Opfindelsen angår hidtil ukendte aromatisk substituerede azacycloalkyl-l-hydroxy-alkan-1, 1-diphosphonsyrer, som er ejendommelige ved, at de har formlen P03H2 5 R' - R" - alk - C - OH (I) P03H2 hvori alk er en bivalent aliphatisk gruppe, R" betyder pyrrolidino, piperidino, 3~aza~bicyclo[ 3. li/5.il,5 ]-hept-3-yl, 1,2,5,6-tetrahydropyridino, piperazino, hexahydroazepino, 3-aza-bicyclo[ 3.2.01/5 ]hept-3-yl eller 10 octahydroazocino, og Rf betyder en usubstitueret eller med lavalkyl, lavalkoxy og/eller halogen mono- eller disubstitueret phenyl-, naphthyl-, pyridyl-, thienyl-, pyrimidinyl-, quinolinyl-, furyl- eller pyrrylgruppe, og deres salte, en fremgangsmåde til fremstilling deraf samt 15 farmaceutiske præparater indeholdende disse forbindelser.
Gruppen R" er forbundet med alk-gruppen over et nitrogenatom.
Grupperne Rf og R" kan tilsammen eksempelvis betyde 3-phenyl- eller 3-(p-chlorphenyl)-pyrrolidino, 4-phenyl-, 20 4-(p-methoxyphenyl)-, 4-(p-methylphenyl)-, 4-(p-chlor phenyl)-, 4-(p-fluorphenyl)- eller 4-(m-fluorphenyl)-piperidino, 3-phenyl-3,5-methylen-piperidino, dvs. 1-phen-yl-3-aza-bicyclo[ 3.ll/5.il,5 ]hept-3-yl, 4-(p-chlorphenyl)- 3,5-methylen-piperidino, dvs. 6-(p-chlorphenyl)-3-aza-25 bicyclo[ 3.1^/5.1^^5 ]hept-3-yl, såsom exo- eller først og fremmest endo-4-(p-chlorphenyl)-3,5-methylen-piperidino, dvs. exo- eller først og fremmest endo-6-(p-chlorphenyl)- 3-aza-bicyclo[3.1^-/5.11,5 ]hept-3-yl, endvidere 1-phenyl-eller 1-(p-chlorphenyl)-3-aza-bicyclo[ 3.1^/5.11,5 Jtiept- 30 3-yl, 4-phenylpiperazino, 1-phenyl- eller 1-(p-chlorphen yl )-3-aza-bicyclo[3.2.ol/5 ]hept-3-yl og 4-phenyl-octa-hydroazocino.

Bivalente aliphatiske alk-grupper er især bivalente, lavere aliphatiske carbonhydrid-grupper, såsom lavalkylen.
I det følgende skal der ved lavere grupper og forbindelser 5 eksempelvis forstås sådannne, der har højst 7, især højst 4, C-atomer. Endvidere har de almene begreber eksempelvis følgende betydning:
Lavalkyl er eksempelvis C(l-4)-alkyl, såsom methyl, ethyl, propyl, isopropyl eller butyl, endvidere iso-, sekundær-10 eller tertiær-butyl, men kan også være en C(5-7)-alkyl, såsom pentyl-, hexyl- eller heptylgruppe.
Lavalkoxy er eksempelvis C(l-4)-alkoxy, såsom methoxy, ethoxy, propyloxy, isopropyloxy eller butyloxy, endvidere iso-, sekundær- eller tertiær-butyloxy.
15 Halogen er eksempelvis halogen med atomnummer på højst 35, såsom chlor, samt fluor eller brom.
Alk-lavalkylen er eksempelvis C(2-6)-alkylen, først og fremmest ligekædet C(2-5)-alkylen, såsom ethylen, 1,3-propylen, 1,4-butylen eller 1,5-pentylen, endvidere 20 methylen, 1,2-propylen, 1,2- eller 1,3-butylen eller 1,4-pentylen, men kan også være en hexylen- eller hept-ylengruppe. Lavalkylen som forbindelsesled til R" er eksempelvis methylen eller i anden række ethylen.
Salte af forbindelser med formlen I er især deres salte 25 med farmaceutisk anvendelige baser, såsom ikke-toksiske, af metaller eller fra grupperne la, Ib, Ila og Ilb afledte metalsalte, f.eks. alkalimetal-, især natrium- eller kaliumsalte, jordalkalimetal-, især calcium- eller magnesiumsalte, kobber, aluminium- eller zinksalte, samt 30 ammoniumsalte med ammoniak eller organiske aminer eller kvaternære ammoniumbaser, såsom eventuelt C-hydroxylerede aliphatiske aminer, især mono-, di- eller trialkylaminer, DK 168630 B1 3 f.eks. methyl-, ethyl-, dimethyl- eller diethylamin, mono-, di- eller tri-(hydroxylavalkyl)-aminer, såsom ethanol-, diethanol- eller triethanolamin, tris(hydroxy-5 methyl )-amino-methan eller 2-hydroxytertiærbutylamin, eller N-(hydroxylavalkyl)-N, N-dilavalkylaminer eller N-(polyhydroxylavalkyl)-N-lavalkylaminer, såsom 2-(di-methylamino)-ethanol eller D-glucamin, eller kvaternære aliphatiske ammoniumhydroxider, f.eks. med tetrabutyl-10 ammoniumhydroxid.
Fra SU patentskrift nr. 1.002.300 kendes piperidino- og morpholino-l-hydroxy-propan-1,1-disphosphonsyrer med regulerende virkning på calcium-stofskiftet. Piperidino- og morpholinogruppen i disse kendte forbindelser er usubsti-15 tuerede, hvorimod de tilsvarende grupper R" i forbindelserne ifølge opfindelsen er substituerede med substituen-ten R' som defineret ovenfor.
Det har overraskende vist sig, at forbindelser ifølge opfindelsen har en endog meget kraftigere regulerende 20 virkning på calcium-stofskiftet end de kendte forbindelser, hvilket illustreres nærmere i det følgende.
