souhaite faire rentrer dans la cellule sous forme marquée, ce groupe fonctionnel, agent de couplage ou substance étant porté par un bras de liaison
Les composés selon l'invention sont capables de traverser les membranes cellulaires, dans la mesure où ils comportent les groupements hydrophobes propres aux cyanmes (polycycles, chaînes polylméthines), et ne comportent pas de groupes polaires tels que ceux proposés dans l'art antérieur pour pallier au problème d'agrégation et de solubilité : les composés selon l'invention ne comportent pas de groupes sulfates, sulfonates, phosphates, phosphonates ou carboxylates.
En revanche, les dérivés de cyanmes selon l'invention comportent au moins un ester (de préférence diester) de phosphate ou phosphonate • ces groupes ont la particularité de subir une hydrolyse dans le cytoplasme de la cellule, catalysée par les enzymes cellulaires, en particulier les phosphodiesterases, et ce malgré la présence de la structure cyanine, dont la taille aurait pu gêner les processus enzymatiques.
Par ailleurs, les dérivés de cyanmes selon l'invention sont fonctionnalisés, ce qui permet à l'homme du métier de les coupler à tout groupe ou molécule qu'il souhaite marquer. Les produits selon l'invention peuvent également comporter un agent de couplage qui va leur permettre de marquer les biomolécules comportant un domaine de couplage approprié.
Ces caractéristiques font des dérivés de cyanine selon l'invention des composés particulièrement adaptés au marquage de molécules biologiques intracellulaires par des composés fluorescents excitables et émettant dans le rouge et le proche infra-rouge, et ce sans forcément perméabiliser les cellules

Les dérivés de cyanine de formule (I) de l'invention comportent un contre-ion « Z » (non représenté) qui contrebalance la ou les charges positives ou négatives du dérivé de cyanine.
La nature du contre-ion n'est pas essentielle selon l'invention ; elle dépend du procédé de synthèse mis en œuvre ou du milieu dans lequel se trouve le dérivé de cyanine.
Par exemple, Z sera :

Composés préférés de l'invention
Les dérivés de cyanines préférés selon l'invention sont les composés répondant aux formules suivantes, dans lesquelles les groupes R1-R6, B1 X et Y sont tels que définis ci-dessus .
Le pont polyméthine B
Les composés de l'invention comportent un pont polyméthine entre les structures cycliques. Comme indiqué plus haut, ces méthines peuvent être substituées, ou encore des méthines adjacentes peuvent former ensemble un cycle hydrocarboné saturé ou insaturé à 4, 5 ou 6 éléments, éventuellement substitué.
Les composés préférés de l'invention comportent des méthines non substituées, ou bien seule la méthine centrale est substituée, ou encore 2 méthines centrales forment un cycle.
A titre illustratif et non limitatif, le pont peut être constitué des chaînes polyméthmes suivantes :

Les esters (de préférence diester) de phosphate ou de phosphonate portés par un bras, présents sur les dérivés de cyanme selon l'invention, offrent plusieurs avantages techniques : ils n'augmentent pas la polarité des composés et favorisent donc leur passage à travers les membranes biologiques lipidiques ; d'autre part, une fois à l'intérieur de la cellule, les esters (de préférence diesters) de phosphate ou de phosphonates sont hydrolyses par les enzymes cellulaires pour donner des groupes phosphates ou phosphonates. Ces groupes phosphates ou phosphonates contribuent à l'efficacité des cyanines selon l'invention pour marquer en particulier des protéines, puisqu'ils permettent de remédier aux phénomènes d'agrégation généralement observés lorsque les cyanines sont utilisées dans cette application
Ces groupes peuvent être greffés sur la cyanme selon des méthodes de synthèse connues de l'homme du métier. Par exemple, l'introduction d'un ester de phosphonate sur un noyau aromatique est réalisée à partir d'un dérivé brome, tel que le 5-bromo-2,3,3-trιméthyl-3H-ιndole comme décrit dans l'exemple 13 en suivant le protocole décrit dans la littérature [A Novel Synthests of Dialkyl Arenephosphonates, Hirao, T. et al. Synthesis, (1), 56-57 (1981 )]. La partie expérimentale ci-après illustre des voies de synthèse de dérivés de cyanines comportant des esters de phosphates ou de phosphonates.
Dans le cas où des esters de phosphate sont introduits au niveau des cycles aromatiques de la cyanme, ces groupes doivent obligatoirement être portés par un bras de liaison

Agent de couplage A
Les dérivés de cyanines selon l'invention peuvent comporter un agent de couplage porté par un bras de liaison. De préférence, cet agent de couplage est capable de traverser la membrane cellulaire
Cet agent de couplage a pour fonction de permettre le couplage covalent ou non-covalent de la cyanme avec une molécule cible, par exemple présente dans une cellule. Pour cela, la molécule cible doit comporter le domaine de couplage approprié Dans le cas où la molécule
cible est présente dans une cellule, il est nécessaire que l'agent de couplage soit lui-même capable de traverser la membrane plasmique
L'agent de couplage A peut ainsi être un anticorps, un fragment d'anticorps ou encore un aptamère peptidique spécifique de la molécule cible Ces anticorps, fragments d'anticorps ou aptarnères peuvent reconnaître un domaine naturellement présent dans la molécule cible, ou encore un domaine introduit dans cette molécule par les techniques bien connues de l'homme du métier, en particulier les techniques de biologie moléculaire permettant I introduction dans une biomolécule d'une étiquette protéique.
L'agent et le domaine de couplage peuvent être des membres d'un couple de partenaires de liaison, les deux membres étant capables de lier de manière non-covalente, comme par exemple les couples suivants :
• avidine (ou streptavιdιne)/bιotιne . ce couple peut être utilisé pour marquer une protéine d'intérêt exprimée par la cellule sous forme de protéine de fusion avec le domaine de liaison de l'avidine ou de la streptavidine. Dans ce cas, les dérivés de cyanine selon l'invention sont conjugués à la biotine en tant qu'agent de couplage. Le couple (strept)avιdιne/bιotιne est connu de l'homme du métier qui n'aura aucun mal à coupler les dérivés de cyanine selon l'invention avec la biotine.
• composés bi-arsenic/motif tétracystéine . Ce couple de partenaires de liaison a été initialement décrit par Adams et al., (« New biarsenical ligands and tétracystéine motifs for protem labeling m vitro and in vivo' synthesis and biological applications », J Am Chem. Soc. 2002 May 29;124(21):6063-76). Un dérivé de cyanine selon l'invention, conjugué à un motif bi-arsenic, permet de marquer une protéine d'intérêt exprimée par la cellule sous forme de protéine de fusion avec un domaine tétracystéine capable de se lier avec le motif biarsenic. La séquence du domaine tétracystéine est : Cys-Cys-aa1-aa2-Cys-Cys (dans laquelle aa1 et aa2 sont tout acide aminé naturel, et de préférence aa1 = Pro et aa2 = GIy).

