Claims (3)
- Procédé pour analyser la présence d'un genre et d'une espèce d'un agent pathogène bactérien dans un échantillon clinique, comprenant les étapes consistant à- isoler, au moins partiellement, un acide nucléique à partir dudit échantillon, caractérisé en ce que ledit acide nucléique est choisi dans un groupe constitué par de l'acide nucléique total, de l'ADN total ou de l'ARN total- quantifier la quantité d'acide nucléique comprenant la région espaceur d'ADNr 16S/23S ou une de partie de celle-ci qui est spécifique du genre dudit agent pathogène bactérien, dans lequel ladite étape de quantification de la quantité dudit acide nucléique est effectuée au moyen d'une réaction en chaîne par polymérase qui est suivie en temps réel en utilisant une sonde d'hybridation choisie de telle manière qu'il existe une complémentarité de séquence maximale par rapport à toutes les séquences d'acides nucléiques cibles dudit genre dudit agent pathogène bactérien, dans lequel un signal seuil défini est déterminé pour toutes les réactions à analyser et le nombre de cycles (Cp) nécessaire pour atteindre cette valeur seuil est déterminé pour l'acide nucléique cible, ainsi que pour les acides nucléiques de référence, dans lequel les nombres de copies absolu ou relatif de la molécule cible sont déterminés sur la base des valeurs de Cp obtenues pour l'acide nucléique cible et l'acide nucléique de référence,- déterminer si ladite quantité d'acide nucléique comprenant la région espaceur d'ADNr 16S/23S ou une partie de celle-ci qui est spécifique dudit agent pathogène bactérien dépasse une première valeur seuil prédéterminée, dans lequel la classe de données dépassant ladite valeur seuil pour la quantité d'acide nucléique qui est détectée quantitativement est clairement indicatrice d'une infection pathogène- déterminer si ladite quantité d'acide nucléique comprenant la région espaceur d'ADNr 16S/23S ou une partie de celle-ci qui est spécifique dudit genre dudit agent pathogène bactérien reste sous une deuxième valeur seuil prédéterminée, dans lequel la classe de données qui restent sous ladite deuxième valeur seuil est indicatrice de l'absence ou de la non-confirmation d'une infection pathogène, de telle sorte qu'une classe de données restant sous ladite deuxième valeur seuil est clairement indicatrice de l'absence d'une infection pathogène, la deuxième classe de données dépassant ladite deuxième valeur seuil mais restant sous ladite première valeur seuil représente ces cas, où une infection pathogène est non confirmée mais par ailleurs ne peut pas non plus être exclue,
ladite réaction en chaîne par polymérase qui est suivie en temps réel, comprenant en outre l'étape de- surveillance de la dépendance de la température de l'hybridation entre la sonde et l'acide nucléique cible amplifié, ladite dépendance de la température étant indicatrice de la présence d'une espèce au sein dudit genre dudit agent pathogène bactérien. - Procédé selon la revendication 1, dans lequel ledit échantillon clinique est le sang total.
- Procédé selon les revendications 1 à 2, dans lequel ledit agent pathogène bactérien spécifique est choisi dans un groupe constitué par les staphylocoques coagulase-négatifs et les entérocoques.
