Description
- Die vorliegende Erfindung betrifft ein Formteil zur Analyse und Präparation von Substanzen.
- Die Entwicklung der Biotechnologie in den letzten Jahren, insbesondere im Bereich der Proteom- und Genom-Forschung verlangt nach effizienter Analytik bei allgemeiner Reduktion der Proben- und Reagenzienvolumina. Aufgrund der zunehmenden Parallelisierung der Untersuchungen bestehen hohe Anforderungen im Bereich einer einfachen, praktikablen und ökonomisch vertretbaren Probenvorbereitung, um eine Charakterisierung von Kleinstmengen zu erlauben.
- An entsprechende Probenvorbereitungsschritte werden vielfältige Anforderungen gestellt, dabei müssen
- a) Verluste an Probenmaterial gering gehalten werden,
- b) Möglichkeiten zu einer ausfallsicheren Automatisierung gegeben sein,
- c) Analysen von Analyten in niedriger Konzentration neben anderen Analyten mit höherer Konzentration möglich sein,
- d) Probenvolumina im µl und sub-µl Bereich verarbeitet werden können,
- e) möglichst universell einsetzbare Einheiten für eine Vielzahl verschiedener Verfahren geschaffen werden.

US 5,833,927 betrifft eine Vorrichtung, mit der eine Komponente aus einer Flüssigkeit gebunden werden kann. Die Vorrichtung ist in Form einer Pipettenspitze ausgebildet. Die Bindung erfolgt an einer Membran, die sich quer zur Flussrichtung in der Pipettenspitze befindet. -
WO 88/09201 - Nachteilig an diesen Vorrichtungen ist, dass sie große Totvolumina aufweisen, die insbesondere beim Umgang mit Probenvolumina im Mikroliter- und Submikroliterbereich inakzeptabel sind und dass sie aus verschiedenen Materialien zusammengesetzt sind.
- Zur Überwindung der Problematik des Totvolumens beschreibt
WO 98/37949 - Diese Probleme versucht die
WO 01/07162 - Das Problem der schlechten Reproduzierbarkeit und des schlechten Auslaufs der
WO 98/37949 US 6,416,716 dadurch zu lösen, dass in die Wand eines Gefäßes Teile eines Separationsmediums eingebunden sind. Nachteilig ist jedoch auch hier, dass unterschiedliche Materialien kombiniert werden müssen. - Bei einer zunehmenden Verkleinerung der Reaktionsgefäße, Pipettenspitzen, etc. ist das Einbringen eines zweiten Materials zunehmend komplizierter.
- Brazilian J. Med. Biol. Res (1994) 27: 1507-1516 beschreibt ein Verfahren, bei dem Nylon-6-Kugeln in Ameisensäure aufgelöst und dann präzipitiert werden, wodurch schwammartige Strukturen und kolloidale Suspensionen erhalten werden können. An diese können dann Enzyme gebunden werden. Entsprechende Schwämme und Suspensionen können nur schwer zu praktikablen Tools für die Mikroanalytik verarbeitet werden.
- Aufgabe der vorliegenden Erfindung war es, Formteile zur Analyse und Präparation bereitzustellen, die die genannten Anforderungen erfüllen und dabei Nachteile des Standes der Technik überwinden.
- Gelöst wird die Aufgabe durch ein einstückiges Formteil aus Kunststoff zur Analyse und Präparation von Substanzen mit mindestens einem Oberflächenbereich und einem Innenbereich, wobei der mindestens eine Oberflächenbereich ein offenporiges, dreidimensionales Netzwerk ist.
- "Einstückig" im Sinne der Anmeldung bedeutet, dass das Formteil aus einem, chemisch einheitlichen Ausgangsmaterial besteht.
- "Innenbereich" ist der Teil des Formteils, der nicht Teil eines Oberflächenbereichs ist.
- Erfindungsgemäß wird ein Formteil aus Kunststoff bereitgestellt, das zur Analyse und Präparation von Substanzen geeignet ist. An der Oberfläche existiert mindestens ein Bereich mit einem offenporigen dreidimensionalen Netzwerk. Die Formteile eignen sich zur Behandlung von Proben im Mikroliter- und Submikroliter-Bereich.
