Claims (10)
1. Fremgangsmåde til isolering af urinstof og fjernelse af C02 fra plasmaprøver omfattende følgende trin a) tilvejebringelse af en plasmaprøve b) tilsætning af en syre for at fjerne C02 delvist c) frysetørring af prøven for at fjerne yderligere C02 og at opnå en tørret prøve d) opløsning af den tørrede prøve igen og neutralisering til en pH-værdi på 4 til 7 med en pufferopløsning.
2. Fremgangsmåde ifølge krav 1, kendetegnet ved, at der inden tilsætningen af en syre i trin b) udføres en filtrering.
3. Fremgangsmåde ifølge krav 1 eller 2, kendetegnet ved, at prøven efter trin d) afgasses under reduceret tryk.
4. Fremgangsmåde ifølge mindst et af kravene 1 til 3, kendetegnet ved, at prøven i trin d) indstilles til en pH-værdi på 4 til 6, fortrinsvis 5 til 6.
5. Fremgangsmåde ifølge mindst et af kravene 1 til 4, kendetegnet ved, at plasmaprøven stammer fra en testperson eller patient, som har indtaget demarkerede urinstofprækursorer inden fjernelsen.
6. Fremgangsmåde til bestemmelse af 13Q-iSotopforholdet af urinstof i en plasmaprøve omfattende trinnene - isolering af urinstof ved hjælp af en fremgangsmåde ifølge et af kravene 1 til 5 - skylning med beskyttelsesgas - tilsætning af urease for at danne C02 - inkubation - tilsætning af en syre for at frigive det dannede C02 - måling af 13C-isotopforholdet af det frigivede C02.
7. Fremgangsmåde til diagnosticering af urinstofmetabolismen omfattende trinnene - tilvejebringelse af en første plasmaprøve fra en patient, - fastlæggelse af 13C-isotopforholdet af urinstof i den første plasmaprøve ifølge krav 6, - tilvejebringelse af mindst en yderligere plasmaprøve, som stammer fra patienten, hvor patienten har indtaget 13C-markerede urinstofprækursorer inden fjernelsen, - fastlæggelse af 13C-isotopforholdet af urinstof i den mindst ene yderligere plasmaprøve ifølge krav 6, - kvantificering af mængden af det dannede urinstof ved hjælp af 13C-isotop-forholdet af urinstoffet i den første og den mindst ene yderligere plasmaprøve.
8. Fremgangsmåde ifølge krav 7, kendetegnet ved, at der anvendes mindst to yderligere plasmaprøver.
9. Fremgangsmåde ifølge krav 8, kendetegnet ved, at de yderligere plasmaprøver er opnået i tidsrummet fra 15 til 240 min, efter patienten havde indtaget 13C-markerede urinstofprækursorer.
10. Fremgangsmåde ifølge et af kravene 8 eller 9, kendetegnet ved, at de yderligere plasmaprøver er opnået i intervaller af 15 min, efter at patienten havde indtaget 13C-markerede urinstofprækursorer.
