(54) Analogifremgangs måde til fremstil* ling af amider af pseudomonsyre.
Den foreliggende opfindelse angår en analogifremgangsmåde til fremstilling af hidtil ukendte amider af pseudomonsyre, hvilke amider har antibakteriel virkning mod visse grampositive og gramnegative organismer, især Haemophilis influenzae og Neisseria gonorrhoeae; forbindelserne har også god antimycoplasmal virkning. Forbindelserne er derfor værdifulde ved behandlingen af veterinærbakterielle infektioner og af speciel værdi for mennesker tt til behandling af bronkitis og veneriske sygdomme.

OO Den gængse behandling af gonorré har i mange år været at anvende ^ penicillinantibiotika. Visse stammer af gonococci er imidlertid mindre følsomme over for penicilliner, og graden af denne resistens er lidt efter lidt blevet større, med det resultat, at det er ble-Ω vet nødvendigt at anvende større doser penicilliner. Endvidere er 2 145823 der blevet rapporteret stammer, der producerer penicillinase, og som således er kraftigt resistente over for penicillinterapi. British Medical Journal (1976), side 963 kommenterer: "Udsigterne til, at gonorré kan kontrolleres, er nu blevet radikalt forringet, efter at der er fremkommet ildevarslende rapporter om eksistensen af kraftigt resistente stammer, hvis resistens skyldes, at de danner penicillinase, det penicillin-ødelæggende enzym, som findes hos mange andre bakterieslægter. Dette er en helt ny udvikling, hvis konsekvenser kan blive katastrofale".
Det har vist sig, at de nedenfor angivne amider af pseudomonsyre har kraftig aktivitet mod mange organismer, herunder Neisseria gonorrhoeae, og da amiderne er helt ubeslægtede med antibiotika af β-lactamtypen (herunder penicilliner og cephalosporiner), forbliver de komplet upåvirkede af penicillinase.
Pseudomonsyre er den E-isomere af strukturen I

“yVvA, T co2(ch2)8co2h OH 1 og i britisk patentskrift nr. 1.395,907 er det beskrevet, at den har antibakteriel virkning. Det har nu vist sig, at de her omhandlede amider af pseudomonsyre har en lavere serumbinding end syren selv og er mere aktive mod visse organismer, især Neisseria gonorrhoeae.
Opfindelsen angår derfor en analogifremgangsmåde til fremstilling af carboxylsyreamider med den almene formel II

Egnede alkylgrupper R og R er methyl, ethyl, n- og isopropyl og n-, iso- og tert.butyl.

