Den foreliggende opfindelse angår et hidtil ukendt atriumpeptid med høj molekylvægt samt et peptidfragment af dette, som har nyttig natriuretisk, diuretisk og vaso-dilataterende aktivitet.

Det er kendt, at cellerne i atriummyocardiet i pattedyr indeholder talrige membranbundne lagergranuler. Disse karakteristiske sekretoriske granuler, som er blevet observeret i rotte-, hunde-, katte- og menneskeatrium, ligner 10 de, der er i peptid-hormonalproducerende celler. Se DeBold et al., J. Histochem. Cytochem. 26, 1094-1102 (1978). Det er blevet rapporteret, at rå vævsekstrakter af atriummyocardium, når de injiceres intravenøst i ikke-diuretiske rotter, producerede et hurtigt og kraftigt na-15 triuretisk respons. Se DeBold et al., Life Science 28, 89-94 (1981). Særlig rensning af rotteatriumhomogenater med et kort kogningstrin og fraktionering på "Sephadex"® blev opnået af Trippodo et al., Proc. Soc. Exp. Biol.
Med. 170, 502-508 (1982). Diuretisk aktivitet fandtes ved 20 disse undersøgelser i det totale molekylvægtsområde 3600 til 44000 dalton i peptidfraktioner med såvel det højere molekylvægtsområde på 36000 til 44000 dalton som i et lavere molekylvægtsområde på 3600 til 5500 dalton.
25 Rotteatriumekstrakter er også blevet fraktioneret i lav molekylvægtsfraktioner (<10000 dalton) og højmolekylvægtsfraktioner (20000-30000 dalton), hvilke ekstrakter begge in vitro afspændte den glatte muskulatur og var kraftige natriuretiske midler, når de blev administreret 30 intravenøst i rotter. 2 (1983) rensningen af et atrium-natriuretisk peptid med en molekylvægt på 5150 dalton og en sekvens af 47 aminosyrer, som de benævnte "Cardionatrin I". Tre yderligere spidser med natriuretisk aktivitet blev opnået ved høj -5 tryksvæskekromatografi (HPLC).
I en sidste publikation omtaler Grammer et al., Biochem. Biophys. Res. Commun. 116(2), 696-703, 31. oktober 1983, den delvise rensning af en rotteatrium-natriuretisk fak-10 tor med en molekylvægt på ca. 3800 og indeholdende 36 aminosyrerester.
I endnu nyere publikationer omtaler Flynn et al., Biochem. Biophys. Res. Commun. 117(3), 859-653, (28. decem-15 ber 1983) og Kangawa og Matsuo, Ibid, 118(1), 131-39 (13. januar, 1984), atrium-natriuretiske peptider for rotte og menneske, der har sekvenser på 28 aminosyrer.
Thibault et al., FEBS Letters 167, 352-56 (1984), omtaler 20 rensningen af et atrium-natriuretisk peptid med en mellemmolekylvægt og 73 aminosyrer, og Kangawa et al., Biochem. Biophys. Res. Commun. 119(3), 933-40 (1984), omtaler rensningen af et beta-rotteatrium-natriuretisk peptid med en middelmolekylvægt og 48 aminosyrer.
25 I ansøgernes verserende ansøgning DK nr. 5334/84 omtales atrium-peptider med lav molekylvægt og ca. 19 til ca. 24 aminosyrer, og sådanne peptider er genstand for patentkrav. Adskillige af disse peptider er yderligere omtalt 30 af en researchgruppe under ledelse af ansøgeren, Currie et al., Science 223, 67-69 (1984).
I overensstemmelse med den foreliggende opfindelse tilvejebringes et hidtil ukendt peptid med høj molekylvægt, 35 som -udviser nyttig natriuretisk, diuretisk og vasodilata-terende aktivitet. Dette biologisk aktive peptid har følgende aminosyresekvens:

Det hidtil ukendte peptid eller peptidfragment ifølge opfindelsen har især terapeutisk anvendelse som diureticum, natriureticum, renal vasodilatator og afspændende middel for den glatte muskulatur. Det vil sige, det er effektivt 5 mod natrium, urinvolumen, renal vasodilatation og den glatte muskulaturs tonus.
Kort sagt er dette hidtil ukendte peptid eller peptidfragment blevet opnået ved fraktionering af rotteatrie-10 ekstrakter ved gelfiltreringskromatografi til tilvejebringelse af en fraktion med høj molekylvægt og en fraktion med lav molekylvægt, der begge havde nyttig natri-uretisk aktivitet. Fraktionen med den lavere molekylvægt blev fraskilt og renset i adskillige atrium-natriuretiske 15 peptider med lav molekylvægt som beskrevet i den i det foregående nævnte verserende ansøgning tilhørende opfinderen.
Den opnåede fraktion med høj molekylvægt (atriopeptigen-20 ATG) ved førnævnte gelfiltreringskromatografi af rotte-atrieekstrakter blev fraktioneret i overensstemmelse med den foreliggende opfindelse ved isoelektrisk fokusering og omvendtfaset HPLC til opnåelse af et delvist renset APG. Rensning af cyanogenbromid-nedbrydninger af den del-25 vist rensede fraktion med høj molekylvægt resulterede i isoleringen af et enkelt biologisk aktivt cyanogenbromid-spaltningspeptid ifølge krav 3 på 93 aminosyrer omfattende aminosyrer 19 til 111 af ovennævnte APG. Sekvensanalyser af disse peptider koblet med nylige rapporter af se-30 kvensanalyser af atriumpeptider med middelmolekylvægt [Thibault et al., FEBS Letters 167, 352-356 (1984) og
Kangawa et al., Biochem. Biophys. Res. Commun. 119, 933-940 (1984)] giver den komplette primære struktur af ovennævnte 111 rest APG.

6 findelsen. For nemheds skyld er de citerede referencer anført efter afslutningen af denne detaljerede beskrivelse. Ved gelfiltreringskromatografi er den natriuretiske og diuretiske aktivitet for rotteatriale ekstrakter ble-5 vet fundet i fraktioner med høj molekylvægt (20000-30000) og lav molekylvægt (mindre end 10000), som også udviser spasmolytisk aktivitet på den glatte muskulatur (in vitro) (1). Det blev konkluderet af opfinderen, at fraktionen med den høje molekylvægt er forstadiet for fraktionen 10 med den lave molekylvægt med henblik på virkningerne af delvis proteolyse med trypsin. Trypsinbehandling af fraktionen med den høje molekylvægt forøgede mærkbart den spasmolytiske aktivitet. De aktive produkter, som vandrer med fraktionen med den lave molekylvægt i gelfiltrering 15 (2), er atriopeptiner I, II og III (3) ved omvendt faset HPLC. Derfor er de aktive komponenter i fraktionen med den høje molekylvægt blevet betegnet som atriopeptigener (APGer), forstadier for atriopeptinerne (APer), de bioaktive atriale peptider med lav molekylvægt. Disse atrio-20 peptiner omfatter følgende delvise sekvens af det fulde 111 aminosyre atriopeptigen som beskrevet i den i det foregående nævnte verserende patentansøgning tilhørende opfinderen: 25 AP I = aminosyrer 88 til 108 AP II = aminosyrer 88 til 110 og AP III = aminosyrer 88 til 111.
Ved rensningen af atriumpeptiderne med den høje molekyl-30 vægt og derivater deraf blev en bioprøve for glat muskulatur for hver fraktion udført efter aktivering af partiel tryptisk proteolyse (1, 2). De aktive fraktioner indeholder sekvensen for atriopeptinerne med lav molekylvægt (3). Det første trin i rensning af fraktionen med 35 den-høje molekylvægt (opnået ved G-75 "Sephadex”-kromatografi) var isoelektrisk fokusering, som viste, at det var en blanding af bioaktive arter (tilsyneladende isoelek-

7 triske punkter ved pH 4,91, 5,01, 5,17, 5,34 og 6,03). Massen af bioaktive arter (pH mellem 4,6 og 5,4) blev efter fjernelse af ampholyter og saccharose underkastet omvendtfaset HPLC.

