Definitions
- the invention relates to an arrangement for contamination-free processing in particular molecular biological reaction sequences with the Features of the preamble of claim 1.
- Subject of the invention also a corresponding closure carrier for such an arrangement, a single closure for such an arrangement and a storage and dispensing arrangement for such individual closures.
- nucleic acid diagnostics A particularly problematic area is nucleic acid diagnostics, in which The prevention of contamination is very important.
- special Importance of modern duplication methods such as the polymerase chain reaction, through which the detection limit significantly reduced becomes.
- the danger of falsification of the measurement results increases through the previously mentioned contaminations.
- nucleic acid diagnostics may total be referred to the EP 0 676 643 A2, from the extensive See further versions.
- the individual closures are here on the closure carrier by held a one-piece or multi-part adhesive film and can so together with the closure carrier all sealing together on the reaction vessels put on, but also together or in groups, together with the closure carrier be deducted from these again. If you remove the adhesive film, Thus, the respective individual closure remains sealingly on the reaction vessel.
- the individual closures are in the previously described prior art as executed comprehensive cover, so that the respective closure portion the open end of the reaction vessel bordered edge.
- the Reaction vessels according to the connection plane of the reaction vessels have protruding edge above. It is therefore in the arrangement with the reaction vessels in this prior art is not a typical Microtiter plate, which is indeed executed plan at the top.
- the starting point for teaching forming arrangement to contamination-free processing of particular molecular biological Reaction processes is a problem in that the selective Opening a single reaction vessel, first the adhesive film for all Reaction vessels are removed and then removed the closure carrier must be, if one wants to ensure that to avoid Contaminations all other reaction vessels remain closed.
- the Teaching therefore sets itself the goal of eliminating this problem.
- the invention moreover, the individual parts of a Arrangement of the type in question, ie in particular the closure carrier and the individual closures.
- the subject matter of the invention is a supply and dispensing arrangement for individual closures, since it has been shown according to the invention that the Single closures also individually, so without the closure carrier, on the reaction vessels can be used like that, of course, from the EP 0 676 643 A 2, EP 0 734 769 A 1).
- the teaching of the invention can be particularly useful in context with an industrial robotic system or for such an industrial robotic system use.
- microtiter plate shown in FIG. 1 for 96 reaction vessels for small and smallest amounts of sample liquid is an example of an arrangement interconnected reaction vessels, not limiting to understand is. Also strip-like arrangements of several together connected reaction vessels are the subject of the invention.
- PCR technology As has been extensively explained in the prior art (see, for example, EP 0 676 643 A2).
- the capacity of a reaction vessel 2 is enclosed this application example at about 200 to 400 ul. That too is not restrictive to understand.
- Fig. 2 shows that in the inventive arrangement each reaction vessel 2 is associated with a single closure 3.
- Fig. 4 shows such a single closure 3 in a side view. It can be seen that the single closure 3 a closure portion 4 for sealing the opening of the reaction vessel 2 and an actuating portion 5 for engaging the single closure 3 for the purpose of managing it.
- FIGS. 2 and 3 further show an all reaction vessels 2 of the microtiter plate 1 covering closure carrier 6, each with a receptacle 7 for the respective Operating section 5 of each single closure 3 for securing the single closure 3 on the closure carrier 6.
- the closure carrier 6 can only cover a group of reaction vessels 2 of the overall arrangement, so that the overall arrangement then has a plurality of closure carrier 6.
- Figs. 2, 3 and 4 can be seen in connection now that in the teaching of Invention is provided that the closure portions 4 of the individual closures 3 are plugged through the receptacles 7 in the closure carrier 6 therethrough, So when inserting and pulling out of the closure carrier 6 not collide with the closure carrier 6.
- the closure carrier 6 is an independent "lid carrier” the individual closures 3 are so releasably attached to them that they too individually taken out in fitting to the reaction vessel 2 closure carrier 6 can be. This allows the single opening of each reaction vessel 2 with otherwise closed other reaction vessels. 2 This has the technical handling explained in the general part of the description Advantages and in particular avoids any cross-contamination.
- the illustrated and In this respect preferred embodiment shows that the individual closures 3 on the closure carrier 6 by means of a releasable positive connection 8, 9 are fixed between the operating section 5 and receptacle 7. This can be seen particularly well in a Zulsanunenschau of Fig. 2, 3 and 4. Die lateral dimensions of the receptacle 7 and the attachment portion. 5 of the single closure 3 are in the illustrated and preferred embodiment chosen and the positive connection 8, 9 is designed so that they are inserted into the receptacle 7 single closure 3 a slight lateral displacement of the single closure 3 in the receptacle 7 allowed.
- a normal bayonet lock reaches its closed position by a Rotary movement in one direction and its opening position by a Rotary motion in the opposite direction.
- Such a bayonet lock can also be used in the invention for the positive connection 8, 9 become.
