Natriuretisk aktivitet ble funnet av disse forskere i det totale molekylvektområde på 3.600 til 44.000 dalton og i peptidfraksjoner av både det høyere molekylvektområde på 36.000-44.000 dalton og det lavere molekylvektområde på 3.600-5.500 dalton.
Rotteatriale ekstrakter er også blitt fraksjonert i lavmolekylære fraksjoner (< 10.000 dalton) og høymolekylære fraksjoner (20.000-30.000 dalton) som begge in vitro avspente glattmuskler og var kraftige natriuretiske midler når de ble administrert intravenøst til rotter. Se Currie et al., Science 221, 71-73 (1983).
I andre nylige publikasjoner har flere forskere beskrevet forskjellige lav- og mellom-molekylære atriale natriuretiske peptider med aminosyresekvenser i området fra 19 til 59 aminosyrer. Således har DeBold et al., Fed.
Proe. 4_2(3), Abstract 1870, s. 611 (1983), beskrevet rensing av et atrialt natriuretisk peptid med en molekylvekt på
5.150 dalton og en sekvens på 47 aminosyrer som forskerne betegnet som "Cardionatrin I". Tre ytterligere topper med natriuretisk aktivitet ble erholdt ved væskekromatografi med høy ytelse (HPLC).
I en senere publikasjon, Grammer et al., Biochem. Biophys. Res. Commun. 116(2), 696-703, 31. oktober 1983,
er det beskrevet delvis rensing av en rotteatrial natriuretisk faktor med en molekylvekt på ca. 3.800 og inneholdende 36 aminosyrerester.
I enda nyere publikasjoner, Flynn et al., Biochem. Biophys. Res. Commun. 117 (3), 859-65 (28. desember 1983),
og Kangawa og Matsuo, Ibid., 118(1), 131-39 (13. jan. 1984), beskrives atriale natriuretiske peptider av rotte og menneske med sekvenser på 28 aminosyrer.
Thibault et al., FEBS Letters 167, 352-56 (1984) beskriver rensing av et mellommolekylært atrialt natriuretisk peptid med 73 aminosyrer, og Kangawa et al., Biochem. Biophys. Res. Commun. 119 (3) , 933-40 (1984), beskriver rensing av et mellommolekylært fi-rotteatrialt natriuretisk peptid med 48 aminosyrer.
I søkerens US søknader 551 372 og 569 684 er lavmolekylære atriale peptider med fra 19 til 24 aminosyrer beskrevet. Flere av disse peptider er ytterligere beskrevet av en forsøksgruppe ledet av Currie et al., Science 223,
67-69 (1984).
Ifølge oppfinnelsen tilveiebringes en fremgangsmåte for fremstilling av et terapeutisk aktivt peptid med følgende aminosyresekvens:
eller et fragment derav, inneholdende sekvensen av aminosyrer 19-111 av ovennevnte peptid.
I peptidstrukturen er aminosyrekomponentene angitt ved konvensjonelle forkortelser som følger:
Peptidmaterialet er isolert i en delvis renset form som ikke eksisterer i rottemycardiet fra hvilket det først ble erholdt. Dvs. det er blitt fremstilt i en form som er hovedsakelig fri for lavmolekylære peptider og fri for andre cellulære komponenter og vevbestanddeler. Dette nye atriale peptid har fysiologiske karakteristika som antyder at det er betydningsfullt innen den medisinske forskning ved studium av det endocrine system av hjerteatriene når det gjelder humorale midler for avpasning av extracellulært volum, natrium og vaskulær resistens.
I særdeleshet har det nye peptid terapeutisk anvendelse som diureticum, natriureticum, renal vasodilator og glatt-muskelrelakserende middel. Dvs. at det utviser en grundig virkning på natrium, urinvolum, renal vasodilasjon og glatt-muskeltonus.
Fremgangsmåten ifølge oppfinnelsen er kjennetegnet
ved at,
a) ekstrakter av hjerteatrier fra pattedyr fraksjoneres for å erholde fraksjoner med høy og lav molekylvekt,
b) fraksjonen med høy molekylvekt fraksjoneres for
å erholde et atriopeptigen med den ovennevnte sekvens av
111 aminosyrer,
c) atriopeptigenet spaltes med cyanogenbromid og fra cyanogenbromidekstraktet isoleres et fragment av atriopeptigenet som inneholder sekvensen av aminosyrene 19-111.