- Bevorzugt ist der Innenbereich frei von offenen Poren.
- Besonders geeignete Kunststoffe für die Formteile sind Polyamide, beispielsweise der Typen PA6, PA66, PA46, PA12, Polysulfone, Polyester und Polycarbonate. Auch Copolymere der entsprechenden Materialien und Mischungen derselben sind möglich. Das gesamte Formteil ist jedoch einstückig aus dem gleichen chemischen Material aufgebaut.
- An die entsprechenden Formteile können Reaktanten gebunden sein.
- Besonders geeignete Reaktanten sind:
- Proteine, Proteinderivate und Peptide, z.B. Antikörper, Antigene, Lektine, Heparine, Protein A, Protein G, Enzyme wie Proteasen, Phosphatasen, Glycosidasen, Lipasen, etc.;
- Nukleinsäuren, z.B. Oligo- und Polynukleotide, DNA- oder RNA-Fragmente;
- Kohlenhydrate wie Monosaccharide, Disaccharide, Glycogen, Stärke, Dextrane und komplexe Kohlenhydrate;
- Lipide wie Triacylglycerine, Steroide, Phosphoglyceride, Ganglioside;
- Affinitätsliganden, z.B. Chelatbildner, Cibacron Blau;
- Effektoren von Enzymen wie Inhibitoren, Aktivatoren, Substrate, Cosubstrate.
- Entsprechende Reaktanten können an einem oder mehreren Oberflächenbereichen des Formteils gebunden sein. Es können auch verschiedene Reaktanten an verschiedenen Oberflächenbereichen gebunden sein.
- Die Herstellung multifunktioneller Schichten durch mehrfach partielle Aktivierung der Oberfläche des Formteils und deren Bestückung mit unterschiedlichen Reaktanten erlaubt eine Abfolge von aufeinander folgenden Reaktionen im Mikroliterbereich ohne aufwendige Zwischenschritte. So könnte beispielsweise in einer Pipettenspitze in der unteren Schicht eine kovalent gebundene Protease Peptide erzeugen, die in der darüber liegenden Schicht durch hydrophobe Bindungen aus der Reaktionslösung entfernt werden und nach Elution im Massenspektrometer untersucht werden können. Dies ist in
Figur 10 dargestellt. - Die Reaktanten können kovalent, ionisch, über Komplexbildung oder durch hydrophobe Wechselwirkung gebunden sein. Die Bindung kann unmittelbar erfolgen oder mit Hilfe von Linkern. Das Formteil kann beispielsweise als Pipettenspitze, Schlauchstück, Stab, Einzel- oder Mehrfachgefäß, Mikrotiterplatte, Tauchkörperkugel oder Platte ausgebildet sein.
- Gegenstand der Erfindung ist auch ein Verfahren zur Herstellung der Formteile. Hierbei wird ein einstückiges Formteil aus Kunststoff an mindestens einem Oberflächenbereich partiell gelöst, so dass ein offenporiger Oberflächenbereich entsteht, der ein dreidimensionales Netzwerk bildet. Dabei kann gleichzeitig, vorher oder nachträglich zur partiellen Lösung des Oberflächenbereiches eine chemische Aktivierung des Oberflächenbereiches erfolgen.
- Das Verfahren ist dadurch gekennzeichnet, dass die Kunststoffoberfläche durch ein flüssiges Reaktionsmedium angelöst wird und durch einen Wechsel des flüssigen Mediums die angelöste Oberflächenstruktur erhalten bleibt bzw. durch Ausfällung eine dreidimensionale Struktur entsteht. Hierdurch erfolgt eine erhebliche Oberflächenvergrößerung, die für die Anbindung von Substanzen benutzt werden kann.