Forbindelserne bevirker i rotter en udpræget hæmning af knogleresorptionen, som viser sig både i forsøget ifølge Acta Endocrinol 78, 613-24 (1975) på grund af den 25 PTH-inducerede stigning af serumcalciumspejlet efter subkutan applikation i doser på fra ca. 0,01 til ca. 1,0 mg/kg, og i TPTX (thyroparathyroidectomised)-forsøget med rotter på grund af ved vitamin D3 udløst eksperimentel hypercalcæmi efter indgift af doser på fra ca. 0,001 til 30 ca. 1,0 mg s.c. Ved anvendelse af denne forsøgsmodel opnås eksempelvis med 3-[3-(4-chlorphenyl)-pyrrolidinyl ]- 1-hydroxy-propan-l,1-diphosphonsyre, fremstillet i det efterfølgende eksempel 4, en EDsQ-værdi på 0,003 mg/kg p.o., hvorimod der med den fra ovennævnte SU patentskrift 35 nr. 1.002.300 kendte forbindelse 3-(piperidin-l-yl)- ri DK 168630 B1 4 1-hydroxy-propan-l,1-diphosphonsyre opnås en EDsg-værdi på 0,009 mg/kg p.o.
Ligeledes hæmmes den med Walker-256-tumorer inducerede 5 tumorhypercalcæmi efter peroral indgift af fra ca. 1,0 til ca. 100 mg/kg. Endvidere har de ved adjuvansarthritis hos rotter i forsøget ifølge Newbould, Brit. J. Pharmacology 21, 127 (1963) samt ifølge Kaibara et al., J. Exp. Med. 159, 1388-96 (1984) i doser på fra ca. 0,01 til ca. 1,0 10 mg/kg s. c. en tydelig hæmning på udviklingen af kronisk-arthritiske processer.
Forbindelserne ifølge opfindelse kan derfor med fordel anvendes som aktive lægemiddelstoffer til behandling af sygdomme, som kan sættes i forbindelse med forstyrrelse af 15 calciumstofskiftet, eksempelvis betændelsesprocesser i leddene, degenerative processer i ledbrusk, osteoporosis, periodontitis, hyperparathyreoidismus og calciumaflejringer i blodkar eller på profetiske implantater. Både sygdomme, hvorved der kan konstateres en anormal 20 aflejring af tungtopløselige calciumsalte, påvirkes gunstigt, såsom sådanne typer af arthritis, f.eks. morbus Bechterew, neuritis, bursitis, periodontitis og tendinitis, fibrodysplasi, osteoarthrosis eller artereosklerose, samt sådanne, hvorved der fremtræder en 25 anormal opløsning af hårdt kropsvæv, såsom arvelig hypophosphatasia, degenerative processer i ledbrusk, osteoporosis af forskellige slags, Pagets sygdom og osteodystrophia fibrosa, samt osteolytiske processer udløst ved tumorer.
30 Opfindelsen angår fortrinsvis forbindelser med formlen I, hvori R" er en 4-R'-piperidinogruppe eller 4-R' -3,5-meth-ylen-piperidino-, dvs. 6-R'-3-aza-bicyclo[ 3.1-^^.11,5 hept-3-yl-gruppe, såsom 4-endo-R'-3,5-methylen- piperidino, dvs. 6-endo-R'-3-aza-bicyclo[ 3.1-^5.li,5 ]- 35 hept-3-yl, hvor R' er usubstitueret eller med C(l-4)- DK 168630 Bl 5 alkyl, C(1-4)-alkoxy eller halogen med atomnummer på højst 35 substitueret phenyl, og alk er ligekædet C(2-5)-alkylen med formlen -(CH2)n-, hvori n betyder et helt tal på fra 2 5 til og med 5, og deres salte, især farmaceutisk anvendelige salte.
Opfindelsen angår i allerførste række forbindelser med formlen I, hvori R” er en 3-R’-pyrrolidino- eller 3- eller 4-Rr-piperidinogruppe, hvor R' er usubstitueret eller med 10 C(l-4)-alkyl, C(l-4)-alkoxy eller halogen med atomnummer på højst 35 substitueret phenyl, og alk er ligekædet C(2-5)-alkylen med formlen -(CH2)n-, hvori n er et helt tal på fra 2 til og med 5, og deres salte, især farmaceutisk anvendelige salte.
15 Opfindelsen angår især nogle af de i eksemplerne nævnte forbindelser med formlen I og deres salte.
Fremgangsmåden ifølge opfindelsen er baseret på i og for sig kendte metoder og er ejendommelig ved, at man a) i en forbindelse med formlen 20 R' - R" - alk - ί - OH · *2 hvori Χχ er en funktionelt omdannet phosphonogrupe, X2 er en fri eller funktionelt omdannet phosphonogruppe, og alk, R' og R" har de ovenfor angivne betydninger, omdanner Χχ og eventuelt X2 til den frie phosphonogruppe eller 25 b) omsætter en forbindelse med formlen PO3H2 H2N - alk - C - OH (III) FO3H2 i DK 168630 B1 6 hvori alk har den ovenfor angivne betydning, eller et salt deraf med en reaktiv ester af en passende aromatisk substitueret aliphatisk dialkohol eller 5 c) omsætter en forbindelse med formlen R' - R" - alk - X3 (IV) hvori X3 er carboxy, carbamoyl eller cyano, og alk, R' og R" har de ovenfor angivne betydninger, med phosphor-syrling og phosphortrichlorid, hydrolyserer primærproduk-10 tet, og i et ud fra forbindelserne med fomlen IV, hvori X3 er cyano eller carbamoyl, dannet mellemprodukt med formlen POjHs R’ - R" - alk - C - KH2 (V) W3H2 hvori alk, R' og R" har de ovenfor angivne betydninger, 15 eller et salt deraf ombytter aminogruppen med hydroxy ved behandling med salpetersyrling; og om nødvendigt omdanner en dannet forbindelse til en anden forbindelse med formlen I og/eller en dannet fri forbindelse til et salt eller et dannet salt til den frie forbindelse eller til et andet 20 salt.
Ifølge fremgangsmåde a) foreligger funktionelt omdannede phosphonogrupper, der skal omdannes til phosphono, eksempelvis i en esterform, især en diesterform med formlen -P(=0)(0R)2 (Ha), hvori OR eksempelvis er 25 lavalkoxy eller en eventuelt med lavalkyl, lavalkoxy, halogen, trifluormethyl og/eller hydroxy substitueret phenyloxygruppe.

Udgangsmaterialerne med formlen Uh, hvori R" er en med R' C-substitueret monocyclisk azacycloalk(en)ylgruppe, kan f.eks. fremstilles ved, at man kondenserer en tilsvarende ved N-atomet intermediært beskyttet med en oxogruppe 15 substitueret azacycloalkan med en forbindelse med formlen R'-M (Hk), hvori M er en metallisk gruppe, f.eks. lithium eller en halogenmagnesiumgruppe, fraspalter vand fra den dannede geminal med R' og hydroxy substituerede. N-beskyttede azacycloalkan ved syrebehandling, fjerner amino-20 beskyttelsesgruppen og, om ønsket, reducerer dobbeltbindingen til enkeltbinding.
