Claims (17)
1. Genmodificeret svin, hvori mindst én allel af et SIGLECl-gen er blevet inaktiveret.
2. Genmodificeret svin, hvori mindst én allel af et CD163-gen er blevet inaktiveret.
3. Genmodificeret svin ifølge krav 1 eller 2, hvori begge alleler af et SIGLECl-gen er blevet inaktiveret.
4. Genmodificeret svin ifølge et hvilket som helst af kravene 1-3, hvori begge alleler af et CD163-gen er blevet inaktiveret.
5. Genmodificeret svin, hvori mindst én allel af et SIGLECl-gen er blevet inaktiveret, og mindst én allel af et CD163-gen er blevet inaktiveret.
6. Fremgangsmåde til fremstilling af et genmodificeret svin, hvori mindst én allel af et SIGLECl-gen er blevet inaktiveret, hvilken fremgangsmåde omfatter: enukleation afen svine-oocyt; fusionering af oocytten med en donor-svine-fibroblastcelle, hvor fibroblastcellens genom omfatter mindst én inaktiveret SIGLECl-allel, og aktivering af oocytten til fremstilling af et embryo.
7. Fremgangsmåde til fremstilling af et genmodificeret svin, hvori mindst én allel af et CD163-gen er blevet inaktiveret, hvilken fremgangsmåde omfatter: enukleation afen svine-oocyt; fusionering af oocytten med en donor-svine-fibroblastcelle, hvor fibroblastcellens genom omfatter mindst én inaktiveret CD163-allel, og aktivering af oocytten til fremstilling af et embryo.
8. Fremgangsmåde ifølge krav 6 eller 7, hvor: den mindst ene allel er blevet inaktiveret ved partiel eller komplet deletion; den mindst ene allel er blevet inaktiveret ved anvendelse af et Cre-lox-rekombinationssystem, og/eller SIGLECl-allelen er blevet inaktiveret ved deletion af en del af exon 1 og hele exon 2 og 3.
9. Fremgangsmåde ifølge et hvilket som helst af kravene 6, 7 og 8, hvor: enukleationen udføres med en mikropipette i et mikromanipulationsmedium; fusioneringen udføres i et fusionsmedium, og/eller aktivering af oocytten omfatter inkubation af oocytten i nærvær af thimerosal.
10. Afkom af et genmodificeret svin fremstillet ved fremgangsmåden ifølge et hvilket som helst af kravene 6, 7, 8 og 9, hvor i afkommet mindst én allel af et SIGLECl-gen er inaktiveret; mindst én allel af et CD163-gen er inaktiveret; mindst én allel af et SIGLECl-gen og mindst én allel af et CD163-gen er inaktiveret; eller begge alleler af et SIGLECl-gen og begge alleler af et CD163 gen er inaktiveret.
11. Afkom af et genmodificeret svin fremstillet ved fremgangsmåden ifølge krav 7, hvor mindst én allel af et CD163-gen er inaktiveret i afkommet.
12. Svin ifølge et hvilket som helst af kravene 1-5, hvor den mindst ene allel er blevet inaktiveret ved partiel deletion.
13. Fremgangsmåde ifølge krav 6 eller 7, hvor den mindst ene allel er blevet inaktiveret ved partiel deletion.
14. Svin ifølge et hvilket som helst af kravene 1-5, hvor den mindst ene allel er blevet inaktiveret ved komplet deletion.
15. Fremgangsmåde ifølge krav 6 eller 7, hvor den mindst ene allel er blevet inaktiveret ved komplet deletion.