Som specifikke amidgrupper -CO.NR R skal nævnes det usubstituerede amid (-CONI^), N-methylamidet, N,N-dimethylamidet og N,N-diethyl-amidet.
Hensigtsmæssigt omsættes et reaktivt derivat af pseudomonsyre med 12 en amin med den almene formel R R .NH. Foretrukne reaktive derivater er blandede anhydrider dannet med f.eks. isobutylchlorformi-at, ethylchlorformiat, pivaloylchlorid og andre reagenser til fremstilling af blandede anhydrider. Andre N-acyleringsderivater af syren med den almene formel I er aktiverede estere såsom estere med cyanomethanol, p-nitrophenol, 2,4-dinitrophenol, thiophenol, halogenphenol, herunder pentachlorphenol, monomethoxyphenol eller 8-hydroxyquino1in; eller amider såsom N-acylsacchariner eller N-acyl-phthalimider} eller en alkylideniminoester fremstillet ved omsætning af syren med den almene formel I med en oxim.
Visse aktiverede estere, f.eks. den ester, der dannes med 1-hydroxy-benztriazol eller N-hydroxysuccinimid, kan fremstilles in situ ved omsætning af syren med den tilsvarende hydroxyforbindelse i nærværelse af et carbodiimid, fortrinsvis dicyclohexylcarbodiimid.
Andre reaktive derivater af pseudomonsyre er det reaktive mellemprodukt, der dannes ved omsætning in situ med et kondenseringsmiddel såsom et carbodiimid, f.eks. Ν,Ν-diethyl-, -dipropyl- eller -diisopropylcarbodiimid, N,N'-dicyclohexylcarbodiimid eller N-ethyl--N'-dimethylaminopropylcarbodiimid; en egnet carbonylforbindelse, f.eks. N,N1-carbonyldiimidazol eller N,Ν'-carbonylditriazol.
Egnede opløsningsmidler til fremgangsmåden ifølge opfindelsen er 4 145823 inerte aprote opløsningsmidler, f.eks. tetrahydrofuran, methylendi-chlorid og Ν,Ν-dimethylacetamid. Reaktionen udføres almindeligvis ved lav temperatur, f.eks. mellem -25°C og 0°C, fortrinsvis ved ca. -10°C.
De ved fremgangsmåden ifølge opfindelsen fremstillede antibiotisk virksomme forbindelser kan til administration formuleres på en hvilken som helst bekvem måde til anvendelse i human- eller veterinærmedicinen, i analogi med andre antibiotika, og der kan således med forbindelserne fremstilles farmaceutiske præparater indeholdende en forbindelse med den almene formel II sammen med en farmaceutisk bærer eller excipiens.
Præparaterne kan formuleres til en hvilken som helst administrations-vej, men den orale administration foretrækkes. Præparaterne kan være i form af tabletter, kapsler, pulvere, granuler, pastiller eller flydende præparater, f.eks. orale eller sterile parentera-le opløsninger eller suspensioner.
..Præparaterne kan indeholde fra 0,1 til 99 vægtprocent, fortrinsvis fra 10 - 60 vægtprocent, af det aktive materiale, afhængigt af administrationsmåden. Når præparaterne omfatter enhedsdoser, vil hver enhed fortrinsvis indeholde 50 - 500 mg af den aktive bestanddel. Den dosis, der anvendes til behandling af voksne mennesker, vil fortrinsvis ligge i området fra 100 - 3000 mg pr. dag, f.eks.
1500 mg pr. dag, afhængigt af administrationsvejen og administrationens hyppighed.