- Die genauen Verfahrensbedingungen sind erwartungsgemäß abhängig vom eingesetzten Kunststoff und können durch einfache Experimente für ein gegebenes Material optimiert werden. Soweit nur eine Anlösung der Oberfläche gewünscht wird, sind z.B. für Polyamide besonders geeignete Lösungsmittel gesättigte wässrige Phenollösung oder 85%ige Ameisensäure. Soweit gleichzeitig eine Freisetzung weiterer reaktiver Gruppen erfolgen soll, eignen sich insbesondere Säuren wie Salzsäure (> 2,5 M).
- Die Einwirkdauer hängt u.a. ab von dem Oberflächenmaterial, möglicher vorheriger Oberflächenbehandlung und der Temperatur. Sie liegt allgemein im Bereich von 10 sec bis 10 min, bevorzugt im Bereich von 30 sec bis 3 min. Im Falle einer gewünschten Freisetzung weiterer reaktiver Gruppen an der Oberfläche durch Spaltung von Bindungen im Kunststoff betragen diese typischerweise zwischen 1 und 10 h, bevorzugt zwischen 2 und 3 h. Nach Einwirken dieser Reaktionsmedien auf den oder die Oberflächenbereiche erfolgt ein Wechsel der Reaktionsmedienbedingungen. Dies kann erfolgen durch einen Austausch des Reaktionsmediums oder durch Veränderung der Konzentration durch Zusatz weiterer Substanzen.
- Im Falle von Formteilen, die zur Aufnahme von Flüssigkeiten geeignet sind (beispielsweise Pipettenspitzen, Reaktionsgefäßen, Mikrotiterplatten, Schläuchen), kann das notwendige Reaktionsmedium in einfacher Weise hineingegeben oder aufgesaugt werden. Im Falle von Tauchkörperkugeln, Stäben, etc. wird der zu behandelnde Oberflächenbereich in das Reaktionsmedium eingetaucht.
- Die erfindungsgemäßen Formteile eignen sich zur Analyse und Präparation von Substanzen, insbesondere zur spezifischen Konzentrierung und Probenvorbereitung und zur Anreicherung einer Substanz in einer Probe, zur Abreicherung einer störenden Substanz aus einer Probe, zur Analytmodifikation, insbesondere zur spezifischen Spaltung oder Entfernung von Modifikation wie Phosphaten, Zuckereinheiten, Fettsäureeinheiten.
- Beispielsweise kann durch die Technik der Extraktion aus, im Verhältnis zueinander gesehen, größeren Gefäßen und der Elution in wesentlich kleineren Gefäßen, eine Konzentrierung der gewünschten Substanz im Verhältnis der Volumina der Extraktions- zu den Elutionsgefäßen erreicht werden, wie dies in
Figur 11 dargestellt ist. - Extraktion aus Volumen V1 = 500 µl

Figur 10 zeigt eine Prinzipsskizze, bei der verschiedene Reaktanten auf verschiedene Oberflächenbereiche eines Formteils gebunden sind. -
Figur 11 zeigt eine Skizze einer Anwendung der erfindungsgemäßen Formteile, bei der Extraktion und Elution in verschiedenen Volumina erfolgt, um eine Konzentrierung der Substanzen zu erreichen. - Eine aus Nylon-6,6 bestehende Pipettenspitze wurde durch Anlösen der inneren Wandung mittels Ameisensäure und nachfolgendes Ausfällen des gelösten Nylon-6,6 an der Innenwand durch Verdünnen der nylonhaltigen Lösung mit Wasser aktiviert.
- Eine Pipettenspitze wurde mit folgenden Schritten behandelt:
- a) Aufziehen von 15 µl 85%iger Ameisensäure,
- b) sofortiges Nachziehen von 4 µl einer gesättigten Natriumchloridlösung zur Verdünnung der Ameisensäure,
- c) Inkubation bei Raumtemperatur für 2 min.,
- d) Ausstoßen der Ameisensäure/Natriumchloridlösung und sorgfältiges Spülen mit Wasser.