Det typiske kromatogram viste en kompleks blanding af bioaktive peptider samlet ved 31 og 34% acetonitril. Af disse var en overvejende komponent med tilstrækkelig renhed ved gelanalyse (tilsyneladende molvægt 17000), og den 10 blev udvalgt som atriopeptigen til yderligere rensning og karakterisering i overensstemmelse med den foreliggende opfindelse.
Med henblik på atriopeptigenernes kompleksitet sammenlig-15 net med den enkle sekvens for atriopeptinerne afledt derfra er det blevet konkluderet af opfinderen, at atriopep-tigenerne igen kan være derivater af nogle større peptider. Følgelig blev virkningerne af kemisk spaltning undersøgt. Cyanogenbromidspaltning blev valgt på grund af 20 manglen på methionin i den lavmolekylære atriopeptinsek-vens (5). Cyanogenbromidspaltning foregår ved methionin i stilling 18 i den fulde 111 aminosyresekvens i atriopep-tigenet til tilvejebringelse af et højmolekylært peptidfragment på 93 aminosyrer. Det indledende trin ved frak-25 tionering af et cyanogenbromid-nedbrydningsprodukt med den totale høje molekylvægtsfraktion gav et bioaktivt materiale (fundet ved 30 til 31% acetonitril), som blev renset ved omvendtfaset HPLC til opnåelse af en enkelt komponent ved HPLC med en tilsyneladende molvægt på 9500 30 ved gelelektroforese.
Sekvensdata for begge peptider er vist i det følgende. Overlapning af sekvenserne for disse peptider og peptiderne ifølge Thibault et al., FEBS Letters 167, 352-6 35 (1984) og Kangawa et al., Biochem. Biophys. Res. Commun.

8 i overensstemmelse hermed. Carboxypeptidasebehandling af APG fjernede hurtigt Phe uden frigørelse af detekterbar Tyr og Arg. På den anden side gav lignende behandling af det cyanogenbromidspaltede peptid hurtig frigørelse af 5 Tyr, Arg og Phe (tabel 1), hvilket indikerede, at dette præparat består af tre peptider, der har de samme sekvenser, som slutter i Tyr, Arg og Phe.
Denne sekvens af 111 aminosyrerester inkorporerer i den 10 C-terminale ende peptiderne med lav molekylvægt, som nyligt er beskrevet (understreget i sekvensen i det følgende) i forbindelse med et par basiske aminosyrerester (Arg-Arg i dette tilfælde), som typisk danner spaltningsstedet i en række forskellige forstadier for sekreterede 15 peptider og proteiner.

EKSEMPEL 1
Fremstilling af et atriopeptigen 35
En -ekstrakt af atria fra 1200 rottehjerter blev fremstillet og underkastet kromatografi på G-75 "Sephadex" ved

11 den almene procedure som beskrevet i det foregående (3). Fraktionen med den høje molekylvægt blev lyofiliseret, opløst i "Ampholine"-bæreampholyter ved pH 4-6 (LBK Instruments, Inc., Rockville, Maryland) og elektrofokuseret 5 i en llO ml saccharosemassefyldegradientsøjle (LKB) ved 1000 V i 40 timer (4). Søjlen derpå tømt med en hastighed på 48 ml pr. time, idet man opsamlede 2 ml fraktioner.
Efter bestemmelse af pH blev aliquoter (50 aliter) af søjlefraktioner indstillet til pH 8 (ved tilsætning af 10 450 aliter 0,1 M tris-puffer, pH 8,4) og inkuberet med trypsin (Sigma Chemical Co., St. Louis, Missouri), en enhed pr. ml i 1 time ved 22 °c. Disse præparater blev prøvet for kyllingerectum-relaxationsaktivitet ved den almene procedure som tidligere beskrevet (1,2). Kombine-15 rede søjlefraktioner (pH 4,6 til 5,4) indeholdende massen af bioaktivitet (~30 ml) blev kromatograferet på G-50 "Sephadex" (80 x 2,7 cm) i 0,5 M eddikesyre til fjernelse af ampholyter og saccharose. Efter frysetørring blev dette materiale underkastet HPLC med omvendt fase under an-20 vendelse af Brownlee RP-300 Aquapore-søjlen og det tidligere beskrevne opløsningsmiddelsystem (3). Gradienten bestod af (a) 0-24% A i 8,8 minutter, (b) 24-28% A i 25 minutter, (c) 28-36% A i 100 minutter. Aliquoter (50 aliter) af HPLC søjlefraktioner (2 ml) blev tørret i va-25 kuum, optaget i 500 aliter 0,1 M tris, pH 8, trypsinise-ret og bioafprøvet som anført i det foregående.
Fremstilling af cyanogenbromidpeptid ud fra fraktionen med høj molvægt 30
Den lyofiliserede fraktion med høj molekylvægt som opnået ved kromatografi på G-75 "Sephadex" fra en ekstrakt af atrium fra 600 rottehjerter (1,3), opløst i 10 ml 70%'s myresyre, blev sat til 500 mg cyanogenbromid (Eastman Or-35 ganic Chemicals, Rochester, N.Y.) i et rør med glasprop (5).· Efter 16 timers forløb (ved 22 °C) blev der tilsat 90 ml vand, og opløsningen blev lyofiliseret. 12 underkastet HPLC med omvendt fase under anvendelse af det i det foregående beskrevne system med (a) 0-27,2% A i 10 minutter efterfulgt af (b) 27,2-32% A i 60 minutter. Bioaktive fraktioner (opnået ved 29,6-31,2% A) blev bragt 5 til tørhed i vakuum og renset ved en anden protokol under anvendelse af en blanding af opløsningsmiddel A’ (0,1% trifluoreddikesyre i propanol-1) og B (0,1% trifluoreddikesyre i vand) ved 0,67 ml pr. minut bestående af (a) 0-15% A' i 5 minutter efterfulgt af (b) 15-24% A' i 90 mi-10 nutter. Bioaktive fraktioner ved 22-22,5% A') blev bragt til tørhed i vakuum og derpå underkastet en tredie protokol under anvendelse af opløsningsmidler A" (0,085% phos-phorsyre i propanol-1) og B" (0,085% phosphorsyre i vand) ved 0,67 ml pr. minut, bestående af 0-50% A' i 50 minut-15 ter. Det bioaktive produkt (opnået ved 32,1% A") blev derpå igen ført to gange gennem HPLC under anvendelse af anden protokol som i det foregående, hvilket til sidst gav 110 ug (protein) af en enkelt bioaktiv komponent (de-tekteret ved 215 nm) ved 22,1% A'.