Fraksjoneringen i punkt a) utføres ved hjelp av gelfiltrering og den høymolekylære fraksjon (atriopeptigen-
APG) fraksjoneres ved hjelp av isoelektrisk fokusering og omvendt fase HPLC under erholdelse av delvis renset APG.
Ved rensing av cyanogenbromidoppslutninger av den delvis rensede høymolekylære fraksjon isoleres et enkelt bio-logisk aktivt cyanogenbromid-spaltet peptid på 93 aminosyrer omfattende aminosyrer 19 til 111 av det ovenfor angitte APG. Sekvensanalyser av disse peptider sammenkoblet med nylige rapporter vedrørende sekvensanalyser av intermediære molekylære atriale peptider (Thibault et al., FEBS Letters 167, 352-
356 (1984) og Kangawa et al., Biochem. Biophys. Res. Commun. 119, 933-940 (1984)) ga den fullstendige primærstruktur av det ovenfor angitte 111 rest-APG.
I det etterfølgende gis en detaljert beskrivelse av foretrukne utførelsesformer ved oppfinnelsen. Av hensikts-messige grunner er de angitte referanser oppført ved slutten av denne detaljerte beskrivelse.
Ved gelfiltreringskromatografi er den natriuretiske og diuretiske aktivitet av rotteatrialekstrakter blitt funnet i høymolekylære (20.000-30.000) og lavmolekylære (mindre enn 10.000) fraksjoner som også utviser spasmolytisk glattmuskel-aktivitet (in vitro) (1). Man har trukket den konklusjon at den høymolekylære fraksjon er forløperen for den lavmolekylære fraksjon i lys av virkningene av partiell proteolyse med trypsin. Trypsinbehandling av den høymolekylære fraksjon øker markert den spasmolytiske aktivitet. De aktive produkter som samvandrer med den lavmolekylære fraksjon ved gelfiltrering (2), er atriopeptin I, II og III (3) ved omvendt fase HPLC. Derfor er de aktive komponenter av den høymolekylære fraksjon blitt betegnet som atriopeptigener (APG), forløpere for atriopeptin (AP), de lavmolekylære bioaktive atriale peptider. Disse atriopeptiner omfatter følgende partielle sekvenser av det komplette 111 aminosyres atriopeptigen som beskrevet i de tidligere angitte patentsøknader:
AP I = aminosyre 88 til 108,
AP II = aminosyre 88 til 110, og
AP III = aminosyre 88 til 111.
Ved rensingen av høymolekylære atriale peptider og deres derivater ble en glattmuskel-biobestemmelse av hver fraksjon etter aktivering ved partiell tryptisk proteolyse utført (1,2). De aktive fraksjoner inneholder sekvensen av de lavmolekylære atriopeptiner (3). Det første trinn i rensingen av den høymolekylære fraksjon (erholdt ved G-75 Sephadex® kromatografi) var isoelektrisk fokusering som viste at denne var en blanding av bioaktive arter (tilsynelatende isoelektriske punkter ved pH 4,91, 5,01, 5,17, 5,34 og 6,03). Hovedmengden av bioaktive arter (mellom pH 4,6 og 5,4) etter fjerning av amfolytter og sucrose ble underkastet omvendt fase HPLC. Det typiske kromatogram utviste en kompleks blanding av bioaktive peptider sammenklumpet ved 31 og 34% acetonitril. Blant disse var en dominerende komponent av tilstrekkelig renhet ved gelanalyse (tilsynelatende molekylvekt 17.000) og ble valgt som et atriopeptigen for ytterligere rensing og karakterisering i henhold til oppfinnelsen.
I lys av kompleksiteten av atriopeptigener sammenlignet med den enkle sekvens av atriopeptiner avledet fra disse,
ble det konkludert med at atriopeptigenene kanskje i sin tur er derivater av et noe større peptid. Følgelig ble effekten av kjemisk splitting undersøkt. Cyanogenbromidsplitting ble valgt på grunn av mangel på methionin i den lavmolekylære atriopeptinsekvens (5). Cyanogenbromidsplitting finner sted ved methioninet i 18-stilling av den hele 111 aminosyresekvens av atriopeptigen under dannelse av et høymolekylært
fragment på 93 aminosyrer. Begynnelsestrinnet ved fraksjoneringen av en cyanogenbromidoppslutning av den totale høy-molekylære fraksjon ga et bioaktivt materiale (funnet ved 30-31% acetonitril) som ble renset ved omvendt fase HPLC under dannelse av en enkel komponent ved HPLC med en tilsynelatende molekylvekt på 9500 ved gelelektroforese.