Des dérivés de cyanines selon l'invention, conjuguées au SLF' comme agent de couplage, seront capable de marquer une protéine d'intérêt exprimée par la cellule sous forme de protéine de fusion avec la FKBP(F36V)
L'agent de couplage A des dérivés de cyanine selon l'invention peut également être un agent permettant un couplage par liaison covalente avec un domaine de couplage présent sur la molécule d'intérêt que l'on souhaite marquer
En particulier, l'agent de couplage peut être le substrat d'une enzyme dite « suicide », présente dans la cellule Dans ce cas, le domaine de couplage est donc une enzyme suicide et est exprimé par la cellule sous la forme d'une protéine de fusion constituée de l'enzyme suicide et de la protéine d'intérêt que l'on souhaite marquer. Les enzymes suicides sont des protéines qui ont une activité enzymatique modifiée par des mutations spécifiques qui leur confèrent une capacité à lier rapidement et de façon covalente un substrat. Ces enzymes sont dites suicides car chacune ne peut lier qu'une seule molécule fluorescente, l'activité de l'enzyme étant bloquée par la fixation du substrat.
Les enzymes suicides ont été récemment utilisées dans des méthodes de marquage de protéines : ces méthodes consistent à fabriquer une protéine de fusion comprenant une protéine que l'on souhaite marquer et une enzyme suicide, et en mettant en contact cette protéine de fusion avec, par exemple, un fluorophore marqueur lié de manière covalente au substrat de ladite enzyme suicide.
Actuellement, deux familles d'enzymes suicides connues permettent ce type de marquage •
Le mutant d'une alkylguanine-DNA alkyltransférase (ou « SnapTag » commercialisé par la société Covalys, décrite dans la demande WO 02/083937 A2), dont le substrat est la benzylguanine ou un dérivé de benzylguanine. Par dérivé de benzylguanine on entend une benzylguanine modifiée mais qui est néanmoins reconnue par l'enzyme suicide. De tels substrats sont décrits dans les demandes WO 2005/085470 et WO 2004/031405 ;
Le mutant d'une haloalcane déhalogénase (« HaloTag » commercialisé par Promega) qui génère également une réaction enzymatique du type suicide (technique décrite dans WO 2004/072232 A2), dont le substrat un haloalcane, de préférence un chloroalcane Des substrats sont décrits dans WO 2004/072232 et WO 2006/093529
Dans le cas ou l'on souhaite marquer de telles enzymes suicides par un dérivé de cyanine selon l'invention, ladite cyanine comportera l'agent de couplage qui sera un groupe benzylguanine (ou un de ses dérivés), ou encore un haloalcane (de préférence un chloroalcane) lié à la cyanine via un bras de liaison
Pour résumer, l'agent de couplage A des dérivés de cyanines selon l'invention peut être un membre d'un couple de partenaires de liaison non-covalente ou un membre d'un couple de partenaires de liaison covalente
En particulier, A peut être choisi parmi les composés suivants un anticorps, un fragment d'anticorps, un aptamère peptidique, la biotine, un composé bi-arsenic, le tπmétropine, le méthotrexate, le SLF', un groupe benzylguanine, un chloroalcane
Groupe réactionnel G
Les dérivés de cyanines selon l'invention peuvent contenir un groupe réactionnel G, qui est un groupe capable de réagir avec un groupe fonctionnel présent sur une autre substance ou molécule, pour former une liaison covalente En l'occurrence, le groupe réactionnel G réagira avec un groupe fonctionnel présent sur la substance ou la molécule que l'on souhaite conjuguer au dérivé de cyanine selon l'invention
Typiquement, le groupe réactionnel est un groupe électrophile ou nucléophile qui peut former une liaison covalente lorsqu'il est mis en présence d'un groupe nucléophile ou électrophile approprié, respectivement. La réaction de conjugaison entre le dérivé de cyanine comportant un groupe réactionnel et une molécule à conjuguer M portant un groupe fonctionnel entraîne la formation d'une liaison covalente comportant un ou plusieurs atomes du groupe réactionnel. A titre d'exemples, les couples de groupes électrophiles/nucléophiles et le type de liaison covalente formée lorsqu'ils sont mis en présence sont listés ci-dessous
• par ester activé, on entend des groupes de formule COY, ou Y est
• un groupe partant, choisi parmi les groupes succinimidyloxy (-C4H4O2), sulfo succinimidyloxy (-OC4H3Oz-SO3H) ;

A titre d'exemple non limitatif, la substance ou molécule à conjuguer M comprend au moins un des groupes fonctionnels suivants, avec lequel réagira le groupe réactionnel G : aminés, amides, thiols, aldéhydes, cétones, hydrazines, hydroxylamines, aminés secondaires, halogénures, époxydes, esters de carboxylate, acides carboxyliques, des groupes comportant des doubles liaisons, ou une combinaison de ces groupes fonctionnels.
Le groupement fonctionnel porté par la molécule M qui réagira avec le groupe réactionnel G sera par exemple un groupe aminé, thiol, alcool, aldéhyde ou cétone. De manière préférée le groupement réactionnel G réagira avec un groupe fonctionnel aminé ou thiol.

Molécule conjuguée M
Bien que les dérivés de cyanine selon l'invention soient particulièrement avantageux pour le marquage fluorescent de composés se trouvant dans la cellule, ils peuvent être couplés à tout type de molécules par les techniques de conjugaison classique, et l'utilisation de groupes réactionnels tels que décrits ci-dessous.
La molécule conjuguée M peut être par exemple une biomolécule. Par biomolécule, on entend une molécule présente dans un organisme vivant, et en particulier les molécules constituant la structure d'un organisme, celles impliquées dans la production et la transformation d'énergie ou dans la transmission des signaux biologiques. Cette définition englobe les acides nucléiques, les protéines, les sucres, les lipides, les peptides, les oligonucléotides, les intermédiaires métaboliques, les enzymes, les hormones et les neurotransmetteurs.
Bras de liaison
Les dérivés de cyanines selon l'invention comporte un groupe réactionnel G, un agent de couplage A, une molécule conjuguée M ou encore un ester (de préférence diester) de phosphate ou phosphonate W. Chacun de ces groupes est lié à la cyanine via un bras de liaison.
