Description
- Proteine, Proteinderivate und Peptide, z.B. Antikörper, Antigene, Lektine, Heparine, Protein A, Protein G, Enzyme wie Proteasen, Phosphatasen, Glycosidasen, Lipasen, etc.;
- Nukleinsäuren, z.B. Oligo- und Polynukleotide, DNA- oder RNA-Fragmente;
- Kohlenhydrate wie Monosaccharide, Disaccharide, Glycogen, Stärke, Dextrane und komplexe Kohlenhydrate;
- Lipide wie Triacylglycerine, Steroide, Phosphoglyceride, Ganglioside;
- Affinitätsliganden, z.B. Chelatbildner, Cibacron Blau;
- Effektoren von Enzymen wie Inhibitoren, Aktivatoren, Substrate, Cosubstrate.

- Inkubation mit 10 µl 2,9 M HCl (2,5 h 37°C),
- Glutaraldehydbindung (10 µl 2,5% Glutaraldehyd in 0,1 M Pa-Puffer pH = 8,0, 18 min bei Raumtemperatur),
- Bindung des PEI-Spacers (10 µl 1%ige PEI-Lösung in 0,1 M Pa-Puffer 1 h bei 37°C),
- Blockung mit einer Aminosäuremischung (10 µl einer Mischung aus je 0,5 mg Phenylalanin, Leucin, Arginin, Glycin und Asparaginsäure / 1 ml Pa-Puffer, 2 h bei 4°C),
- Glutaraldehydbindung (wie oben)
- 9 ml Bisoxiran
- 1 ml 100% Ethanol
- 1 ml 25 mM wässrigen Natriumcarbonatlöusng (pH= 11)
- 1 g IDA gelöst in
- 10 mL einer 25 mM wässrigen Natriumcarbonatlösung (pH = 12) inkubiert und anschließend wiederum gründlich mit Wasser gewaschen.
- 10 mM Kupfer-(II)-Chlorid gelöst in
- 50 mM Na-Acetatpuffer (pH = 5)
Claims (13)
- Einstückiges Formteil aus Kunststoff zur Analyse und Präparation von Substanzen mit mindestens einem Oberflächenbereich und einem Innenbereich,
wobei der mindestens eine Oberflächenbereich ein offenporiges, dreidimensionales Netzwerk ist. - Formteil gemäß Anspruch 1, dadurch gekennzeichnet, dass der Innenbereich keine offenen Poren aufweist.
- Formteil nach einem der Ansprüche 1 oder 2 dadurch gekennzeichnet, dass der Kunststoff ausgewählt wird aus Polyamiden, Polysulfonen, Polyestern, Polycarbonaten sowie Copolymeren und Mischungen derselben.
- Formteil nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass an den mindestens einem Oberflächenbereich mindestens teilweise Reaktanten gebunden sind.
- Formteil nach Anspruch 4, dadurch gekennzeichnet, dass die Reaktanten ausgewählt sind aus Proteinen, Nukleinsäuren, Kohlenhydraten, Lipiden, Affinitätsliganden, Effektoren von Enzymen.
- Formteil nach einem der Ansprüche 4 oder 5, dadurch gekennzeichnet, dass die Reaktanten über reaktive Seitengruppen des Kunststoffs gebunden sind.
- Formteil nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass das Formteil als Pipettenspitze, Mikroplatte, Schlauchstück, Stab, Einzel- oder Mehrfachgefäß, Tauchkörperkugel oder Platte ausgebildet ist.
- Verfahren zur Herstellung eines Formteils nach einem der Ansprüche 1 bis 7, bei dem ein einstückiges Formteil aus Kunststoff an mindestens einem Oberflächenbereich partiell gelöst wird, um mindestens einen offenporigen Oberflächenbereich zu erzeugen, der ein dreidimensionales Netzwerk ist.
- Verfahren nach Anspruch 8, dadurch gekennzeichnet dass, vor, gleichzeitig oder nachträglich zur partiellen Lösung des Oberflächenbereiches eine chemische Aktivierung des Oberflächenbereiches erfolgt.
- Formteil erhältlich durch ein Verfahren nach Anspruch 8 oder 9.
- Verwendung eines Formteils nach mindestens einem der Ansprüche 1 bis 7 zur Analyse und Präparation von Substanzen.
- Verwendung nach Anspruch 11, dadurch gekennzeichnet, dass das Formteil zur Identifikation und Quantifikation von Analyten, insbesondere zur spezifischen Konzentrierung und Probenvorbereitung eingesetzt wird.
- Verwendung nach Anspruch 11, dadurch gekennzeichnet, dass das Formteil zur Anreicherung einer Substanz in einer Probe, zur Abreicherung einer störenden Substanz aus einer Probe, zur Analytmodifikation, insbesondere zur spezifische Spaltung oder Entfernung von Modifikation wie Phosphatgruppen, Zuckereinheiten, Fettsäureeinheiten eingesetzt wird.