Fremgangsmåden ifølge opfindelsen belyses nærmere ved følgende eksempler:
Eksempel 1.
Pseudomonsyre-N-methylamid.
522 mg (1 millimol) natriumpseudomonat opløses i tørt, destilleret N/N-dimethylacetamid (10 ml), og opløsningen afkøles til -10°C. Der tilsættes isobutylchlorformiat (0,13 ml; massefylde = 1,04; 1 millimol), og opløsningen holdes ved -10°C i 10 minutter. En 1% w/v opløsning af methylamin i vand (3,1 ml; 1 millimol) tilsættes hurtigt, og opløsningen omrøres i 1 time, medens temperaturen langsomt stiger til stuetemperatur. Blandingen hældes ud i ca. 50 ml vand, mættes med natriumchlorid og ekstraheres med 4 x 20 ml ethylacetat. Ethylacetatekstrakten vaskes med en mættet saltopløsning, tørres o-ver magnesiumsulfat og filtreres, og filtratet inddampes til tørhed i vakuum (304 mg). Tyndtlagschromatografi i chloroform/methanol 9:1 viser én hovedplet, R^-værdi = 0,3, med et spor af pseudomonsyre, R^-værdi = 0,21, og meget små spor ved R^-værdi 0,40 og 0,47. Dette materiale chromotograferes på en 2 mm tyk siliciumdioxidplade HF 254 20 x 20 cm og elueres med 15% methanol i chloroform. Båndet ved R^-værdi = 0,35 (under U.V.) fjernes og elueres fra sili-ciumdioxidet med 20% methanol i chloroform. Det resulterende olie-agtige produkt opløses i ethylacetat og vaskes grundigt med vandig natriuiribicarbonatopløsning. Den organiske fase tørres over magnesiumsulfat, filtreres og inddampes i vakuum, hvorved fås produktet i form af en olie (127 mg) .
Tyndtlagschromatografi viser 1 komponent, R^-værdi = 0,33 i chloroform/methanol 9 : 1, <5H(CDC13) 0,92 (3H,d, 12-Me) , 1,20 (3H,d, 13-Me) , 9,30 (methylenkonvolut); 2,18 (3H,s, vinylisk CH^), 2,70 (m, epoxid-protoner), 2,76 (3H,d,N-Me), 5,75 (IH, bred s, vinylisk H), 6,23 (-NH-); C (CDC13) 174,20 (l'-C,s), 166,95 (1-C,s), 156,95 (1-C,s), 156-97 (3-C,s), 117,71 (2-C,d), 75,07 (5-C,d), 71,47 (13-C,d), 70,60 (7-C,d), 69,11 (6-C,d), 65,43 (16-C,t), 63,83 (9'-C,t), 61,38 (11-C,d), 55,710 (10-C,d), 42,94 (4-C og 12-C, tod's), 39,68 (8-C,d), 36,74 (2'-C,t), 31,72 (9-C,d), 29,139 (4',5' og 6'-C), 6 145823 28,63 (8'-CO), 26,35 (N-Me,q), 26,00 (7'-C,d), 25,73 (3'-C), 20,84 (14-C,q), 19,23 (15-C,q) og 12,75 (17-C,q); Umav (CHCI3), 3400 (OH, kraftig), 3300 (NH, kraftig) 1690 (C = O, kraftig, ester), 1650 og 1540 (C = O, amid I, og C = C), 1510 (amid II) cm"1 Xm 222 nm (ε = m, 10.700); m/e 513 (M ), 495 (-H20) , 269 (^-CH2C02-(CH2) 8CONHCH3) , 227. Eksempel 2.
Ps eudomonsyreamid.
522 mg (1 millimol) natriumpseudomonat opløses i 10 ml tørt destilleret Ν,Ν-dimethylacetamid, og opløsningen afkøles til -15°C. Der tilsættes 0,13 ml isobutylchlorformiat (massefylde = 1,04, 1 millimol), og temperaturen holdes i 10 minutter ved -15°C. Der tilsættes 2,0 ml af en 1%'s vandig ammoniakopløsning (>1 millimol), og blandingen omrøres i 1 1/2 time ved stuetemperatur. Blandingen fortyndes med vand til ca. 75 ml, mættes med natriumchlorid og ekstraheres med 4 x 15 ml ethylacetat. Ethylacetatfasen vaskes med saltopløsning og vandig natriumbicarbonatopløsning, tørres over magnesiumsulfat og filtreres, og filtratet inddampes til en svagt farvet olie (269 mg). Tyndtlagschromatografi i chloroform/methanol 9:1 viser 1 hovedkomponent ved R^-værdi = 0,25 og spor af urenheder ved 0,18 og 0,3. Dette materiale påføres på en 20 x 20 cm, 2 mm tyk præparativ siliciumdioxidplade og elueres med 15% methanol i chloroform.
Båndet svarende til hovedkomponenten fjernes og elueres fra sili-ciumdioxidet med 50% methanol i chloroform. Efter inddampning fås produktet i form af en farveløs olie (164 mg). Tyndtlagschromatografi viser 1 komponent i chloroform/methanol 9:1 ved R^-værdi 0,4. δΗ (CDC13) 0,90 (3H,d,12-Me), 1,19'(3H,d,13-Me), 1,31 (me-thylenkonvolut), 2,19 (3H,s, vinylisk Me), 2,70 (multiplets, epoxid-protoner og 4-CH2), 4,08 (2H,t,9'-CH), 5,76 (IH,s, vinylisk H) og 5,83 (2H, bred C0NHo) ; i> v (CHCU) 3450 (OH, kraftig), 3350 (NH, iucix «3 kraftig), 1700 (C = O, ester), 1650 - 1670 (amid I, og C = C), 1600 (amid II)cm-1; λ 222 nm (em 9.230); [a]i?0 = -5,02° (C,0,6