Dans le cas où la cyanine comporte plusieurs de ces groupes, par exemple 2 groupes phosphonates et un agent de liaison, ces groupes peuvent être portés par des bras identiques ou différents.
Le bras de liaison L qui porte les groupe G, M, A ou W, peut être une liaison covalente simple ou un bras d'espacement comprenant de 1 à 20 atomes différents de l'hydrogène, choisis parmi les atomes de carbone, azote, phosphore, oxygène et soufre, ce bras de liaison étant linéaire ou ramifié, cyclique ou hétérocyclique, saturé ou insaturé, et constitué d'une combinaison de liaisons choisies parmi : les liaisons carbone-carbone qui peuvent être simples, doubles, triples ou aromatiques; les liaisons carbone-azote; les liaisons azote-azote; les liaisons carbone- oxygène; les liaisons carbone-soufre; les liaisons phosphore-oxygène; les liaison phosphore- azote ; les liaisons éther ; les liaisons ester ; les liaisons thioéther; les liaisons aminé , les liaisons amide ; les liaisons carboxamide; les liaisons sulfonamid; les liaisons urée; les liaisons uréîhane ; les liaisons hydrazme, les liaisons carbamoyle
De préférence, le bras de liaison L comprend de 1 à 20 atomes différents de l'hydrogène et choisis parmi C, N, O, P et S et peut comprendre des combinaisons de liaisons éther, thioéther, carboxamide, sulfonamide, hydrazine, aminé, ester et des liaisons aromatiques ou hétéroaromatiques.
De manière préférée, L est constitué d'une combinaison de liaisons simples carbone-carbone et de liaisons carboxamide ou thioéther.
A titre d'exemple, L peut être choisi parmi les chaînes suivantes : polyméthylène, arylène, alkylarylène, arylènealkyle, ou arylthio.

La synthèse de dérivés de cyanine est largement décrite dans la littérature et l'homme du métier peut s'y référer pour préparer les dérivés selon l'invention. La synthèse de cyanine repose en général sur l'utilisation de 3 réactifs de départ . les groupes polycycliques et la chaîne méthine Ces 3 réactifs sont modifiés avant ou après leur condensation pour leur faire porter les substituants voulus

Exemple 1 : Synthèse du bromure de 1-[2-(Diéthoxyphosphoryl)-éthyl]-2,3,3-triméthyl-3W- indolium
1 ,59 g (10,0 mmol) de 2,3,3-triméthyl-3H-indole et 2,94 g (12,0 mmol) de bromoéthyl-diéthyl- phosphonate sont mélangés et chauffés à 7O0C pendant 40 h sous agitation. Après refroidissement du mélange réactionnel, 10 ml de méthanol sont ajoutés puis ensuite 30 ml d'eau. Le composé recherché est isolé par chromatographie sur colonne (silice RP-18) avec un gradient de méthanol/eau allant de 1 :1 to 5:1. MS (ESI+): 324,2 .Rendement: 775 mg (19 %).
Exemple 2 : Synthèse du chlorure de 3-(3-Carboxypropyl)-1-[2-(diéthoxyphosphoryl)- éthyl]-2,3-diméthyl-3H-indolium
2,31 g (10,0 mmol) de 3-(3-Carboxypropyl)-2,3-diméthyl-3W-indole et 2,94 g (12,0 mmol) de bromoéthyl-diéthylphosphonate sont dissous dans 10 ml d'éthanol et agités à 7O0C pendant 64 h. Après retour à température ambiante, le liquide restant est extrait par deux fois avec 20 ml d'eau. Les deux phases aqueuses réunies sont extraites deux fois par 20 ml de diéthyléther. Le produit désiré resté dans la phase aqueuse est isolé par chromatographie sur colonne (silice RP-18) avec un gradient de méthanol/eau allant de 1 :3 to 3:2 (chaque solvant contenant 2% en volume d'acide chlorhydrique 3M). La solution contenant le produit désiré est neutralisée par du bicarbonate de sodium, le mélange est concentré puis déposé sur une colonne courte de silice RP-18. Après avoir éliminé les sels inorganiques par lavage à l'eau, le produit sous forme de sel de sodium est élue avec du méthanoi. MS (ESI+): 396,2. Rendement: 575 mg (12 %)
Exemple 3 : Synthèse du chlorure de 1-[2-(diéthoxyphosphoryl)-éthyI]-3,3-diméthyl-2-
((1 E,3E)-4-phénylamino-buta-1 ,3-diényl)-3W-indolium
404 mg (1 ,0 mmol) de bromure de 1-[2-(diéthoxyphosphoryl)-éthyl]-2,3,3-triméthyl-3H-indolium et 285 mg (1 ,1 mmol) de chlorhydrate du dianilide de l'aldéhyde malonique sont dissous dans 10 ml d'un mélange d'acide acétique et d'anhydride acétique (v/v = 3:2) et le mélange est agité 8 h à 60 0C. After retour à température ambiante 150 ml water d'eau sont ajoutés. La solution est déposée sur une colonne de silice RP-18. After lavage avec 200 ml d'eau, le produit recherché est élue par un gradient acétonitrile/eau allant de 1:2 à 3:2 (chaque solvant contenant 2% en volume d'HCI 3M), La solution contenant le produit désiré est neutralisée par du bicarbonate de sodium, le mélange est concentré puis déposé sur une colonne courte de silice RP-18. Après avoir éliminé les sels inorganiques par lavage à l'eau le produit sous forme de sel de sodium est élue avec du méthanol. MS (ESI+): 453,2 Rendement: 76 mg (14 %).
Exemple 4 : Synthèse du chlorure de 2-{(1 E,3E)-5-[3-(3-Carboxypropyl)-1-[2- (diéthoxyphosphoryl)-éthyl]-3-méthyl-1 ,3-dihydro-indol-(2E)-ylidène]-penta-1 ,3-diényl}-1- [2-(diéthoxyphosphoryl)-éthyl]-3,3-dirnéthyl-3W-indoliurn (cyanine 1 sous forme de diéthylphosphono ester)
48 mg (100 μmol) chlorure de 3-(3-carboxypropyl)-1 -[2-(dιéthoxyphosphoryl)-éthyl]-2,3- dιméthyl-3/-/-ιndolιum obtenu à l'exemple 2, et 53 mg (100 μmol) de chlorure de 1 -[2- (dιéthoxyphosphoryl)-éthyl]-3,3-diméthyl-2-((1 E,3E)-4-phénylamιno-buta-1 ,3-dιényl)-3H- indolium obtenu à l'exemple 3, et 25 mg (300 μmol) d'acétate de sodium sont dissouts dans 5 ml d'un mélange d'acide acétique et d'anhydride acétique (v/v = 3:2) et le mélange est agité 3 h à 60 0C. Après concentration sous vide, le résidu est d'abord dissous dans 1 ml de méthanol, et 2 ml d'eau sont ajoutés. Le composé recherché est isolé par chromatographie sur une colonne de silice RP-18 en éluant par un gradient méthanol/eau, gradient allant de 1 :1 à £M MS (ESI+)- 755,3. Rendement: 32 mg (38 %).
