Claims (15)
1. Isoleret polypeptidvariant med glycosidhydrolaseaktivitet, som har en grad af identitet med aminosyreme 1 til 513 ifølge SEQ ID NO: 2 på mindst 90%, endnu mere fortrinsvis mindst. 95%, og mest fortrinsvis mindst 95%, hvor polypeptidets aminosyresekvens afviger fra SEQ ID NO: 2 i mindst én position svarende til position 94 blandt aminosyreme 1 til 513 ifølge SEQ ID NO: 2, hvor forskellen i mindst én position er en substitution i position 94, og varianten har forbedrede egenskaber sammenlignet med stamglycosidhydrolasen, hvor de forbedrede egenskaber er valgt fra gmppen bestående af temperaturafhængig aktivitetsprofil, termostabilitet, pH-aktivitet, pH-stabilitet og substratspecificitet, produktspecificitet og kemikaliebestandighed.
2. Polypeptidvariant ifølge krav 1, hvor substitutionen i polypeptidets aminosyresekvens i forhold til SEQ ID NO: 2 i en position svarende til position 94 er Ser, Ala, Arg or Gin.
3. Polypeptidvariant ifølge krav 1, hvor substitutionen i polypeptidets aminosyresekvens i forhold til SEQ ID NO: 2 i en position svarende til position 94 er Gly94Ser, Gly94Ala, Gly94Arg or Gly94Gln.
4. Polypeptidvariant ifølge et hvilket som helst af kravene 1-3, hvor polypeptidvarianten omfatter substitutioner i positioner svarende til position 94 og 226 blandt aminosyreme 1 til 513 ifølge SEQ ID NO: 2.
5. Po 1 ypeptidvariant ifølge krav 4, hvor varianten omfatter substitutionerne Gly94Ser (eller Gly94Ala eller Gly94Arg eller Gly94Gln) + Thr226Ala.
6. Isoleret nukleotidsekvens, der koder for polypeptidet ifølge et hvilket som helst af kravene 1-5.

12. Fremgangsmåde til nedbrydning af cellulose- og hemicelluloseholdig biomasse, hvilken fremgangsmåde omfatter behandling af biomassen med en effektiv mængde af et polypeptid ifølge et hvilket som helst af kravene 1-5 og indvinding af den nedbrudte biomasse.
13. Fremgangsmåde ifølge krav 12, som yderligere omfatter behandling af biomassen med en effektiv mængde af endo-l,4-beta-glucanase og exo-l,4-beta-D-glucanase.
14. Fremgangsmåde til nedbrydning af cellulose- og hemicelluloseholdig biomasse, hvilken fremgangsmåde omfatter behandling af biomassen med en værtscelle ifølge krav 9 og indvinding af den nedbrudte biomasse.
15. Fremgangsmåde ifølge krav 14, som yderligere omfatter behandling af biomassen med en effektiv mængde af endo-l,4-beta-glucanase og exo-l,4-beta-D-glucanase.