Sekvensdata for begge peptider er vist i det etter-følgende. Overlapping av sekvensene av disse peptider og de ifølge Thibault et al., FEBS Letters 167, 352-6 (1984) og Kangawa et al., Biochem. Biophys. Res. Commun. 119, 933-40
(1984) ga primærstrukturen av et 111 rest-atriopeptigen. Resultatene av C-terminalanalyse er i overensstemmelse med dette. Carboxypeptidasebehandling av APG fjernet hurtig Phe uten frigivelse av påvisbar Tyr eller Arg. På den annen side ga lignende behandling av det cyanogenbromidsplittede peptid en hurtig frigivelse av Tyr, Arg og Phe (tabell 1)
som derved indikerte at dette preparat består av tre peptider med samme sekvenser som ender i Tyr, Arg og Phe.
Denne sekvens av 111 aminosyrerester inkorporerer ved dets C-ende de lavmolekylære peptider som nylig er blitt beskrevet (understreket i sekvensen i det etterfølgende) i forbindelse med et par av basiske aminosyrerester (Arg-Arg i dette tilfelle) som typisk danner splittingssetet i et utall av forløpere av utskilte peptider og proteiner.
Den komplette primærstruktur av det 111 aminosyres atriopeptigen ble utledet fra: A aminosyresekvens av de første 34 rester av atriopeptigenet, B aminosyresekvens av de første 39 rester av det cyanogenbromidsplittede peptid, C aminosyresekvens av "intermediær Mr form atrial natriuretisk faktor" (11) og D komplett aminosyresekvens av "(J-rotte atrialt natriuretisk polypeptid" (12) .
De etterfølgende eksempler beskriver rensing, delvis karakterisering og natriuretisk, diuretisk og vasodilaterende aktivitet av den ovenfor angitte komponent (111 aminosyrer) av den høymolekylære fraksjon (20.000-30.000) og av et cyanogenbromid-peptidfragment (93 aminosyrer) avledet fra denne fraksjon.
Eksempel 1
Fremstilling av et atriopeptigen - Et ekstrakt av atrier fra 1.200 rottehjerter ble fremstilt og underkastet kromatografi på G-75 Sephadex®ved den tidligere beskrevne generelle metode (3). Den høymolekylære fraksjon ble lyofilisert, oppløst i pH 4-6 'Ampholine" bærer-amfolytter og ble elektro-fokusert i en 110 ml sucrosedensitets-gradientkolonne ved 1.000 V i 40 timer (4). Kolonnen ble deretter tømt med 48 ml pr. time idet det ble oppsamlet 2 ml fraksjoner.
Etter bestemmelse av pH ble aliquoter (50^ul) av kolonnefraksjoner justert til pH 8 (ved tilsetning av 4 50^ul 0,1 M tris-buffer, pH 8,4) og inkubert med trypsin (en enhet pr. ml) i 1 time ved 22°. Disse preparater ble bestemt med hensyn til kyllingrectum-avspenningsaktivitet ved den generelle prosedyre som tidligere er beskrevet (1,2). Kombinerte kolonnefraksjoner (pH 4,6 til 5,4) inneholdende hovedmengden av bioaktivitet (~30 ml) ble kromatografert på G-50 Sephadex® (80 x 2,7 cm) i 0,5 M eddiksyre for å fjerne amfolytter og sucrose. Etter frysetørking ble dette materiale underkastet omvendt fase HPLC under anvendelse av "Brownlee RP-300 Aquapore" kolonne og løsningsmiddelsystem som tidligere beskrevet (3). Gradienten besto av (a) 0 til 24% A i 8,8 minutter, (b) 24 til 28% A i 25 minutter, (c) 28 til 36% A i 100 minutter. Aliquoter (50^ul) av HPLC kolonnefraksjonene (2 ml) ble tørket i vakuum, tatt opp i 500yUl 0,1 M tris, pH 8, ble trypsinert og biobestemt som ovenfor angitt.