Exemple 5 : Synthèse du dérivé aminoéthylamide de la cyanine 1 sous forme de bis- diéthylphosphono ester.
Afin de pouvoir greffer une molécule d'intérêt tel qu'un substrat d'une enzyme permettant un marquage intra-cellulaire, on introduit un groupement aminé primaire.
Une solution de la cyanine 1 sous forme de bis-diéthylphosphono ester obtenu à l'exemple 4 (160 nmol) dans 100 μl de DMF est traitée par 5 équivalents de dnsopropyléthylamine, 2,5 équivalents de tert-butyl N-(2-amιnoéthyl)carbamate (N-boc-éthylènediamine) et 2 équivalents de TBTU après 24 h à température ambiante le produit de départ (tR = 15,7 mn) est transformé en un nouveau produit (tR = 16,8 mn).
Conditions HPLC : colonne Lichrospher* 100, RP18, 5 μm, 125mm x 4mm ; gradient d'acétonitπle (ACN) dans l'acide trifluoracétique (TFA) à 0,1% dans l'eau à 1 ml/mn. Isocratique
5% ACN 3 min, gradient linéaire de 5% à 100% en 12 min.
Le produit intermédiaire obtenu, dissous dans de l'acétonitrile, est traité par l'acide tπfluoroacétique (ACN/TFA : 2/1 (v/v)). Le produit possédant Pamine déprotégée (tR=1 1 ,5 mn) est isolé par HPLC semi-préparative (colonne Grace-Vydac Protein & Peptide C18, 218TP510) débit 4 ml/mn avec un gradient ACN / TFA à 0,1% dans l'eau identique au gradient analytique.
Exemple 7 : Synthèse du dérivé N-Hydroxysuccinimide de la cyanine 1 sous forme de bis-diéthylphosphono ester.
La cyanine sous forme de bis-diéthylphosphono ester obtenue dans l'exemple 4 (1 ,5 mg ,1 ,79 μmole) est dissoute dans 70 μl de DMF anhydre et on ajoute 1 ,25 μl de DIPEA et 0,55 mg de

La O6-[4-(13-Amιno-2,5,8,11 -tétraoxa-tπdécyl)-oxyméthyl-benzyl]guanιne préparée selon le protocole de la littérature [Gendreizig S et al (2003) "Induced protem dimenzation in vivo through covalent labeling " J Am Chem Soc 125(49). 14970] (0 46 μmole) dans 70 μl de DMF anhydre est traitée par 0 3 μl de DIPEA et 0 46 μmole de dérivé N-Hydroxysuccinimide de la cyanine bis-diethylphosphono ester préparée selon l'exemple 7. Par analyse en HPLC (colonne Lichrospher® 100, RP18, 5 μm, 125mm x 4mm ; gradient d'acétonitπle (ACN) dans l'acide tnfluoracétique (TFA) à 0,1% dans l'eau à 1 ml/mn. Isocratique 0% ACN 5 min, gradient linéaire de 0% à 70% en 13 min puis de 85% à 100% en 2 min, détection à 280 nm) on observe la disparition du pic correspondant au dérivé guanine de départ (tR=12,1 mn) et formation d'un pic présentant un temps de rétention plus long (tR=16,4 mn). Exemple 11 : Test de perméabilité cellulaire de conjugués de la Benzylguanine avec le DY647, le CY5 et la cyanine 1 sous forme de bis-diéthylphosphono ester.
Les cellules utilisées sont issues d'une lignée stable CHO exprimant le SnapTag au niveau nucléaire grâce à la séquence NLS. Les composés sont ajoutés dans le milieu de culture (milieu DMEM complémenté par 10% de SVF inactivé 30mιn à 600C) à une concentration de 5 μM et laissés en contact une heure avec les cellules. Après l'incubation, trois lavages sont effectués avec du milieu de culture. Une heure supplémentaire d'incubation uniquement dans milieu est faite pour permettre le marquage du SnapTag par le substrat Une coloration du noyau est ensuite réalisée avec le DAPI avant l'analyse microscopique. L'incubation des cellules avec les conjugués BG-DY647 et BG-CY5 produisent des images peu intenses , en augmentant le gain on observe de la fluorescence uniquement sous forme d'un bruit de fond ce qui montre que les conjugues testés n'on pas traverse la membrane plasmique
des cellules Au contraire l'incubation des cellules avec le conjugue BG-CEP (BG-Cyanine Ethyl Phosphonate), préparé selon l'exemple 8, produit une fluorescence intense localisée à l'intérieur des cellules montrant que le composé a traversé la membrane plasmique. Letrahedron Lett 18, 2 (1977), 155-158.] ; après 1 h30 on observe un précipité, on évapore la solution, reprend le résidu dans un mélange d'eau et d'acétonitnle et purifie par RP-HPLC (détecteur barette de diodes Shimadzu SPD-M10A, Pompe Merck L6200A, colonne Vydac 218TP510) A : H2O contenant 0 1% TFA B acétonitπle. Débit = 4 ml/min. Gradient linéaire de 5% ACN à 100 % d'acétonitnle en 15 min. Le produit de départ (tR=11 ,9 min) donne majoritairement le composé recherché portant deux fonctions éthylphosphono ester (tR= 0,1 min) On observe également des pics à tR=10,6 min et à tR=9,7 min correspondant respectivement à la diphosphono cyanine portant trois groupes éthyle et un groupe éthyle.
Exemple 13 : Préparation des dérivés 2-{(1£,3E,5£)-5-[3-(5-carboxypentyl)-4-éthyl-3- méthyI-5-phophono-1 ,3-dihydro-2W-indol-2-ylidène]penta-1,3-dienyl}-5-phosphono-3,3- diméthyl-1-(4-éthyl)-3W-indolium.
Le procédé de synthèse de ce composé est représenté sur le schéma réactionnel 1.
