Den foreliggende opfindelse angår en analogifrem-gangsmåde til fremstilling af hidtil ukendte 1-(3-mercap-to-2-methylpropanoyl)-proly1-aminosyrederivater, optiske isomere deraf eller farmaceutisk acceptable salte deraf.
5 De fremstillede forbindelser har antihypertensiv virkning og anvendes som antihypertensive midler.
Ondettf et al. syntetiserede forskellige mer-captoalkanoylaminosyrer og afprøvede dem in vitro for hæmning af det angiotensin I-omdannende enzym.
10 Herved konstateredes det, at 1-(D-3-mercapto-2- -methylpropanoyl)-L-prolin (som de betegnede SQ-14225) er en særlig god inhibitor af det angiotensin I-omdannende enzym, jvf. Biochemistry, 16, 5484 (1977).
Ondetti et al. undersøgte også SQ-14225 og beslægtede 15 forbindelser og anførte, at forbindelsen SQ-14225 også har en kraftig inhiberende virkning in vivo på det angiotensin I-omdannende enzym samt har fremragende antihypertensiv virkning på hypertensive forsøgsdyr baseret på dens inhiberende virkning, jvf. Science 196, 20 441 (1977). SQ-14225 og beslægtede forbindelser er be skrevet i US patentskrift nr. 4.046.889 og DE offentliggørelsesskrift nr. 2.703.828.
Det har nu vist sig, at der fremkommer farmakologisk bedre derivater af SQ-14225 og beslægtede forbindelser, hvis 25 der dannes en ny peptidbinding ved indføring af visse aminosyrer i carboxylgruppen i prolindelen af SQ-14225 eller dens derivater, idet der herved tilvejebringes forbindelser, som har en mildere antihypertensiv begyndelsesvirk-ning og en mere langvarig antihypertensiv virkning end SQ-30 -14225 eller dens derivater.
Ved den foreliggende opfindelse tilvejebringes i overensstemmelse hermed en analogifremgangsmåde til fremstilling af hidtil ukendte 1-(3-mercapto-2-methylpropanoyl)- -prolyl-aminosyrederivater med den almene formel 35

R3 5 hvor R betyder et hydrogenatom eller en alkylgruppe med 1-2 carbonatomer, R^ betyder en alkanoylgruppe med 2-7 carbonatomer, R betyder en 2-methylthioethylgruppe, en benzylgruppe, en p-hydroxybenzylgruppe eller en 3-indo-lylmethylgruppe, optiske isomere deraf eller farmaceutisk 10 acceptable salte deraf.
Forbindelserne med formlen (I) og de farmaceutisk acceptable salte deraf har fremragende antihyper-tensiv virkning, hvilken hovedsagelig er baseret på forbindelsernes evne til at inhibere det angiotensin I-om-15 dannende enzym. Denne antihypertensive virkning er karakteriseret ved en svag begyndelse og en lang varighed. Følgelig har de her omhandlede forbindelser stor klinisk anvendelighed som antihypertensive midler med en stabil virkningsgrad. På grund af deres karakteristiske virk-20 ning, vil forbindelserne kunne fjerne kliniske problemer, som er forårsaget af akut hypotension fremkaldt af en hurtig begyndelsesvirkning, og reducere ulemper i forbindelse med hyppig dosering, som er nødvendiggjort af en kortvarig virkning.
25 Forbindelserne med formlen (I) indeholder tre a- symmetriske carbonatomer. Asymmetriske carbonatomer er carbonatomet i 2-stillingen i 3-mercapto-2-methylpropa-noylgruppen, carbonatomet i 2-stillingen i prolindelen og 3 carbonatomet, hvortil R er knyttet. Disse carbonatomer er 30 vist med asterisk i formel (I) anført ovenfor. Følgelig foreligger de her omhandlede forbindelser som stereoiso-mere eller blandinger deraf. Disse stereoisomere og blandinger deraf er omfattet af de her omhandlede forbindelser med den almene formel (I).

acetyl, propanoyl, butanoyl, 2-methylpropanoyl, pentanoyl, 3-methylbutanoy1, 2-methylbutanoyl, 2,2-dimethylpropanoyl, hexanoyl, 4-methylpentanoyl og heptanoyl.
Gruppen -NH-CHCOOR er afledt af en a-aminosyre, ' 3 5 R·5 som kan være phenylalanin, tyrosin, tryptophan og methio-nin.
Som anført ovenfor har forbindelserne med formlen (I) tre asymmetriske carbonatomer, dvs. carbonatomet i 10 2-stillingen i 3-mercapto-2-methylpropanoylgruppen, carbonatomet i 2-stillingen i prolindelen og carbonatomet, 3 hvortil R er knyttet. Konfigurationerne af forbindelserne ved disse asymmetriske carbonatomer kan enten være i D-, L- eller DL-formen. Generelt er konfigurationerne af 3 15 prolindelen og R fortrinsvis i L-formen, og konfigurationen af methylgruppen i 2-stillingen i 3-mercapto-2-me-thylpropanoylgruppen er fortrinsvis i D-formen.
Eksempler på anvendelige forbindelser med formlen (I) er anført nedenfor. De første to forbindelser foretræk-20 kes.

Når R i formel (I) er et hydrogenatom, kan forbindelserne med formlen (I) også foreligge i form af salte 35 med uorganiske eller organiske baser. Saltene med uorganiske baser omfatter alkalimetalsalte, f.eks. natrium- el-

Reaktionen (a) mellem forbindelsen med formlen (II) eller et ved carboxylgruppen reaktivt derivat deraf og forbindelsen med formlen (III) og reaktionen (b) mellem forbindelsen med formlen (IV) eller et ved carboxylgrup-10 pen reaktivt derivat deraf og forbindelsen med formlen (V) kan gennemføres i overensstemmelse med kendte amide-ringsfremgangsmåder inden for peptidfremstillingen. Disse fremgangsmåder er f.eks. beskrevet detaljeret i Methoden der Organischen Chemie (Houben-Weyl), Vol. XV, del I og 15 li (1974).
Reaktionerne (a) og (b) gennemføres sædvanligvis ved, at de to udgangsforbindelser kontaktes i et indifferent opløsningsmiddel. Det indifferente opløsningsmiddel bør vælges afhængigt af typen af 20 udgangsforbindelserne o.s.v. Som eksempler kan nævnes tetrahydrofuran, dioxan, chloroform, dichlormethan, ethylacetat, acetone, acetonitril, dimethylformamid, dimethylsulfoxid, pyridin og vand. Disse opløsningsmidler anvendes enten alene eller i blanding. Hvis det er nødvendigt gennemføres reaktionerne i nærværelse af baser. Specifikke eksempler på baser er alkalimetal-hydrogencarbonater såsom natrium- og kaliumhydrogencar-bonat, alkalimetalcarbonater såsom natrium- og kalium- carbonat, alkalimetalhydroxider såsom natrium- og kalium-30 hydroxid og organiske baser såsom triethylamin, tri-n--butylamin, N-methylmorpholin og dicyclohexylamin.
Forbindelsen med formlen (ΙΙΙ) eller (V) kan anvendes i form af et syreadditionssalt. I dette tilfælde kan forbindelsen med formlen (III) eller (V) fås i den frie form ved tilsætning af en ækvivalent mængde af den førnævnte base til reaktionssystemet.

Reaktionstemperaturen afhænger af typen af udgangsforbindelserne o.s.v. men ligger sædvanligvis i området fra ca. -50 til ca. 60°C.
Forholdet mellem forbindelsen med formlen (III) 5 og forbindelsen med formlen (II) eller et reaktivt derivat deraf og forholdet mellem forbindelsen med formlen (V) og forbindelsen med formlen (IV) eller et reaktivt derivat deraf er ikke kritisk og kan varieres bredt afhængigt af typen af de anvendte udgangsmaterialer.
10 Generelt kan der med fordel anvendes ca. 1 til ca. 1,5 mol af forbindelsen med formlen (III) pr. mol af forbindelsen med formlen (II) eller et reaktivt derivat deraf og ca. 1 til ca. 1,5 mol af forbindelsen med formlen (IV) eller et reaktivt derivat deraf pr. mol af forbin-15 .delsen med formlen (V).
Når forbindelsen med formlen (II) eller (IV) som sådan skal omsættes med forbindelsen med formlen (III) eller (V), gennemføres reaktionen hensigtsmæssigt i nærværelse af et koblingsmiddel såsom 20 dicyclohexylcarbodiimid, Ν,Ν'-carbonyldiimidazol,
Woodward-reagens K eller l-ethoxycarbonyl-2--ethoxy-1,2-dihydroquinolin.
Reaktive carboxylgruppederivater af forbindelserne med formlen (II) eller (IV) omfatter alle 25 reaktive derivater, som normalt anvendes inden for peptidfremstilling. Som specifikke eksempler kan nævnes blandede anhydrider dannet med syrer såsom alkylcarbonsyrer, f.eks. ethylcarbonsyre eller iso-butylcarbonsyre, arylcarbonsyrer, f.eks. phenylcar-30 bonsyre, alkylcarboxylsyrer, f.eks. isovaleriane- eller pivalinsyre, phosphorsyrer, f.eks. diethylphos-phorsyre, og sulfonsyrer, f.eks. methansulfonsyre eller p-toluensulfonsyre, reaktive estere såsom p--nitrophenyl-, 2,4,5-trichlorphenyl-, pentachlorphe-35 nyl-, p-nitrophenylthio-, cyanomethy1-, N-hydroxy- piperidin-, N-hydroxysuccinimid- og N-hydroxyphthalimid- 7