Udgangsmaterialerne med formlen Uh, hvori R" er en med R' N-substitueret diazacycloalkylgruppe, kan f.eks. fremstilles ved, at man kondenserer en tilsvarende α,ω-ΰϊ-25 halogenazaalkan med en amin med formlen R'-NH2 (III).
Udgangsmaterialerne med formlen Uh, hvori R" er en med R' substitueret 3-aza-bicyclo[ 3,1,1 ]hept-3-ylgruppe, fås f.eks. ved, at man cycliserer en tilsvarende ved N-atomet intermediært beskyttet 2-R'-3,5-dioxo-4-aza-hepta- 30 1,6-dien eller 6-R'-2-Br-3,5-dioxo-4-aza-hepta-l,6-dien under sure betingelser, fra det dannede R'-2,4-dioxo- DK 168630 B1 9 3-aza-bicyclo[ 3,1,1 ]heptan-derivat afspalter N-beskyt telsesgruppen, og erstatter oxogruppen samt det eventuelle bromatom reduktivt med hydrogen.
5 Som aminbeskyttelsesgrupper anvendes f.eks. a-aryllav-alkyl, såsom benzyl eller p-methoxybenzyl, a, or, a-triaryl-lavalkyl, såsom trityl, eller trilavalkyl-, såsom tri-methylsilyl. cc-Aryl- og a, a, α-triaryllavalkyl kan nemt fjernes igen hydrogenolytisk, trilavalkylsilyl og 10 or, a, of-triaryllavalkyl hydrolytisk.
Forbindelser med formlen Uh, hvori R" er en eventuelt i 2-, 3- og/eller 4-stilling lavalkyleret 3-R'-pyrrolidino-gruppe, kan endvidere fremstilles særdeles elegant, idet man omsætter en tilsvarende 3-R*-lavalk-2-en-carboxylsyre-15 ester med en nitrolavalkan, hydrogenerer nitrogruppen til aminogruppen i den dannede 3-R'-3-(1-nitrolavalkyl)-lavalkancarboxylsyreester, hvorved der finder en ringslutning sted til den tilsvarende 3-R'-pyrrolidin-5-on, og erstatter oxogruppen reduktivt med hydrogen, f.eks. ved 20 behandling med lithiumaluminiumhydrid eller diboran.
Reaktive mono- eller diestere af aromatisk substituerede aliphatiske dialkoholer, der skal anvendes ifølge fremgangsmåde b), er eksempelvis hydrogenhalogenidsyre-estere, såsom hydrogenchloridsyreester, hydrogenbromid-25 syreester eller hydrogeniodidsyreester, sulfonsyrestere, såsom alkan- eller eventuelt substituerede benzensulfon-syreestere, såsom methan- eller p-toluensulfonater, eller svovlsyreestere.
Omsætningen med de nævnte reaktive diestere af aromatisk 30 substituerede aliphatiske dialkoholer gennemføres eksempelvis i nærværelse af en base, såsom et alkalimetal-eller jordalkalimetalhydroxid, f.eks. af natriumhydroxid, eller et kvaternært ammoniumhydroxid, f.eks. af tetra-butyl ammoniumhydroxid, med fordel i nærværelse af et DK 168630 B1 10 opløsnings- eller fortyndingsmiddel.
Udgangsmaterialerne med formlen III kan eksempelvis fremstilles ved, at man omsætter en forbindelse med formlen 5 H2N - alk - C00H (Illa) på sædvanlig måde, eksempelvis i chlorbenzen, med phosphorsyrling og phosphortrichlorid og dernæst foretage hydrolytisk oparbejdning.
I en variant af fremgangsmåde b) kan man også kondensere 10 en forbindelse med formlen III med en reaktiv mono- eller diester af en med R ’ aromatisk substitueret aliphatisk alkohol og cyclisere primærproduktet med formlen ?°3H2 (Q)
Xu-NH-alk-C-OH
F03H2 hvori X4 er med eventuelt reaktionsdygtigt forestret 15 hydroxy substitueret R’-aliphatyl.
Omsætningen af forbindelser med formlen IV med phosphorsyrling og phosphortrichlorid ifølge fremgangsmåde c) gennemføres på kendt måde, idet phosphorsyrlingkomponen-ten fortrinsvis dannes in situ ved reaktion af over-20 skydende phosphortrichlorid med vandig phosphorsyre, f.eks. med gængs fra ca. 75%'s til ca. 95%'s, fortrinsvis ca. 85%'s phosphorsyre. Omsætningen gennemføres med fordel under opvarmning, f.eks. ved fra ca. 70° til ca. 120°C, i et egnet opløsningsmiddel, såsom tetrachlorethan, 25 trichlorethan, chlorbenzen, chlortoluen eller paraffinolie, under hydrolytisk oparbejdning.
Behandlingen af mellemprodukter med formlen V med salpetersyrling gennemføres på kendt måde under frigørelse DK 168630 B1 11 af denne i vandig opløsning af et af dens salte, f.eks. af natriumnitrit, ved syrebehandling, f.eks. indvirkning af saltsyre, hvorved der intermediært dannes et tilsvarende, 5 instabilt diazoniumsalt, f.eks. -chlorid, som under indførelse af α-hydroxygruppen fraspalter nitrogen.
Når udgangsmaterialerne med formlen IV ikke er kendte, kan de eksempelvis fremstilles ved, at man omsætter en tilsvarende forbindelse med formlen 10 R' - R" - H (Uh) med en forbindelse med formlen Y - alk - X3 (Ili) hvori Y er halogen, såsom brom, eller til fremstilling af forbindelser med formlen IV, hvori alk er C(2-7)-alkylen, 15 hvis frie valenser går ud fra nabostillede C-atomer, f.eks. ethylen, med en forbindelse med formlen alk' - X3 (Hf) hvori alk' er en C(2-7)-alkenylengruppe, og, om ønsket, hydrolyserer det dannede primærprodukt til syre.
20 Forbindelser med formlen I, som fås ifølge fremgangsmåden, kan omdannes til andre forbindelser med formlen I på i og for sig kendt måde.
Således kan man substituere gruppen R', eksempelvis indføre halogen ved omsætning med et gængs kerne-25 halogeneringsmiddel, f.eks. med chlor eller brom i nærværelse af en Lewissyre, såsom jern-III-trichlorid.
De nye forbindelser kan alt efter anvendelse af udgangs- DK 168630 B1 12 materialer og arbejdsmåde foreligge i form af et af de mulige isomerer eller som blanding deraf, f.eks. ifølge antallet af asymmetriske carbonatomer som rene optiske 5 isomerer, såsom antipoder eller som isomerblandinger, såsom racemater, diastereoisomerblandinger eller racematblandinger.
Dannede diastereomerblåndinger og racematblandinger kan på grund af bestanddelenes fysisk-kemiske forskelle opdeles 10 på kendt måde i de rene isomerer, diastereomerer eller racemater, eksempelvis ved kromatografi og/eller fraktioneret krystallisation.
Dannede racemater kan endvidere adskilles på kendt måde i de optiske antipoder, eksempelvis ved omkrystallisation af 15 et optisk aktivt opløsningsmiddel, ved hjælp af mikroorganismer, eller ved omsætning af et surt slutprodukt med en optisk aktiv base, der danner salte med den racemiske syre, og deling af de på kendt måde dannede salte, f.eks. på grund af deres forskellige opløselighedsgrader, i 20 diastereomerer, hvoraf antipoderne kan frigøres ved indvirkning af egnede midler. Man isolerer med fordel den mest virksomme af de to antipoder.
Dannede frie forbindelser med formlen I inklusive deres indre salte med formlen I kan omdannes ved partiel eller 25 fuldstændig neutralisation med en af de indledningsvis nævnte baser til basesalte. På tilsvarende måde kan man også omdanne syreadditionssalte til de tilsvarende frie forbindelser eller deres indre salte.
Omvendt kan man omdanne dannede frie forbindelser med 30 formlen I ved behandling med en af de indledningsvis nævnte protonsyrer til syreadditionssalte.
Dannede salte kan på i og for sig kendt måde omdannes til de frie forbindelser, f.eks. ved behandling med et surt DK 168630 B1 13 reagens, såsom en mineralsyre, eller en base, f.eks. en alkalimetalhydroxidopløsning.
Forbindelserne inklusive deres salte kan også fås i form 5 af deres hydrater eller omfatte det til krystallisation anvendte opløsningsmiddel.
Opfindelsen angår også de udførelsesformer af fremgangsmåden, ifølge hvilke man anvender et udgangsmateriale i form af et salt og/eller racemat eller antipoder eller 10 især danner dette under reaktionsbetingelserne.
Ved fremgangsmåden i den foreliggende opfindelse anvendes fortrinsvis sådanne udgangsmaterialer, som fører til de indledningsvis beskrevne særdeles værdifulde forbindelser.
15 De farmaceutiske præparater ifølge opfindelsen er ejendommelige ved, at de indeholder en forbindelse med formlen I eller et salt deraf samt gængse farmaceutiske hjælpestoffer. De kan anvendes til enteral, såsom oral eller rektal, og parenteral indgift.
20 De farmaceutiske præparater indeholder f.eks. fra ca. 10 til ca. 80%, fortrinsvis fra ca. 20 til ca. 60% af det aktive stof. De farmaceutiske præparater ifølge opfindelsen til enteral eller parenteral indgift er f.eks. sådanne i dosisenhedsformer, såsom dragéer, tabletter, 25 kapsler eler suppositorier samt ampuller. Disse fremstilles på i og for sig kendt måde, f.eks. ved hjælp af konventionelle blandings-, granulerings-, dragérings-, opløsnings- eller lyofilisationsfremgangsmåder. Således kan man fremstille farmaceutiske præparater til oral 30 anvendelse ved, at man kombinerer det aktive stof med å DK 168630 B1 14 faste bærestoffer, eventuelt granulerer en dannet blanding og komprimerer blandingen eller granulatet, om ønsket eller nødvendigt, efter tilsætning af egnede hjælpe-5 stoffer, til tabletter eller dragékerner.
Egnede bærestoffer er især fyldstoffer, såsom sukker, f.eks. lactose, saccharose, mannitol eller sorbitol, cellulosepræparater og/eller calciumphosphater, f.eks. tricalciumphosphat eller calciumhydrogenphosphat, samt 10 bindemidler, såsom stivelsesklister under anvendelse af f.eks. majs-, hvede-, ris- eller kartoffelstivelse, gelatine, tragant, methylcellulose og/eller polyvinyl-pyrrolidon, og/eller, om ønsket, disintegreringsmidler, såsom de ovennævnte stivelser, samt carboxymethyl-15 stivelse, tværbundet polyvinylpyrrolidon, agar, alginsyre eller et salt deraf, såsom natriumalginat. Hjælpemidler er i første række strømningsregulerende midler og smøremidler, f.eks. kiselsyre, talkum, stearinsyre eller salte deraf, såsom magnesium- eller calciumstearat, og/eller 20 polyethylenglycol. Dragékerner forsynes med egnede, eventuelt mavesaftresistente overtræk, idet man bl.a. anvender koncentrerede sukkeropløsninger, som eventuelt indeholder arabisk gummi, talkum, polyvinylpyrrolidon, polyethylenglycol og/eller titandioxid, lakopløsninger i 25 egnede organiske opløsningsmidler eller opløsningsmiddel-blandinger eller til fremstilling af mavesaftresistente overtræk, opløsninger af egnede cellulosepræparater, såsom acetylcellulosephthalat eller hydroxypropylmethyl-cellulosephthalat. Til tabletterne eller dragéovertræk-30 kene sættes farvestoffer eller pigmenter, f.eks. til identifikation eller til karakterisering af forskellige doser aktivt stof.
Yderligere oralt anvendelige farmaceutiske præparater er gelatinestikkapsler samt bløde, lukkede kapsler af gela- DK 168630 B1 15 tine og et blødgøringsmiddel, såsom glycerin eller sorbitol. Stikkapslerne kan indeholde det aktive stof i form af et granulat, f.eks. i blanding med fyldstoffer, 5 såsom lactose, bindemidler, såsom stivelser, og/eller glittemidler, såsom talkum eller magnesiumstearat, og eventuelt stabilisatorer. I bløde kapsler er det aktive stof fortrinsvis opløst eller suspenderet i egnede væsker, såsom fede olier, paraffinolie eller flydende polyethylen-10 glycoler, hvorved der ligeledes kan være tilsat stabilisatorer.
Som rektalt anvendelige farmaceutiske præparater anvendes f.eks. suppositorier, som består af en kombination af det aktive stof med en suppositoriegrundmasse. Som 15 suppositoriegrundmasse anvendes f.eks. naturlige eller syntetiske triglycerider, paraffincarbonhydrider, poly-ethylenglycoler eller højere alkanoler. Endvidere kan der også anvendes gelatine-rektalkapsler, som indeholder en kombination af det aktive stof med en grundmasse. Som 20 grundmassematerialer anvendes f.eks. flydende triglycerider, polyethylenglycoler eller paraffincarbonhydrider.
Til parenteral indgift anvendes i første række vandige opløsninger af et aktivt stof i vandopløselig form, f.eks. et vandopløseligt salt, samt suspensioner af det aktive 25 stof, såsom tilsvarende olieagtige injektionssuspens ioner, idet man anvender egnede lipofile opløsningsmidler eller hjælpestoffer, såsom fede olier, f.eks. sesamolie, eller syntetiske fedtsyreestere, f.eks. ethyloleat eller triglycerider, eller vandige injektionssuspensioner, som 30 indeholder viskosistetsforøgende stoffer, f.eks. natrium-carboxymethylcellulose, sorbitol og/eller dextran og eventuelt også stabilisatorer.
Doseringer under 0,01 mg/kg kropsvægt påvirker kun i ringe grad den patologiske forkalkning eller opløsningen af DK 168630 B1 16 hårde væv. Ved doseringer på over 100 mg/kg kropsvægt kan der optræde langvarige toksiske bivirkninger. Forbindelserne med formlen I og deres salte kan indgives både oralt 5 og i hypertonisk opløsning, subkutant, intramuskulært eller intravenøst. De foretrukne dagsdoser til disse anvendelser ligger ved oral indgift på fra ca. 0,1 til ca.
5 mg/kg, ved subkutan og intramuskulær indgift på fra ca.
0,1 til ca. 1 mg/kg og ved intravenøs indgift på fra ca.
10 0,01 til ca. 2 mg/kg, eksempelvis på fra ca. 0,013 til ca.
0,67 mg/kg.
Doseringen af de anvendte forbindelser er dog variabel og afhænger af de tilstedeværende betingelser, såsom sygdommens art og sværhedsgrad, behandlingens varighed og 15 den anvendte forbindelse. Enkeltdoser indeholder eksempelvis fra 0,01 til 10 mg, dosisenhedsformer til parenteral, såsom intravenøs, applikation f.eks. fra 0,01 til 0,1 mg, fortrinsvis fra 0,02 til 0,08 mg, orale dosisenhedsformer f.eks. fra 0,2 til 2,5 mg, fortrinsvis fra 0,3 til 1,5 mg 20 pr. kg kropsvægt. Den foretrukne enkeltdosering indeholder ved oral indgift fra 10 til 100 mg og ved intravenøs applikation fra 0,5 til 5 mg og kan indgives op til fire gange dagligt. De højere doseringer ved oral indgift er nødvendige som følge af den begrænsede resorption. Ved 25 længerevarende behandlinger kan der efter indledningsvis højere dosering normalt omstilles til lavere doseringer for at opretholde den ønskede virkning.
Opfindelsen illustreres nærmere ved hjælp af de følgende eksempler.
30 Eksempel 1 26,98 g (0,1 mol) 3-(4-phenylpiperidino)propionsyre-hydrochlorid opvarmes til 100°C med 13,4 ml 85%'s phosphorsyre og 50 ml chlorbenzen under omrøring og tilbagesvaling. Dernæst tilsættes dråbevis ved 100°C 27 ml DK 168630 B1 17 phosphortrichlorid, hvorved der udvikles gas. Reaktionsblandingen udskiller i løbet af 30 minutter en tyk masse.
Man opvarmer endnu 3 timer til 100°C og fradekanterer 5 dernæst den ovenstående chlorbenzen. Den tilbageblevne seje masse opvarmes til kogepunktet med 100 ml 9 N saltsyre i 3 timer under omrøring og tilbagesvaling. Man filtrerer varmt under tilsætning af kul og fortynder filtratet med acetone, hvorved 3-(4-phenylpiperidino)-10 1-hydroxy-propan-l,1-diphosphonsyre udskilles krystallinsk, smp. 243-245°C (sønderdeling) (udbytte 57% af det teoretiske).
Det som udgangsmateriale anvendte 3-(4-phenylpiperidino)-propionsyre-hydrochlorid kan fremstilles på følgende måde: 15 25,0 g 4-phenylpiperidin (0,15 mol) blandes først i 50 ml diethylether og dertil sættes under omrøring 15,1 g acryl-syreethylester. Der dannes en klar opløsning under let temperaturstigning. Efter henstand natten over ved stuetemperatur afdestilleres etheren. Den tilbageblevne olie 20 er rå 3-(4-phenylpiperidino)-propionsyreethylester (udbytte ca. 95%).
39,4 g 3-(4-phenylpiperidino)-propionsyreethylester opvarmes med 600 ml 4 N saltsyre i 24 timer til tilbagesvaling. Der inddampes under formindsket tryk, og 25 den krystallinske remanens udrives med acetone. Efter frafiltrering, vask og tørring af krystallerne får man 3-(4-phenylpiperidino)-propionsyre-hydrochlorid, smp.
216-217°C. (Udbytte 90% af det teoretiske).
Eksempel 2 30 På tilsvarende måde som beskrevet i eksempel 1 kan man ud fra 0,1 mol af 3-(3-phenylpyrrolidino)propionsyre-hydrochlorid, 3-[ 4-(p-methoxyphenyl)piperidino ]propionsyre-hydrochlorid, DK 168630 B1 18 3-[4-(p-chlorphenyl)piperidino ]propionsyre-hydrochlorid, 3- [4-(p-fluorphenyl)piperidino ]propionsyre-hydrochlorid, 4- (4-phenylpiperidino)smørsyre-hydrochlorid og 5 6-(4-phenylpiperidino)hexansyre-hydrochlorid også fremstille 3-(3-phenylpyrrolidino)-1-hydroxy-propan-1,1-diphosphon-syre, smp. 221°C (sønderdeling), 3-[4-(p-methoxyphenyl)piperidino ]-l-hydroxy-propan-l,1-10 diphosphonsyre, smp. 243-245°C (sønderdeling), 3-[4-(p-chlorphenyl)piperidino ]-1-hydroxy-propan-1,1-diphosphonsyre , 3- [4-(p-fluorphenyl)piperidino ]-l-hydroxy-propan-l,1-di-phosphonsyre, 15 4- (4-phenylpiperidino) -1-hydroxy-butan-l, 1-diphosphonsyre, smp. 230°C (sønderdeling) og 5- (4-phenylpiperidino)-1-hydroxy-hexan-l,1-diphosphonsyre, smp. 236-237°C (sønderdeling) samt deres salte, f.eks. dinatriumsalte.
