Den foreliggende opfindelse angår en syntetisk vaccine mod mund- og klovsyge som angivet i krav 1-7, en fremgangsmåde til fremstilling af vacinen som angivet i krav 8 og et farmaceutisk eller veterinærmedicinsk præparat som angivet 5 i krav 9.
Mund- og klovsygen forårsager store tab ved kvægavlen til trods for, at der allerede i lang tid har været disponible vacciner. En grund til at mund- og klovsyge forekommer i vor tid er usikkerheden ved klassiske vacciner, der 10 indeholder dræbte eller inaktiverede MKS-vira: inaktiveringen af vira er undertiden ikke fuldstændig, således at "post-vaccinale" udbrud af MKS kan forekomme (jf. Bohm, Stroh-' maier, Tierårztl. Umschau 39, 3-8 (1984). Denne fare forekommer ikke ved syntetiske MKS-vacciner, da der ved de sidst-15 nævnte kun anvendes delsekvenser af bestemte virusproteiner, der ikke har et intakt virus’ funktion.
Der eksisterer ganske vist allerede syntetiske MKS-vacciner (jf. EP patentansøgning 0.204.480), der dog trænger til en forbedring.
20 Det har nu vist sig, at man ved anvendelse af membran- ankerforbindelser og bestemte delsekvenser af MKS-virus kan fremstille særligt aktive MKS-vacciner. I DE-offentliggørel-sesskrift nr. 3.546.150 Al nævnes ganske vist fremstillingen af syntetiske vacciner som en af mange anvendelsesmuligheder 25 for membrananker-aktiv forbindelse-konjugater, man kunne dog ikke forvente, at konjugater af membranankerforbindelser og delsekvenser af MKS-virus (membrananker-aktiv forbindelse-konjugater) udviser den fundne usædvanlige aktivitet ved anvendelse af relativt ringe mængder vaccine.
30 Endvidere udmærker de nævnte vacciner sig overraskende ved, at de allerede efter en enkelt overførsel giver en tilstrækkelig beskyttelse ved vaccination. I forhold til tilgængelige vacciner har de desuden den fordel, at de uden køling har en praktisk talt ubegrænset holdbarhed.

Membranankerforbindelsen kan i princippet forekomme som R, S-, R,R-diastereomere eller som diastereomerblanding.
Det har vist sig, at de vacciner, der indeholder en R,R-diastereomer membranankerforbindelse, udviser en særlig 5 høj aktivitet.
Opfindelsen angår endvidere en fremgangsmåde til fremstilling af en syntetisk vaccine, hvilken fremgangsmåde er ejendommelig ved, at MKS-virusets delsekvenser bindes til membranankerforbindelsen ved hjælp af en konjugationsreak-10 tion. Konjugationsreaktionen kan f.eks. være en kondensation, addition, substitution, oxidation eller en disulfidbinding. Foretrukne konjugationsmetoder er anført i eksempel 1. Yderligere konjugationsmetoder er beskrevet i det allerede nævnte DE offentliggørelsesskrift 3.546.150.
15 Fremstillingen af membranankerforbindelserne er lige ledes udførligt beskrevet i det sidstnævnte DE offentliggørelsesskrift.
Den eventuelt nødvendige spaltning af diastereomerene kan ske efter forskellige metoder som det f .eks. er beskrevet 20 i Hoppe-Seyler' s 2. Physiolog. Chem. 364 (1983) 593. I eksempel 2 er en foretrukket spaltningsfremgangsmåde beskrevet.
Opbygningen af del sekvenserne af de hver gang forekommende MKS-proteiner kan udføres på forskellige i litteraturen kendte måder, jf. f.eks. Wunsch et al. i Houben-25 Weyl, bd. 15/1,2, Stuttgart, Thieme-Verlag eller Wunsch i Angew. Chem. 83/ (1971), 773, E. Gross og J. Meienhofer (udgiver), The Peptides, bind 1 (1979), 2 (1979), 3 (1981) og 5 (1983) Academic Press, New York eller DE offentliggørelsesskrift 3.546.150. I eksempel 3 illustreres en foretrukket 30 fremgangsmåde til fremstilling af en delsekvens og et kon-jugat nærmere.
Endvidere angår opfindelsen farmaceutiske eller veterinærmedicinske præparater, der har et indhold af konjugat af membranankerforbindelse og delsekvens af MKS-virus. Nor-35 malt er det ikke nødvendigt foruden et opløsningsmiddel at tilsætte yderligere hjælpe- og bærestoffer eller adjuvanser

