Abstract

Die Erfindung betrifft die Verwendung von LTB4 Antagonisten zur Herstellung eines Arzneimittels zur Behandlung oder Prävention von respiratorischen Erkrankungen bei Begleit- und Nutztieren.

Description

Verwendung von LTB4 Antagonisten in der Tiermedizin
Die Erfindung betrifft die Verwendung eines LTB4 Antagonisten oder eines Prodrugs davon zur Herstellung eines Arzneimittels zur Behandlung oder Vorbeugung von respiratorischen Erkrankungen bei Begleit- und Nutztieren.
Hintergrund der Erfindung LTB4 Antagonisten, sind Verbindungen mit pharmakologisch wertvollen Eigenschaften. LTB4 Antagonisten werden in der Humanmedizin bei der Behandlung von Arthritis, Asthma, chronisch obstruktiven Lungenerkrankungen, Psoriasis, Colitis ulcerosa, Alzheimer Krankheit, Schock, Reperfusions Schäden /Ischaemie, Cystischer Fibröse, Atherosklerose und Multipler Sklerose eingesetzt.
LTB4 Antagonisten sind bekannt z.B. aus den Internationalen Patentanmeldungen WO 93/16036, WO 94/11341, WO 96/02497, WO 97/21670, WO 98/11062, WO 98/11119, WO 01/25186, WO 01/51457 und WO 02/34715.
Erkrankungen der Atemwege, insbesondere chronische Erkrankungen bei Begleit- und Nutztieren weisen in der Regel andere Ursachen auf als Asthma oder chronisch obstruktiven Lungenerkrankungen beim Menschen. Aufgrund dieser Atem Wegserkrankungen fallen insbesondere bei Nutz- oder Sporttieren die Erträge bzw. Leistungen stark ab und führen so zu hohen wirtschaftlichen Verlusten. Die bisher eingesetzten Medikamenten wie beta-Mimetika oder Steroide führen bei Tieren, die der Fleischerzeugung dienen zu langen Wartezeiten und bei Tieren, die im Sport eingesetzt werden, zu Ausschlüssen von Wettkämpfen wegen Dopingverdachts. Außerdem weisen diese Medikamente teilweise geringe Wirksamkeit und/oder unerwünschte Nebenwirkungen auf.
Es ist daher Aufgabe der vorliegenden Erfindung, eine Methode zur Behandlung oder Verbeugung von respiratorischen Erkrankungen bei Begleit- oder Nutztieren aufzuzeigen, welche zu einer schnellen und nachhaltigen Verbesserung des Gesundheitszustands und der Leistungsfähigkeit dieser Tiere führt, ohne die Nachteile der bisher eingesetzten Behandlungsmethoden aufzuweisen.' Es wurde nun überraschenderweise gefunden, dass sich LTB4 Antagonisten hervorragend zur Behandlung oder Vorbeugung respiratorischer Erkrankungen bei solchen Tieren eignen.
Detaillierte Beschreibung der Erfindung
Vorstehend genannte Aufgaben werden durch die im Folgenden detailliert beschriebene Verwendung gelöst.
Gegenstand der Erfindung ist somit die Verwendung eines LTB Antagonisten oder eines Prodrugs davon zur Herstellung eines Arzneimittels zur Behandlung oder Vorbeugung von respiratorischen Erkrankungen bei Begleit- und Nutztieren.
Bevorzugt sind solche LTB Antagonisten, welche eine Benzamidingruppe der Formel ( 1 )

R9 H oder Cι-C6-Alkyl, vorzugsweise H oder d-C4-Alkyl, insbesondere Isopropyl bedeutet; oder R8 und R9 zusammengenommen eine C4-C6-Alkylene Gruppe bedeuten; R10 Ci-Cö-Alkyl, Cs-C7-Cycloalkyl, Aryl, Heteroaryl, Aralkyl oder Heteroaryl-(Cι- C6-alkyl), vorzugsweise d-C4-Alkyl bedeutet wobei die in den Resten R2 und R10 aufgeführten Arylgruppen für Phenyl oder Naphthyl stehen, die Heteroaryl Gruppen für Pyrrol, Pyrazol, Imidazol, Furanyl, Thienyl, Pyridin oder Pyrimidin stehen und jeweils ein- oder mehrfach durch Cl, F, CF3, d-C4- Alkyl, OH, HSO3-O- oder d-C4-Alkoxy, vorzugsweise durch OH oder HSO3-O- substituiert sein können, sowie im Falle der durch OH substituierten Verbindungen die entsprechenden Glycoside davon.
Ein weiterer Gegenstand der Erfindung sind pharmazeutische Zubereitungen enthaltend mindestens einen LTB4 Antagonisten der Formel I sowie mindestens ein Beta-
Mimetikum oder mindestens ein Steroid.
Weiterhin Gegenstand der Erfindung ist ein gebrauchsfertiges Zweikomponentensystem, wobei (a) eine Komponente mindestens einen LTB4 Antagonisten der Formel I enthält; und
(b) die andere Komponente mindestens ein Beta-Mimetikum oder mindestens ein Steroid enthält.
Besonders bevorzugt sind LTB4 Antagonisten der Formel IA,