20 Det som udgangsmateriale anvendte 4-(4-phenylpiperidino)-smørsyre-hydrochlorid og 6-(4-phenylpiperidino)hexan-carboxylsyre-hydrochlorid kan fremstilles på følgende måde: 1,61 g (0,01 mol) 4-phenylpiperidin, 2,76 g kaliumcarbonat 25 og 2,15 g bromsmør syr eethyl ester opvarmes i 20 ml 2-butanon under omrøring og tilbagesvaling i 24 timer. Dernæst frafiltreres de uorganiske salte, og filtratet inddampes. Den tilbageblevne, rå 4-(4-phenylpiperidino)-smørsyreethylester giver efter 24 timers kogning med 30 40 ml 4 N saltsyre og efterfølgende inddampning, digerering med acetone og frafiltrering af krystallerne 4- (4-phenylpiperidino)smørsyre-hydrochlorid, smp.
217-220°C (sønderdeling).
På tilsvarende måde får man ved anvendelse af 6-brom-35 hexancarboxylsyreethylester 6-(4-phenylpiperidino)hexan- DK 168630 B1 19 carboxylsyre-hydrochlorid, smp. 197-198°C.
På tilsvarende måde får man også ud fra 3- phenylpyrrolidin, 5 4-(4-methoxyphenyl)piperidin, 4- (4-chlorphenyl)piperidin eller 4-(4-fluorphenyl)piperidin, ved omsætning med acrylsyreester og efterfølgende hydrolyse med saltsyre 10 3-(3-phenylpyrrolidino)propionsyre-hydrochlorid, smp.
139-140°C, 3-[ 4-(4-methoxyphenyl)piperidino]-propionsyre-hydrochlorid, smp. 214°C (sønderdeling), 3-[4-(4-chlorphenyl)piperidino]-propionsyre-hydrochlorid, 15 og 3- [ 4-(4-fluorphenyl)piperidino]-propionsyre-hydrochlorid.

25 Endo-6-(p-chlorphenyl)-3-aza-bicyclo[ 3.1-^-/5.11,5 jheptan- (hydrochlorid), der skal anvendes som udgangsmateriale, kan fremstilles på følgende måde: a) Til en opløsning af 72 g endo-l-brom-6-(p-chlorphenyl )- 3-(p-methoxybenzyl)-3-aza-bicyclo[ 3.1^'5.il,5 ]heptan-2,4-30 dion og 1,65 g bisazoisobutyronitril i 1,66 1 tetrahydro-furan sættes under nitrogenatmosfære 49 g tri-n-butyltin-hydrid. Reaktionsopløsningen opvarmes 1 time til tilbagesvaling og inddampes dernæst. De således dannede krystaller oparbej des som beskrevet i eksempel 4dl). Man får 35 endo-6-(p-chlorphenyl)-3-(4-methoxybenzyl)-3-aza-bicyclo-[3.1^-/5.1^/5 ]heptan-2,4-dion i form af hvide krystaller DK 168630 B1 21 med snip. 156-158°C.
b) Til en omrørt suspension af 47,7 g endo-6-(p-chlor-phenyl )-3-( p-methoxybenzyl) -3-aza-bicyclo[ 3.1^/5.11/5^-5 heptan-2,4-dion i 0,5 1 acetonitril sættes dråbevis ved stuetemperatur en opløsning af 280 g cer(IV)-ammoniumnitrat i 390 ml vand. Man efterrører efter endt tilsætning i 4 timer ved stuetemperatur, destilierer ved vandstrålevakuum 200 ml acetonitril fra reaktionsbian-10 dingen og fortynder dernæst med 800 ml vand. Der omrøres 1 time i isbad og filtreres. De dannede lysegule krystaller vaskes med vand og ether. Mellemproduktet opløses i 1 1 methylenchlorid, og der tilsættes 6,8 g n-propylamin, og blandingen henstår natten over. Dernæst filtreres og 15 den lysebrune opløsning inddampes til 70 ml. Der fortyndes med 70 ml ether og filtreres. Det således danne gråbrune krystallinske endo-6-(p-chlorphenyl)-3-aza-bicyclo- [ 3.11/5,il,5 ]heptan-2,4-dion vaskes med 50 ml methylen-chlorid-ether (1:1): smp. 227-228°C.
20 c) 5,9 g endo-6-(p-chlorphenyl)-3-aza-bicyclo-[3.ll/5.il,5 ]heptan-2,4-dion omsættes i 130 ml toluen med 24 ml natriumdihydrobis-(2-methoxyethoxy)-aluminat-toluenopløsning (70%, FLUKA) og oparbejdes med 24 ml koncentreret natriumhydroxidopløsning analogt med eksempel 25 4d2). Man får endo-6-(p-chlorphenyl)-3-aza-bicyclo- [3.1^/5.11,5 Jheptan-hydrochlorid som hvide krystaller med smp. 236-237°C.

Exo-6-(p-chlorphenyl)-3-aza-bicyclo[ 3.il/5.li/5 ]heptan-30 (hydrochlorid), der skal anvendes som udgangsmateriale, kan fremstilles på følgende måde: al) Til en omrørt suspension af 100 g phosphorpentachlorid i 2 1 benzen sættes under omrøring 131,6 g N-(4-methoxy-benzyl)-4-chlorkanelsyreamid. Der efterrøres 30 minutter 35 ved stuetemperatur og dernæst 30 minutter ved 50°C. Efter inddampning optages den mørkebrune olie i 600 ml toluen og DK 168630 B1 23 inddampes. Remanensen opløses i 1,2 1 tetrachlorkulstof, som tilsættes kiselgur ("HYFLO-Super-Cel"®), filtreres, inddampes og tørres under vakuum.
5 Det således dannede orange, krystallinske mellemprodukt opløses i 1,2 1 methylenchlorid og sættes dråbevis til en opløsning af 430 ml 1 N natriumhydrogencarbonatopløsning, 6,2 g tetra-n-butylammoniumbromid og 600 ml vand. Efter endt tilsætning tilsættes dråbevis 100 ml 1 N natrium-10 hydrogencarbonatopløsning, og der efterrøres i 2 timer.
