Description
- Die vorliegende Erfindung betrifft bestimmte Benzimidazol- Anthelminthika, die ziemlich unerwarteterweise topisch und parenteral wirksam und daher zur transdermalen und parenteralen (insbesondere intramuskulären) Verabreichung geeignet sind.
- Die FR-2 046 114-A offenbart Benzimidazol-Derivate, die durch eine Carbamat-Gruppe substituiert sind. Die Verbindungen werden u.a. als Anthelminthika angegeben.
- Die Benzimidazol-Derivate der vorliegenden Erfindung werden dargestellt durch die Formel:
- und ihre nicht-toxischen Salze, worin R¹, das in Stellung 5(6) vorliegt, bedeutet: entweder (i) H, Benzoyl, Phenyloxy, Phenylthio, Phenylsulfinyl, Phenylsulfonyl, Phenylsulfonyloxy, C&sub1;-C&sub6;-Alkyl, C&sub1;-C&sub6;-Alkoxy, C&sub1;-C&sub6;-Alkylthio, C&sub1;-C&sub6;-Alkylsulfinyl, C&sub1;-C&sub6;-Alkylsulfonyl oder (C&sub3;-C&sub7;-Cycloalkyl)-carbonyl, wobei die Phenyl-Gruppen und der Phenyl-Teil der Benzoyl-Gruppe gegebenenfalls 1 bis 3 Substituenten aufweisen, die jeweils unabhängig ausgewählt sind aus Halogen, C&sub1;-C&sub4;-Alkyl, Halogen-(C&sub1;-C&sub4;-alkyl), C&sub1;-C&sub4;-Alkoxy, C&sub1;-C&sub4;-Alkylthio, C&sub1;-C&sub4;-Alkylsulfinyl, C&sub1;-C&sub4;-Alkylsulfonyl, C&sub2;-C&sub4;-Alkanoyl, Nitro, Isothiocyanato und cyano; oder (ii) eine Gruppe der Formel:
- worin X O, S, SO, SO&sub2; oder NR&sup4; darstellt, wobei R&sup4; bedeutet: Wasserstoff, C&sub1;-C&sub4;-Alkyl, Phenyl oder Phenyl-(C&sub1;-C&sub4;-alkyl), wobei die Phenyl-Gruppen gegebenenfalls substituiert sind durch 1 oder 2 Substituenten, jeweils ausgewählt aus C&sub1;-C&sub4;-Alkyl, Halogen, Hydroxy und C&sub1;-C&sub4;-Alkoxy; und R&sup5; H, C&sub1;-C&sub4;-Alkyl, Halogen, Hydroxy oder C&sub1;-C&sub4;-Alkoxy bedeutet; oder (iii) eine Gruppe der Formel: worin R C&sub1;-C&sub4;-Alkyl, Allyl oder Phenyl darstellt,
- "alk" eine Methylen- oder Ethylen-Gruppe ist; R² Cyano, C&sub2;-C&sub4;-Alkenyl, C&sub2;-C&sub4;-Alkinyl, C&sub2;-C&sub4;-Alkanoyl, C&sub1;-C&sub4;-Alkoxy, C&sub2;-C&sub4;-Alkenoxy, Aryl, Heteroaryl, Aryl-(C&sub1;-C&sub4;-alkoxy) oder Heteroaryl-(C&sub1;-C&sub4;-alkoxy) bedeutet, wobei "Aryl" Phenyl, gegebenenfalls substituiert durch 1 bis 3 Substituenten, jeweils unabhängig ausgewählt aus Halogen, C&sub1;-C&sub4;-Alkyl, C&sub1;-C&sub4;-Alkoxy, C&sub1;-C&sub4;-Alkylthio, C&sub1;-C&sub4;-Alkylsulfinyl und C&sub1;-C&sub4;-Alkylsulfonyl, darstellt, und wobei "Heteroaryl" Thienyl, Furyl oder Pyridinyl, alle gegebenenfalls substituiert durch 1 bis 3 Substituenten, jeweils unabhängig ausgewählt aus Halogen, C&sub1;-C&sub4;-Alkyl, C&sub1;-C&sub4;-Alkoxy, C&sub1;-C&sub4;-Alkylthio, C&sub1;-C&sub4;-Alkylsulfinyl und C&sub1;-C&sub4;-Alkylsulfonyl, bedeutet; und R³ eine C&sub1;-C&sub4;-Alkyl-Gruppe ist.
- Bevorzugte Alkyl- und Alkoxy-Gruppen haben 1 bis 4 Kohlenstoffatome.
- R¹ ist vorzugsweise H, C&sub1;-C&sub4;-Alkyl, C&sub1;-C&sub4;-Alkoxy, C&sub1;-C&sub4;-Alkylthio, C&sub1;-C&sub4;-Alkylsulfinyl, Phenylthio, Phenylsulfinyl, Benzoyl, gegebenenfalls substituiert durch Halogen, Phenylsulfonyloxy, gegebenenfalls substituiert durch Halogen, oder 1,2-Benzisoxazol-3-yl.
- Bevorzugter bedeutet R¹ H, Benzoyl, 4-Fluorbenzoyl, 4-Fluorphenylsulfonyloxy, n-P.ropylthio, n-Propylsulfinyl, Phenylthio, Phenylsulfinyl, n-Butyl, n-Propyloxy oder 1,2-Benzisoxazol-3-yl.
- Am meisten bevorzugt stellt R¹ n-Propylthio dar.
- "alk" ist vorzugsweise Methylen.
- R² bedeutet vorzugsweise Methoxy, Ethoxy, Allyloxy, Benzyloxy, Phenyl, gegebenenfalls substituiert durch 1 bis 3 Methoxy- Substituenten, Cyano, Ethinyl, Vinyl oder Acetyl.