De blandede anhydrides eller reaktive estere er særligt foretrukne som de reaktive derivater af forbindelsen med formlen (II) og 5 syrehalogeniderne (især syrechloriderne), de blandede an-hydrider og reaktive estere som de reaktive derivater af forbindelsen med formlen (IV). Disse reaktive derivater kan fremstilles i overensstemmelse med konventionelle fremgangsmåder.
10 Forbindelser med formlen (I), hvor R er erstattet af en tert.butylgruppe, kan nemt omdannes til de tilsvarende forbindelser med formlen (I), hvor R er hydrogen, f.eks. ved behandling af forbindelserne med trifluoreddikesyre og anisol.
15 Forbindelserne med formlen (I) fremstillet ved de ovennævnte reaktioner kan isoleres og adskilles ved konventionelle fremgangsmåder såsom ekstraktion, chromatografering og omkrystallisation.
Udgangsforbindelsen (II), (III), (IV) eller 20 (V) anvendt ved de ovenfor beskrevne fremgangsmåder kan være en racemisk blanding eller en af de stereoiso-mere. For at opnå den ønskede stereoisomer bør kon- 3 figurationerne af prolindelen og R i udgangsforbindelsen (II) , (III) eller (V) fortrinsvis være identiske 25 med konfigurationerne i de ønskede slutprodukter.
Imidlertid kan konfigurationen af methylgruppen i 2-stillingen i 3-mercapto-2-methylpropanoylgruppen i udgangsforbindelsen (II) eller (IV) være i DL-formen, og stereoisomerene dannet i produktet kan adskilles 30 ved en konventionel chromatografisk eller fraktioneret krystallisationsfremgangsmåde.
Forbindelser med formlen (I), hvor R er et hydrogenatom, danner salte med forskellige uorganiske eller organiske baser. Saltdannelsen kan gennemføres på gængs 35 måde.
Mere specifikke fremgangsmåder til fremstilling af de her omhandlede forbindelser vil fremgå af de nedenstående eksempler.

Nogle af forbindelserne med formlen (II) anvendt som udgangsmateriale i reaktion (a) er kendte. Andre forbindelser, som er omfattet af formlen (II) kan f.eks. nemt fremstilles i overensstemmelse med fremgangsmåderne 5 beskrevet i US patentskrift nr. 4.046.889, DE offentliggørelseskrift nr. 2.703.828 og Biochemistry 16, 5484 (1977). Odgangsforbindelserne med formlen (III) er kommercielt tilgængelige eller kan nemt fremstilles ved kendte fremgangsmåder.
10 Nogle af forbindelserne med formlen (IV) an vendt som udgangsmateriale i reaktion (b) er kendte, og andre, hidtil ukendte forbindelser, som er omfattet af formlen (IV), kan f.eks. fremstilles i overensstemmelse med fremgangsmåderne beskrevet i US patentskrift nr. 4.046.889 15 og DE offentliggørelsesskrift nr. 2.703.828. Udgangsforbindelserne med formlen (V) kan nemt fremstilles ud fra prolin- og α-aminosyrer ifølge gængse fremgangsmåder inden for peptidfremstillingen.
De her omhandlede forbindelser med formlen (I) 20 og de farmaceutisk acceptable salte deraf har fremragende antihypertensiv virkning, hvilken hovedsagelig er baseret på forbindelsernes evne til at inhibere det angiotensin I-omdannende enzym.
Resultaterne af farmakologiske tests på nogle 25 typiske eksempler af de her omhandlede forbindelser er anført nedenfor. Følgende forbindelser er anvendt i forsøgene:

Kontrolforbindelser 1: 1-(D-3-mercapto-2-methylpropanoyl)-L- -prolin (SQ-14225) 2: 1-(D-3-mercapto-2-methylpropanoyl)-L- 10 -prolinamid beskrevet i US patentskrift nr. 4.046.889 3: 1-(D-3-acetylthio-2-methylpropanoyl)-L- -prolinamid (hidtil ukendt forbindelse, som er omfattet af den almene formel i US patent-15 beskrivelse nr. 4.046.889).
Test 1; Antihypertensiv virkning
Hanrotter (stamme STD Wistar), som er ca. 8 uger gamle og vejer 185-195 g, bedøves med ether, 20 og en sølvklemme, som er 0,20 mm bred, anbringes på den venstre nyrearterie gennem et sidesnit. Den kontralaterale nyre og nyrearterien holdes intakt.
Dyrene forsynes med et kommercielt rottefoder og ledningsvand ad libitum.
25 Disse forsøgsdyr (Goldblatt dobbelt-nyre renal- hypertensive rotter) menes at have et typisk tilfælde af hypertension, som afhænger af den forøgede aktivitet i det renin-angiotensine system, jvf. Science, 188, 1316 (1975).
30 Dyrene anvendes 10-21 dage efter afklemningen, når blodtrykkene nærmer sig 180-220 mm Hg. Forsøgsforbindelserne indgives oralt til dyrene i en dosis på 0,1 mmol/kg legemsvægt. Blodtrykkene måles lige før og 1, 2, 3, 5, 7, 12, 14, 16, 18, 20, 22 og 24 timer 35 efter indgiften ved haleplethysmografimetoden uden bedøvelse.
I den nedenstående tabel er anført forsøgsdyrenes begyndelsesblodtryk, tidspunktet, på hvilket den mak- 148711 ίο O simale hypotensive virkning konstateres (tidspunktet for optimal virkning) og forandringerne i blodtryk både ved tidspunktet for optimal virkning og 7 timer efter indgiften. Omfanget af den hypotensive virkning ved 7 timer anses for at være 5 en god målestok til bedømmelse af varigheden af forsøgsforbindelsernes virkning.

Det fremgår af den ovenfor anførte tabel, at for 3 bindelserne A-F fremkalder et maksimalt hypotensivt re 4 spons (en formindskelse af begyndelsesblodtrykket på 15-18%) 5 5 timer efter indgiften, hvorimod forbindelse 1 (SQ-14225) 6 udviser et maksimalt respons (18% formindskelse) 1 time 7 efter indgift. 7 timer efter indgiften giver forbin 8 delserne A-F betydelige hypotensive responser 9 med en formindskelse af begyndelsesblodtrykket på 10 ca. 13-17%, medens forbindelse 1 (SQ-14225) ud- 11 viser en formindskelse af begyndelsesblodtrykket på kun 8%. Disse data indicerer, at den hypotensive 11 148711 o styrke af forbindelserne A-F kan sammenlignes med styrken af forbindelse 1 (SQ-14225), og at forbindelserne A-F har en mildere hypotensiv begyndelsesvirkning og en længere varighed end 5 forbindelse 1 (SQ-14225). Derimod reducerer for bindelse 2 (et simpelt amidderivat af SQ-14225) og forbindelse 3 (et simpelt amidderivat af S--acetyl SQ-14225) ikke blodtrykket.
Fig. 1 og 2 på tegningen viser tidsforløbene 10 med hensyn til forandringer i blodtrykket hos de dobbelt-nyre renalhypertensive rotter efter en enkelt oral indgift (0,015 mmol/kg og 0,1 mmol/kg) af forbindelserne A og B og forbindelse 1 (SQ-14225).
I hver fig. viser ordinaten forandringer i blod-15 trykket (mm Hg), og abscissen angiver tiden (timer), som er forløbet efter den orale indgift. Hvert punkt angiver en middelværdi baseret på seks dyr, og de lodrette linjer indicerer standardfejl.
Disse grafiske afbildninger viser klart, at 20 forbindelserne A og B har en mildere og mere pro- traheret antihypertensiv virkning end forbindelse 1 (SQ-14225).
Test 2: Toksicitet
Der anvendes hanmus (stamme STD-ddY), som vejer 25 21-24 g. Forbindelserne A og B har, når de indgives oralt til musene, LD50-værdier på mere end 6 g/kg legemsvægt og, når de indgives intravenøst, LD,^-værdier på mere end 1 g/kg legemsvægt. Disse data viser, at toksiciteten af forbindelserne A og B er 30 meget svag.
De ovennævnte forsøgsresultater påviser, at forbindelserne med formlen (I) og de farmaceutisk acceptable salte deraf har fremragende antihypertensiv virkning med svag toksicitet, og for-35 bindeiserne kan derfor anvendes som antihyper tensive midler til behandling af hypertension hos •t.