Fremstilling av et cyanogenbromidpeptid fra den høymolekylære fraksjon
Den lyofiliserte høymolekylære fraksjon erholdt ved kromatografi på G-75 Sephadex® fra et ekstrakt av atrier fra 600 rottehjerter (1,3), oppløst i 10 ml 70% maursyre, ble tilsatt til 500 mg cyanogenbromid i et glassrør med kork (5). Etter 16 timer (ved 22°C) ble 90 ml vann tilsatt, og løs-ningen ble lyofilisert. Residuet ble underkastet omvendt fase HPLC under anvendelse av det ovenfor angitte system med (a) 0 til 27,2% A i 10 minutter etterfulgt av (b) 27,2 til 32% A i 60 minutter. Bioaktive fraksjoner (erholdt ved 29,6 til 31,2% A) ble bragt til tørrhet i vakuum og renset ved
en andre prosedyre under anvendelse av en blanding av løs-ningsmiddel A' (0,1% trifluoreddiksyre i propanol-1) og B (0,1% trifluoreddiksyre i vann) til 0,67 ml pr. minutt bestående av (a) 0 til 15% A<1> i 5 minutter etterfulgt av
(b) 15 til 24% A' i 90 minutter. Bioaktive fraksjoner (ved 22 til 22,5% A') ble bragt til tørrhet og deretter underkastet en tredje prosedyre under anvendelse av løs-ningsmidler A" (0,085% fosforsyre i propanol-1) og B"
(0,085% fosforsyre i vann) ved 0,67 ml pr. minutt, bestående av 0 til 50% A" i 50 minutter. Det bioaktive produkt
(erholdt ved 32,1% A") ble deretter Igjen underkastet HPLC to ganger, igjen under anvendelse av den andre prosedyre, som til slutt ga llCyug (protein) av en enkel bioaktiv komponent (påvist ved 215 nm) ved 22,1% A'.
Analyse av peptider
Aminosyreanalyser ble utført ved utført ved hydrolyse av 1 nmol peptid i 6~N HCl i 22 timer ved 110°C. Hydrolysatet ble lyofilisert og anbragt i et Waters aminosyre-analyse-system under anvendelse av o-fthalaldehyd-prekolonne-derivatisering (6) etterfulgt av omvendt fase HPLC.
Peptidsekvensering - N-terminal sekvensering
ble utført ved Edman sekvensnedbrytning av 2-4 nmol peptid under anvendelse av en "Applied Biosystems model 470A" gass-fasesekvenser (7) som påviser fenylthiohydantoinderivater ved HPLC (8). Gjennomsnittlige gjentatte utbytter overskred 93%. C-terminal sekvensering ble utført ved tilsetning av carboxypeptidase Y (2^ug, 2Cyu]) til 1-2 nmol peptid oppløst i 28Cyul 50 raM natriuraacetatbuffer, pH 5,5. Ved intervaller ble 50^ul aliquoter tilsatt til 25^ul 1% trifluoreddiksyre (9), og de frigitte aminosyrer ble bestemt ved aminosyre-analyse (som ovenfor angitt).
Gelanalyse - elektroforese av peptider (l-2^ug) ble foretatt med en 15% polyacrylamidgel (0,4% bis-acrylamid)
i henhold til Laemmll (10). Gelen ble beiset med 0,8% sølv-nitrat i 20 minutter, ble vasket og fremkalt med en løsning av 0,005% sitronsyre og 0,2% formaldehyd.
Eksempel 2
Det høymolekylære rotteatriale, cyanogenbromid-behandlede peptid renset som beskrevet i eksempel 1, ble testet med hensyn til natriuretisk aktivitet i hunder.
Det høymolekylære peptid ble enten injisert alene eller etter trypsinbehandling. Trypsin-(1 enhet/ml) inkuber-ingen ble utført i 60 minutter ved romtemperatur med 100^,ug av det cyanogenbromid-behandlede høymolekylære peptid. Atriopeptin I og III ble anskaffet fra Peninsula Labs,
San Carlos, California. Atriopeptin II og Ser-leu-arg-arg-Atriopeptin III (Flynn et al. peptid, henvisning 13) ble syntetisert ved en automatisert peptidsyntese.