- R³ stellt vorzugsweise Methyl dar.
- Alkyl-, Alkoxy-, Alkenyl- und Alkinyl-Gruppen, wenn geeignet, können gerad- oder verzweigtkettig sein. "Halogen" bedeutet F, Cl, Br oder J.
- Geeignete nicht-toxische Säureadditionssalze, die zur veterinären Verwendung geeignet sind, sind beispielsweise Hydrochlorid-, Hydrobromid- und Sulfatsalze. Diese können alle herkömmlich hergestellt werden.
- Die Benzimidazole der Formel (I) und ihre Salze sind insbesondere Anthelminthika, die zur Bekämpfung von parasitären Erkrankungen sowohl bei Menschen als auch bei Tieren, wie Schafen, Rindern und Haustieren, geeignet sind. Die Verbindungen zeigen Wirksamkeit gegen reife und unreife Parasitenformen von beispielsweise Nematoden, Trematoden und Cestoden, wie z.B. durch die Arten Trichostrongylus, Dictyocaulus, Ostertagia, Nematodirus, Stronglyoides, Trichuris, Heamonchus, Cooperia, Dirofilaria, Toxocara, Trichuris, Fasciola und Monezia vertreten.
- Eine effiziente Bekämpfung dieser Arten wird erzielt, indem in das Kreislaufsystem der Tiere oder der Menschen eine anthelminthisch wirksame Menge einer Verbindung (I) oder eines Salzes hievon eingeführt wird. Im Fall dieser Verbindungen wurde unerwartet gefunden, daß diese Bekämpfung durch perkutane Absorption und/oder Penetration aus einer flüssigen oder cremigen Formulierung, die direkt auf die Haut des Tieres oder der Menschen aufgebracht wird, erreicht werden kann. Derartige flüssige Formulierungen sind als "Aufgießformulierungen" bekannt. Derartige Aufgießformulierungen sind dadurch gekennzeichnet, daß der aktive Bestandteil, d.h. die Verbindung (I) oder ein Salz hievon, in einem geeigneten Lösungsmittel oder einer Lösungsmittelmischung, die hautverträglich und für das Tier nicht-toxisch ist, gegebenenfalls mit bestimmten Hilfsbestandteilen, gelöst ist.
- Zur Herstellung von Aufgießformulierungen werden die Verbindungen der Formel (I) und ihre Salze auf herkömmliche Weise formuliert, indem sie mit Trägern gemischt werden, die durch das Eindringen in die Haut wirksam sind, wobei die Verbindung (I) dann vom Tier durch die Haut absorbiert und systemisch durch das Tier transportiert wird.
- Die Aufgießformulierung enthält:
- 1. ein nicht-flüchtiges Arzneilösungsmittel oder eine Lösungsmittelmischung, das bzw. die normalerweise als transdermale Penetrationsverstärker klassierte Lösungsmittel enthalten kann, und gegebenenfalls eines oder mehrere der folgenden:--
- 2. ein Lösungsmittel mit der spezifischen Rolle der Verstärküng der transdermalen Penetration, wenn ein derartiges Lösungsmittel nicht bereits zur Durchführung der Funktion 1 als Hauptarzneilösungsmittel vorliegt;
- 3. ein Verteilungshilfsmittel, wenn diese Hilfsfunktion nicht bereits adäquat vom Arzneilösungsmittel (1) und irgendeinem transdermalen Penetrationsverstärker (2) erfüllt wird;
- 4. ein flüchtiges Lösungsmittel. Dieses flüchtige Lösungsmittel kann die Verteilung und Distribution der Komponenten 1 und 2 unterstützen, die Formulierung auf, ein zweckmäßiges Dosierungsvolumen einstellen und die Löslichkeit und Mischbarkeit der Formulierung unter extremen Lagerungsbedingungen sicherstellen; und
- 5. weitere Adjuvantien, wenn notwendig, um die chemische Stabilität bei der Lagerung und Verwendung sicherzustellen, die Viskosität der Formulierung zur Verhinderung eines Ablaufens zu erhöhen, andere Tiere daran zu hindern, die Zusammensetzung vom behandelten Tier abzulecken, und die Haut gegen eine unerwünschte Reizung zu schützen.
- Um eine adäquate Solubilität zu erzielen, werden geeignete Arzneilösungsmittel (1) ausgewählt aus:
- Verteilungsölen - Silikonölen, Isopropylmyristat, Isopropylpalmitat, Capryl/Caprinsäuretriglycerid, gesättigten Triglyceriden von natürlich vorkommenden Fettsäuren, Fettsäure estern (z.B. Ethyloleat) und Fettsäureestern, die synthetischem Anatinuropygialdrüsenfett entsprechen.
- Aliphatischen Kohlenwasserstoffen - z.B. Leichtparaffinöl.
- Hydroxyl-Lösungsmitteln - weniger flüchtigen Alkoholen (z.B. Hexanol, Octanol), Propylenglykol, Polypropylenglykolen, Ethylenglykol, Diethylenglykol, Glycerinen und Ether- oder Ester-Substituenten dieser Lösungsmittel (z.B. Triacetin), Benzylalkohol und Carbonsäureestern (z.B. Benzylbenzoat), Butylacetat, Propylencarbonat und Ethyllactat.
- Polyalkoxylierten Lösungsmitteln - Polyethylenglykolen, Polyglykolethern und/oder -estern, z.B. 2-(2-Alkoxy)-ethoxyethanolen und 2-(2-Alkoxy)-ethoxyethylalkanoaten.