Efter tilsætning af 5 ml EDC (1-(3-dimethyl-aminopropyl)-3-ethyl-carbodiimid-hydrochlorid), der er opløst i 100 μΐ H20, omrøres der i 18 timer ved 5 stuetemperatur, hvorefter der dialyseres to gange mod hver gang 1 liter destilleret H20. Indholdet i dialyseslangen lyofiliseres.
Eksempel 2 10
Spaltning af N-Palmitoyl-S-[2>3-(bispalmitoyloxy)propyl)-cystein-tert.butylester (PamjCys-OBut)'s diastereomere 2 g Pan^Cys-OBu1- opløses i 10 ml. elueringsmiddel, 15 dichlormethan/ethylacetat (20:1), hvorefter opløsningen sættes på en søjle (længde 120 cm, diameter 4 cm), der er fyldt MN-kiselgel 60, 0,063-0,2 mm/70-230 mesh ASTM. Ved en dråbehastighed på 2 dråber pr. sekund opsamles 350 fraktioner af 10 ml, og en alikvot af hver fraktion undersøges efter 20 chromatografi på kiselgel 60-plader i dichlormethan/ethylacetat (20:1) og farvning med chlor/TDM-reagens for Pan^Cys-OBut.
Fraktionerne 280-315 indeholder R,R-diastereomeren, fraktionerne 316-335 indeholder en blanding af R,R og R,S 25 og fraktionerne 336-354 indeholder R,S-diastereomeren af Pam3Cys-OBut. Efter inddampning af opløsningsmidlet på en rotationsfordamper optages remanensen i varm tert.butanol, og der lyof iliseres, hvorved der fås 600 mg R,R-, 370 mg ' blanding af R,R- og R,S- og 540 mg R,S-PamjCys-OBu^.

Eksempel 4
Aktivicetsanalyse 5 Tilfældigt udvalgte marsvin med en vægt på 450 til 500 g vaccineres intramuskulært eller subcutant. 0,5 mg af det lyofiliserede vaccine N-palmitoyl-S-[(2R,R)-2,3-(bispal-mitoyloxy) -propyl] -cysteinyl-seryl-seryl- (135-154) emulgeres i 500 μΐ af en l:l-blanding af 0,05 M phosphatpuffer 10 og "Intralipid" (Kabi Vitrum, Sverige). Blandingen behandles med ultralyd i 10 sekunder. Fire dyr inficeres med MKS-virus, idet' de injiceres subcutant med mindst 500 marsvinenheder af et virulent O^K FMD-virus i deres venstre bagpote 21 dage efter vaccinationen. Som kontrol foretrækkes dyr, der 15 i stedet for vaccinen er blevet injiceret med membrananker-forbindelsen eller phosphatpuffer. Ved alle vaccinerede dyr fandt man en højere titer for neutraliserende antistoffer log10SN50 på 0,36. Kontroldyrene har ingen antistof-titer (blindværdi 0,17) . Titeren for de neutraliserende antistoffer 2 0 bestemmes som logaritmen af den serumfortynding, der er nødvendig, for at neutralisere 50% af de virusceller, der er indlejret i lag BHK-(Baby Hamster Kidney)-celler. Ved de vaccinerede dyr kunne ved hjælp af anti-peptid-ELISA-analyser (A492) påvises antistoffer, hvilket ikke er muligt ved de 25 ikke-vaccinerede dyr. Vaccinerede dyr har ingen sekundære læsioner, medens alle ikke-vaccinerede dyr viser et fuldt-. stændigt billede på en mund- og klovsygeinfektion.