Bastardhunder av begge kjønn ble bedøvet (i.v. med
30 mg/kg pentobarbitalnatrium. Et sideinnsnitt for retro-peritoneal eksponering av høyre nyre ble utført. Urinlederen ble kanylert med PE160 slange som ble koblet til en fraksjons-oppsamler for uringjenvinning. En elektromagnetisk strøm-ningssonde (8 mm i diameter) ble anbragt rundt den høyre nyrearterie for måling av renal blodstrømning. En nål nr. 22 (festet til PE50-slangen og sprøyte) ble innsatt i nyrearterien over strømningssonden. Hunden ble kontinuerlig infusert med saltvann (0,9% NaCl) med 1,3 ml/min. Urinprøve ble oppsamlet ved 5 minutters intervaller og analysert med hensyn til volum, natrium, kalium og osmolaritet. Peptidene ble injisert (i.a. i nyrearterien) ved 30 minutters intervaller.
Resultater
Det høymolekylære (cyanogenbromid) peptid fremkalte en konsentrasjonsavhengig diuresis som ikke signifikant ble forandret ved trypsinering. Terskelresponsen (en 50% økning i urinvolum) ble oppnådd med 0,3 nmol, mens en 450% økning i urlnstrømning ble oppnådd med 3 nmol. En sammen-ligning av mengden av peptid som var nødvendig for å gi en 200% økning i urinvolum, indikerte at ser-leu-arg-arg-APIII krever 0,3 nmol; høymolekylært peptid + trypsin krever 0,5 nmol, høymolekylært peptid alene 0,7 nmol; atrio-
peptin II og III krever 10 nmol; og atriopeptin I i dose på 30 nmol øket urinvolumet bare med 50%. Graderingsrekke-følgen når det gjelder styrken av vasodilatering idet man sammenligner den dose som er nødvendig for å fremkalle en økning på 20 ml/min i renal blodstrømning, er som følger: ser-leu-arg-arg-APIII krevet 1 nmol; det cyanogenbromid-behandlede høymolekylære peptid (i nærvær eller fravær av trypsin-forbehandling) krevet 3 nmol; atriopeptin III
krevet 8 nmol; atriopeptin II krevet 17 nmol; mens 30 nmol av atriopeptin I bare ga 6 ml/min forandring i renal blod-
strømning. Hvert peptid ble testet i 3-5 separate hunder.
En direkte injeksjon av renset cyanogenbromidfragment av høymolekylært atriopeptigen fremkalte en uttalt diurese som ikke kan skjelnes fra de mest kraftige av de lavmolekylære peptider. Dette kan gjenspeile en øyeblikkelig omdannelse av peptidet i nyren eller direkte gjenkjennelse av nyren av det intakte peptid. Ulikheten i rekkefølgen når det gjelder styrken av peptidene med hensyn til renal vasodilatering mot natriurese antyder at disse responser formidles av separate reseptorer. Hovedsakelig lignende dluresis ble oppnådd når det delvis rensede 111 aminosyres atriopeptigen (før cyanogenbromidsplitting) ble testet i hunder.

1. Fremgangsmåte for fremstilling av et terapeutisk aktivt peptid med følgende aminosyresekvens:
eller et fragment derav, inneholdende sekvensen av aminosyrer 19-111 av ovennevnte peptid, karakterisert ved at a) ekstrakter av hjerteatrier fra pattedyr fraksjoneres for å erholde fraksjoner med høy og lav molekylvekt, b) fraksjonen med høy molekylvekt fraksjoneres for å erholde et atriopeptigen med den ovennevnte sekvens av 111 aminosyrer, c) atriopeptigenet spaltes med cyanogenbromid og fra cyanogenbromidekstraktet isoleres et fragment av atriopeptigenet som inneholder sekvensen av aminosyrene 19-111.
2. Fremgangsmåten ifølge krav 1, karakterisert ved at fraksjoneringen i punkt a) utføres ved hjelp av gelfiltrering.
3. Fremgangsmåte ifølge krav 1, karakterisert ved at fraksjoneringen i punkt b) utføres ved hjelp av isoelektrisk fokusering og omvendt fase HPLC.