R6 d-C -Alkyl bedeutet;

CONHRio, SO2-(d-C6-alkyl), SO2-Phenyl, wobei die Phenyl Gruppe ein- oder zweifach durch Cl, F, CF3, Cι-C - Alkyl, OH und/oder C[-C4-Alkoxy substituiert sein kann, bedeutet;
R9 H oder d-C6- Alkyl bedeutet; oder
Rio Ci-Cβ-Alkyl bedeutet wobei die im Rest R2 aufgeführten Arylgruppen für Phenyl stehen und jeweils einfach durch OH, HSO3-O- oder Cι-C4-Alkoxy substituiert sein können, sowie deren pharmakologisch akzeptablen Säureadditionssalze, Glykoside und
O-Sulfate.
Der jeweilige Wirkstoff der Formel I bzw. IA kann in der erfindungsgemäßen Formulierung in Form eines physiologisch verträglichen Säureadditionssalzes vorliegen. Als physiologisch verträgliche Säureadditionssalze werden erfindungsgemäß pharmazeutisch verträgliche Salze verstanden, die ausgewählt aus den Salzen der Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Phosphorsäure, Methansulfonsäure, Essigsäure, Fumarsäure, Bernsteinsäure, Milchsäure, Zitronensäure, Weinsäure und Maleinsäure sind. Gegebenenfalls können zur Herstellung der Salze auch Mischungen der vorgenannten Säuren eingesetzt werden. Erfindungsgemäß bevorzugt sind die Salze der Formel I ausgewählt aus der Gruppe bestehend aus Hydrochlorid, Hydrobromid, Sulfat, Phosphat, Fumarat und Methansulfonat. Besonders bevorzugt sind die Salze ausgewählt aus Hydrochlorid, Hydrobromid und Fumarat. Der Wirkstoff kann gegebenenfalls in Form eines Hydrats vorliegen. Erfindungsgemäß bevorzugt wird die Verbindung der Formel I allerdings in Form der freien Base und in der wasserfreien Form der Tablette zugesetzt.
Besonders bevorzugte Verbindungen der Formel I sind die der Formeln IA1 bis IA8, , insbesondere IA1:

(IA8) sowie im Falle der durch OH substituierten Verbindungen die entsprechenden Sulfate oder Glycoside davon.
Die Verbindungen der Formel I, worin R! von Wasserstoff verschieden ist, sind in der Regel Prodrugs, die in-vivo in die entsprechenden Verbindungen der Formel I, worin Ri Wasserstoff ist, umgewandelt werden. Aus der Verbindung IA entsteht beispielsweise in-vivo die Verbindung der Formel IA1:
(IA1) worin X OH, HSO3-O- oder einen Kohlenhydratrest der Formel C6HπO5-O- bedeutet.
Vorzugsweise wird der jeweilige Wirkstoff in kristalliner, unvermahlener Form oder gemahlener Form, insbesondere in strahl gemahlener Form eingesetzt, bei dem die Teilchengrößenverteilung innerhalb folgender Grenzen liegt: Dι0 < 3 μm, D50 3 bis 8 μm, D90 < 8 bis 30 μm. Bei den vorstehend genannten Zahlenangaben für D10, D5o und D90 in μm (Mikrometer) handelt es sich um die Teilchengrößenbereiche innerhalb derer eine
Durchgangssumme, von 10 Volumen-%, 50 Volumen-% bzw. 90 Volumen-% der gemessenen Partikel (kumulative Volumenverteilung) erreicht wird. Diese Werte wurden mit dem Verfahren der Laserdiffraktometrie bestimmt, im vorliegenden Fall speziell unter Anwendung einer sogenannten Trockendispergierung bei 2 bar Dispersionsdruck sowie einer Brennweite f = 500 mm, z.B. mit Hilfe eines Sympatec/RODOS Gerätes. Diese Methodik ist im Stand der Technik bekannt.
Wird im Rahmen der vorliegenden Erfindung auf Salze der Verbindungen der Formel I Bezug genommen, wird dies durch die Bezeichnung P kenntlich gemacht. Ein explizite Bezugnahme auf die freie Base der Formel I erfolgt dagegen durch Verwendung der Bezeichnung I.
Der Begriff „Begleittiere (companion animals)" steht in der Regel für Tiere, die aus sozialen Gründen, insbesondere zur Freizeitgestaltung gehalten werden, insbesondere Freizeit- oder Sportpferde, Hunde und Katzen.
Der Begriff „Nutztiere" steht in der Regel für solche Tiere, die aus wirtschaftlichem Interesse, zur Produktion von Lebensmitteln oder Wolle oder zur Verrichtung von Arbeit, vorzugsweise in landwirtschaftlichen Betrieben gehalten werden. Es handelt sich dabei überwiegend um paarhufige Säugetiere, wie zum Beispiel Rinder, Schafe und Ziegen oder um Schweine.
Der jeweilige Wirkstoff wird in der Regel oral appliziert, vorzugsweise als Tablette, Granulat, Suspension oder als Futterzusatz.
Bezogen auf die Gesamtmasse der Darreichungsform ist der LTB Antagonist, vorzugsweise die Verbindung der Formel I, insbesondere IA erfindungsgemäß zu 0,2 bis 95 Gew-%, bevorzugt 0,7 bis 60 Gew-%, besonders bevorzugt etwa 5 bis 50 Gew-% enthalten. Besonders bevorzugt liegt der Anteil der freien Base von I zwischen 6 und 40 Gew-% bezogen auf die Gesamtmasse der Darreichungsform.
Die erfindungsgemäße Arzneimittelformulierung enthält neben dem Wirkstoff ferner wenigstens einen Hilfsstoff als Füllstoff/Trockenbindemittel.
Im Rahmen der vorliegenden Erfindung kommt dabei Kohlenhydraten wie Laktose oder Mannose, insbesondere feinverteilte Laktose oder Zuckeralkoholen wie Mannitol, Sorbit oder Xylit, insbesondere Mannitol als Hilfsstoff eine besondere Bedeutung zu. Diese Hilfsstoffe haben sich bei der erfindungsgemäßen Formulierung als besonders vorteilhaft erwiesen. Ein bevorzugter Aspekt der vorliegenden Erfindung betrifft daher eine mindestens eine Verbindung der Formel I enthaltende Tablette, die neben dem Wirkstoff und dem Netzmittel Laktose, insbesondere feinverteilte Laktose, besonders bevorzugt Laktosemonohydrat oder Mannitol als Hilfsstoff enthält.
Die erfindungsgemäße Darreichungsform kann neben Laktose, einem Netzmittel und dem Wirkstoff ferner weitere Hilfsstoffe bzw. Füllstoffe enthalten. Erfindungsgemäß bevorzugt können solche Verbindungen eingesetzt werden, die als Bindemittel fungieren können.
Der Begriff „Bindemittel" steht vor- und nachstehend für solche Hilfsmittel, die geeignet sind, andere Komponenten miteinander abzubinden. Erfindungsgemäß bevorzugte Bindemittel sind ausgewählt aus der Gruppe bestehend aus:
Pulvercellulose, mikrokristalline Cellulose, Sorbitol, Stärke, Polyvinylpyrrolidon (Povidon), Copolymerisaten von Vinylpyrrolidon mit anderen Vinylderivaten (Copovidone), Cellulosederivaten, insbesondere Methylhydroxypropylcellulose, z.B. Methocel A 15 LV, und Gemische dieser Verbindungen. Bevorzugt sind als Bindemittel Pulvercellulose, besonders mikrokristalline Cellulose und/oder Copovidone enthalten. Am meisten bevorzugt ist ein Gemisch aus mikrokristalliner Cellulose und einem Co-polymerisat aus Vinylpyrrolidon und Vinylacetat, nämlich Copovidone VA 64, wobei das Verhältnis von Vinylpyrrolidon und Vinylacetat etwa 3:2 (m/m) ist. In der Regel weist die erfindungsgemäße Tablette ein Gewichtsverhältnis von mikrokristalliner Cellulose zu Copovidone VA 64 von 20:1 bis 1:1, vorzugsweise 15:1 bis 2:1, insbesondere etwa 10:1 bis 3:1 auf. Aufgrund dieser besonders bevorzugten Bindemittel-Kombination aus mikrokristalliner Cellulose und Copovidone werden Tabletten mit erhöhter Bioverfügbarkeit der Verbindungen der Formel I erhalten.
Die erfindungsgemäße Darreichungsform kann als weitere Bestandteile ferner Fließ- bzw. Fließregulierungsmittel sowie Schmiermittel enthalten. Hierzu kommen im Rahmen der vorliegenden Erfindung beispielsweise in Betracht Siliciumdioxid, Talkum, Stearinsäure, Natrium-Stearylfumarat, Magnesiumstearat und Glyceroltribehenat. Erfindungsgemäß bevorzugt wird Magnesiumstearat verwendet. Werden vorstehend genannte Fließ- bzw. Fließregulierungsmittel bzw. Schmiermittel verwendet, liegt ihr Gewichtsanteil bezogen auf die Gesamtmasse der erfindungsgemäßen Tablette bevorzugt in einem Bereich von etwa 0,1 - 10 Gew-%, bevorzugt bei etwa 0,5 - 5 Gew-%, besonders bevorzugt zwischen 0,6 und 1,5 Gew-%.
Weiterhin kann die erfindungsgemäße Darreichungsform einen oder mehrere synthetische oder natürliche, pharmazeutisch verträgliche Farbstoffe enthalten, vorzugsweise Indigo carmine. Werden vorstehend genannte Farbstoffe verwendet liegt ihr Gewichtsanteil bezogen auf die Gesamtmasse der erfindungsgemäßen Tablette bei 0,01 bis 0,5 Gew-%.
Weiterhin kann die erfindungsgemäße Darreichungsform einen oder mehrere synthetische oder natürliche, pharmazeutisch verträgliche Geruchs- oder Aromastoffe enthalten. Die Art dieser Geruchs- und Aromastoffe richtet sich nach der Spezies des zu behandelnden Tieres. Werden vorstehend genannte Geruchs- oder Aromastoffe verwendet liegt ihr Gewichtsanteil bezogen auf die Gesamtmasse der erfindungsgemäßen Tablette bei 0,01 bis 0,5 Gew-%. Die erfindungsgemäße Darreichungsform kann durch direktes Mischen der Bestandteile, Verpressen und Pelletieren oder durch Granulieren, Verpressen und Pelletieren hergestellt werden.
In der Regel wird der LTB Antagonist in einem täglichen Dosisbereich, insbesondere in Verbindung mit der Fütterung, von 0,1 bis 20 mg kg, vorzugsweise von 0,5 bis 8,0 mg/kg,
Das nachfolgende Beispiel dient der Illustration einer erfindungsgemäßer Verwendung. Es ist lediglich als mögliche, exemplarisch dargestellte Vorgehensweise zu verstehen, ohne die Erfindung auf deren Inhalt zu beschränken.
Beispiel: Beschreibung des durchgeführten Versuchs:
Ein Pferd (Warmblüterstute, 12 Jahre) weist aufgrund einer chronischen Bronchitis folgende Symptome auf: Husten, Atemlosigkeit, Lethargie.
Diesem Pferd werden 20 Tage, täglich einmal 10 g der Verbindung der Formel IA1 in Form einer Methylcellulose (Thylose)-Suspension, mit dem Futter (Hafer) verabreicht.
Nach etwa 7 Tagen Behandlung tritt eine deutliche Besserung der Symptome ein: Reduziertes Husten, mit einer Phase ohne Husten von mehreren Tagen, verbesserte Leistungsfähigkeit (exercise capacity), Vitalität und Agilität. Die vorherige Symptomatik stellte sich etwa 2 Wochen nach Behandlung wieder ein.
Der LTB4 Antagonist der Formel IA1 wurde gut vertragen ohne irgendwelche Hinweise auf Nebenwirkungen.