Den organiske fase isoleres, og den vandige fase ekstra-heres to gange med methylenchlorid. De blandede organiske faser tørres over magnesiumsulfat, inddampes, og råproduktet tørres under højvakuum. 4-Aza-2-brom-7-(p-chlor-15 phenyl)-4-(4-methoxybenzyl)-1,6-heptadien-3,5-dion fås som mørkebrun olie og videreomsættes straks.
bl) En opløsning af 111,8 g 4-aza-2-brom-7-(4-chlor-phenyl)-4-(4-methoxybenzyl)-1,6-heptadien-3,5-dion og 0,6 g 2,6-di-tert-butyl-p-kresol i 1,4 1 xylen opvarmes 20 2 timer ved tilbagesvaling. Efter afkøling isoleres det mørkebrune bundfald fra den sorte reaktionsopløsning, omrøres med ether, filtreres og vaskes med ether. Ved fraktioneret krystallisation af acetonitril får man først exo-l-brom-6-(4-chlorphenyl)-3-(4-methoxybenzyl)-3-aza-bi-25 cyclo[3.ll/5.il,5 ]heptan-2,4-dion som lysegule krystaller med et smp. på 177-180°C og som andet produkt den dia-stereomere forbindelse endo-l-brom-6-(p-chlorphenyl)- 3-(p-methoxybenzyl)-3-aza-bicyclo[ 3.11'5.11'5 ]heptan-2,4-dion med smp. 142,5-143,5°C.
30 cl) Til en omrørt suspension af 52,1 g exo-l-brom-6-(p-chlorphenyl)-3-(4-methoxybenzyl)-3-aza-bicyclo[ 3.11'5.11'5 ]_ heptan-2,4-dion i 2,1 1 acetonitril sættes dråbevis ved stuetemperatur en opløsning af 249 g Cer(IV)-ammoniumnitrat i 330 ml vand. Man efterrører efter endt tilsæt-35 ning i 1 time ved stuetemperatur, opvarmer til 40°C, lader i DK 168630 B1 24 opløsningen afkøle langsomt til stuetemperatur og omrører endnu 3 timer. Der inddampes til en tredjedel af volumenet, hvorefter der fortyndes med 1,6 1 vand. Det 5 udfældede produkt filtreres, vaskes med vand, ether og eddikesyreethylester og tørres under vakuum. Man får exo-l-brom-6-(p-chlorphenyl)-3-aza-bicyclo[ 3.li/5.il,5 ]_ heptan-2,4-dion som gule krystaller med smp. 231-232°C.
dl) Til en opløsning af 58,5 g exo-l-brom-6-(p-chlor-10 phenyl)-3-aza-bicyclo[ 3.1-^/^.1^^5 jheptan-2,4-dion og 1,9 g bisazoisobutyronitril ill tetrahydrofuran sættes under nitrogenatmosfære 60 g tri-n-butyltinhydrid. Reaktionsopløsningen opvarmes 4 timer til tilbagesvaling og inddampes dernæst under vakuum. Den krystallinske 15 remanens optages i cyclohexan, filtreres og vaskes med ether. Man får exo-6-(p-chlorphenyl)-3-aza-bicyclo-[ 3.1^/5.11,5 ]heptan-2,4-dion som hvide krystaller med smp. 179-181°C.
e) 3,5 g exo-6-(p-chlorphenyl)-3-aza-bicyclo[ 3.1^/5.11,5 y.
20 heptan-2,4-dion i 75 ml toluen omsættes med 24 ml natrium-dihydrobis(2-methoxyethoxy)-aluminat-toluenopløsning (70%, FLUKA) og oparbejdes med 24 ml koncentreret natriumhydroxidopløsning som beskrevet i eksempel 4d2). Man får exo-6- (p-chlorphenyl)-3-aza-bicyclo[ 3.li/5.il,5 ]heptan-25 hydrochlorid med smp. 201-203eC.
l-Phenyl-3-aza-bicyclo[ 3.li/5.il, 5]heptan(-hydrochlorid), der skal anvendes som udgangsmateriale, kan fremstilles på følgende måde: a2) Til en omrørt opløsning af 88,8 g 2-phenylacrylsyre i 30 7 ml dimethyl formamid og 1,6 1 methylenchlorid sættes dråbevis i løbet af 2 1/2 time ved stuetemperatur en opløsning af 103 ml oxalylchlorid i 400 ml methylenchlorid. Efter endt tilsætning efterrøres endnu 2 timer og dernæst inddampes under vakuum. Det brune, olieagtige DK 168630 B1 25 produkt optages i 600 ml ether og isoleres fra den klæbrige remanens, filtreres over kiselgur ("HYFLO-Super-Cel"®) og inddampes under vakuum. Den brune 5 olie opløses i 0,8 1 methylenchlorid og sættes dråbevis til en til 0-5^ afkølet opløsning af 95,6 g N-(p-methoxy-benzyl)-acrylamid, 6,43 g 4-dimethylaminopyridin og 63,1 g triethylamin i 1 liter methylenchlorid. Efter endt tilsætning efterrøres endnu 3 timer ved stuetemperatur.
10 Efter inddampning af reaktionsopløsningen til 250 ml tilsættes 1 liter ether. Den organiske fase dekanteres fra den klæbrige remanens. Remanensen optages tre gange med 500 ml ether,og den organiske fase dekanteres. De blandede organiske faser inddampes til 200 ml og filtreres over 15 "HYFLO-Super-Cel"® (s.o.). Ved inddampning til tørstof får man 4-aza-4-(4-methoxybenzyl)-2-phenyl-l,6-heptadien- 3,5-dion som brun olie. Denne videreomsættes straks.
b2) En opløsning af 65,7 g 4-aza-4-(p-methoxybenzyl)-2-phenyl-1,6-heptadien-3,5-dion og 0,5 g 2,6-di-tert-butyl-20 p-kresol i 1 liter 1,3-dichlorbenzen omrøres 6 timer ved 170°C. Efter inddampning kromatograferes remanensen med toluen-ether (9:1) ved 2,5 kg kiselgel. Den dannede brune olie opløses ved 70°C i 550 ml diisopropylether og afkøles under omrøring i isbad. Bundfaldet filtreres. Man får 25 efter tørring under højvakuum 3-(4-methoxybenzyl)-l-phenyl-S-aza-bicyclofS.l^'S.U-'ij ]heptan-2,4-dion som hvide krystaller med smp. 87-88°C.
c2) Til en omrørt opløsning af 53 g 3-(4-methoxybenzyl)-l-phenyl-3-aza-bicyclo[ 3.1^-r 5. li, 5 ]heptan-2,4-dion i 560 ml 30 acetonitril sættes dråbevis ved stuetemperatur en opløsning af 344 g cer(IV)-ammoniumnitrat i 1,1 1 acetonitril.
Efter 1 time tilsættes 515 ml vand, og der omrøres endnu 2 timer. Ved afdestillation af acetonitril inddampes til den halve volumen og dernæst fortyndes med 1 liter vand.