- Pflanzenölen - die in der Definition von Verteilungsölen nicht eingeschlossen sind, z.B. Mais-, Sesam-, Oliven-, Kiefern-, Leinsamen-, Baumwollsamen- und Erdnußöl.
- Penetrationsverstärkern (2) - (a) z.B. Dimethylsulfoxid, Dimethylformamid und Dimethylacetamid; (b) Pyrrolidonen, insbesondere 2-Pyrrolidon, N-Methylpyrrolidon und 1- oder 5- und 1,5-Alkyl-substituierten Pyrrolidonen, z.B. 1,5-Dimethyl-2- pyrrolidon, oder Carbonsäure-substituierten Pyrrolidonen; (c) Alkylsulfoxiden, Zuckerestern und Phosphinoxiden; und (d) Azacycloalkan-2-onen.
- Während diese Lösungsmittel in mischbaren Kombinationen verwendet werden können, kann die Mischbarkeit-durch den Einschluß eines ternären Lösungsmittels erzielt werden, wenn notwendig, um eine adäquate Arzneimittellöslichkeit vorzusehen.
- Verteilungshilfsmittel (3) umfassen Verteilungsöle (wenn diese nicht als Hauptarzneilösungsmittel verwendet werden, wie vorstehend in (1) angegeben) oder grenzflächenaktive Stoffe, wobei der verwendete Ausdruck "grenzflächenaktiver Stoff" Materialien abdeckt, die verschiedentlich als Netzmittel, Emulgatoren und Dispergiermittel bezeichnet werden. Sie schließen ein:
- nicht-ionische wasserlösliche Emulgatoren, wie, Alkylaryl polyglykolether, Polyoxyethylenalkylarylether, Alkylpolyglykol ether, Polyoxyethylenester und -ether, Polyoxyethylensorbitanmonofettsäureester, Sorbitanmonofettsäureester, ethoxylierte Nonylphenole, Isooctylphenol, Polyethoxyethanol und polyethoxyliertes Castoröl (Cremophor EL ). Anionische grenzflächenaktive Stoffe schließen ein: Seifen, Fettsäuresulfatester (z.B. Dodecyl-Na-sulfat), aromatische Fettsäuresulfonate (z.B. Alkylbenzolsulfonate), komplexere Fettsäuresulfonate, wie das Amid- Kondensationsprodukt von Oleinsäure und N-Methyltaurin, Natriumsulfonat von Dioctylsuccinat und den ethoxylierten Nonylphenol- Dinatriumhalbester von Sulfonbernsteinsäure. Kationische Mittel, wie Cetyltrimethylammoniumbromid, können ebenso wie ampholytische grenzflächenaktive Stoffe, wie Di-Na-N-lauryl-β-iminodipropionat oder Lecithin, verwendet werden. Geeignete flüchtige Lösungsmittel (4) schließen ein: Ketone, wie Aceton, Methylethylketon, Methylisobutylketon und Cyclohexanon; einfache Alkohole (insbesondere Methanol, Ethanol und [spezifisch] Isopropylalkohol); gerade und verzweigte Alkylether (z.B. Dibutyldiisopropylether), Tetrahydrofuran, Glykolether und gerad- und verzweigtkettige Alkylacetate (z.B. Isopropylacetat) und andere Ester, wie Milchsäureethylester; aromatische Kohlenwasserstoffe, wie Xylol, Benzol, Toluol, Alkylnaphthaline und Chlorbenzole; und aliphatische Kohlenwasserstoffe, wie Paraffine mit einer Kettenlänge von 6 bis 20, und halogenierte aliphatische Kohlenwasserstoffe.
- Geeignete Hilfsadditive (5) schließen ein:
- Stabilitätsverstärker - Antioxidantien, z.B. Ascorbinsäure, butyliertes Hydroxyanisol und butyliertes Hydroxytoluol.
- Färbemittel - anorganische Pigmente, Eisen(II)-oxid, Titandioxid, Preußischblau; organische Farbstoffe, z.B. Farbstoffe auf der Basis von Alizarin, Azo-Farben oder Metallphthalocyanin.
- Haftvermittler - Carboxymethylcellulose, Methylcellulose und andere Gellulose- und Stärke-Derivate, Polyacrylate, Alginate, Gelatinen, Akaziengummi, Polyvinylpyrrolidon, Copolymere von Methylvinylether und Maleinsäureanhydrid, Polyethylenglykole, Paraffine, Öle und Wachse, hydriertes Castoröl, Lecithine und synthetische Phospholipide.
- Orale Deterrentien - wie bittere Aien.
- Emollentien - wie Lanolin.
- Das Arzneimittel wird in typischen Formulierungen gelöst, die 1 bis 100 % der Hauptarzneilösungsmittel, üblicherweise nicht mehr als 70 % und idealerweise nicht mehr als 20 %, enthalten. Der Rest der Formulierung besteht vorwiegend aus dem flüchtigen Lösungsmittel, das 0 bis 99 % der Formulierung, vorzugsweise nicht weniger als 30 %, umfassen kann. Ferner werden nach Bedarf transdermale penetrationsverstärker (0 bis 33 %), Verteilungshilfsmittel (0 bis 25 %) und Adjuvantien (0 bis 5 %) zugesetzt.
- Bevorzugte Formulierungen für die aktiven Stoffe wurden aus typischen Formulierungen ausgewählt.

- Die Aufgießformulierungen enthalten typischerweise die aktive Verbindung (I) oder ein Salz hievon in einer Menge von 0,5 % M/V bis 60 % M/V. Eine typische Aufgießformulierung enthält 1 bis 50 mg des aktiven Bestandteils pro kg Körpergewicht des Tieres in etwa 0,01 bis 1 ml pro kg Körpergewicht des Tieres. Typischerweise wird die Formulierung einfach auf das Tier gegossen. Konzentrierte Formulierungen werden häufig als Formulierungen "zum punktförmigen Auftragen" bezeichnet, die punktförmig auf das Tier aufgetragen werden.
- Die Verbindungen der Formel (I) werden als Formulierung verabreicht, die für die spezifische beabsichtigte Verwendung, die bestimmte behandelte Wirtstierart und den involvierten Parasiten geeignet ist. Zur Verwendung als Anthelminthika können die Verbindungen oral in Form einer Kapsel, eines Bolus, einer Tablette oder eines Tranks oder als Aufgießformulierung verabreicht werden, oder alternativ dazu können sie durch Injektion (z.B. subkutan, intramuskulär oder intravenös) oder als Implantat verabreicht werden. Derartige Formulierungen werden auf herkömmliche Weise gemäß der Standard-Veterinärpraxis hergestellt. So können Kapseln, Boh oder Tabletten hergestellt werden, indem der aktive Besandteil mit einem geeigneten fein zerteilten Verdünnungsmittel oder Träger, das bzw. der zusätzlich ein Desintegriermittel und/oder Bindemittel.enthält, wie Stärke, Lactose, Talkum, Magnesiumstearat, etc., gemischt wird. Ein oraler Trank wird durch das Lösen oder Suspendieren des aktiven Bestandteils in einem geeigneten Medium hergestellt. Injizierbare Formulierungen können in Form einer sterilen Lösung hergestellt werden, die andere Substanzen, beispielsweise ausreichend Salze oder Glucose, um die Lösung isotonisch mit Blut zu machen, enthalten kann. Annehmbare flüssige Träger schließen ein: Pflanzenöle, wie Sesamöl und dgl., Glyceride, wie Triacetin und dgl., Ester, wie Benzylbenzoat, Isopropylmyristat und Fettsäure-Derivate von Propylenglykol und dgl., sowie organische Lösungsmittel, wie Pyrrolidon, Glycerin, Formal und dgl. Die Formulierungen werden durch das Lösen oder Suspendieren des aktiven Bestandteils im flüssigen Träger hergestellt, so daß die Endformulierung 0,5 bis 60 Masse-% des aktiven Bestandteils enthält.
- Diese Formulierungen variieren hinsichtlich der Masse der aktiven Verbindung in Abhängigkeit von der Art des zu behandelnden Wirtstieres, dem Schweregrad und Typ der Infektion und dem Körpergewicht des Wirts. Zur parenteralen und oralen Verabreichung betragen typische Dosisbereiche des aktiven Bestandteils 1 bis 50 mg pro kg Körpergewicht des Tieres. Aufgießformulierungen wurden vorstehend beschrieben.
- Als Alternative können die Verbindungen mit der Tiernahrung verabreicht werden, und zu diesem Zweck kann ein konzentriertes Nahrungsmitteladditiv oder eine Vormischung zum Mischen mit der normalen Tiernahrung hergestellt werden.
- Die Verbindungen der Erfindung sind hochwirksame Antiparasitika, die nicht nur als Anthelminthika, sondern auch als Ektoparasitenmittel, Insektizide, Akarizide sowie Antipilzmittel und Antiprotozoenmittel nützlich sind.
- Daher sind die Verbindungen bei der Behandlung verschiedenster durch Endoparasiten verursachter Zustände wirksam, die insbesondere einschließen: Helminthose, welche am häufigsten durch eine Gruppe von Parasitenwürmern verursacht wird, die als Nematoden, Cestoden und Trematoden beschrieben werden, und welche schwere wirtschaftliche Verluste bei Schweinen, Schafen, Pferden und Rindern hervorrufen sowie Haustiere und Geflügel befallen kann. Die Verbindungen sind auch gegen andere Nematoden wirksam, die verschiedene Tierarten befallen, einschließlich beispielsweise Dirofilaria, Toxocara, Ancyclostoma, Dipylidium, Echinococcus und Taenia bei Hunden und verschiedener Parasiten, die Menschen infizieren können, einschließlich gastromtestinaler Parasiten, wie Acylostoma, Necator, Ascaris, Strongybides, Trichinella, Capillaria, Trichuris, Enterobius, und Parasiten, die im Blut oder arideren Geweben und Organen zu finden sind, wie Fadenwürmer und die extra-intestinalen Stadien von Strongyloides und Trichinella.
- Die Verbindungen sind auch bei der Behandlung von Ektoparasiteninfektionen wertvoll, die insbesondere arthropode Ektoparasiten von Tieren und Vögeln einschließen, wie Zecken, Milben, Läuse, Flöhe, Schmeißfliegen, stechende Insekten und migrierende Zweiflüglerlarven, die Rinder und Pferde befallen können.
- Die Verbindungen sind auch Insektizide, die gegen Haushaltsschädlinge wirksam sind, wie Küchenschaben, Kleidermotten, Teppichkäfer und Stubenfliegen, und können gegen arthropode Schädlinge von gelagertem Getreide und landwirtschaftlichen Pflanzen, wie Blattspinnmilben, Blattläuse, Raupen und gegen migrierende Geradflügler, wie Heuschrecken, wirksam sein.
- Zur Verwendung als Insektizid und zur Behandlung von landwirtschaftlichen Schädlingen werden die Verbindungen als Sprays, Pulver, Emulsionen und dgl. gemäß der landwirtschaftlichen Standard-Praxis aufgebracht.
- Die Verbindungen der Formel (I) können hergestellt werden durch das Alkylieren eines gegebenenfalls geschützten 5-(6)Benzimidazols der Formel:
- worin R¹ in Stellung 5 (6) vorliegt, wobei R' wie für Formel (I) definiert ist, und X H oder eine Amino-Schutzgruppe bedeutet, mit einem Alkylierungsmittel der Formel:
- R²-alk-Q (III),
- worin Q eine geeignete Abgangsgruppe, wie Chlor, Brom oder Jod, ist, und alk und R² wie für Formel (I) definiert sind.
- Die neuen Verbindungen (II), worin X eine Amino-Schutzgruppe ist, bilden auch einen Teil der Erfindung.
- Eine bevorzugte Amino-Schutzgruppe ist tert.Butoxycarbonyl.
- Die Reaktion wird vorzugsweise in Anwesenheit einer Base, wie Kaliumcarbonat, durchgeführt. Die Reaktion wird typischerweise in einem organischen Lösungsmittel, wie Dimethylformamid, etwa bei Raumtemperatur vorgenommen. Die Amino-Schutzgruppe x, sofern vorhanden, wird dann herkömmlich entfernt: beispielsweise wird tert.Butoxycarbonyl typischerweise durch Behandlung mit Trifluoressigsäure etwa bei Raumtemperatur entfernt.
- Da in den Benzimidazol-Endprodukten (I) und Ausgangsmatenahen (II) die Position des Wasserstoffatoms am Stickstoffatom des Imidazolrings nicht bestimmt werden kann (Tautomene), ist es klar, daß die Verbindungen korrekt als 5(6)-substituierte Benzimidazole bezeichnet werden.
- Die Ausgangsmaterialien (II), worin X eine Amino-Schutzgruppe ist, können durch das N-Schützen der entsprechenden Benzimidazole mit einem Wasserstoffatom in Stellung 1 mit einem geeigneten Reagens, z.B. Di-tert .butyldicarbonat, hergestellt werden. Diese "ungeschützten" Benzimidazole sind bekannte Verbindungen und haben in vielen Fällen ungeschützte Freinamen, wie folgt: Oxibendazol Parbendazol Albendazol Fenbendazol Lobendazol (C-arbendazim) Mebendazol Flubendazol Luxobendazol Albendazoloxid (Ricobendazol) Oxfendazol

- C&sub1;-C&sub6;-Alkylthio-Gruppen,die R¹ darstellt oder in R² vorliegen, können zu C&sub1;-C&sub6;-Alkylsulfinyl oder Alkylsulfonyl-Gruppen durch herkömmliche Techniken oxidiert werden, z.B. unter Verwendung von 1 oder mehreren Äquivalenten von m-Chlorperbenzoesäure, wie geeignet, in einem geeigneten Lösungsmittel, wie Methylenchlorid.
- Die Nützlichkeit der Verbindungen als Anthelminthika bei transdermaler Verabreichung kann beispielsweise durch die folgende Technik festgestellt werden:
- Männliche Cobb-Wistar-Ratten mit einem Gewicht von 40 bis 50 g (3 bis 4 Wochen alt) werden verwendet. Die Tiere werden mit normaler würfelförmiger Nagetiernahrung gefüttert, die 60 ppm des Immunsuppressivums Hydrokortisonacetat enthält. Die Immunsuppression beginnt eine Woche vor der Infektion und wird bis zum Ende aufrechterhalten. Sowohl Nahrung als auch Wasser werden ad lib zur Verfügung gestellt.
- Jede Ratte erhält oral 1500 infektive T. colubriformis- Larven. Von Schafen stammende Larven werden aus Vorratskulturen unmittelbar vor der Dosierung entnommen und mikroskopisch auf ihre Lebensfähigkeit untersucht. Nur sich bewegende, lebensfähige Larven werden verwendet. Die Parasiten werden in 0,25 ml Wasser verabreicht.
- Ein bis drei Wochen nach der Infektion werden die Tiere zufällig entweder den Behandlungs- (allgemein 5 Ratten) oder Kontroll- (allgemein 10 Ratten) Gruppen zugeteilt. Lösungen der Testverbindungen in geeigneten Trägern (siehe unten) werden topisch mit einer Pipette auf eine rasierte Fläche (ca. 6,5 cm²/ 1 Quadratzoll) am Rücken nahe beim Hals aufgebracht. Die Arzneimittelkonzentrationen im Träger werden derart eingestellt, daß jedes Tier die gewünschte Dosis bei < 0,25 ml/100 g Körpergewicht erhält. Zur Zeit der Dosierung liegt das Gewicht der Tiere im Bereich von 90 bis 110 g. Alle Tiere werden drei Tage nach der Dosierung getötet.
- Danach wird der Dünndarm jedes Tieres entfernt und in eine Kunststoffschale gegeben, enthaltend 20 ml Pepsin-Verdaumischung, umfassend 8 g Pepsin A-Pulver, 8,5 g NaCl plus 16 ml konzentrierte HCl in 1 l destilliertem Wasser. Die Verdaue werden 4 Stunden lang bei 37ºC inkubiert, bevor sie über einem 75 µm Sieb mit einem Hochdruckwasserstrahl gewaschen werden. Auf dem Sieb zurückgehaltene Würmer werden durch Auswaschen in frischen Schalen gesammelt und unter Verwendung einer Jod/ Kaliumjodid-Lösung, umfassend 30 g Jod, 40 g Kaliumjodid plus 30 ml methylierter Spiritus in 70 ml destilliertem Wasser, angefärbt. Der Inhalt jeder Schale wird dann mit destilliertem Wasser auf ein Endvolumen von 500 ml verdünnt und eine aliquote 50 ml Menge zur Wurmzählung entnommen. Die Effizienz jeder Arzneimittelbehandlung wird als perzentuelle Verringerung der mittleren Wurmbelastung unbehandelter Kontrollen bestimmt.
- Die folgenden Beispiele veranschaulichen die Herstellung der Verbindungen der Formel (I). Die Herstellungsweise zeigt die Herstellung eines Ausgangsmaterials.
- Einer gerührten Suspension von 150 g (0,57 M) Methyl-5(6)- n-Propylthiobenzimidazol-2-carbamat (Albendazol) in 2 l Tetrahydrofuran wurden 123 g (0,57 M) Di-tert.butyldicarbonat zugesetzt. Die Mischung wurde 18 h lang bei Raumtemperatur und dann 5,5 h lang bei 50ºC gerührt. Dann wurden weitere 62 g Di- tert.butyldicarbonat in 150 ml Tetrahydrofuran zugesetzt. Die Mischung wurde 18 h lang bei Raumtemperatur, anschließend weitere 2 h lang bei 50ºC gerührt. Nach dem Eindampfen wurde der Rückstand mit 1 l Diethylether aufgeschlämmt und filtriert. Das Filtrat wurde eingedampft und aus n-Hexan kristallisiert, wobei 66 g einer Mischung der Titelverbindungen erhalten wurden. Der Filterkuchen wurde mit Methylenchlorid aufgeschlämmt, filtriert und das Filtat eingedampft. Der Rückstand wurde aus n-Hexan kristallisiert, wobei weitere 61 g der Titelverbindungen erhalten wurden. Fp. 85-87ºC.
- Analyse:
- Berechnet für C&sub1;&sub7;H&sub2;&sub3;N&sub3;O&sub4;S: C,55,87%; H,6,34%; N,11,50%.
- Gefunden: C,55,91%; H,6,41%; N,11,58%.
- 7,3 g (0,02 M) 2-(Methoxycarbonylamino)-1-(tert.butoxycarbonyl)-(5- und 6-n-propylthio)-benzimidazol, 1,5 ml (0,02 M) Chlormethylmethylether, 5,6 g (0,04 M) wasserfreies Kaliumcarbonat und 50 ml Dimethylformamid wurden gemeinsam 72 h lang bei Raumtemperatur gerührt, dann zur Trockene eingedampft.
- Der Rückstand wurde in Ethylacetat gelöst und mit Wasser, gesättigter Kochsalzlösung gewaschen und dann über Magnesiumsulfat getrocknet. Die Mischung wurde filtriert und das Filtrat eingedampft. Dem Rückstand wurden 20 ml Trifluoressigsäure zugesetzt, und die Mischung wurde 2 h lang bei Raumtemperatur gerührt, dann mit festem Kaliumbicarbonat neutralisiert. Anschließend wurde die Mischung in Ethylacetat gelöst, mit Wasser und Kochsalzlösung gewaschen, dann über Magnesiumsulfat getrocknet, filtriert und eingedampft. Das Produkt wurde durch Chromatographie auf Kieselerde gereinigt, wobei mit 1 % Methanol in Methylenchlorid eluiert wurde. Die relevanten Fraktionen wurden kombiniert und eingedampft, dann wurde der Rückstand aus Ether und Hexan kristallisiert, wobei 1,5 g der Titelverbindung erhalten wurden. Fp. 62 ºC.
- Analyse:
- Berechnet für C&sub1;&sub4;H&sub1;&sub9;N&sub3;O&sub3;S: C,54,35%; H,6,19%; N,13,58%.
- Gefunden: C,54,89%; H,6,12%; N,13,69%.
- Das Verfahren von Beispiel 1 wurde wiederholt, wobei Chlormethylethylether anstelle von Chiormethylmethylether verwendet wurde. Fp. 59-63ºC.
- Analyse:
- Berechnet für C&sub1;&sub5;H&sub2;&sub1;N&sub3;O&sub3;S: C,55,71%; H,6,54%; N,12,99%.
- Gefunden: C,55,98%; H,6,56%; N,13,09%.
- Das Verfahren von Beispiel 1 wurde wiederholt, wobei Chlormethylallylether (siehe Hurd et al., JACS, 74, 5128, 1952) anstelle von Chlormethylmethylether verwendet wurde. Das Produkt war ein Öl.
- NMR (ppm, CDCl&sub3;,300 MHz) 1,0 (3H,t), 1,65 (2H,dt), 2,85 (2H,t), 379 (3H,s), 4,2-6,0 (5H,m), 5,3 (2H,s), 7,5 (3H,m), 11,0 (1H,br S).
- Das Verfahren von Beispiel 1 wurde wiederholt, wobei Chlormethylbenzylether anstelle von Chlormethylmethylether verwendet wurde. Fp. 59-63ºC.
- Analyse:
- Berechnet für C&sub2;&sub0;H&sub2;&sub3;N&sub3;O&sub3;S: C,62,32%; H,6,01%, N,10,90%.
- Gefunden: C,61,96%; H,5,95%; N,10,67%.
- Das Verfahren von Beispiel 1 wurde wiederholt, wobei 3,4,5-Trimethoxybenzylchlorid anstelle von Chlormethylmethylether verwendet wurde. Fp. 133-136ºC.
- Analyse:
- Berechnet für C22H&sub2;&sub7;N&sub3;O&sub5;S: C,59,31%; H,6,11%, N,9,43%
- Gefunden: C,59,67%; H,6,08%, N,9,72%.