12 148711 pattedyr, herunder mennesker. Forbindelserne er især anvendelige til behandling af hypertension, som er forårsaget af en forøget aktivitet i det renin-angiotensine system.
5 De her omhandlede forbindelser kan indgives oralt, parenteralt eller intrarektalt, dog fortrinsvis oralt. Dosismængden af forbindelserne med formlen (I) eller de farmaceutisk acceptable salte deraf kan varieres afhængigt af typen af 10 den antihypertensive forbindelse, indgiftsmetoden, og patientens tilstand, alder o.s.v. Den samlede dagsdosis er sædvanligvis på 0,1-200 mg/kg legemsvægt, fortrinsvis 0,2-50 mg/kg legemsvægt ved indgift til mennesker. Lægemidlet indtages én eller to gange 15 dagligt i de anførte samlede doser.
Sædvanligvis indgives forbindelsen med formlen (I) eller et farmaceutisk acceptabelt salt deraf til et individ i form af et farmaceutisk præparat indeholdende en terapeutisk effektiv og ikke-toksisk 20 mængde af den nævnte forbindelse. Det farmaceutiske præparat formuleres ved blanding af forbindelsen med formlen (I) eller et farmaceutisk acceptabelt salt deraf med et farmaceutisk acceptabelt bærestof. Egnede bærestoffer er sådanne, som sædvanligvis anvendes ved 25 formuleringen af lægemidler, og som ikke reagerer med forbindelserne med formlen (I) eller saltene deraf.
Som specifikke eksempler på sådanne bærestoffer kan nævnes lactose, stivelse, saccharose, mikrokrystallinsk cellulose, natriumcarboxymethylcellulose, calcium-30 carboxymethylcellulose, methylcellulose, gelatine, aka- sia, hydroxypropylcellulose, hydroxypropylmethylcellu-lose, polyvinylpyrrolidon, lys vandfri kiselsyre, mag-nesiumstearat, talkum, titandioxid, sorbitan— fedtsyreestere, glycerider af mættede fedtsyrer, 35 makrogol, propylenglycol og vand. Det farmaceutiske præparat kan foreligge i forskellige dosis- J9

13 148711 former såsom tabletter, kapsler, granulat, fint granulat, pulvere, sirupper, suppositorier og injek-tionspræparater, som formuleres på gængs måde. Præ’-· parater på flydende form skal kunne opløses 5 eller suspenderes i vand eller andre egnede bærestoffer umiddelbart før anvendelsen. Tabletter kan overtrækkes på kendt måde. Om ønsket, kan det farmaceutiske præparat indeholde smagsstoffer, aromastoffer, konserveringsmidler, puffere og salte, 10 som gør præparatet isotonisk.
Sædvanligvis indeholder det farmaceutiske præparat mindst 0,5%, fortrinsvis 1-60% af forbindelsen med formlen (I) eller et farmaceutisk acceptabelt salt deraf som aktiv bestanddel. Præparatet kan også 15 indeholde andre terapeutisk virksomme forbindelser.
Opfindelsen illustreres mere detaljeret i de nedenstående eksempler. Forbindelserne i parentes efter angivelsen af smeltepunkter er omkrystallisationsopløsningsmidler .