35 Det udfældede produkt filtreres, vaskes med vand og tørres under vakuum. Det brungullige, krystallinske stof optages _i DK 168630 B1 26 i 800 ml methylenchlorid, hvortil der sættes 11 ml n-propylamin, som henstår natten over. Den sorte opløsning inddampes, og til den brune, krystallinske masse 5 sættes 80 ml methylenchlorid-ether (1:1), filtreres og tørres under højvakuum. Man får l-phenyl-3-aza-bicyclo-[ 3.1^5.11^ ]heptan-2,4-dion i form af hvide krystaller med smp. 217-218°C.
d2) Til en omrørt suspension af 3 g l-phenyl-3-aza-10 bicyclo[3.ll'5.il,5 ]heptan-2,4-dion i 150 ml toluen sættes dråbevis under nitrogenatmosfære 25,5 ml natrium-dihydrobis- (2-methoxy ethoxy) -aluminat-toluenopløsning (70%, FLUKA). Under tilsætningen holdes temperaturen ved ydre afkøling i isbad inden for 25-35°C. Efter endt 15 tilsætning omrøres 15 minutter ved stuetemperatur, og der opvarmes dernæst 1 time til tilbagesvaling. Efter afkøling i isbad tilsættes dråbevis ved 10-15°C 25,5 ml koncentreret natriumhydroxidopløsning. Den organiske fase dekanteres, og vandfasen vaskes med toluen. De blandede 20 organiske faser vaskes to gange med 100 ml vand og én gang med 70 ml mættet natriumchloridopløsning. Efter tilsætning af magnesiumsulfat filtreres den organiske fase, og der inddampes under vandstrålevakuum. Den brunlige olie opløses i 50 ml ether. Der fås l-phenyl-3-aza-bicyclo-25 [ 3.1^/S^il, 5 ]heptan-hydrochlorid ved tilførelse af hydrogenchlorid som et krystallinsk produkt, som yderligere opslæmmes i ether efter filtreringen, og filtreres på ny og tørres til sidst under højvakuum natten over: hvide krystaller med smp. 248-249°C.

De faste bestanddele sigtes først gennem en si med 0,6 mm maskevidde. Dernæst blandes det aktive stof, lactose, talkum, magnesiumstearat og halvdelen af stivelsen grundigt. Den anden halvdel af stivelsen suspenderes i 15 65 ml vand, og denne suspension sættes til en kogende opløsning af polyethylenglycol i 260 ml vand. Det dannede klister sættes til de pulverformede stoffer, det hele blandes og granuleres, om nødvendigt under tilsætning af vand. Granulatet tørres natten over ved 35°C, sigtes 20 gennem en si med 1,2 mm maskevidde og komprimeres til dobbeltkonkave tabletter med ca. 10 mm diameter med delekærv på oversiden.
På tilsvarende måde kan man også fremstille tabletter indeholdende 75 mg med en af de andre i eksemplerne 1-4 25 nævnte forbindelser med formlen I, idet disse også kan foreligge i form af salte med baser, f.eks. som dinatriumsalt.

Natriumlaurylsulfatet sigtes gennem en si med en maskevidde på 0,2 mm i det aktive stof (lyofiliseres), og begge bestanddele blandes grundigt i 10 minutter. Dernæst 10 sigtes den mikrokrystallinske cellulose gennem en si med en maskevidde på 0,9 mm deri, og der blandes på ny grundigt i 10 minutter. Til sidst sigtes magnesium-stearatet gennem en si med en maskevidde på 0,8 mm deri, og efter 3 minutters yderligere blanding fyldes 390 mg af 15 blandingen i gelatinestikkapsler med størrelsen 0 (elongeret).
På tilsvarende måde kan man også fremstille kapsler indeholdende 100 mg af en anden forbindelse med formlen I ifølge eksemplerne 1-4, idet disse også kan foreligge i 20 form af salte med baser, f.eks. som dinatriumsalt.

En 0,2%'s injektions- eller infusionsopløsning kan eksempelvis fremstilles på følgende måde:

1. Aromatisk substituerede azacycloalkyl-l-hydroxy- alkan-1,1-diphosphonsyrer, kendetegnet ved, at de har 5 formlen !03H2 - OH (I) 03H2 hvori alk er en bivalent aliphatisk gruppe, R" betyder pyrrolidino, piperidino, 3-aza-bicyclo[ 3.11'1 2 3.I1*3]-hept-3-yl, 1,2,5,6-tetrahydropyridino, piperazino, hexa-10 hydroazepino, 3-aza-bicyclo[3.2.01/5 ]hept-3-yl eller octahydroazocino, og R' betyder en usubstitueret eller med lavalkyl, lavalkoxy og/eller halogen mono- eller disubsti-tueret phenyl-, naphthyl-, pyridyl-, thienyl-, pyrimidin-yl-, quinolinyl-, furyl- eller pyrrylgruppe, og deres 15 salte.

9. Farmaceutisk præparat, kendetegnet ved, at det indeholder en forbindelse ifølge et af kravene 1-8 samt gængse farmaceutiske hjælpestoffer.

Fremgangsmåde til fremstilling af forbindelser 25 ifølge krav 1, kendetegnet ved, at man a) i en forbindelse med formlen R' - R" - alk - C1- OH » *2 DK 168630 B1 hvori X]_ er en funktionelt omdannet phosphonogrupe, X2 er en fri eller funktionelt omdannet phosphonogruppe, og alk, R' og R" har de i krav 1 angivne betydninger, omdanner X^ 5 og eventuelt X2 til den frie phosphonogruppe eller b) omsætter en forbindelse med formlen PO3H2 HjN - alk - C - OH (IH) PO3H2 hvori alk har den i krav 1 angivne betydning, eller et salt deraf med en reaktiv ester af en passende aromatisk 10 substitueret aliphatisk dialkohol eller c) omsætter en forbindelse med formlen R' - R" - alk - X3 (IV) hvori X3 er carboxy, carbamoyl eller cyano, og alk, R' og R" har de i krav 1 angivne betydninger, med phosphor-15 syrling og phosphortrichlorid, hydrolyserer primærproduktet, og i et ud fra forbindelserne med formlen IV, hvori X3 er cyano eller carbamoyl, dannet mellemprodukt med formlen PO3H2 R' - R” - *lk - t - »Hz (V) PO3H2 20 hvori alk, R’ og R" har de i krav 1 angivne betydninger, eller et salt deraf ombytter aminogruppen med hydroxy ved behandling med salpetersyrling; og om nødvendigt omdanner en dannet forbindelse til en anden forbindelse med formlen I og/eller en dannet fri forbindelse til et salt eller et 25 dannet salt til den frie forbindelse eller til et andet salt.