- the preferred embodiment shows a construction in which the bayonet lock in both directions can be actuated is and has no stop, but a surmountable detent 8a. It can be seen the detent 8a on each pin part 8 in Fig. 5. With this construction the operator is free, in which direction they the bayonet lock wants to operate, it always reaches the other functional position of the bayonet lock.
- Fig. 2 indicates and Fig. 5 also shows the single closure 3 more clearly that the single closure 3, in particular and shown here on the operating section 5, a closure handling means 10 for attacking one of the handling a single single closure 3 serving operating tool 11 has.
- the actuating tool 11 indicated as a pin-like hand tool. But one is for more extensive Handling often also automatically moved and operated Insert actuation tools 11, how to stand out from the stand Technique is known.
- the closure handling means 10 is designed as a form-locking element, to which a corresponding Positive locking element 12 corresponds to the actuating tool 11. More specifically, in the illustrated embodiment, the shutter handling means 10 as a part, namely here as a sliding part, a bayonet closure executed.
- the form-locking element 12 on the actuating tool 11 is as the other part, in particular the pin part of the bayonet closure executed.
- closure handling means 10 and the corresponding Positive locking element 12 shows the illustrated embodiment a standard, classic bayonet closure construction. It should be so far expediently provided that after production of the bayonet closure engagement a rotation of the individual closure used in the receptacle 7 3 is effectively possible in both directions, so that the present invention realized in the preferred embodiment Right / left insensitivity of the positive connection 8, 9 also exploited can be.
- the illustrated and preferred embodiment shows in particular FIGS. 4 and 5, that the single closure 3, the positive connection 8, 9 or frictional connection is arranged on the outside of the actuating section 5, while the closure handling means 10 of the single closure 3 inside is arranged.
- closure portion 4 of a single closure 3 With regard to the interaction of the closure portion 4 of a single closure 3 with the open end of the associated reaction vessel. 2 can, as in the prior art, be provided that the closure portion 4 of the single closure 3, the reaction vessel 2 overlapped.
- the illustrated and preferred embodiment shows an embodiment which characterized in that the closure portion 4 of the single closure 3 enters the open end of the reaction vessel 2 like a plug.
- This is known per se from the prior art. That allows one particularly expedient, realized here in the embodiment also Conception in such a way that the fastening portions 5 of the individual closures 3 consist of a relatively hard, rigid plastic material, the Closure sections 4 are integrally formed materially and from a consist of relatively soft, rubber-elastic plastic material.
- the prior art has been explained that even individual closures. 3 are known, which have a thin membrane through which the interior of the each reaction vessel can be reached with a needle.
- the illustrated and preferred embodiment shows a similar construction, namely a construction in which the closure portion 4 is a pierceable Membrane section 4a has. This one is through a central one Passage 5a reachable in the attachment section 5. Both show Fig. 5.
- the Execution of the closure portion 4 of a relatively soft, rubber-elastic Plastic material, in particular of a thermoplastic elastomer offers the convenient possibility that a puncture in the Diaphragm section 4a of the closure section 4 closes again by itself, because the thermoplastic elastomer has a sufficient restoring force.
- Fig. 6 shows an enlarged view
- the operating tool 11 with the therein form-locking element 12 and a possibly spring-loaded Thrust pin 13 the fixation of the bayonet lock for Closure handling means 10 of the single closure 3 makes.
- the spring force for fixing the Bayonet closure by the inherent elasticity of the closure portion. 4 is applied. In the illustrated embodiment is to the Thrust pin 13 in the passage 5a directly on the material of the closure portion 4, so that this additional function is performed.
- Fig. 3 shows the closure carrier 6 of an inventive arrangement for 2 shows the closure carrier 6 equipped with individual closures 3.
- Such closure carrier are also self-negotiable elements, the therefore be subject to independent protection. The same applies to the Single closures 3.
- Fig. 7 shows schematically an example of a supply and dispensing arrangement 14 for individual closures 3 of the type in question.
- This is equipped with its own form-locking formation 15, with the form-locking education, namely the pin parts 8, on the operating section. 5 the single closure 3 cooperates in such a way that the individual closures 3 in exactly predetermined, certain lateral orientation of the Supply and dispensing arrangement 14 can be dispensed. So you can specifically a Equipping either the closure carrier 6 with the individual closures. 3 accomplish or even directly equipping the reaction vessels. 2 without closure carrier 6.
- the arrangement according to the invention can be particularly useful within of an industrial robot system, with the reaction sequences of in Speech type in series and essentially automatically checked become.
- the operating tool 11th Be part of the industrial robot system for example on an actuating arm arranged, whereby the industrial robot system also the reaction vessels 2 and the closure carrier 3 automatically handles, at least operable fixed.
- the teaching is also a use of an arrangement of in Talking style in an industrial robot system.