Claims

Patentansprüche
1. Verwendung eines LTB4 Antagonisten oder eines Prodrugs davon zur Herstellung eines Arzneimittels zur Behandlung oder Vorbeugung von respiratorischen Erkrankungen bei Begleit- und Nutztieren.
2. Verwendung nach Anspruch 1, dadurch gekennzeichnet, dass der LTB4 Antagonist eine Verbindung der Formel I ist,
wobei A eine Gruppe der Formel
-O-CmH2m-O-(PHE)n- ( π ) bedeutet, worin m eine ganze Zahl von 2 bis 6 ist, n 0 oder 1 ist, PHE für eine gegebenenfalls durch ein oder zwei d-C6 Alkylgruppen substituierte 1,4-Phenylengruppe steht; oder A eine Gruppe der Formel
bedeutet,
Ri H, OH, CN, COR10, COOR10 oder CHO bedeutet;
R2 H, Br, Cl, F, CF3, CHF2, OH, HSO3-O-, d-C6-Alkyl, Cι-C6-Alkoxy, C5-C7-
Cycloalkyl, CONR8R9, Aryl, O-Aryl, CH2-Aryl, CR5R6-Aryl, oder C(CH3)2-R7 bedeutet, R3 H, d-C6 -Alkyl, d -C6-Alkoxy, OH, Cl oder F bedeutet,
R4 H oder d-C6-Alkyl bedeutet; R5 d-C4-Alkyl, CF3, CH2OH, COOH oder COO(d-C4-Alkyl) bedeutet;
R6 H, C C4- Alkyl oder CF3 bedeutet;