13.1. 5-bromo-2,3,3-trιméthyl-3H-ιndole (14a):
Le 4-bromophénylhydrazιne.HCI 13 (6,5 g) et la 3-méthyl-2-butanone (6,4 ml) sont dissous dans du benzène contenant une quantité catalytique d'acide acétique et le mélange est porté à reflux (16h) avec entraînement azeotropique de l'eau. Le mélange concentré est repris par de l'acide acétique (70 ml) et chauffé au bain d'huile (1200C) pendant 12 heures ; on observe la formation d'un produit solide. L'analyse par HPLC (RP-18 gradient acétonitπle dans l'eau à 1 % de TFA) montre la formation d'un nouveau produit. Après évaporation sous vide le résidu est repris par de l'acétate d'éthyle et lavé avec une solution saturée d'hydrogénocarbonate de sodium, et la phase organique est séchée (MgSO4) et évaporée, le produit brut est purifié par chromatographie-Flash sur silice en éluant par un gradient d'acétate d'éthyle dans l'hexane. On obtient 5,5 g (80%) de produit pur. 1H-RMN (CDCI3)O= 7,5(1 H,d) , 7,2-7,3 (2H, m), 2,3 (3H,s) , 1 ,3 (6H, s).[Letcher, R.M. et al., J. Chem. Soc. Perkin Trans., 1 (1993) 939-944].
13.2. 5-(dιéthoxyphosphono)-2,3,3-trιméthyl-3H-ιndole (15a) :
Le 5-bromo-2,3,3-tπméthyl-3/-/-ιndole 14a (5g , 21 mmol) est dissous dans du toluène (5ml) ; on dégaze puis on ajoute de la tπéthylamine (3,2 ml), du diethylphosphite (3ml) et du tétrakιs(tπphénylphosphιne)-palladιum(0) (1 ,2 g) ; on chauffe à 800C pendant 3h sous azote et sous agitation. On chromatographie (chromatographie flash) sur silice en éluant avec un gradient d'acétate d'éthyle dans l'hexane. On obtient 3 g (48%) de produit pur. 1H-RMN (CDCI3) δ = 7,7 (2H,m) , 7,6 (1 H, m), 4,13 (4H, m), 2,3 (3H,s) , 1 ,3 (9H, m).
13.3. 5-(dιéthoxyphosphono)-1 -éthyl-2,3,3-trιméthyl-3/-/-ιndolιum iodure (16a):
Le 5-(dιéthoxyphosphono)-2,3,3-trιméthyl-3/-/-ιndole 15a (3g) est traité par de l'iodure d'éthyle (10 ml) à reflux 16h sous azote. L'iodure d'éthyle est évaporé sous vide et le résidu est trituré avec de l'hexane, collecté par centπfugation et séché. Il est utilisé tel quel pour la réaction suivante.
13.4. 5-(phosphono)-1-éthyl-2,3,3-trιmethyl-3W-ιndolιum bromure : Le 5-(dιéthoxyphosphono)-2,3,3-trιméthyl-3H-ιndolιum 16a est traité par un excès de bromure de tnméthylsilyle dans le dichlorométhane (5 ml) selon le protocole de la littérature [McKenna CE. et al. Letrahedron Lett 18, 2 (1977), 155-158.]. On obtient ainsi le dérivé complètement déprotégé sous la forme « acide phosphonique » 17a.
13.5. Préparation du 3,3-dιméthyl-i -éthyl-2-((1 E,3E)-4-phénylamιno-buta-1 ,3-dιényl)-5-
(phosphono)-3H-ιndolium chlorure (18):
Le bromure de 1 -éthyl-5-(phosphono)-2,3,3-trιméthyl-3W-ιndolιum 17a (10 mmol) est traité par de l'anhydride acétique (10 ml) et du dianilide de l'aldéhyde malonique (20 mmol) ; on laisse réagir 8h à 600C. On refroidit, ajoute de l'éther (50 ml) et de l'hexane (50 ml), puis on sépare le produit huileux (qui est l'ami recherché) par décantation, on purifie par chromatographie (RP-18 exemple 3) pour donner l'ami 18. Rendement: g (40%).
13.6. Préparation du 5-bromo-3-(5-carboxypentyl)-2,3-dιméthyl-3W-ιndole (14b) : Le 4-Bromophénylhydrazιne, HC1 1_3 (6,5 g) et l'acide 7-méthyl-8-oxo-nonanoιque (6,7 g)
[préparé à partir du 2-méthylacetoacetate d'éthyle et du 6-bromohexanoate d'éthyle selon le procédé décrit par LEUNG, Wai-Yee dans le WO 02/26891] sont portés à reflux dans de l'acide acétique (50 ml) pendant 5 heures. Après évaporation le produit est purifié par chromatographie-Flash sur silice, (rendement 1 1g).
13.7. Préparation du 3-(5-Carboxypentyl)-5-(dιéthoxyphosphono)-2,3-dιméthyl-3H-ιndole
Le 5-bromo-3-(5-carboxypentyl)-2,3-dιméthyl-3H-ιndole (14b) est traité par le diéthylphosphite comme décrit dans l'exemple 13.2 pour donner le dérivé 15b.
13.8. Préparation du iodure de 3-(5-Carboxypentyl)-5-(dιéthoxyphosphono)-2,3-dιméthyl-1- éthyle-3H-ιndolιum (16b) :
Le 3-(5-Carboxypentyl)-5-(dιéthoxyphosphono)-2,3-dιméthyl-3H-ιndole (15b) est traité par Piodure d'éthyle (voir exemple 13.3) pour donner le composé 16b.
13.9. Préparation du bromure de 3-(5-Carboxypentyl)- 2,3-dιméthyl-1-éthyl-5-(phosphono)- 3H-ιndolιum (17b) :
Le iodure de 3-(5-Carboxypentyl)-5-(dιéthoxyphosphono)-2,3-dιméthyl-1 -ethyle-3H-ιndolιum (16b) est traité par le bromure de tπméthylsilyl (voir exemple 13.4) pour donner le composé 17b.
