Claims (5)
1. Fremgangsmåde til fremstilling af en eller flere peptidligander, omfattende polypeptider kovalent bundet til et molekylært stillads ved to eller flere aminosyrerester, omfattende trinnene at: (a) vise en eller flere peptidligander i et genetisk displaysystem, hvor polypeptidet omfatter to eller flere reaktive grupper som danner en kovalent binding til det molekylære stillads, og mindst en sløjfe som omfatter en sekvens med to eller flere aminosyrer liggende mellem to af de reaktive grupper; (b) udsætte peptidliganderne for mere end en protease; og (c) screene peptidliganderne for binding mod et mål, og at vælge liganderne som binder til målet og er modstandsdygtige over for spaltning af den mere end ene protease.
2. Fremgangsmåde til fremstilling af en eller flere peptidligander, omfattende polypeptider kovalent bundet til et molekylært stillads ved to eller flere aminosyrerester, omfattende trinnene at: (a) vise en eller flere peptidligander i et genetisk displaysystem, hvor polypeptidet omfatter to eller flere reaktive grupper som danner en kovalent binding til det molekylære stillads, og mindst en sløjfe som omfatter en sekvens med to eller flere aminosyrer liggende mellem to af de reaktive grupper; (b) screene peptidliganderne for binding mod et mål uden en protease, og at vælge liganderne som binder til målet; (c) udsætte peptidliganderne som binder til målet for en eller flere proteaser; og (d) endvidere at screene for peptidligander som er modstandsdygtige over for protease-spaltning og binder til målet.
3. Fremgangsmåde ifølge krav 1 eller krav 2, hvor det genetiske displaysystem er fag-display.
4. Fremgangsmåde ifølge krav 1, hvor peptidliganderne som binder til målet endvidere modificeres med (a) aminosyresidekæde-modifikation eller -substitution; eller (b) polypeptidskelet-modifikation.
5. Fremgangsmåde ifølge krav 4, hvor skelet-modifikationen vælges fra gruppen bestående af at introducere, nær stedet for protease-spaltning, en D-aminosyre, en reduceret peptidbinding, og N-methylering.