Eksempel 3.
Pseudom0nsyre-N,N-dimethylamid.
2,09 g (4 millimol) natriumpseudomonat opløses i 20 ml tørt destilleret Ν,Ν-dimethylacetamid, og opløsningen afkøles under omrøring til -10°C. Der tilsættes 0,53 ml (4 millimol) isobutylchlor-formiat, idet temperaturen holdes på -10°C i 10 minutter. En opløsning af dimethylamin i vand (1M, 4,2 ml) tilsættes, og opløsningen omrøres ved 0°C i en halv time og ved stuetemperatur i 2 timer. Opløsningen inddampes til tørhed i vakuum, og remanensen opløses i ethylacetat, som vaskes med natriumbicarbonatopløsning og saltopløsning, tørres over magnesiurasulfat, filtreres og inddampes i vakuum til en olie (1,197 g). Tyndtlagschromatografi og højtryks-væskechromatografi bekræfter, at forbindelsen er homogen. Olien begynder at krystallisere, og ved triturering med tør ether fås et hvidt fast stof (776 mg), smeltepunkt 82 - 85°C. δΗ (CDCl^) 5,70 (IH,s, vinylisk H), 4,03 (2H,t,9'-CH.2) , 2,98 og 2,90 (to 3H,s,CON(CH3)2), 2,70 (multiplets, epoxidprotoner, H^q og 4-CH2) , 2,17 (3H,s, vinylisk CH3), 1,31 (methylenkonvolut), 1,19 (3H,d, 13-CH3) og 0,91 (3H,s, 12-CH,); (CHC1,), 3450 (OH, kraftigt), 2950, 1710 (ester C = O) 1615 (bred, C = C og C = O amid)cm ; λ λ λ ΙΪΙαΧ (EtOH) 219 (em = 14.700)nm; [α]ρ = -6,88° (c = 1,0 i CHC13); m/e 527 (M+), 509 (-H20), 465 (509 - N(CH3)2). Fundet: M+ 527.347132; beregnet for C2gH4gN08 M+ 527.347132.

c) Aktivitet mod mycoplasma.
Tabel III viser antimycoplasmale aktiviteter (udtrykt som MlC-vær-dier [minimale inhiberende koncentrationer]) for de ifølge eksempel 2 og 3 fremstillede forbindelser og viser, at de begge besidder god antimycoplasmal aktivitet in vitro mod mycoplasmaer fra humane og veterinære kilder.
Den metode, der anvendes til bedømmelse af den antimycoplasmale aktivitet/ er følgende:
Den minimale inhiberende koncentration (MIC), som pseudomonsyre-amiderne udviser, bestemmes i "Microtitre"-plader ved en modifikation af metaboli-inhibitionstesten (Taylor-Robinson, 1967). Forbindelserne fortyndes i serie i sterilt afioniseret vand, således at der fås en række koncentrationer fra 250 - 0,5 ^ug/ml. Mycoplasmabouillon indeholdende 1% w/v glucose og 0,005¾ w/v phe-nolrødt tilsættes i en sådan styrke, at der kompenseres for dens 4 fortynding forårsaget af den vandige lægemiddelopløsning. Ca. 10 kolonidannende enheder af mycoplasma sættes til hver koncentration af lægemiddel. Lægemiddelfrie inficerede, ikke-inficerede og pH-kon-trolhuller medtages på hver plade. Pladerne tillukkes med cellofantape og inkuberes ved 37°C i 7 dage. MIC er den laveste koncentration af forbindelsen, som hindrer den af mycoplasmaernes metabolisering af glucose forårsagede farveændring i mycoplasmabouillonen.

Den grad af binding til humant serum, som de ifølge eksempel 1 og 2 fremstillede forbindelser udviser, sammenlignet med pseudomonsy-res serumbinding, bestemmes på følgende måde:
Testprøver blandes med humant serum og anbringes i visking-rør. Serumbindingen bestemmes ved ultrafiltrering, idet det resulterende filtrat styrkebestemmes efter en hul-i-plade-metode under anvendelse af Bacillus subtilis ATCC 6633 mod standarder, der er fremstillet i 0,05M phosphatpuffer, pH-værdi 7,0. Resultaterne fremgår af nedenstående tabel IV:
