- Die Peptide der vorliegenden Erfindung weisen stärkere antimikrobielle Wirkungen als herkömmliche Peptide auf und weisen die Wirkung bei hohen Salzkonzentrationen auf.
- TECHNISCHER HINTERGRUND
- Die vorliegende Erfindung betrifft biologisch aktive Peptide. Jedes Tier bzw. jeder Mensch auf der Erde besitzt Biophylaxe-Systeme, um sich gegen eine Infektion durch Viren oder Bakterien zu verteidigen oder davor zu schützen. Ein derartiges System ist eine unspezifische Immunität unter Verwendung von antimikrobiellen Peptiden.
- Antimikrobielle Peptide werden aufgrund der folgenden herausragenden Eigenschaften als neue Art Arzneistoff angesehen. Erstens zeigen antimikrobielle Peptide stärkere antimikrobielle Wirkungen als herkömmliche Antibiotika gegen ein breites Spektrum von Mikroorganismen. Zweitens weisen antimikrobielle Peptide eine hohe industrielle Anwendbarkeit auf, die für den menschlichen Körper vorteilhaft ist, da die antimikrobiellen Peptide eine antimikrobielle Wirkung gegen fremde Pathogene ohne Zerstörung der Wirtszellen zeigen. Drittens ist die Möglichkeit kleiner, eine Mikrobenresistenz zu entwickeln, da die antimikrobiellen Peptide ihre Wirkung durch einen Mechanismus zeigen, der vollständig von demjenigen herkömmlicher Antibiotika verschieden ist, welche ernste Probleme bei der Entwicklung von Resistenz aufweisen. Untersuchungen an antimikrobiellen Peptiden begannen durch Isolierung von Cecropin aus einem Insekt, das ein unterentwickeltes Immunsystem aufweist. Nach dem ersten Auffinden wurden Magainin, Bombinin aus Amphibien, Defensine aus Säugern isoliert. Die Untersuchungen an antimikrobiellen Peptiden werden aktiv vorgenommen, und bis heute sind etwa 2000 antimikrobielle Peptide aus Spezies im Bereich von Mikroorganismen bis Mensch identifiziert und mitgeteilt worden.
- Jedoch gibt es mehrere Hindernisse, um die oben erwähnten antimikrobiellen Peptide als Arzneistoffe zu entwickeln. Erstens wirken die herkömmlichen antimikrobiellen Peptide bei relativ hohen Konzentrationen. Beispielsweise beträgt im Fall von Magainin, einem antimikrobiellen Peptid, das aus der Epidermis von Amphibien isoliert wurde, die aktive Konzentration 50 – 200 μg/ml (Zasloff M. (1987), Proc. Natl. Acad. Sci. USA, 84: 5449–5453), obwohl es gegen gram-positive und gram-negative Bakterien und Pilze wirksam ist. Dieser Konzentrationsbereich ist ziemlich hoch, wenn man bedenkt, dass herkömmliche Antibiotika gegen einen spezifischen Mikroorganismus im Bereich von 0,1 – 1 μg/ml wirken. Zweitens ist die antimikrobielle Wirkung der antimikrobiellen Peptide gegenüber der Salzkonzentration empfindlich. Im Fall von Mukoviszidose, welche die menschliche Lunge befällt, war beispielsweise das antimikrobielle Peptid aufgrund einer abnormalen Erhöhung der Salzkonzentrationen an der Stelle des Befalls nicht wirksam (Goldmann, M. J. et al. (1997), Cell, 88: 553–560).
- Die vorliegenden Erfinder berichteten in Biochemical and Biophysical Research Communications 218, 408–413 (1996) über antimikrobielle Peptide, die aus der koreanischen Kröte isoliert wurden. Diese antimikrobiellen Peptide, die als Buforin I und Buforin II bekannt sind, zeigten starke antimikrobielle Wirkungen gegen ein breites Spektrum von Mikroorganismen, einschließlich gram-positiver und gram-negativer Bakterien und Pilze. Buforin I und Buforin II weisen auch antimikrobielle Wirkungen bei einer Konzentration von 1–4 μg/ml auf, was stärker ist als diejenige herkömmlicher antimikrobieller Peptide. Diese antimikrobiellen Peptide sind jedoch ebenfalls gegen Salzkonzentrationen empfindlich. Deshalb war es ein Wunsch, antimikrobielle Peptide zu entwickeln, die verstärkte antimikrobielle Wirkungen aufweisen und nicht gegen Salzkonzentrationen empfindlich sind, so dass sie antimikrobielle Wirkungen in vivo aufweisen.