- Eine Mischung von 2,65 g (0,01 M) 5(6)-n-Propylthiobenzimidazol, 1,67 g (0,01 M) Jodacetonitril, 3 g (0,22 M) Kaliumcarbonat und 30 ml Dimethylformamid wurde 2 Tage lang bei Raumtemperatur gerührt, wonach das Lösungsmittel abgedampft wurde. Der Rückstand wurde zwischen Ethylacetat und Wasser verteilt. Die wässerige Schicht wurde abgetrennt und mit Ethylacetat reextrahiert. Die kombinierten organischen Schichten wurden mit Wasser gewaschen, getrocknet (MgSO4) und eingedampft. Das Rohprodukt wurde durch Säulenchromatographie auf Silikagel ("Silica Gel 60", Merck [Warenzeichen], 100 g) gereinigt, wobei anfänglich mit Dichlormethan und dann mit 1 % Methanol enthaltendem Dichlormethan eluiert wurde. Durch Kombinieren und Eindampfen geeigneter Fraktionen wurden 1,46 g (48 %)
- 2-[(N-Methoxycarbonyl-N-cyanomethyl)-amino]-[5(6)-n-propylthio]benzimidazol erhalten. Fp. 151-152&sup4;C.
- Analyse:
- Berechnet für C&sub1;&sub4;H&sub1;&sub6;N&sub4;O&sub2;S: C,55,25%, H,5,30%; N,18,41%.
- Gefunden: C,55,31%; H,5,01%, N,18,12%.
- Das Verfahren von Beispiel 6 wurde wiederholt, wobei Propargylbromid anstelle von Jodacetonitril verwendet wurde. Fp. 127-128 ºC.
- Analyse:
- Berechnet für C&sub1;&sub5;H&sub1;&sub7;N&sub3;O&sub2;S: C,59,39%; H,5, 65%; N,13,85%.
- Gefunden: C,59,21%; H,5,54%, N,13,76%.
- Das Verfahren von Beispiel 6 wurde wiederholt, wobei Allylbromid anstelle von Jodacetonitril verwendet wurde. Analyse: Fp. 85-87ºC.
- Berechnet für C&sub1;&sub5;H&sub1;&sub9;N&sub3;O&sub2;S: C,58,99%; H,6,27%; N,13,76%.
- Gefunden: C,59,29%; H,6,22%; N,13,74%.
- Das Verfahren von Beispiel 6 wurde wiederholt, wobei Chloraceton anstelle von Jodacetonitril verwendet wurde.
- NMR; H (ppm, CHCl&sub3;, 300 MHz) = 1,0 (3H,t), 1,65 (2H,dt), 2,62 (3H,5), 2,88 (2H,t), 3,9 (3H,s), 5,05 (2H,s), 7,25-7,7 (3H,m), 11,0 (1H,brs). Beispiel 4 Beispiel 5 Beispiel 6 Beispiel 7 Beispiel 8 Beispiel 9 Beispiel 10: Eine zur transdermalen Verabreichung geeignete Zusammensetzung ist wie folgt: 2-[(N-Methoxycarbonyl)-N-(methoxymethyl)-amino]-[5(6)-n-propylthio]-benzimidazol (Produkt von Beispiel 1) Butyldigol-[(2-(2-butoxyethoxy)-ethanol] 2-Pyrrolidon Isopropylacetat ) gleiche ) Mengen ) (V) Beispiel 11: Eine zur parenteralen Verabreichung geeignete Zusammensetzung ist wie folgt: 2-[(N-Methoxycarbonyl)-N-(methoxymethyl)-amino]-[5(6)-n-propylthio]-benzimidazol (Produkt von Beispiel 1) Benzylbenzoat Sesamöl
Claims (14)
1. Benzimidazol-Derivat der Formel:
oder ein nicht-toxisches Salz hievon, worin R , das in Stellung
5(6) vorliegt, bedeutet: entweder (i) H, Benzoyl, Phenyloxy,
Phenylthio, Phenylsulfinyl, Phenylsulfonyl, Phenylsulfonyloxy,
C&sub1;-C&sub6;-Alkyl, C&sub1;-C&sub6;-Alkoxy, C&sub1;-C&sub6;-Alkylthio, C&sub1;-C&sub6;-Alkylsulfinyl,
C&sub1;-C&sub6;-Alkylsulfonyl oder (C&sub3;-C&sub7;-Cycloalkyl)-carbonyl, wobei die
Phenyl-Gruppen und der Phenyl-Teil der genannten Benzoyl-Gruppe
gegebenenfalls 1 bis 3 Substituenten aufweisen, die jeweils
unabhängig ausgewählt sind aus Halogen, C&sub1;-C&sub4;-Alkyl,
Halogen(C&sub1;-C&sub4;-alkyl), C&sub1;-C&sub4;-Alkoxy, C&sub1;-C&sub4;-Alkylthio,
C&sub1;-C&sub4;-Alkylsulfinyl, C&sub1;-C&sub4;-Alkylsulfonyl, C&sub2;-C&sub4;-Alkanoyl, Nitro,
Isothiocyanato und Cyano; (ii) eine Gruppe der Formel:
worin X O, S, SO, SO&sub2; oder NR&sup4; darstellt, wobei R&sup4; bedeutet:
Wasserstoff, C&sub1;-C&sub4;-Alkyl, Phenyl oder Phenyl-(C&sub1;-C&sub4;-alkyl),
wobei die Phenyl-Gruppen gegebenenfalls substituiert sind durch
1 oder 2 Substituenten, jeweils ausgewählt aus C&sub1;-C&sub4;-Alkyl,
Halogen, Hydroxy und C&sub1;-C&sub4;-Alkoxy; und R&sup5; H, C&sub1;-C&sub4;-Alkyl,
Halogen, Hydroxy oder C&sub1;-C&sub4;-Alkoxy bedeutet; oder (iii) eine