- Eine entsprechend behandelte Pipettenspitze ist in der
Figur 1 dargestellt. -
- 2a) Ein Nylonschlauch bestehend aus Nylon-6 wurde durch Zugabe einer wässrigen Phenollösung und nachfolgendes Ausfällen des gelösten Nylon-6 an der Innenwand durch Acetonzugabe behandelt. Die Aktivierung erfolgte wie folgt:
- a) Aufziehen von 15 µl einer gesättigten wässrigen Phenollösung,
- b) Inkubation für 10 min bei Raumtemperatur,
- c) Nachziehen von 25 µl Aceton,
- d) 5 min warten,
- e) Ausstoßen der Phenol-/Acetonlösung und mehrfaches Spülen mit Aceton.
Figur 2 dargestellt. - 2b) Alternativ erfolgte eine Aktivierung des Schlauchstückes durch
- a) Aufziehen mit 10 µl 85%iger Ameisensäure,
- b) Inkubation für 60 sec bei Raumtemperatur,
- c) Ausstoßen der Ameisensäure und wiederholtes Spülen mit Wasser.
- Eine Nylon-6,6-Spitze wurde mit Dimethylformamid, 80%igem Acetonitril und 0,1 M Phosphatpuffer pH = 8,0 absorbanzkontrolliert bei 215 nm bis zu einer maximalen Absorbanzdifferenz von 0,04 gewaschen. Anschließend wurde die Spitze mit einer Lösung von 0,5 mg Hämoglobin/ml Phosphatpuffer 2 Stunden inkubiert. Dann wurde die verbleibende Konzentration des Hämoglobins in der Lösung photometrisch bei 215 nm gemessen und aus der Absorbanzdifferenz die gebundenen Menge berechnet. Unter diesen Bedingungen konnte keine Bindung nachgewiesen werden (0 µg/cm2 Nylonoberfläche in sechs Versuchen).
- In einem zweiten Experiment wurde eine Nylonspitze zunächst mit 15 µl 85%iger Ameisensäure 2 min bei Raumtemperatur, wie im Beispiel 1 beschrieben, aktiviert. Anschließend wurde wie oben gewaschen. Dann wurde die Spitze - wie oben beschrieben - mit Hämoglobin inkubiert. Aus der Absorbanzabnahme des Hämoglobins bei 215 nm ergab sich aus vier Versuchen eine Bindung von 18,4 µg/cm2 Oberfläche.
- In einem dritten Experiment wurde eine Nylonspitze wie oben beschrieben mit Ameisensäure aktiviert. Dann wurde die Spitze mit 2,9 M Salzsäure, 2,5 h bei 37°C inkubiert und wie oben gewaschen. Hämoglobin wurde kovalent an Aminogruppen des Nylons über Polyethyleniminspacer mit Glutaraldehyd gebunden. Die Bindung betrug 26,5 µg/cm2 Oberfläche (aus einer Doppelbestimmung).
- Eine vierte Probe wurde wie Probe drei behandelt. An Stelle von Hämoglobin wurde Trypsin kovalent an die Aminogruppen des Nylons gebunden. Die gebundene Menge von 26,8 µg Trypsin/cm2 wurde aus einer Aktivitätsmessung mit fluorogenem Substrat bestimmt.
- Nylon-6,6 Spitzen (25 µl) wurden mit 85%iger Ameisensäure 30 sec angelöst und die Säure durch mehrfaches Waschen mit Wasser entfernt. Aus humanem Serumalbumin wurde durch Spaltung mit Trypsin eine Mischung definierter Peptide hergestellt. In die aktivierte Spitze wurden 7 µl einer Lösung mit 1,4 µg Peptidmischung in 20 mM Ammoniumbicarbonat mit einem Zusatz von Trifluoressigsäure einpipettiert und nach 5 min Kontaktzeit ausgestoßen. Anschließend wurde die Spitze mit 50 µl einer wässrigen, 0,1% Trifluoressigsäurelösung gewaschen. Danach erfolgte die Elution der gebundenen Peptide mit je 10 µl wässriger Acetonitrillösung, wobei der Acetonitrilgehalt von 10 auf 50% schrittweise erhöht wurde. Die Elutionslösung wurde mit MALDI-MS untersucht. Die Sequenzen der Peptide konnten durch Abgleich mit einer Datenbank bestimmt werden. Aus der so erhaltenen Aminosäuresequenz der identifizierten Peptide wurde als Maß für die Hydrophobizität der Peptide der sogenannte "Gravy-Index" berechnet.
