Definitions
- the invention relates to nucleic acid molecules which code for proteins with the enzymatic activity of an acyl hydrolase, in particular a triacylglycerol lipase (TAG lipase), methods for producing transgenic plants which contain these nucleic acid molecules and their content of acyl hydrolase, in particular TAG lipase compared to Wild type plants is changing, these new plants, their parts and products and plant cells, as well as the use of the TAG lipase
- Nucleic acid molecules to influence the content of polyunsaturated fatty acids in transgenic plants are provided.
- TAG lipases catalyze a large number of reactions, many of which have industrial potential.
- TAG lipases catalyze the conversion of triacylglycerol and water to diacylglycerol and a fatty acid anion.
- amino acid sequences of lipases from the human stomach, from the rat tongue and from the human hepatic lysosome are homologous, but apart from a sequence of six amino acids around the essential amino acid
- Serine 152 of the pig pancreas lipase is not related to the lipase from the pig pancreas. These enzymes are glycosylated, contain a hydrophobic signal peptide and belong to the family of acid lipases (D. Ameis et al. (1994) Eur. J. Biochem. 219: 905-914). Lysosomal acid lipase (LAL) is a hydrolase essential for the intracellular breakdown of cholesteryl esters and triacylglycerols and plays a role in the mobilization of seed oil during germination.
- LAL Lysosomal acid lipase
- Neutral triacylglycerol lipases have been extensively studied in fungi, bacteria, mammals and insects. Nucleotide sequences similar to the neutral triacylglycerol lipases in Arabidopsis thaliana and Ipomea nil have been described, but their function has not been demonstrated. Furthermore, the Crystal structure of Mucor miehei triacylglycerol lipase has been reported, in which a trypsin-like catalytic triad Ser-His-Asp with an active serine residue was found below a short helical fragment of a long surface loop (L. Brady et al. (1990) Nature 343 : 767-770).

- Triacylglycerols whose acyl residues consist of 13 -hydro (pero) xylinolic acid, are then hydrolyzed by a TAG lipase, which is highly specific for oxidized polyene fatty acid residues. These modified fatty acids serve as an energy reserve for the growing seedling.
- TAG lipases Isolation of vegetable TAG lipases can be useful for processing vegetable oils that contain fatty acids with unusual oxygenated structures. - Nucleic acid molecules and vectors in plant cells is the availability of suitable transformation systems.
- a wide range of transformation methods have been developed and established here over the past two decades. These techniques include the transformation of plant cells with T-DNA using Agrobacterium tumefaciens or Agrobacterium rhizogenes as a transformation agent, diffusion of protoplasts, the direct gene transfer of isolated DNA to protoplasts, the injection and electroporation of DNA into plant cells, the introduction of DNA by means of the biolistic Methods and other possibilities, whereby the person skilled in the art can easily determine the most suitable method. All transformation processes have been well established for many years and are undoubtedly part of the standard repertoire of the specialist in plant molecular biology, plant biotechnology and cell and tissue culture.
- plasmids When injecting and electroporation of DNA into plant cells, there are no special requirements per se for the plasmids used. The same applies to direct gene transfer. Simple plasmids, such as pUC derivatives, can be used. However, if whole plants are to be regenerated from such transformed cells, the presence of a selectable marker gene is recommended.
- the usual selection markers are known to the person skilled in the art and it is not a problem for him to select a suitable marker.
- the plant cell uses the Ti or Ri plasmid, at least the right boundary, but often the right and left boundary of the T-DNA contained in the Ti or Ri plasmid, must be connected as a flank region to the genes to be introduced.
- Agrobacteria are used for the transformation, the DNA to be introduced must be cloned into special plasmids, either in an intermediate or in a binary vector.
- the intermediate vectors can be integrated into the Ti or Ri plasmid of the agrobacteria by means of sequences which are homologous to sequences in the T-DNA by homologous recombination. This also contains the vir region necessary for the transfer of the T-DNA. Intermediate vectors cannot replicate in agrobacteria.