C6-alkyl) bedeutet, wobei die in den Resten R2 und Rι0 aufgeführten Arylgruppen für Phenyl oder Naphthyl stehen, die Heteroaryl Gruppen für Pyrrol, Pyrazol, Imidazol, Furanyl, Thienyl, Pyridin oder Pyrimidin stehen und jeweils ein- oder mehrfach durch Cl, F, CF3, Cι-C4- Alkyl, OH, HSO3-O- oder d-C4-Alkoxy substituiert sein können, sowie deren pharmakologisch akzeptablen Säureadditionssalze, Glykoside und O-Sulfate.
Verwendung nach Anspruch 2 dadurch gekennzeichnet, dass der LTB4 Antagonist eine Verbindung der Formel IA ist,

bedeutet, R] COOR10 bedeutet; R2 CF3, CHF2, OH, HSO3-O-, d-C6-Alkyl, d-C6-Alkoxy, CONR8R9 oder
CR5R6-Aryl bedeutet, R3 H oder Cι-C6-Alkoxy bedeutet,
R4 H bedeutet;
R5 Cι-C4- Alkyl bedeutet; R6 C C -Alkyl bedeutet;
R8 H, Cj-Cό-Alkyl, Phenyl, Phenyl-(d-C6-alkyl), CORio, COORJO, CHO, CONH2,
CONHR10, SO2-(Cι-C6-alkyl), SO2-Phenyl, wobei die Phenyl Gruppe ein- oder zweifach durch Cl, F, CF3, Cι-C4-Alkyl, OH und/oder Cι-C -Alkoxy substituiert sein kann, bedeutet; R9 H oder Cι-C6-Alkyl bedeutet; oder
R10 d-C6-Alkyl bedeutet wobei die im Rest R2 aufgeführten Arylgruppen für Phenyl stehen und jeweils einfach durch OH, HSO3-O- oder C]-C4-Alkoxy substituiert sein können, sowie deren pharmakologisch akzeptablen Säureadditionssalze, Glykoside und O-Sulfate.
4. Verwendung nach einem der Ansprüche 1 bis 3, dadurch gekennzeichnet, dass der LTB4 Antagonist eine Verbindung ausgewählt aus den Formeln IA1 bis IA8 ist:

(IA8) sowie im Falle der durch OH substituierten Verbindungen die entsprechenden Sulfate oder Glycoside davon.
5. Verwendung nach einem der Ansprüche 1 bis 4, dadurch gekennzeichnet, dass das Begleit- oder Nutztier ausgewählt ist aus der Gruppe bestehend aus Hunden, Katzen,
Pferden, Rindern und Schweinen.
6. Verwendung nach einem der Ansprüche 1 bis 5, dadurch gekennzeichnet, dass das Arzneimittel eine zur oralen Applikation geeigneten Darreichungsform aufweist.
7. Verwendung nach einem der Ansprüche 1 bis 6, dadurch gekennzeichnet, dass das Arzneimittel in einer täglichen Dosisbereich von 0,1 mg/kg bis 10,0 mg/kg verabreicht wird.
8. Verwendung nach einem der Ansprüche 1 bis 7, dadurch gekennzeichnet, dass der LTB4 Antagonist in Kombination mit einem Wirkstoff ausgewählt aus der Gruppe bestehend aus: beta-Mimetika und Steroide.
9. Verwendung nach einem der Ansprüche 1 bis 8, dadurch gekennzeichnet, dass das Medikament zur Prävention oder Behandlung des chronischen Hustens, der chronisch obstruktiven Lungenerkrankung oder der chronischen Bronchitis bei Pferden eingesetzt wird.
10. Pharmazeutische Zubereitung enthaltend mindestens einen LTB4 Antagonisten der Formel I gemäß einem der Ansprüche 1 bis 4 sowie mindestens ein beta-Mimetikum oder mindestens ein Steroid.
11. Gebrauchsfertiges Zweikomponentensystem, wobei
(a) eine Komponente mindestens einen LTB4 Antagonisten der Formel I gemäß einem der Ansprüche 1 bis 4 enthält; und
(b) die andere Komponente mindestens ein beta-Mimetikum oder mindestens ein Steroid enthält.