Analyse af peptider
Aminosyreanalyse blev foretaget ved hydrolyse af 1 nmol peptid i 6 N HC1 i 22 timer ved 110 °C. Hydrolysatet blev 25 lyofiliseret og påført et Waters aminosyreanalysesystem under anvendelse af o-phthalaldehyd-forkolonnederivatise-ring (6) efterfulgt af HPLC med omvendt fase.
Peptidsekvensering - N-terminal sekvensering blev foreta-30 get ved sekventiel Edman-nedbrydning af 2-4 nmol peptid under anvendelse af en Applied Biosystems model 470A gasfasesekvenser (7), detektering af phenylthiohydantoinde-rivater ved HPLC (8). Gennemsnitlige gentagne udbytter overskred 93%. C-terminal sekvensering blev foretaget ved 35 addition af carboxypeptidase Y (2 ug, 20 uliter, Perce Chemical Co., Rockford, Illinois) til 1-2 nmol peptid opløst i 280 uliter 50 mM natriumacetatpuffer, pH 5,5. Med 13 mellemrum blev aliquoter på 50 uliter sat til 25 uliter 1%'s trifluoreddikesyre (9), og de frigjorte aminosyrer blev bestemt ved aminosyreanalyse (se i det foregående).
5 Gelanalyse - Elektroforese af peptider (1-2 ug) blev foretaget med en 15%'s polyacrylamidgel (0,4% bis-acryl-amid) efter Laemmli (10). Gelen blev farvet med 0,8%'s sølvnitrat i 20 minutter, vasket og udviklet med en opløsning af 0,005%'s citronsyre og 0,2%'s formaldehyd.
10 EKSEMPEL 2
Cyanogenbromidpeptidet med høj molekylvægt fra rotteatrium som beskrevet i eksempel 1 i det foregående blev 15 afprøvet for natriuretisk aktivitet i hunde.
Peptidet med høj molekylvægt blev enten injiceret alene eller efter trypsinbehandling. Trypsininkuberingen (1 enhed/ml) blev foretaget i 30 minutter ved stuetemperatur 20 med 100 ug af det rensede høj molekylære cyanogenbromid-peptid, Atriopeptin I og II blev leveret af Peninsula Labs, San Carlos, California. Atriopeptin III og Ser-Leu-Arg-Arg-Atriopeptin III (Flynn et al., peptidet, reference 13) blev syntetiseret ved automatiseret peptidsyntese. 25
Bastardhunde af begge køn blev anæstetiseret (i.v.) med pentobarbital-natrium (30 mg/kg). Der blev foretaget et flankeindsnit til retroperitoneal frilæggelse af den højre nyre. Urinlederen blev kanyleret med PE160 (Clay 30 Adams) rør, som blev forbundet til en fraktionsopsamler for urin. En elektromagnetisk strømningsprobe (8 mm diameter Carolina Instrument) blev anbragt rundt om den højre renale arterie for måling af den renale blodstrømning. En nål nr. 22 (knyttet til PE50 rør og sprøjte) 35 blev- indsat i den renale arterie over strømningsproben.
Hunden blev kontinuerligt infuseret med saltvand (0,9% NaCl) ved 1,3 ml/min. Urinprøver blev samlet med 5 minut-

14 ters mellemrum og analyseret for volumen, natrium, kalium og osmolaritet. Peptiderne blev injiceret (bl.a. i den renale arterie) med 30 minutters mellemrum.
5 Resultater
Peptidet med den høje molekylvægt (cyanogenbromid) gav en koncentrationsafhængig diurese, som ikke blev væsentligt ændret ved trypsinisering. Grænserespons (en 50%'s for-10 øgelse i urinvolumen, FU5Q) blev opnået med 0,3 nmol, mens en 450%"s i urinstrøm (FU^gg) blev opnået med 3 nmol. Sammenligning af mængden af peptid påkrævet til at producere en 200%'s forøgelse i urinvolumen (FU200^ ln^l“ kerer, at Ser-Leu-Arg-Arg-APIII kræver 0,3 nmol; høj mo-15 lekylvægt + trypsin kræver 0,5 nmol, høj molekylvægt ale ne 0,7 nmol; a triopep tin II og III kræver 10 nmol; og atriopeptin I forøgede ved 30 nmol kun urinvolumenet med 50%. Styrkerækkefølgen for vasodilatation i sammenligning med dosis påkrævet for at give en forøgelse på 10 ml/min 20 i renal blodstrømning (RB^q) er som følger: Ser-Leu-Arg-Arg-APIII krævede 1 nmol; det højmolekylære cyanogenbro-midpeptid (med eller uden trypsinforbehandling) krævede 3 nmol; atriopeptin III krævede 8 nmol; atriopeptin II krævede 17 nmol; mens 30 nmol atriopeptin I kun gav en 6 25 ml/min ændring i renal blodstrøm (RDg). Hvert peptid blev afprøvet på 3-5 forskellige hunde.

Sammenfattende kan det siges, at direkte injektion af det 20 rensede cyanogenbromidfragment af atriopeptigen med høj molekylvægt gav en udtalt diurese, der ikke kunne skelnes fra de kraftigste polypeptider med lav molekylvægt. Dette kan genspejle en øjeblikkelig overføring af peptidet i nyren eller direkte genkendelse af nyren for det intakte 25 peptid. Denne væsensforskellighed i styrkerækkefølgen for peptiderne i henseende til renal vasodilatation mod na-triurese antyder, at disse respons er medieret af separate receptorer.