Gruppe der Formel:
worin R&sup6; C&sub1;-C&sub4;-Alkyl, Allyl oder
Phenyl darstellt,
"alk" eine Methylen- oder Ethylen-Gruppe ist; R Cyano,
C&sub2;-C&sub4;-Alkenyl, C&sub2;-C&sub4;-Alkinyl, C&sub2;-C&sub4;-Alkanoyl, C&sub1;-C&sub4;-Alkoxy,
C&sub2;-C&sub4;-Alkenoxy, Aryl, Heteroaryl, Aryl-(C&sub1;-C&sub4;-alkoxy) oder
Heteroaryl-(C&sub1;-C&sub4;-alkoxy) bedeutet, wobei "Aryl" Phenyl,
gegebenenfalls substituiert durch 1 bis 3 Substituenten, jeweils
unabhängig ausgewählt aus Halogen, C&sub1;-C&sub4;-Alkyl, C&sub1;-C&sub4;-Alkoxy,
C&sub1;-C&sub4;-Alkylthio, C&sub1;-C&sub4;-Alkylsulfinyl und C&sub1;-C&sub4;-Alkylsulfonyl,
darstellt, und wobei "Heteroaryl" Thienyl, Furyl oder Pyridinyl,
alle gegebenenfalls substituiert durch 1 bis 3 Substituenten,
jeweils unabhängig ausgewählt aus Halogen, C&sub1;-C&sub4;-Alkyl,
C&sub1;-C&sub4;-Alkoxy, C&sub1;-C&sub4;-Alkylthio, C&sub1;-C&sub4;-Alkylsulfinyl und
C&sub1;-C&sub4;-Alkylsulfonyl, bedeutet; und R eine C&sub1;-C&sub4;-Alkyl-Gruppe
ist.
2. Verbindung nach Anspruch 1, worin R¹ H, C&sub1;-C&sub4;-Alkyl,
C&sub1;-C&sub4;-Alkoxy, C&sub1;-C&sub4;-Alkylthio, C&sub1;-C&sub4;-Alkylsulfinyl, Phenylthio,
Phenylsulfinyl, Benzoyl, gegebenenfalls substituiert durch
Halogen, Phenylsulfonyloxy, gegebenenfalls substituiert durch
Halogen, oder 1,2-Benzisoxazol-3-yl bedeutet.
3. Verbindung nach Anspruch 2, worin R¹ H, Benzoyl,
4-Fluorbenzoyl, 4-Fluorphenylsulfonyloxy, n-Propylthio,
n-Propylsulfinyl, Phenylthio, Phenylsulfinyl, n-Butyl, n-Propyloxy oder
1,2-Benzisoxazol-3-yl bedeutet.
4. Verbindung nach Anspruch 3, worin R¹ n-Propylthio bedeutet.
5. Verbindung nach einem der vorhergehenden Ansprüche, worin
"alk" Methylen darstellt.
6. Verbindung nach einem der vorhergehenden Ansprüche, worin
R² Methoxy, Ethoxy, Allyloxy, Benzyloxy, Phenyl, gegebenenfalls
substituiert durch 1 bis 3 Methoxy-Substituenten, Cyano,
Ethinyl, Vinyl oder Acetyl bedeutet.
7. Verbindung nach einem der vorhergehenden Ansprüche, worin
R³ Methyl bedeutet.
8. Veterinär-Zusammensetzung, welche eine Verbindung der
Formel (I) oder ein nicht-toxisches Salz hievon nach einem der
vorhergehenden Ansprüche und ein nicht-toxisches Verdünnungsmittel
oder einen nicht-toxischen Täger umfaßt.
9. Zusammensetzung nach Anspruch 8, welche in Form einer
Formulierung zum punktförmigen Auftragen oder Aufgießen oder eine
parenterale Formulierung ist.
10. Verbindung der Formel (I) oder ein nicht-toxisches Salz
hievon nach einem der Ansprüche 1 bis 7 zur Verwendung als
Medikament
11. Verbindung der Formel:
worin R¹ und R³ wie in Anspruch 1 definiert sind, und X¹
tert. Butoxycarbonyl bedeutet.
12. Verfahren zur Herstellung einer Verbindung der Formel (I)
oder eines nicht-toxischen Salzes hievon nach Anspruch 1,
gekennzeichnet durch das Umsetzen einer Verbindung der Formel:
worin R¹ und R³ wie in Anspruch 1 definiert sind, und X H oder
eine Amino-Schutzgruppe bedeutet&sub1; mit einem Alkylierungsmittel
der Formel:
R²-alk-Q (III),
worin R² und alk wie in Anspruch 1 definiert sind, und Q eine
Abgangsgruppe ist, gefolgt von der Entfernung einer durch X
dargestellten Amino-Schutzgruppe, und gegebenenfalls Oxidation
einer durch R¹ dargestellten C&sub1;-C&sub6;-Alkylthio-Gruppe zu
C&sub1;-C&sub6;-Alkylsulfinyl oder C&sub1;-C&sub6;-Alkylsulfonyl, und/oder
Überführen des Produkts (I) in ein nicht-toxisches Salz.
13. Verfahren nach Anspruch 12, dadurch gekennzeichnet, daß X
tert.Butoxycarbonyl bedeutet, und daß X durch Behandlung mit
Trifluoressigsäure entfernt wird.
14. Verfahren nach Anspruch 12 oder 13, dadurch gekennzeichnet,
daß Q die Bedeutung Cl, Br oder J hat, und daß die Reaktion in
Anwesenheit einer Base durchgeführt wird.
