Description

5-Halo-4-fluor-4,7,7-trimethyl-3-oxabicyclo[4.1.0]heptan-2-one, Verfahren zu ihrer Herstellung sowie ihre Verwendung zur Herstellung von Cyclopropancarbonsäuren.
Pyrethroide sind eine wichtige Klasse von Insektiziden, deren Wirksamkeit auf einer starken Beeinflussung der Natriumkanäle in den Nervenmembranen von Insekten beruht.
In der WO-A 99/32426 sind Derivate der 2,2-DimethyI-3-(2-fluorvinyl)cyclopropan- carbonsäure als hochwirksame Pyrethroide beschrieben. Die Herstellung dieser Säure erfolgt durch mehrstufige reduktive Transformation von 2,2-Dimethyl-3-(2- fluor-3-hydroxy-3-oxo-1-propenyl)cyclopropancarbonsäureestern.
Aufgabe war es, neue vorteilhafte Synthesewege für die Säure und damit die genannten Pyrethroide zu finden.
Es wurde nun überraschend gefunden, daß sich 2,2-Dimethyl-3-(2- fluorvinyl)cyclopropancarbonsäure in einfacher Weise in hoher Reinheit und guten Ausbeuten durch Halofluorierung von 4,7,7-Trimethyl-3-oxabicylo[4.1.0]hept-4-en-2- on zu den neuen 5-Halo-4-fluor-4,7,7-trimethyl-3-oxabicyclo [4.1.0]heptan-2-onen mit anschließender reduktiver Eliminierung herstellen läßt.
Die Halofluorierung von Enollactonen ist im Prinzip bekannt (siehe M. Brand u. S. Rozen, J. of Fluorine Chem. (1982), 20, 419 oder Houben-Weyl, Methoden der organischen Chemie, Bd. E. 10).
Allerdings ist die Halofluorierung nur auf solche Substrate beschränkt, die unter Halofluorierungsbedingungen stabil sind. Ferner ist bekannt, daß Cyclopropan oder dessen Derivate mit HF oder sogar mit HF/Py schon bei Raumtemperatur unter Ringöffnung und Bildung von Fluorpropanen oder Cyclobutanderivaten reagieren (siehe z.B. J. Org. Chem. (1979), 44, 3873 oder Tet. Lett. (1987), 28, 6313).
Dreiringe enthaltende bicydische Enollactone sind sehr sensibel gegenüber Säuren, Basen und Oxidationsmitteln und reagieren normalerweise unter Lacton- oder Cyclopropanringöffnung. Deswegen ist es überraschend, daß die Chloro-, Bromo- oder lodofluorierung von Enollactonen zu Chlorfiuor-, Bromfluor- oder lodfluorverbindungen in guten Ausbeuten und hohen Reinheiten führt.
Gegenstand der Erfindung sind halogenierte Lactone der Formel (I),
wobei Hai Cl, Br oder I bedeutet.
Die Verbindungen der Formel (I) umfassen alle möglichen Stereoisomeren.
Verbindungen der Formel (I) eignen sich in hervorragender Weise als Zwischenprodukte zur Herstellung von Pyrethroiden, wie sie in WO-A 99/32426 beschrieben sind.
Die Herstellung der Verbindungen der Formel (I) erfolgt durch Umsetzung eines Enollactons (II) mit einem Halogenfluorierungsmittel („HalF"),
wobei Hai Cl, Br oder I bedeutet.
Dieses Verfahren ist ebenfalls Gegenstand der Erfindung.
Unter dem Begriff Halogenfluorierungsmittel versteht man Regenzien oder Kombinationen daraus, die positive Halogenatome und Fluoridanionen auf organische Verbindungen übertragen können.
Die Umsetzung kann mit CIF, BrF (siehe z.B. M. Brand u. S. Rozen, I. of Fuorine Chem. (1982), 20, 419) oder vorzugsweise einem Äquivalent, enthaltend eine Verbindung, die ein oder mehrere positivierte Halogenatome (Hal+) enthält, sowie Flußsäure oder ihren Salzen (siehe z.B. F. Camps et al., J. Org. Chem. (1989), 54, 4294) erfolgen.
Als Hal+-Quelle sind bevorzugt N-Chloracetamid, Hexachlormelamine.N- Bromacetamid, N-Chlorsuccinimid (NCIS), N-Bromsuccinimid (NBS), N- lodsuccinimid, 1-Brom-3,5,5-trimethylhydantoin, N,N-Dibrom-5,5-dimethylhydantoin DBH), daneben sind andere Halogenierungsmittei möglich. Man kann auch ein Gemisch von zwei oder mehreren Halogenverbindungen einsetzen. Besonders bevorzugt als Halogenierungsmittei sind NBS und DBH, ganz besonders bevorzugt ist DBH.
Als Fluoride werden im allgemeinen Fluoride der Formel (III) eingesetzt:

worin KAT für ein stöchiometrisches Äquivalent eines Erdalkalimetallions, eines Alkalimetallions, eines Tetraalkylammonium-lons, eines Tetralkylphosphonium-Ions, eines Tetraarylphosphonium-Ions oder eines Tetrakis(dialkylamino)phosphonium- lons steht, wobei Alkyl eine Alkylgruppe mit vorzugsweise 1 bis 6 Kohlenstoffatomen, Aryl eine Arylgruppe mit 6 bis 12 Kohlenstoffatomen und n 0, 1 , 2, 3, 4 oder 5 bedeutet.
Ohne Anspruch auf Vollständigkeit zu erheben, seien folgende Stoffe als Fluoride genannt:
Kaliumhydrogendifluorid, Kaliumtetrahydrogenpentafluorid, Tetramethylammoniumfluorid, Tetramethylammoniumhydrogendifluorid, Tetramethylammoniumdihydrogen-trifluorid, Tetraethylammoniumhydrogendifluorid, Tetrabutylammoniumhydrogen-difluorid, Tetrabutylammoniumdihydrogentrifluorid, Benzyltrimethylammoniumhydrogendifluorid, Tetraphenylphosphoniumhydrogen- difluorid, Tetrakis(diethylamino)phosphoniumhydrogendifluorid, HF, HF/Puridin- Komplex, Et3N x 3HF, Bu3N x 3HF. Man kann auch ein Gemisch von zwei oder mehreren Fluoriden einsetzen. Es ist auch möglich, Systeme aus fluorhaltigen Verbindungen und Wasser einzusetzen, z.B SiF4/H2O, SbF3/H2O, AIF3/H2O. Ferner es ist möglich, [CIF] aus Cl2 und F2 oder [BrF] aus Br2 und F2 einzusetzen (siehe z.B.M. Brand u. S. Rozen, J. of Fluorine Chem. (1982), 20, 419).
Das Verfahren läßt sich in Anwesenheit oder Abwesenheit eines Lösungsmittels durchführen. Werden Lösungsmittel verwendet, so sind sowohl polare als auch nichtpolare Lösungsmittel geeignet. Geeignete Lösemittel sind beispielsweise: Hexan, Cyclohexan, Dioxan, Diethylether, Diisopropylether, Toluol, Dichlormethan, Dichlorethan und Polyether.
Die Reaktionstemperatur liegt üblicherweise zwischen -30 und + 30°C, bevorzugt zwischen -15 und +20°C und hängt auch in der dem Fachmann bekannten Weise von der Art des Halofluorierungsreagenzes ab.
Man setzt die Fluoride im allgemeinen in Mengen von 0.2 bis 2, insbesondere 0.25 bis 1.5, bevorzugt 0.4 bis 1 Mol, bezogen auf 1 Mol Enollacton ein. Die Menge an Fluorid hängt von der Anzahl Fluoridatome im Molekül ab. So erfordert die Anwendung von HF die doppelte Menge im Vergleich zu Tetrabutylammonium - hydrogendifluorid. Es ist zu berücksichtigen, daß es Fälle geben kann, in denen ein Überschuß an Fluorid zu unerwünschten Nebenreaktionen führen kann. In diesen Fällen empfiehlt es sich, das Fluorid im Unterschuß einzusetzen.
Das Halogenierungmittel wird im allgemeinen in einer Menge von 0.5 bis 2 Mol, bevorzugt 0.4 bis 1 Mol bezogen auf 1 Mol Enollacton eingesetzt. Die Menge an Halogenierungsmittei hängt von der Anzahl Halogenatome im Molekül ab. So erfordert die Anwendung von N-Bromsuccinimid die doppelte Menge im Vergleich zu Dibromhydantoin.
Das Enollacton der Formel (II) umfaßt alle möglichen stereoisomeren Formen. Es ist bekannt und läßt sich nach literaturbekannten Methoden synthetisieren (siehe z.B. D. Bakshi et al., Tetrahedron (1989), 45, 767).
Die Halofluorierung von Enollactonen der Formel (II) führt zu einem Isomerengemisch von mehreren Stereoisomeren, z.B. entsteht im Fall der Bromofluorierung von (1 R,6S)-4,7,7-Trimethyl-3-oxabicyclo[4.1.1]hept-4-en-2-on ein Isomerengemisch von hauptsächlich drei Stereoisomeren:

Allerdings ist es in einfacher, dem Fachmann geläufiger Weise möglich, durch Reaktionsbedingungen (Zeit, Temperatur etc.) die Isomerenverhältnisse zu beeinflussen (siehe die Tabelle 1 in den Beispielen). Isomer A läßt sich durch Kristallisation aus Alkohol in reiner Form zu isolieren. Die Isolierung von anderen Isomeren gelingt durch Chromatographie, beispielsweise durch Mitteldruckflüssigchromatographie (MPLC) an Hexan/Essigester Gemischen. Die Verbindungen der Formel (I) lassen sich durch Reaktion mit einem Reduktionsmittel weiter zu cis-Cyclopropancarbonsäuren der Formel (IV) umsetzen:

Dieses Verfahren, sowie ein Verfahren zur Herstellung von Cyclopropancarbonsäuren der Formel (IV) durch sukzessive Halofluorierung und Reduktion des Enollactons (II) sind ebenfalls Gegenstand der Erfindung.
Als Reduktionsmittel kommen alle herkömmlichen Reduktionsmittel, wie Mg, Fe, Zn, Sn , AI, Bu3SnH, LiAIH4, in Frage (siehe z.B. J. Am. Chem. Soc, 121 , 4155, 1990). Die Cyclopropancarbonsäure der Formel (IV) kann als Gemisch von zwei geometrischen Isomeren entstehen.
Die Säure der Formel (IV) oder ein funktionelles Derivat der Säure können mit einer Verbindung der Formel (V),

worin X eine Abgangsgruppe, vorzugsweise OH, Cl, Br, I, OTosyl (Tosyl: p- Toluolsulfonyl) oder OMesyl (Mesyl: Methansulfonyl), und R den Rest eines Alkohols aus der Pyrethroidreihe bedeutet, zu einer Verbindung der Formel (VI) umgesetzt werden,
(VI) wobei R die oben genannte Bedeutung hat.
Bevorzugt hat R dabei die in Formel (I) in WO-A 99/32426 genannten Bedeutungen. Auf diese Schrift wird hiermit ausdrücklich Bezug genommen; sie gilt durch Zitat als Bestandteil dieser Beschreibung.
Die Ester der Formel (VI) können durch Veresterung entsprechender Carbonsäuren (oder ihrer reaktionsfähigen Derivate) mit Alkoholen (oder ihren reaktionsfähigen Derivaten) nach der DCC-Methode (DCC = Dicyclohexylcarbodiimid) oder analog DE-A 44 27 198 erhalten werden. Die entsprechenden Carbonsäuren und Alkohole sind bekannt oder können in Analogie zu bekannten Verfahren hergestellt werden.
Als reaktionsfähige Derivate der genannten Carbonsäuren eignen sich insbesondere die Säurehalogenide, vor allem die Chloride und Bromide, ferner die Anhydride, z.B. auch gemischte Anhydride, Azide oder Ester, insbesondere Alkylester mit 1-4 C- Atomen in der Alkylgruppe.
Als reaktionsfähige Derivate von Alkoholen kommen insbesondere die entsprechenden Metallalkoholate, vorzugsweise eines Alkalimetalls, wie Natrium oder Kalium, in Betracht.
Die Veresterung wird vorteilhaft in Gegenwart eines inerten Lösungsmittels durchgeführt. Gut geeignet sind insbesondere Ether, wie Diethylether, Di-n- butylether, THF (Tetrahydrofuran), Dioxan oder Anisol, Ketone, wie Aceton, Butanon oder Cyclohexanon, Amide, wie DMF (NiN-Dimethylformamid) oder Phosphorsäurehexamethyltriamid, Kohlenwasserstoffe, wie Benzol, Toluol oder Xylol, Halogenkohienwasserstoffe, wie Tetrachlorkohlenstoff, Dichlormethan oder Tetrachlorethylen, und Sulfoxide, wie Dimethylsulfoxid oder Sulfolan. Gegenstand der Erfindung ist daher auch ein Verfahren zur Herstellung einer Verbindung der Formel (VI),

a) Umsetzung eines Enollactons der Formel (II) mit einem Halogenfluorierungsmittel zu einer Verbindung der Formel (I), b) Umsetzung der Verbindung der Formel (I) mit einem Reduktionsmittel zu einer Cyclopropancarbonsäure der Formel (IV) und c) Veresterung der Cyclopropancarbonsäure der Formel (VI) mit einer Verbindung