- Die WO 97/18826 ist auf antimikrobielle Peptide gerichtet, die mit natürlich vorkommenden Protegrin-Peptiden verwandt sind. Die WO 96/28468 betrifft amphiphile Peptide mit verbesserten antimikrobiellen Eigenschaften. SWISSPROT:H2A1-YENLA (PO6897) offenbart eine Peptidsequenz, die das Histon H2A.1 von Xenopus laevis repräsentiert. Yi et al. (1996), FEBS letters, 398: 87–80, offenbaren die Struktur von Buforin II, wie durch NMR-Spektroskopie bestimmt. Kim et al. (1996), Biochemical and Biophysical Research Communications, 229: 381–387, offenbaren die Identifikation einer cDNA kontaktierenden Codierungsinformation für Buforin I mittels PCR. Park et al. (1995), Biochemical and Biophysical Research Communications, 218: 408–413, offenbart die Isolierung von Buforin I und die Isolierung von Buforin II, das von Buforin I abgeleitet ist, und teilt mit, dass diese Peptide gegen Salzkonzentrationen empfindlich sind. Die FR 2715847 offenbart Zusammensetzungen, die Nucleinsäuren und Oligopeptide enthalten, und ihre Verwendung in der Therapie und Gentherapie. Han et al. (1996), FASEB J10: 1621–1626, offenbarten ein Verfahren für die Überwachung spezifischer Peptidase-Aktivitäten in biologischen Proben auf der Grundlage der Konstruktion von synthetischen Polypeptid-Substraten, in denen Di- oder Triarginin-Sequenzen miteinander über eine oder mehrere andere Aminosäuren verknüpft sind, welche spezifisch von der zu bestimmenden Peptidase erkannt werden.
- OFFENBARUNG DER ERFINDUNG
- Es ist ein Ziel der vorliegenden Erfindung, neue biologisch aktive Peptide bereitzustellen.
- Ein weiteres Ziel der vorliegenden Erfindung ist es, Peptide bereitzustellen, die antimikrobielle Wirkungen gegen eine große Vielfalt von Mikroorganismen mit stärkeren antimikrobiellen Wirkungen aufweisen.
- Es ist ein weiteres Ziel der vorliegenden Erfindung, Peptide bereitzustellen, die bei einer Potenzierung der antimikrobiellen Wirkung unempfindlich gegen Salzkonzentrationen sind.
- KURZE BESCHREIBUNG DER ZEICHNUNGEN
-
1 ist eine Sekundärstruktur von Buforin II, die durch NMR-Spektrometrie in Anwesenheit von 50% Trifluorethanol als strukturbildendem Mittel bestimmt wurde. -
2 ist eine graphische Darstellung, welche die minimale Inhibierungskonzentration als Funktion einer Salzkonzentration zeigt. - DETAILLIERTE BESCHREIBUNG DER ERFINDUNG
- Die vorliegenden Erfinder haben gezeigt, dass die Sekundärstruktur von Buforin II eine statistische Knäuel- (1.–4. Rest), eine ausgestreckte Helix- (5.–10. Rest) und eine normale α-Helix- (11.–21. Rest) Struktur aufweist, ausgehend vom N-Terminus.
- In der Struktur von Buforin II weist die Peptidsequenz mit normaler α-Helix-Struktur (11.–21. Rest), d. h. PVGRVHRLLRK, eine starke antimikrobielle Wirkung auf. Die vorliegenden Erfinder haben identifiziert, dass ein Peptid, insbesondere ein Peptid, bei dem mindestens die Sequenz entfernt ist, welche die statistische Knäuel-Struktur bildet (1. bis 4. Rest), eine sehr starke antimikrobielle Wirkung aufweist. Deshalb besteht die Gruppe von Peptiden gemäß der vorliegenden Erfindung aus Peptiden mit der PVGRVHRLLRK-Sequenz. Die Sequenz PVGRVHRLLRK kann zusätzlich einen Gly-Rest am N-Terminus oder ein amidiertes Peptid am C-Terminus aufweisen.
- Eine weitere Gruppe von Peptiden gemäß der vorliegenden Erfindung umfasst ein Peptid mit einer Wiederholungseinheit [RLLR]n (n ist eine ganze Zahl zwischen 2 und 6) (wobei RLLR das spezielle Wiederholungsmuster ist, das in der Aminosäuresequenz von Buforin II gefunden wird) und vorzugsweise Peptide, in denen n = 3 – 5.
- Die Gruppe von Aminosäuresequenzen umfasst weiter RAGLQFPVG[RLLR]1, RAGLQFPVG[RLLR]2, RAGLQFPVG[RLLR]3, [RLLR]3, [RLLR]4, [RLLR]5 und RKGLQKLVGRVHRLLRK.
- Die Peptide gemäß der vorliegenden Erfindung können durch wohlbekannte Techniken auf dem Gebiet synthetisiert werden, beispielsweise unter Verwendung eines automatischen Peptid-Synthetisierers oder durch Verwendung von Gentechnik. Beispielsweise kann das Peptid erzeugt werden, indem man ein Fusionsgen konstruiert, das aus dem Fusionspartner und den Peptidgenen zusammengesetzt ist, dieses in einen Wirtsorganismus transformiert, das Fusionsprotein im Wirt exprimiert, das Fusionsprotein mit proteolytischem Enzym oder einem chemischen Mittel spaltet und das antimikrobielle Peptid reinigt. Für diesen Zweck kann beispielsweise eine DNA-Sequenz zwischen den Fusionspartner und die Peptidgene inseriert werden, um eine Sequenz einzuführen, die für Verarbeitungsstellen codiert, welche durch Proteasen, wie Faktor Xa und Enterokinase, oder durch chemische Mittel, wie CNBr und Hydroxylamin, gespaltet werden kann.
- Um eine DNA-Sequenz einzuführen, die für eine CNBr-Spaltungsstelle codiert, können beispielsweise der Fusionspartner und antimikrobielle Peptidgene im Raster durch Ligieren des Fusionspartner-Gens, das an seinem 3-Ende mit einem Restriktionsenzym verdaut worden ist, dessen Erkennungssequenz das Met-Codon (ATG) in seiner Erkennungssequenz enthält, wie AffIII, BsmI, BspHI, BspLU11I, NcoI, NdeI, NsiI, Ppu10I, SphI, StyI oder deren Isoschizomere, und des Peptidgens, das an seinem 5-Ende mit einem Restriktionsenzym verdaut worden ist, dessen Spaltungsstelle mit der Spaltungsstelle des Fusionspartners kompatibel ist, fusioniert werden. In einem weiteren Beispiel kann, um eine DNA-Sequenz einzuführen, die für eine Hydroxylamin-Spaltungsstelle codiert, eine DNA-Sequenz, die für Asn-Gly codiert, zwischen den Fusionspartner und die Peptidgene eingeführt werden. Beispielsweise können der Fusionspartner und die Peptidgene im Raster fusioniert werden, indem man das Fusionspartner-Gen, das an seinem 3-Ende mit einem Restriktionsenzym oder dessen Isoschizomer verdaut worden ist, dessen Erkennungssequenz das Asn-Codon in seiner Erkennungssequenz enthält, und das Peptidgen fusioniert, das an seinem 5-Ende mit einem Restriktionsenzym verdaut worden ist, dessen Spaltungssequenz, die das Gly-Codon enthält, im Raster mit dem 3-Ende des Fusionspartners durch ein kompatibles klebriges oder glattes Ende fusioniert werden kann.
- Die Genstruktur in der vorliegenden Erfindung kann durch Klonen derselben in einen Expressionsvektor, wie ein Plasmid, Virus oder anderes herkömmliches Vehikel, in welches das Gen inseriert oder einverleibt werden kann, in eine Wirtszelle eingeführt werden.
- Die Peptide gemäß der vorliegenden Erfindung enthalten C-terminate amidierte Formen.
- Die Peptide gemäß der vorliegenden Erfindung zeigen starke antimikrobielle Wirkungen gegen eine große Vielfalt von Mikroorganismen, einschließlich gram-negativer und gram-positiver Bakterien, Pilze und Protozoen.
- Die Peptide gemäß der vorliegenden Erfindung können mit anderen biologisch aktiven pharmazeutischen Präparaten, wie biologisch aktiven Chemikalien oder anderen Peptiden, verabreicht werden.
- Die Aminosäuren in der vorliegenden Erfindung werden gemäß der IUPACIUB-Nomenklatur wie nachstehend abgekürzt.