- the intermediate vector can be transferred to Agrobacterium tumefaciens (conjugation).
- Binary vectors can replicate in E. coli as well as in Agrobacteria. They contain a selection marker gene and a linker or polylinker, which are framed by the right and left T-DNA border region. They can be transformed directly into the agrobacteria.
- the agrobacterium serving as the host cell is said to contain a plasmid which carries a vir region. The vir region is necessary for the transfer of the T-DNA into the plant cell. Additional T-DNA may be present.
- the agrobacterium transformed in this way is used to transform plant cells.
- the use of T-DNA for the transformation of Plant cells have been intensively examined and have been adequately described in well-known overview articles and manuals on plant transformation.
- plant explants can expediently be cultivated with Agrobacterium tumefaciens or Agrobacterium rhizogenes. Whole plants can then be regenerated from the infected plant material (e.g. leaf pieces, stem segments, roots, but also protoplasts or suspension-cultivated plant cells) in a suitable medium, which may contain antibiotics or biocides for the selection of transformed cells.
- Agrobacterium tumefaciens e.g. leaf pieces, stem segments, roots, but also protoplasts or suspension-cultivated plant cells
- suitable medium which may contain antibiotics or biocides for the selection of transformed cells.
- the introduced DNA is integrated in the genome of the plant cell, it is generally stable there and is also retained in the progeny of the originally transformed cell. It normally contains a selection marker which gives the transformed plant cells resistance to a biocide or an antibiotic such as kanamycin, G 418, bleomycin, hygromycin, methotrexate, glyphosate, streptomycin, sulfonylurea, gentamycin or phosphinotricin and others.
- the individually selected marker should therefore allow the selection of transformed cells from cells that lack the inserted DNA.
- Alternative markers are also suitable for this, such as nutritive markers and screening markers (such as GFP, green fluorescent protein).
- selection markers can also be dispensed with entirely, but this is accompanied by a fairly high need for screening. If the selection marker used is to be removed again after the transformation and identification of successfully transformed cells or plants has been carried out, various strategies are available to the person skilled in the art.
- sequence-specific recombinases can be used, for example in the form of retransformation of a starting line expressing recombinase and outcrossing of the recombinase after removal of the selection marker (see, for example, Reiss et al. (1996) Proc. Natl. Acad. Sei. USA 93: 3094-3098; Bayley et al. (1992) Plant Mol. Biol.
- the selection marker can also be removed by cotransformation followed by outcrossing.
- the regeneration of the transgenic plants from transgenic plant cells is carried out according to customary regeneration methods using conventional nutrient media and phytohormones.
- the plants thus obtained can then, if desired, be carried out by conventional methods, including molecular biological methods, such as PCR, blot analysis, or biochemical methods for the presence of the introduced DNA which encodes a protein with the enzymatic activity of a TAG lipase specific for oxygenated polyene fatty acids , or be examined for the presence of TAG lipase enzyme activity.
- the detection of the enzymatic activity of the TAG lipase according to the invention can also be determined by a person skilled in the art using protocols available in the literature. Furthermore, one can, for example, lay out the seed obtained by selfing or crossings on medium which contains the selection marker transferred to the TAG lipase DNA sequence contains suitable selection agents, and based on the germination capacity and growth of the daughter generation (s) and the segregation pattern, conclusions can be drawn about the genotype of the respective plant.
- Another object of the invention is to show uses of the nucleic acid sequences according to the invention and of the nucleic acid molecules containing them.
- This object is achieved by the novel uses of the new DNA sequences and molecules for producing plant cells and plants which are distinguished by a change in the content of polyene fatty acids, particularly preferably of oxygenated polyene fatty acids in seed oil, in comparison with wild type cells or wild type plants.