wobei X und R die oben genannteen Bedeutungen haben.
Bevorzugt nach dem erfindungsgemäßen Verfahren hergestellte Verbindungen der Formel (VI) sind die Beispiele 1 bis 33 der WO-A 99/32426, auf die hier ausdrücklich Bezug genommen wird; sie gelten durch Zitat als Bestandteil dieser Beschreibung.
Die Synthese der entsprechenden Alkoholreste findet sich in WO-A 99/32426 und der dort aufgeführten Literatur.
Weiterhin Gegenstand der Erfindung ist die Verwendung von Verbindungen der Formel (I) als Zwischenprodukte bei der Herstellung von Pyrethroiden, vorzugsweise solcher der Formel (VI). Auf den Inhalt der deutschen Patentanmeldung 199 54 160.4, deren Priorität die vorliegende Anmeldung beansprucht, sowie der beiliegenden Zusammenfassung wird hiermit ausdrücklich Bezug genommen; er gilt durch Zitat als Bestandteil dieser Beschreibung.
Die Erfindung wird durch die Beispiele näher erläutert, ohne sie dadurch einzuschränken.
Beispiele
Halofluorierung (allgemeine Methode)
In einem Dreihalskolben mit Blasenzähler, Thermometer, mechanischem Rührer und Tropftrichter wurden unter N2 Halogenierungsmittei und Fluorid in einem Lösemittel vorgelegt und das Enollacton bei einer bestimmten Temperatur zugegeben. Nach beendeter Zugabe wurde die Lösung noch bestimmte Zeit nachgerührt, dann wurde die Reaktion durch Zugabe von Wasser und Na2SO3 beendet. Das Produkt wurde mit Diethylether extrahiert. Die Etherphase wurde mit Wasser gewaschen, mit MgSO4 getrocknet, filtriert, eingeengt und der Rückstand gereinigt. Gemäß dieser Methode wurden erhalten:
Isomer A

Isomer B
(1R,4S,5S,6S)-5-Brom-4-fluor-4,7,7-trimethyl-3-oxabicyclo[4.1.0]heptan-2-on
1H- NMR (CDCI3, 300 MHz): 1.30 (s, 3H, CH3); 1.4 (s, 3H, CH3); 1.84 (d, 3J =19 Hz ,

Beispiel 12 (1 R-cis)-3-(E)-2-Fluorpropenyl)-2,2-dimethylcyclopropancarbonsäure und (1 R,cis)-3-[(Z)-2-Fluorpropenyl)-2,2- dimethylcyclopropancarbonsäure.
In einem Dreihalskolben mit Blasenzähler, Thermometer und mechanischem Rührer wurden 25.2 g (0.1 mol) Isomerengemisch von 5-Brom-4-Fluor-4,7,7- trimethyl-7-fluor-3-oxabicyclo[4.1.0]heptan-2-on und 150 ml Essigsäure vorgelegt, und 12.6 g (0.2 Mol) Zinkstaub wurden portionsweise während 1 h zugegeben. Nach beendeter Zugabe wurde die Lösung noch 4 h nachgerührt, restliches Zn abfiltriert und die Lösung mit 200 ml Eiswasser versetzt. Das Produkt wurde mit Diethylether extrahiert. Die etherische Phase wurde mit Wasser gewaschen, getrocknet (MgSO4), filtriert und eingeengt. Die Isolierung der Säuren erfolgte durch Chromatographie. Ausbeute 21 %, Öl.

Claims

Patentansprüche
1. Verbindung der Formel (I),
wobei Hai Cl, Br, oder I bedeutet.
2. Verbindung der Formel (I) nach Anspruch 1 , wobei Hai Br bedeutet.
3. Verwendung von Verbindungen der Formel (I) nach Anspruch 1 oder 2 als Zwischenprodukte bei der Synthese von Pyrethroiden.
4. Verfahren zur Herstellung einer Verbindung der Formel (I) nach Anspruch 1 oder 2, wobei man ein Endlacton der Formel (II) mit einem Halogenfluorierungsmittel („HalF") umsetzt,
wobei Hai Cl, Br oder I bedeutet.
5. Verfahren zur Herstellung einer cis-Cyclopropancarbonsäure der Formel (IV),
(I) (IV)
wobei man die Verbindung der Formel (I) gemäß Anspruch 1 mit einem Reduktionsmittel umsetzt.
6. Verfahren zur Herstellung einer Verbindung der Formel (VI),
wobei R den Rest eines Alkohols aus der Pyrethroidreihe bedeutet, enthaltend die Schritte:
a) Umsetzung eines Endollactons der Formel (II) mit einem Halogenfluierungsmittel zu einer Verbindung der Formel (I) gemäß Anspruch 1 oder 2,
b) Umsetzung der Verbindung der Formel (I) mit einem Reduktionsmittel zu einer Cyclopropyancarbonsäure der Formel (IV) gemäß Anspruch 5 und
c) Veresterung der Cyclopropancarbonsäure der Formel (IV) mit einer Verbindung (V),
R - X (V) wobei X eine Abgangsgruppe und R den Rest eines Alkohols aus der Pyrethroidreihe bedeutet.