- Gemäß der in Tabelle 1 angegebenen Sequenz wurde eine Anzahl von Peptiden unter Verwendung eines aromatischen Peptid-Synthetisierers synthetisiert und unter Verwendung einer C18-Umkehrphasen-Hochleistungsflüssigkeits-Chromatographie (Waters Associates, USA) gereinigt.
- BEISPIEL 2. Abschätzung der antimikrobiellen Wirkung
- Unter Verwendung der Peptide wie in Beispiel 1 wurde die minimale Inhibierungskonzentration der Peptide gegenüber einer Anzahl von Mikroorganismen bestimmt. Bakterien und Pilze wurden über Nacht in Müller-Hinton- bzw. Saboraud-Medium bei 37 bzw. 30°C inkubiert und wurden 2 Stunden lang zu einer halblogarithmischen Phase in Medien eingeimpft. Nach Verdünnen der Bakterien und Pilze auf 104–105 pro 1 ml wurden sie in eine Platte mit 96 Vertiefungen eingesät, die serielle verdünnte Peptide enthielt, und weitere 18 Stunden inkubiert. Die minimale Inhibierungskonzentration wurde bei einer Konzentration, welche das Wachstum der Mikroorganismen inhibiert, durch Messen der Extinktion bestimmt. Die Ergebnisse sind in Tabelle 2 gezeigt.
- Das Peptid RAGLQFPVG(RLLR)3, das eine fusionierte Aminosäuresequenz aufweist, die eine ausgestreckte <alpha>-Helix am N-Terminus von (RLLR)3 bildet, zeigte eine besonders potente antimikrobielle Wirkung, und die Peptide (RLLR)4 und (RLLR)5, die 4 bzw. 5 Wiederholungen von RLLR aufweisen, zeigten ebenfalls starke antimikrobielle Wirkungen.
- Das Peptid, das eine Deletion der Sequenz aus Buforin II aufwies, welche eine statistische Knäuelstruktur bildet, zeigte ebenfalls eine potente antimikrobielle Wirkung, und das Peptid, das eine Amidierung am N-Terminus aufwies, zeigte ein potentere antimikrobielle Wirkung.
- BEISPIEL 3. Abschätzung der antimikrobiellen Wirkung als Funktion von Salzkonzentrationen
- Die minimale Inhibierungskonzentration der Peptide wurde als Funktion von Salzkonzentrationen gemessen, um festzustellen, ob die antimikrobielle Wirkung von den Salzkonzentrationen abhängt. Das Verfahren der Abschätzung der minimalen Inhibierungskonzentration war identisch wie in Beispiel 2, außer dass die Konzentration an NaCl geändert wurde. Die Ergebnisse sind in
2 gezeigt. Die antimikrobielle Wirkung der Peptide gemäß der Erfindung variierte nicht als Funktion der Salzkonzentration, wohingegen diejenige von Buforin und Magainin sich empfindlich als Funktion der Salzkonzentrationen änderte. Claims (10)
- Peptid, bestehend aus der Aminosäuresequenz [RLLR]n, worin n eine Zahl im Bereich von 2 bis 6 ist.
- Peptid nach Anspruch 1, bestehend aus [RLLR]3, [RLLR]4 und [RLLR]5.
- Peptid, umfassend eine Aminosäuresequenz RKGLQKLVGRVHRLLRK.
- Peptid, umfassend eine Aminosäuresequenz RAGLQFPVG[RLLR]m, worin m eine ganze Zahl im Bereich von 1 bis 3 ist.
- Peptid nach irgendeinem der vorangehenden Ansprüche, in dem der C-Terminus des Peptids amidiert ist.
- Peptid nach irgendeinem der Ansprüche 1 bis 5, in dem das Peptid einen zusätzlichen Gly-Rest an seinem N-Terminus kondensiert umfasst.
- Peptid, das aus einer Aminosäuresequenz besteht, die ausgewählt ist aus der Gruppe bestehend aus PVGRVHRLLRK und RAGLQFPVGRVHRLLRK.
- Peptid, wie in irgendeinem der Ansprüche 1 bis 7 definiert, zur Verwendung als antimikrobielles Mittel.
- Verwendung eines Peptids, wie in irgendeinem der Ansprüche 1 bis 7 definiert, für die Herstellung eines Mittels zur Inhibierung des Wachstums von Mikroorganismen.
- Verwendung eines Peptids nach Anspruch 9, in dem der Mikroorganismus ausgewählt ist aus der Gruppe bestehend aus gram-negativen Bakterien, gram-positiven Bakterien, Pilzen und Protozoen.