- Das Verhältnis zwischen "Gravy-Index" und ansteigender Acetonitrilkonzentration ist in
Figur 3 dargestellt. - Ein Nylon-6 Schlauchstück wurde wie unter 2b) beschrieben aktiviert. Der aktivierte Nylonschlauch mit einer Länge von 15 mm und einem Innendurchmesser von 1,9 mm wurde auf eine Pipettenspitze gesteckt und intensiv mit Dimethylformamid, 100% Acetonitril und 80% Acetonitril gewaschen. Dann wurde der Schlauch mit 0,1%iger wässriger Trifluoressigsäure äquilibriert und 10 µl einer Peptidmischung (4,8 nmol/ml, 100 mM Natriumchlorid, 50 mM Phosphat, angesäuert mit TFA) pipettiert. Nach 5 min Kontaktzeit wurde die Lösung auspipettiert und der Schlauch mit 10 µl wässriger 0,1%iger TFA gewaschen. Die gebundenen Peptide wurden mit 10 µl 80%igem Acetonitril in Wasser eluiert.

Figur 4A , oben zeigt das MALDI MS Spektrum vor der Entsalzung. DieFigur 4B zeigt das MALDI Spektrum nach Elution der Peptide aus dem Schlauch. - Eine Nylon-6,6-spitze wurde wie folgt aktiviert: 10 µl 80%iger Ameisensäure und danach 1 µl gesättigte Kochsalzlösung wurden in die Spitze pipettiert und nach 2 min ausgestoßen. Dann wurde die Spitze mehrmals mit Wasser gespült. In die so aktivierte Nylonspitze wurden 10 µl einer 2,9 M Salzsäure pipettiert und 2,5 h bei 37°C in einer Wasserdampf gesättigten Atmosphäre inkubiert, um die Säureamidbindungen des Nylons partiell zu hydrolysieren. Nach Waschen der Spitze mit Wasser und 0,1 M Phosphatlösung pH = 8,0 (Pa-Puffer) wurde mit Glutaraldehyd umgesetzt. Dazu wurde die Spitze mit 10 µl 2,5%iger Glutaraldehydlösung (v/v) in Pa-Puffer 18 min bei Raumtemperatur inkubiert. Nach intensivem Waschen mit Wasser und Pa-Puffer erfolgt die Bindung des Spacers Polyethylenimin (PEI) durch Inkubation mit 10 µl einer 1%igen PEI-Lösung in Pa-Puffer (w/v) für 1 h bei 37°C. Nach dem Waschen mit Wasser und Pa-Puffer wurden übrig gebliebene Bindungsstellen durch Inkubation bei 4°C für mindestens 2h, besser über Nacht, mit 10 µl einer Aminosäuremischung aus je 0,5 mg Phenylalanin, Leucin, Arginin, Glycin und Asparaginsäure / 1 ml Pa-Puffer inkubiert. Dann erfolgt eine zweite Inkubation mit Glutaraldehyd (2,5% in Pa-Puffer, 18 min bei Raumtemperatur). Nach intensivem Waschen mit Wasser und Pa-Puffer werden 10 µl einer Trypsinlösung (1mg/ml Pa-Puffer) in die Spitze pipettiert und 23 h bei 4°C inkubiert. Dann erfolgt nach Waschen mit Pa-Puffer die Reduktion der gebildeten Doppelbindungen zwischen den Aldehydgruppen des Glutaraldehyds und den Aminogruppen des Nylons, des PEI bzw. des Trypsins mit einer 0,1 M NaBH4-Lösung in Pa-Puffer (30 min bei Raumtemperatur). Die Nylonspitze wurde zum Schluss mit Wasser, Pa-Puffer, 1 M NaCl-Lösung in Pa-Puffer, 0,5% Octylglycosid in Pa-Puffer und Pa-Puffer gewaschen. Die gebundene Trypsinmenge wurde mit dem fluorogenen Trypsinsubstrat Bz-Arg-AMC·HCl bestimmt. Dazu wurden über einen Zeitrum von je 2 min jeweils 10 µl aus einem Gesamtvolumen von 150 µl einer Substratlösung (25 µM Bz-Arg-AMC·HCl in 20 mM NH4HCO3, das 0,5% n-Octylglycosid enthält) mit dem CyBi-Well Multipipettor aufgezogen und unter Schütteln auspippetiert. Das gebildete AMC wurde bei 360/440 nm mit einem Fluoreszenzreader gemessen und mit mitgeführten AMC Eichwerten verglichen. Der Test ergab eine gebundene Trypsinmenge von 5,9 µg.
- Die Spitze wurde wiederholt zum Verdau unterschiedlich konzentrierter Humanserumalbuminlösungen (HSA) eingesetzt. Die gespaltenen Peptide wurden im MALDI-MS nachgewiesen.
Figur 5 zeigt zwei MALDI-Spektren von HSA-Peptiden nach jeweils 10 min Trypsinverdau.Figur 5A zeigt das Spaltungsergebnis von humanem Albumin bei der ersten (oben) undFigur 5B bei der sechsten Benutzung. Die Spitzen sind also für mehrfachen Einsatz hinreichend stabil. - An eine Nylon-6,6 Spitze, aktiviert durch Anlösen mit 10 µl 85%iger Ameisensäure 30 sec und Ausfällen mit Wasser, wurde gereinigte alkalische Phosphatase (AP) aus Kälberdarm mit geringen Änderungen der Prozedur analog Trypsin (Beispiel 6) kovalent gebunden. Nach der zweiten Glutaraldehydaktivierung wurde die Spitze intensiv mit 20 mM NaHCO3-Lösung an Stelle des Phosphatpuffers gewaschen. Die AP wurde zur Bindung als Lösung von 1.38 mg/ml in 60 mM NaHCO3 eingesetzt. 10 µl dieser Lösung wurden in die Nylonspitze pipettiert und 23 h bei 4°C inkubiert. Nach der Inkubation wurde die AP-Lösung auspipettiert und später mit den Waschlösungen vereinigt. Dann wurde die Spitze mit 8 x 10 µl 25 mM Diethanolamin/HCl, pH = 9,8 (DEA) gewaschen. In den vereinigten Waschlösungen wurde die Aktivität der AP mit p-Nitrophenylphosphat als Substrat in 1 M DEA, pH = 9,8 photometrisch bestimmt und mit der eingesetzten AP-Menge verglichen. Aus der Abnahme der AP-Menge wurde in Kenntnis der spezifischen Aktivität auf die an die Spitze gebundene Menge AP von 6,5 µg geschlossen. Die Reduktion der Doppelbindungen mit NaBH4 erfolgte in 0,1 M NaHCO3. NaHCO3 wurde auch für die nachfolgenden Waschschritte an Stelle von Phosphatpuffer eingesetzt. Die Aktivität der gebundenen AP in der Spitze wurde durch Spaltung eines Phosphopeptids mit Hilfe der MALDI-MS untersucht. 8 µl einer Lösung von 1,6 µg eines Phosphopeptids in 20 mM DEA pH = 9,8 wurden in die Spitze pipettiert und 5 min bei Raumtemperatur inkubiert. Das MALDI Spektrum der negativen Ionen nach 5 min Inkubation zeigt, dass das Phosphopeptid (monoisotopische Masse 1238,35 Da) nicht mehr nachweisbar ist. Das Erscheinen eines neuen Peaks (monoisotopische Masse 1158,35 Da) mit einer um 80 Da reduzierten Masse zeigt die Abspaltung einer Phosphatgruppe (
Figur 6 ). - Eine'Nylon-6,6 Spitze - analog Beispiel 7 - wurde wie folgt behandelt:
- Inkubation mit 10 µl 2,9 M HCl (2,5 h 37°C),
- Glutaraldehydbindung (10 µl 2,5% Glutaraldehyd in 0,1 M Pa-Puffer pH = 8,0, 18 min bei Raumtemperatur),
- Bindung des PEI-Spacers (10 µl 1%ige PEI-Lösung in 0,1 M Pa-Puffer 1 h bei 37°C),
- Blockung mit einer Aminosäuremischung (10 µl einer Mischung aus je 0,5 mg Phenylalanin, Leucin, Arginin, Glycin und Asparaginsäure / 1 ml Pa-Puffer, 2 h bei 4°C),
- Glutaraldehydbindung (wie oben)
- Hieran wurde PNGase F der Firma Boehringer (10 µl mit 0,2 U/ml, 23 h bei 4°C) gebunden. Danach erfolgte die Reduktion mit NaBH4 und Waschungen wie im Beispiel 6 für Trypsinbindung beschrieben. Die Nylonspitze wurde nach jeder Inkubation gründlich mit Pa-Puffer gewaschen.