Eksempel 1 1- (D-3-Acetylthio-2-methylpropanoyl) -L-prolyl-L-phenyl-alan.inethylester 25 0,39 g N-methylmorpholin sættes til en opløs ning af 1,0 g 1-(D-3-acetylthio-2-methylpropanoyl)--L-prolin i 20 ml tørt tetrahydrofuran. Den dannede opløsning omrøres og afkøles ved -20 til -15°C. Opløsningen henstilles ved stuetemperatur i 1 time, hvorpå den koncentreres til tørhed under for-20 mindsket tryk. Remanensen krystalliseres fra diethyl-ether. De frafiltrerede krystaller omkrystalliseres fra ethanol/n-hexan,hvorved fås 1,8 g af den ovennævnte forbindelse med smp. 155-156°C.

Remanensen chromatograferes på 5 silicagel med chloroform-methanol (99:1), hvorved fås 4,2 g 1-(D-3-acetylthio-2-methylpropanoyl)-L-prolyl-L--tyrosin-tert.-butylester som en viskos olie. cm_1: 3250' 1715» 1670, 1610.
2,0 g 1-(D-3-acetylthio-2-methylpropanoyl)-L-prolyl-10 -L-tyrosin-tert.butylester opløses i en blanding af 18 ml anisol og 37 ml trifluoreddikesyre. Opløsningen henstår ved stuetemperatur i 1 time og koncentreres derpå til tørhed ved formindsket tryk. Den fremkomne remanens chromatograferes på silicagel med chloroform-methanol (98:2), 15 hvorved fås 1,4 g af den ovennævnte forbindelse.

Eksempel 7 20 1-(D-3-Acetylthio-2-methylpropanoyl)-L-prolyl-L- -phenylalanin
Til en omrørt opløsning af 1,0 g L-prolyl--L-phenyl-alanin-tert.-butylester (fremstillet ved fremgangsmåden beskrevet i Ann. Chem. 680, 132-141 25 (1964)) i dichlormethan ved ca. -30°C sættes 0,32 g N-methylmorpholin, hvorpå der dråbevis tilsættes 0,57 g 3-DL-acetylthio-2-methylpropanoylchlorid.
Blandingen omrøres ved den samme temperatur i 30 minutter og derpå ved stuetemperatur i 1 time. Reak-30 tionsblandingen vaskes successivt med 4% natriumhydro gencarbonat, vand, 10% citronsyre og vand, tørres over natriumsulfat og koncentreres under formindsket tryk.
Remanensen chromatograferes på silicagel med chloroform, hvorved fås 1-(3-DL-acetylthio-2-methylpropanoyl)-35 -L-prolyl-L-phenylalanin-tert.-butylester.

Den således fremstillede forbindelse opløses i en blanding af 10 ml anisol og 20 ml trifluoreddikesyre. Opløsningen henstilles ved stuetemperatur i 1 time, hvorpå den koncentreres til tørhed under for-5 mindsket tryk.
Remanensen opløses i en lille mængde ethanol, og der tilsættes en passende mængde n-hexan. De udfældede krystaller samles og omkrystalliseres gentagne gange fra ethanol/ii-hexan, hvorved fås 0,36 g af oven- o 25 10 nævnte forbindelse med smp. 155-156 C. Eet]D *= -81,3° (c = 1,02, ethanol).
Eksempel 8 15 1-(D-3-Acetylthio-2-methylpropanoyl)-L-prolyl- -L-phenylalanin
Til en opløsning af 0,52 g L-prolyl-L-phenyl-alanin i 2 ml IN natriumhydroxid afkølet i isbad sættes 0,36 g 3-DL-acetylthio-2-methylpropanoylchlorid 20 og 1 ml 2N natriumhydroxid med kraftig omrøring.
Efter omrøring i 3 timer ved stuetemperatur vaskes opløsningen med diethylether, syrnes og ekstraheres med ethylacetat. Det organiske lag tørres over natriumsulfat og koncentreres til tørhed under for-25 mindsket tryk. Den fremstillede 1-(3-DL-propanoylthio-2-methyl-propanoyl)-L-prolyl-L-phenylalanin-tert.-butylester 5 opløses i en blanding af 10 ml anisol og 20 ml trifluor- eddikesyre. Opløsningen henstilles ved stuetemperatur i 1 time og koncentreres til tørhed under formindsket tryk. Remanensen chromatograferes på silicagel med chloroform-methanol (98:2) til dannelse af l-(3-DL-propa-10 noylthio-2-methylpropanoyl)-L-prolyl-L-phenylalanin.
Den således fremstillede diastereomere blanding opløses i en passende mængde diethylether. Opløsningen podes med krystaller af den ovennævnte forbindelse og henstilles i køleskab. De udfældede 15 krystaller samles og omkrystalliseres gentagne gange fra diethylether/petroleumsether, hvorved fås 0,15 g af ovennævnte forbindelse med smp. 107-114°C.

Eksempel 10 1-(D-3-Propanoylthio-2-methylpropanoyl)-L-prolyl--L-phenylalanin 0,54 g L-prolyl-L-phenylalanin-tert.-butyl-25 ester sættes til en opløsning af 0,30 g 3-DL-propanoyl- thio-2-methylpropansyre og 0,35 g dicyclohexylcar-bodiimid i 10 ml dichlormethan afkølet i isbad.
Blandingen omrøres i isbadet i 1 time og derpå ved stuetemperatur natten over, hvorpå den koncentreres 30 under formindsket tryk. Ethylacetat sættes til remanensen. Efter afkøling frafiltreres bundfaldet.
Filtratet vaskes, tørres og koncentreres som i eksempel 7. Remanensen chromatograferes som i eksempel 7, hvorved fås 1-(3-DL-propanoylthio-2-methyl-35 propanoyl)-L-prolyl-L-phenylalanin-tert.-butylester.

Den således fremstillede forbindelse behandles som beskrevet i det 2. afsnit af eksempel 9, hvorved fås 0,13 g af ovennævnte forbindelse.
Udgangsforbindelserne anvendt i eksempel 9 og 5 10 fremstilles som følger.
Fremstilling af 3-propanoylthio-2-methylpropansyre
En blanding af 50 g thiopropansyre og 34,1 g ίο methacrylsyre opvarmes på et dampbad i 3 timer og henstilles ved stuetemperatur natten over. Reaktionsblandingen destilleres under formindsket tryk, hvorved fås den ovennævnte forbindelse med kp. 135-138°C (2 mm Hg).
15 Fremstilling af 3-propanoylthio-2-methylpropanoylchlorid
En blanding af 19,7 g 3-propanoylthio-2-methyl-propansyre og 16 g thionylchlorid omrøres ved stuetemperatur natten over. Reaktionsblandingen destille-20 res under formindsket tryk, hvorved fås den ovennævnte forbindelse med kp. 135-137°C (35 mm Hg).
Forbindelse 3, som anvendes ved den farmakologiske test, fremstilles som følger.

Fremstilling af 1-(D-3-acetylthio-2-methylpropanoyl)-L-prolinamid 2,0 g 1-(D-3-acetylthio-2-methylpropanoyl)-L-30 -prolin og 0,78 g N-methylmorpholin opløses i dichlor-methan, og opløsningen afkøles ved ca. -30°C. Der tilsættes 1,2 g phenylchlorformiat med omrøring, og efter 5 minutter ledes ammoniak gennem blandingen i løbet af 2 minutter. Blandingen omrøres ved ca.
35 -30°C i yderligere 15 minutter, hvorpå der tilsættes

21 148711 10% saltsyre under afkøling. Den dannede blanding omrøres grundigt. Det organiske lag isoleres/ vaskes med 10% saltsyre og derpå med vand, tørres og koncentreres under formindsket tryk. Remanensen chromatogra-5 feres på silicagel med chloroform-methanol (99:1), hvor ved fås den ovennævnte forbindelse.

Claims (3)
1. Analogifremgangsmåde til fremstilling af l-(3--mercap to-2-methylpropanoy1)-prolyl-aminosyrederivater med den almene formel i “3 n R1_S_CH -CHCO-N-L· CONH-CHCOOR
2 P (I) 10 hvor R betyder et hydrogenatom eller en alkylgruppe med 1-2 carbonatomer, R1 betyder en alkanoylgruppe med 2-7 3 carbonatomer, R betyder en 2-methylthioethylgruppe, en ben-zylgruppe, en p-hydroxybenzylgruppe eller en 3-indolylme-thylgruppe, optiske isomere deraf eller farmaceutisk accep-15 table salte deraf, kendetegnet ved, at (a) en forbindelse med den almene formel ! O (II) Rx-S-CH,-CHCO-N-1— COOH 20 Δ hvor R1 har ovennævnte betydning, eller et ved carboxyl-gruppen reaktivt derivat deraf omsættes med en forbindelse med den almene formel H~N - CHCOOR' (III) ^ I O
