Abstract

Ce procédé, qui permet de mesurer des caractéristiques physiques, telles que la taille et le poids, de molécules individuelles, consiste à soumettre une molécule, déformable ou non, à une force externe qui produit des changements de conformation ou de position, puis à mesurer ces changements. Des moyens préférés de mesurer de tels changements consistent à 1) déterminer la vitesse à laquelle une molécule s'oriente dans une nouvelle direction quand on modifie la direction de la force perturbatrice, 2) déterminer la vitesse à laquelle une molécule déformable revient à un état relâché après disparition de force externe, 3) déterminer la vitesse àlaquelle une molécule pivote, 4) mesurer la longueur d'une molécule quand elle est partiellement étirée, ou 5) mesurer le diamètre d'une molécule sphérique ou ellipsoïdale. On peut réaliser ces mesures avec un microscope optique connecté à un dispositif spectroscopique. Ce procédé sert en particulier à mesurer des molécules polymères telles que des acides nucléiques et permet de déterminer la taille et la cartograhie de produits de digestion de restriction.