13.10. Diphosphonocyanine fonctionnalisée carboxylate (19)
2-{(1 E,3E,5£)-5-[3-(5-carboxypentyl)-3-méthyl-4-éthyl-5-phophono-1 ,3-dιhydro-2/-/-ιndol-2- ylιdène]penta-1 ,3-dιényl}-5-phosphono-3,3-dιméthyl-1 -(4-éthyl)-3W-ιndolιum : L'ami 1J3 (g, 7 mmol), et le bromure de 3-(5-Carboxypentyl)- 2,3-dιméthyl-1 -éthyl-5- (phosphono)-3H-ιndolιum (17b) sont dissous dans le diméthylformamide (10 ml), on ajoute de la tπethylamine (1 ml) et de l'anhydride acétique (1 ml) et chauffe à 60° C pendant 1 heure. On refroidit, ajoute de l'éther éthylique (50 ml), et le précipité est collecté repris par de l'acide chlorhydπque dilué (2 N) puis purifié par chromatographie (Lichroprep Merck C18) en éluant avec 1 °) une solution d'acétate de sodium (1 ,5 m M) 2") de l'eau distillée , le produit recherché
19 est ensuite élue par un mélange éthanol-eau (4/6). On obtient ainsi la diphosphonocyanine
19 sous la forme de sel de sodium. A côté de la diphosphonocyanine 19 recherchée on observe aussi la présence d'une diphosphocyanine non fonctionnalisée 20 provenant du couplage de |8 sur le composé 17a contaminant 1§, ces deux composés étant séparables par chromatographie en phase inverse. HPLC (détecteur barette de diodes Shimadzu SPD-M10A, Pompe Merck L6200A, colonne Vydac 218TP510) A : H2O contenant 0 1% TFA B : acétonitπle. Débit = 4 ml/min. Gradient linéaire de 5% ACN à 60% acétonitnle en 23 min. Composé 19 : tR=14,8 min. (N.B. le composé 20 présente un tR = 14 min dans les mêmes conditions) Rendement: 0,5 g (8%). Masse (ES-) m/z = 669,3 (100%) (ES+) m/z = 671 ,2 (60%). Spectre U V- Vis : λmax (tampon PO4 0,1 M pH 7) = 649 nm (ε649 98 000 M 1.cm ~1) ratio A649ZA604 = 2,6.
Spectre de fluorescence : Mesure sur appareil Perkin-Elmer LS50 , les échantillons ont été dilués dans du tampon phosphate 0,1 M pH 7 contenant 0.1 % BSA (absorbance maximum = 0,04 à la longueur d'onde d'excitation). Excitation : 600 nm (10nm). Emission (max) = 670, 5nm (rendement quantique = 13%).
Exemple 14 : Diphosphonocyanine tetraester fonctionnalisée aminé (24): (voir schéma réactionnel 2)
14.1. Dérivé N-Boc (21) :
La Diphosphonocyanine fonctionnalisée carboxylate Jj) (24 mg, 32.6 μmol) est dissoute dans le DMF (2 ml) et on ajoute 9 μl de DIPEA et une solution de TSTU (35 mg dans 500μl de DMF). Après 20 min on ajoute le dérivé mono-BOC de Péthylène diamine. Après 1 h de réaction on purifie par RP-HPLC comme décrit dans l'exemple 12. On obtient ainsi le dérivé N-BOC 2J.. MS (ES+) m/z = 81 1 ,6 (100%) calculé pour C41H55N4O9P2 .
14.2. Dérivé N-Boc de la Cyanine di-(phosphono-mono-ester méthylglycolique) (22) :
Le dérivé N-BOC (21) (3 mg, 4,7 μmol) est repris par du DMF anhydre (550 μl), on ajoute de la tπéthylamine anhydre (distillée sur hydrure de calcium, 14 μl, 100μmol) du méthyl bromoacétate
(1 Oμl, 105μmol). On chauffe à 700C pendant 16h sous argon. Le mélange réactionnel est purifié par RP-HPLC comme décrit dans l'exemple 12 (colonne Vydac 218TP510 , A : H2O contenant
0.1% TFA B : acétonitπle. Débit = 4 ml/min. Gradient linéaire de 5% ACN à 100 % d'acétonitnle en 40 mm). Le produit de départ (IR= W mm) a donné majoritairement le composé recherché 22 portant deux fonctions phosphono ester glycolique (tR = 22.6 min) Rendement 15 %. MS (ES+) m/z = 957,5 (100%) calculé pour C47H66N4O13P2.
14.3. Dérivé N-Boc de la Cyanine dî-(phosphonodiester méthylique et méthylglycolique)
(23) :
Méthode A. Le dérivé phosphono ester monoglycolique (22) est traité par Piodure de méthyle, on obtient ainsi le dérivé 23 portant ayant chaque groupe phosphonate sous la forme phosphono diester méthyle et méthyle glycolique
Méthode B. Le dérivé phosphono ester monoglycolique (22) dans le DMF est traité par du BOP en présence de méthanol pour donner le produit 23 Le produit est purifié par RP-HPLC (colonne Vydac 218TP510 , A H2O contenant 0.1 % TFA B acétonitπle. Débit = 4 ml/min. Gradient linéaire de 5% ACN à 100 % d'acétonitπle en 40 min). On observe majoritairement le composé recherché 23 (tR = 31 ,7 min) Rendement 50 % UV(H2O/ACN , 1 :1 ) ; ?vmax = 646,5 nm, ratio A646/ A600 = 3,7. MS (ES+) m/z = 985,6 (100%) calculé pour C49H71 N4OIqP?.
14.4. Cyanine di-(phosphonodiester méthylique et méthylglycolique) fonctionnalisée aminé (24): La cyanine dι-(phosphonodιester méthylique et méthylglycolique) 23 est traitée par l'acide tπfluoracétique dans le dichlorométhane (30 min à 200C) ; après évaporation du solvant le produit est purifié par RP-HPLC On obtient ainsi la cyanine tétraester fonctionnalisée aminé 24.
Exemple 15 : Synthèse d'un dérivé benzylguanine de diphosphonocyanine tétraester (26)
(schéma réactionnel 2) :
La cyanine 24 ainsi obtenue est reprise par du DMF contenant de la DIPEA et traitée par l'ester de NHS 25 [obtenu par réaction d'un excès de DSS (di-succmimidyl suberate) sur la O6-[4- (amιno-méthyl)-benzyl]guanιne (Keppler, S. et al. (2003) Nature Biotechnology 21 (1 ): 86 - 89) dans le DMF, le produit 25 est purifié par RP-HPLC dans des conditions semblables à celles utilisées pour la purification des cyanines dans les exemples précédents]. Le composé recherché 26 portant deux fonctions mixtes phosphono esters méthylique et méthylglycolique et fonctionnalisé par un groupe benzyl-guanine est obtenu après purification par RP-HPLC (colonne Vydac 218TP510, A : H2O contenant 0.1 % TFA B • acétonitrile. Débιt=4 ml/min Gradient linéaire de 5% ACN à 100 % d'acétonitπle en 40 min). Rendement 46 %. UV(H2O/ACN , 1 :1 ) , λmax = 650 nm, ratio A647/A600 = 3,4. MS (ES+) m/z = 1293,8 (M-H)+ calculé pour
Exemple 16. Dérivé benzyl-guanine de Ia Cyanine di-(phosphonodiester méthylglycolique) (29) : (voir schéma réactionnel 3)
Le dérivé phosphono ester monoglyeolique (22) dans le DMF est traité par du BOP en présence de méthyl glycolate pour donner le produit 27 Le produit est purifie par RP-HPLC (colonne Vydac 218TP510, A H2O contenant 0 1% TFA B acétonitπle Débit = 4 ml/mm Gradient linéaire de 5% ACN à 100 % d'acétonitnle en 40 min) On observe le composé portant trois
fonctions esters méthylglycolique (tR=29,9 min) ainsi que le composé recherché 27 portant quatre fonctions esters methylglycolique recherché Rendement 20 %. MS (ES+) m/z = 1102 Calculé pour C53H75N4O17P2.
Le dérivé 27 est traité 15 min dans l'acide tπfluoracétique pur pour enlever le groupe protecteur BOC, puis, après évaporation sous vide et coévaporation au toluène, le produit 28_aιnsι obtenu est repris par du DMF contenant de la DIPEA et traité par l'ester de NHS 25 obtenu par réaction d'un excès de DSS (di-succinimidyl suberate) sur la O6-[4-(amιno-méthyl)-benzyl]guanιne dans le DMF (le produit 25 est purifié par RP-HPLC dans des conditions semblables à celles utilisées pour la purification des cyanines dans les exemples précédents) Le composé recherché 29 portant quatre fonctions esters méthylglycolique et fonctionnalisé par un groupe benzyl-guanine est obtenu après purification par RP-HPLC (colonne Vydac 218TP510, A : H2O contenant 0,1% TFA B . acétonitnle. Débιt=4 ml/min. Gradient linéaire de 5% ACN à 100 % d'acétonitπle en 40 min)
Le produit 29 est obtenu après évaporation des fractions issues de la purification HPLC et coévaporation au toluène, il est ensuite repris par du DMSO et stocké à -2O0C. Rendement 70 %. UV(H2O/ACN, 1 :1 ) ; Gmax = 647 nm, ratio A647/A600 = 3,5. MS (ES+) m/z = 1409,6 (M-H)+ calculé pour C69H9IN10O18P2.
Exemple 17 : Comparaison des rendements quantiques de différents dérivés de Cyanine :
Les fonctions esters de phosphate/phosphonate des dérivés de cyanine selon l'invention, sont hydrolysées dans la cellule par les enzymes cellulaires en fonction phosphate/phosphonate ; Les rendements quantiques des dérivés de cyanine portant des groupes phosphonates, correspondant donc aux composés selon l'invention tels qu'ils sont présents dans la cellule, après hydrolyse des fonctions esters, ont été mesurés, afin de déterminer si la position des groupes esters de phosphate/phosphonate a une influence sur les propriétés photophysiques des composés.
Ces résultats montrent que les cyanines comportant des fonctions phosphonates au niveau des cycles indoles ont un rendement quantique 3 fois plus élevé que celles comportant des phosphonates au niveau des azotes des cycles indoles.
Exemple 18 : Dérivé benzyl-guanine de la Cyanine di-(phosphonomonoester méthylglycoϋque) (31) : (voir schéma réaction nel 4)
Le dérivé phosphono ester monoglycolique (22), décrit dans l'exemple 14, est traité 15 min dans l'acide trifluoracétique pur pour enlever le groupe protecteur BOC, puis, après évaporation sous vide et coévaporation au toluène, le produit 30 ainsi obtenu est repris par du DMF contenant de la DIPEA et traité par l'ester de NHS 25 préparé comme décrit dans l'exemple 16. Le composé recherché 3J. portant deux fonctions esters méthylglycolique et fonctionnalisé par un groupe benzyl-guanine est obtenu après purification par RP-HPLC (colonne Vydac 218TP510, A : H2O contenant 0.1% TFA B : acétonitrile. Débit=4 ml/min. Gradient linéaire de 5% ACN à 100 % d'acétonitrile en 40 min). Le produit est obtenu après évaporation des fractions issus de la purification HPLC et coévaporation au toluène, il est ensuite repris par du DMSO et stocké à -200C. Rendement 53 %. UV(H2O/ACN, 1 :1) ; λmax = 648 nm, ratio A648/A600 = 3,2. MS (ES-) m/z = 1263.6 (M-2H)" calculé pour C63H82N10O14P2.
Exemple 19 : Marquage d'une protéine intracellulaire par le composé 29 ajouté sur cellule vivantes intactes, détection du marquage par mesure HTRF:
Cet exemple vise à montrer que les composés selon l'invention comportant un ester de phosphonate et conjugués à un substrat (Benzylguanine, BG) d'une enzyme suicide « snaptag » (ST26) sont capables de traverser Ia membrane cellulaire, et de marquer une
protéine de fusion exprimée par la cellule et comprenant l'enzyme snaptag (ST12), par opposition à l'utilisation d'une cyanine ne comportant pas d'esters de phosphonate (DY-647)
La technique HTRF est utilisée pour tester le marquage intracellulaire d'une protéine par les dérivés de cyanine selon l'invention, sur des cellules vivantes, en utilisant la protéine recombinante GST-ST26-Flag produite par les cellules et marquée indirectement via un anticorps anti-GST conjugué avec un cryptate d'europium tπs-Bipyπdine (anti-GST-EuTBP)
Après mise en culture, des cellules CHO-K1 sont transfectées en présence de lipofectamine 2000 (0.8 μl_ pour 50 μl_ solution de transfection, Lipofectamine™ Transfection Reagent
Invitrogen, Inc., Carlsbad, Calif ) à raison de 50 000 cellules par puits dans une microplaque noires 96 puits. Les cellules transfectées sont ensuite incubées 24h à 37°C, 5% CO2. Le plasmide utilisé pour la transfection est un plasmide pSEM-GST-ST26-Flag (80ng / puits) obtenu en introduisant les séquences de la GST et du Flag dans le plasmide pSEMS- SNAP26m-Gateway en suivant le protocole du kit commercial Covalys (Mammalian Expression
Plasmid Kit pSEMSSNAP26m-Gateway® PL218), de façon à induire la synthèse d'une protéine de fusion GST-ST26-Flag par les cellules. Le protocole est semblable à celui décrit dans la littérature [Engineering Substrate Specificity of O6-Alkylguanιne-DNA Alkyltransferase for
Spécifie Protein Labehng in Living CeIIs, K. Johnsson et al. ChemBioChem. 2005, 6(7) 1263 - 1269].
Le lendemain, les cellules sont incubées avec le BG-DY647 ou le composé 29 (préparé selon l'exemple 16) ajoutés sous forme de solution dans le DMSO dans 50μl de milieu de culture et de façon à avoir une concentration finale en substrat de 5 μfvl Les cellules sont incubées 3h à 200C, puis lavées à l'aide d'un milieu de culture.