- the present invention further encompasses any possible form of use of the nucleic acid molecules according to the invention, the presence and expression of which, if appropriate, in plants causes a change in the content of polyene fatty acids, in particular of oxygenated polyene fatty acids in seed oil, and the use of the proteins or fragments thereof, the enzymatic activity of which brings about such a change.
- the invention further relates to the use of a DNA sequence according to the invention or parts thereof for the identification and isolation of homologous sequences from plants or other organisms.
- the nucleic acid molecules according to the invention can thus be used according to the invention to generate transgenic plants in which the content of the acyl hydrolases, in particular TAG lipases, is higher or lower than natural, or in cell types or development stages in which the acyl hydrolases, in particular TAG, are present -Lipasen normally not found. This causes a change in the levels of triacylglycerol and cholesteryl esters in these cells.
- Triacylglycerol lipases can also be useful in the processing of vegetable oils and the development of new seed oils, since the shelf life of vegetable seed oils for the food industry can be prolonged by a selective cleavage of oxidized polar fatty acid residues by the TAG lipases according to the invention.

Keegstra (1989) Cell 56: 247-253), signal sequences and the like ,
- nucleic acid molecules that code for TAG lipases especially for TAG lipases that are specific for oxygenated polyene fatty acids.
- a nucleic acid molecule developed for the cosupression of the TAG lipase according to the invention can be linked by linking a gene or a gene fragment which is one for oxygenated
- RNA-specific TAG lipase encoded with plant promoter sequences Polyen fatty acid-specific TAG lipase encoded with plant promoter sequences.
- a nucleic acid molecule which has been developed for the expression of antisense RNA for all or part of the nucleic acid molecule according to the invention can be generated by linking the gene or gene fragment in a reverse orientation to plant promoter sequences. Both the Genes for cosuppression as well as the antisense genes can be introduced into the plants by means of transformation, as a result of which the expression of the corresponding endogenous genes is reduced or eliminated.
- the invention is based on the successful isolation of cDNA clones coding for an acyl hydrolase, in particular for a TAG lipase, particularly preferably for a TAG lipase coding for oxygenated polyene fatty acids from a cDNA library from Arabidopsis thaliana.
- the sequences of these cDNA clones which comprise a complete reading frame, are shown in SEQ ID NO: 1, SEQ ID No: 3 and SEQ ID No: 5.
- transgenic plants which, owing to the transmission and expression of a TAG lipase, in particular a TAG lipase specific for oxygenated polyene fatty acids, have an oxygenated polyene fatty acid content in the seed oil which is different from that of wild type plants.
- cDNA sequences coding for an Arabidopsis thaliana TAG lipase were identified by screening of Arabidopsis thaliana cDNA libraries prepared as described below in accordance with a standardized protocol also described below. Three of the annotated sequences (Lipase 1: SEQ ID No: 1, Lipase 2: SEQ ID No: 3, Lipase 3: SEQ ID No: 5) were cloned, expressed and tested for activity. I. Preparation of various cDNA banks from Arabidopsis thaliana
- cDNA libraries were prepared from a mixture of various Arabidopsis organs and a cDNA library from Arabidopsis flowers.
- RNA from Arabidopsis The following reagents were used to isolate RNA from Arabidopsis:
- RNA from Arabidopsis is carried out according to the following protocol: - Mortaring 20 g of plant material in liquid nitrogen
- RNA in the photometer at a dilution of 1: 100.
- the ratio of A 60 to A 280 should be greater than or equal to 1.8.
- the measured value is multiplied by the dilution factor and a factor of 40, which results in the concentration of the RNA in ⁇ g / ml.
- a wavelength scan of 200-300 nm results in a typical RNA curve with a maximum at 260 nm.
- the absolute amount of RNA is then calculated and an RNA agarose gel is used to determine the quality of the RNA.
- the mRNA isolation is carried out with the mRNA isolation kit Poly-Attract from Promega (Mannheim, Germany).
- the reverse transcriptase-PCR was carried out with 1 ⁇ g RNA, 200 pmol oligo-dT with Superscript II from Gibco (Eggenstein, Germany) according to the manufacturer's instructions.
- the PCR amplification was carried out using the Expand TM high fidelity PCR system in accordance with the manufacturer's instructions (Boehringer Mannheim, Germany).
- the Arabidopsis cDNA mixture was used as the template for the lipases 1 and 2 and the cDNA from the flower of Arabidopsis for the lipase 3.
- the following sequences were used as primers:
- Lipase 1 (3 x methionine in the beginning of the sequence) 5 ': - CCC GCATGC ATG CAG TTG TCT CCG GAA CGA TGC (AI) 5 *: - GGG GCATGC ATG TCT GAA AAC AGA GAG GCT TGG (A2) 5 ': - AAA GCATGC ATG GAG CTG CTT CAC GGC TCC AAC (A3) 3 *: - AAA GTCGAC TCAAAAAGG GCT GAC CCT GCC AGC C
- Lipase 2 5 ': - CCC CGC ATG CAT GGC TTC TTCACT GAA GAA
- the open reading frame for the complete unprocessed protein was determined by means of the above oligo-nucleotide primers from the DNA sequences according to the sequences SEQ ID No: 1, 3 and 5 amplified by PCR.
- the PCR conditions were as follows: 2 minutes denaturation at 94 ° C, then for 10 cycles: 30 seconds denaturation at 94 ° C, 30 seconds annealing at 65 ° C, 3 minutes elongation at 72 ° C; then for 15 cycles: 30 seconds denaturation at 94 ° C, 30 seconds annealing at 65 ° C, 3 minutes elongation at 72 ° C plus a time increment of 5 seconds per cycle; finally another 2 minutes elongation at 72 ° C.
- the amplified PCR fragment was ligated into the expression vector QIAexpress pQE 30 and pre-cloned with transformation in XLl-Blue cells using the p GEM R- Easy Vector System II kit (Promega, Madison, USA). The cloning was then carried out into the expression strain E. coli SG13009 [pREP4].
- the cleaning was carried out according to the following steps: - Preparation of a membrane fraction of the E.coli SG13009 [pREP4]
- the protein eluate obtained as described above was extracted with a lipid extract (chloroform: methanol, Bligh / Dyer 1959) from four-day-old seedlings of Cucumis sativa for 40 minutes at 25 ° C. with the addition of buffer concentrate (1 M Tris pH 8.5; 0.5 M NaCl; 50 mM CaCl 2 ) to adjust the pH to 8.0. It was then acidified with acetic acid and extracted with hexane. The analysis was carried out by means of HPLC (LiCHrosphere 100 RP-18 (MERCK, Darmstadt, Germany); mobile phase: acetonitrile: H 2 O: acetic acid 70: 30: 0.1).
- Example 3 Transformation of Arabidopsis thaliana and Nicotiana t ⁇ bacum plants and regeneration of intact plants
- the DNA sequences according to SIQ ID No: 1, 3 and 5 were each converted into a suitable one binary vector (e.g. pPCVOOl, pPCV002 (Koncz & Schell 1986 MGG 204: 383-396) cloned.
- a suitable one binary vector e.g. pPCVOOl, pPCV002 (Koncz & Schell 1986 MGG 204: 383-396) cloned.
- any vector suitable for plant transformation can be used for the production of a chimeric gene, consisting of a fusion of the CaMV 35S promoter or another constitutive, seed-specific, germination-specific or flower-specific promoter which promotes transcription and, if desired, translation guaranteed in plant cells, and DNA sequences coding for TAG lipase are used.
- the binary vector was then transformed into Agrobacterium tumefaciens (strain GV2260; Horsch et al. (1985), Science 227, 1229-1231) and for the transformation of Arabidopsis or tobacco plants (SNN) by means of the leaf disc transformation technique (Horsch et al. , supra) used.