- Der Nachweis der Bindung von enzymatisch aktiver PNGase F wurde durch Deglycosylierung eines Glycopeptides geführt, das aus einem tryptischen Verdau von alkalischer Phosphatase chromatographisch gereinigt wurde.
- 10 µl einer Lösung von 0,5 µg des Glycopeptids in 10 mM Tris/HCl pH = 8,3 wurden in die Nylonspitze pipettiert und 20 h bei Raumtemperatur inkubiert.

Figur 7 zeigt das Massenspektrum vor (7A) bzw. nach der Inkubation (7B). Die Kohlenhydratreste sind abgespalten. - Eine, wie im Beispiel 1 beschriebene, aktivierte Nylonspitze wurde mit einer wässrigen 0,1% TFA-Lösung äquilibriert. Dann wurden 10 µl einer Lösung eines 17-mer Oligonucleotids (25 pmol/1µl) in 5 mM EDTA, 100 mM NaCl, 1 mM MgCl2 und 10% Glycerol, pH = 5,2 in die Spitze pipettiert. Nach 10 min Kontaktzeit wurde die Lösung ausgestoßen und die Spitze mit 3 x 10 µl wässriger 0,1% TFA-Lösung und 3 x 10 µl 0,1% TFA / 40% Acetonitril-Lösung gewaschen. Die Elution erfolgte mit 6 µl einer Lösung von 50% Acetonitril 50 mM NH4OH pH = 9,0.
Figur 8 zeigt die MALDI Spektren der Ausgangslösung (A) und (B) nach Entsalzung an der Nylonspitze. - Das Modul, das aus acht Nylon-6 Stäben besteht, wurde zunächst mit Chloroform entfettet und danach 15 mm tief für 10 min in eine gesättigte wässrige Phenollösung getaucht.
- Anschließend wurde das angelöste Nylon durch Eintauchen für 5 min in 100% Aceton an den Stäben ausgefällt und danach mehrmals gründlich mit 100% Ethanol gespült.
- Nach dem Spülen erfolgte eine weitere Freisetzung reaktiver Gruppen durch Inkubation für 2,5 Stunden bei 37°C mit 2,9 M HCl. Anschließend wurde mehrmals gründlich mit Wasser gewaschen.
- Das zuvor mit Phenol und Salzsäure aktivierte Modul wurde für 20 h bei 80°C unter ständigem Schütteln in 10 ml einer Lösung aus:
- 9 ml Bisoxiran
- 1 ml 100% Ethanol
- 1 ml 25 mM wässrigen Natriumcarbonatlöusng (pH= 11)
- Das zuvor mit Bisoxiran aktivierte Modul wurde für 20 h bei 50°C unter ständigem Schütteln in 10 ml einer gesättigten Lösung aus:
- 1 g IDA gelöst in
- 10 mL einer 25 mM wässrigen Natriumcarbonatlösung (pH = 12) inkubiert und anschließend wiederum gründlich mit Wasser gewaschen.
- Das Modul mit immobilisiertem IDA wurde für 1 h bei Raumtemperatur unter ständigem Schütteln in 10 ml einer Lösung aus:
- 10 mM Kupfer-(II)-Chlorid gelöst in
- 50 mM Na-Acetatpuffer (pH = 5)
- Ein Nylonstab des so hergestellten Moduls wurde etwa 10 min in 300 µl einer Lösung aus tryptischen Peptiden von Humanserumalbumin (1,28 nmol) und einem Phosphopeptid (1,8 nmol) in 50 mM MES-Puffer (N-mopholinoethansulfonsäure) pH = 5,5 mit 10% Acetonitril (ACN) getaucht. Der Stab wurde dreimal mit je 170 µl 45,5 mM MES-Puffer pH = 5,5 in 40% ACN und zweimal mit Wasser gewaschen. Danach wurde mit 30 µl einer 1% NH4OH-Lösung in 40% ACN eluiert.

Figur 9 zeigt die negativen MALDI Spektren A) der Mischung von Phosphopeptid mit HSA-Peptiden und B) nach Elution vom Metall-Chelat-Nylonstab.
Claims (13)
- Einstückiges Formteil aus Kunststoff zur Analyse und Präparation von Substanzen mit mindestens einem Oberflächenbereich und einem Innenbereich,
wobei der mindestens eine Oberflächenbereich ein offenporiges, dreidimensionales Netzwerk ist. - Formteil gemäß Anspruch 1, dadurch gekennzeichnet, dass der Innenbereich keine offenen Poren aufweist.
- Formteil nach einem der Ansprüche 1 oder 2 dadurch gekennzeichnet, dass der Kunststoff ausgewählt wird aus Polyamiden, Polysulfonen, Polyestern, Polycarbonaten sowie Copolymeren und Mischungen derselben.
- Formteil nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass an den mindestens einem Oberflächenbereich mindestens teilweise Reaktanten gebunden sind.
- Formteil nach Anspruch 4, dadurch gekennzeichnet, dass die Reaktanten ausgewählt sind aus Proteinen, Nukleinsäuren, Kohlenhydraten, Lipiden, Affinitätsliganden, Effektoren von Enzymen.
- Formteil nach einem der Ansprüche 4 oder 5, dadurch gekennzeichnet, dass die Reaktanten über reaktive Seitengruppen des Kunststoffs gebunden sind.
- Formteil nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass das Formteil als Pipettenspitze, Mikroplatte, Schlauchstück, Stab, Einzel- oder Mehrfachgefäß, Tauchkörperkugel oder Platte ausgebildet ist.
- Verfahren zur Herstellung eines Formteils nach einem der Ansprüche 1 bis 7, bei dem ein einstückiges Formteil aus Kunststoff an mindestens einem Oberflächenbereich partiell gelöst wird, um mindestens einen offenporigen Oberflächenbereich zu erzeugen, der ein dreidimensionales Netzwerk ist.
- Verfahren nach Anspruch 8, dadurch gekennzeichnet dass, vor, gleichzeitig oder nachträglich zur partiellen Lösung des Oberflächenbereiches eine chemische Aktivierung des Oberflächenbereiches erfolgt.
- Formteil erhältlich durch ein Verfahren nach Anspruch 8 oder 9.
- Verwendung eines Formteils nach mindestens einem der Ansprüche 1 bis 7 zur Analyse und Präparation von Substanzen.
- Verwendung nach Anspruch 11, dadurch gekennzeichnet, dass das Formteil zur Identifikation und Quantifikation von Analyten, insbesondere zur spezifischen Konzentrierung und Probenvorbereitung eingesetzt wird.
- Verwendung nach Anspruch 11, dadurch gekennzeichnet, dass das Formteil zur Anreicherung einer Substanz in einer Probe, zur Abreicherung einer störenden Substanz aus einer Probe, zur Analytmodifikation, insbesondere zur spezifische Spaltung oder Entfernung von Modifikation wie Phosphatgruppen, Zuckereinheiten, Fettsäureeinheiten eingesetzt wird.