Marquage total : Sur une partie des puits servant à déterminer le « 100% de marquage» on ajoute le substrat à 10OnM final (BG-DY647 ou le composé 29_suιvant le puits) dans un tampon de lyse (cAMP kit , Cisbio ) contenant également un anticorps anti-GST couplé à un cryptate d'europium (EuTBP), l'anti-GST-EuTBP (1 nM final, GST tag check kit réf. 62GSTPEB, Cis bip).
Marquage passif Sur les autres puits servant à déterminer le pourcentage de marquage intracellulaire (qui ont donc été incubés soit avec DY647 soit avec le composé 29) on ajoute seulement l'anticorps anti-GST-EuTBP (1 nM final) dans le même tampon de lyse
On prépare également des puits contenant des cellules transfectées avec un plasmide « vide » c'est-à-dire ne codant pas pour la séquence d'intérêt, pour constituer des puits de contrôle
La fluorescence à 665 nm est mesurée sur un lecteur HTRF (RUBYstar, BMG Labtechnologies)
. Le pourcentage de marquage intracellulaire, obtenu en divisant le signal à 665 nm obtenu en marquage passif par le signal à 665 nm obtenu en marquage total est représenté sur la figure 2.
On observe que le pourcentage de marquage intracellulaire en présence du substrat 29 est beaucoup plus important que le pourcentage de marquage obtenu en présence du substrat de référence DY-647 qui pénètre mal dans les cellules.
Cet exemple confirme donc que le composé 29 traverse la membrane cellulaire et qu'il permet le marquage d'une protéine intracellulaire, ce qui n'est pas le cas du composé BG-DY-647 ne portant pas d'esters de phosphates ou phosphonates.

7. Dérivé de cyanine selon l'une quelconque des revendications 1 à 3, caractérisé en ce que un ou deux RrR4 représentent un groupe L1 -W et en ce que R15 ou R16 représente un groupe L2- A, L2-G ou L2-M.
8. Dérivé selon l'une quelconque des revendications 1 à 7, caractérisé en ce que le groupe W est un diester de phosphonate.
9. Dérivé de cyanine selon l'une quelconque des revendications 1 à 8, caractérisé en ce que W est un diester de phosphonate de formule :

12 Dérivé de cyanine selon l'une quelconque des revendications 1 à 1 1 » caractérisé en ce que les bras de liaison L1 et L2 sont choisis parmi : une liaison covalente simple ou un bras d'espacement comprenant de 1 à 20 atomes différents de l'hydrogène choisis parmi les atomes de carbone, azote, phosphore oxygène et soufre, ce groupe de liaison étant linéaire ou ramifié, cyclique ou hetérocyclique, saturé ou insaturé et constitué d'une combinaison de liaisons choisies parmi • les liaisons carbone-carbone qui peuvent être simples, doubles, triples ou aromatiques, les liaisons carbones-azote, les liaisons azotes-azote; les liaisons carbone- oxygène, les liaisons carbone-soufre, les liaisons phosphore-oxygène, les liaison phosphore- azote , les liaisons éther , les liaisons ester , les liaisons thioéther, les liaisons aminé , les 5 liaisons amide ; les liaisons carboxamide, les liaisons sulfonamide, les liaisons urée; les liaisons urethane ; les liaisons hydrazine , les liaisons carbamoyle.

Dérivé de cyanme selon l'une quelconque des revendications 1 à 12, caractérisé en ce que le groupe de liaison L comprend de 1 à 20 atomes, différents de l'hydrogène, choisis parmi I O les atomes de carbone, azote, phosphore, oxygène et soufre et comprend en outre au moins une liaison choisie parmi les liaisons éther, thioéther, carboxamide, sulfonamide, hydrazine, aminé, ester et des liaisons aromatiques ou hétéroaromatiques
14 Dérivé de cyanme selon l'une quelconque des revendications 1 à 13, caractérisé en ce 15 que le groupe de liaison L est choisi parmi les chaînes substituées ou non substituées suivantes : polyméthylène arylène, alkylarylène, arylènealkyle, ou arylthio.
15 Dérivé de cyanme selon l'une quelconque des revendications 1 à 14, caractérisé en ce que le groupe réactionnel G est choisi parmi les groupes dérivés des composés suivants : une 0 acrylamide, une aminé activée (par exemple une cadavenne ou une éthylènediamine), un ester activé, un aldéhyde, un halogénure d'alkyle, un anhydride, une aniline, un azide, une aziπdine, un acide carboxylique, un diazoalkane, une haloacétamide, une halotnazine, telle que monochlorotnazine, la dichlorotπazine, une hydrazine (y compris les hydrazides), un imido ester, un isocyanate, un isothiocyanate, un maléimide, un halogénure de sulfonyle, ou thiol, une5 cétone, une aminé, un halogénure d'acide, un hydroxysuccinimidyl ester, un hydroxysulfosucαnimidyl ester, un azidonitrophényl, un azidophényl, une 3-(2-pyπdyl dithio)- propionamide, glyoxal et en particulier les groupes de formule
- NH -4- (CH2)n - NH — Ar
où n varie de 0 à 8 et p est égal à O ou 1 , et Ar est un hétérocycle à 5 ou 6 chaînons comprenant 1 à 3 hétéroatomes, éventuellement substitué par un atome d'halogène.

Dérivé de cyanine selon la revendication 15, caractérisé en ce que le groupe A est choisi parmi : la benzylguanine ou un de ses dérivés substrat de Palkylguanyl transférase ; un haloalcane substrat de l'haloalcane déhalogénase en particulier un chloroalcane ; un anticorps ; un fragment d'anticorps ; un aptamère protéique ; la biotine, un composé bi-arsenic, le tnméthopnm, le méthotrexate, SLF'.
17. Dérivé de cyanine selon l'une quelconque des revendications 1 à 16, caractérisé en ce que le groupe M est une biomolécule choisie parmi . les acides nucléiques, les protéines, les sucres, les lipides, les peptides, les oligonucléotides, les intermédiaires métaboliques, les enzymes, les hormones, les neurotransmetteurs
18. Utilisation d'un dérivé de cyanine selon l'une quelconque des revendications 1 à 17 en tant que marqueur fluorescent.
19. Méthode de marquage d'une biomolécule présente dans une cellule, caractérisée en ce qu'elle consiste à introduire dans le milieu extracellulaire un composé selon l'une quelconque des revendications 1 à 17, comportant un agent de couplage A, ladite biomolécule comportant un domaine de couplage.
