Description
- Die vorliegende Erfindung betrifft neue Inhibitoren der N-Myristoyltransferase, Verfahren zu ihrer Herstellung und sie enthaltende pharmazeutische Zusammensetzungen.
- Es ist aus dem Stand der Technik bekannt, daß die N-terminale Aminogruppe der Proteine durch Acetylgruppen, Pyroglutamylgruppen und Formylgruppen blockiert wird. SHOJI et coll. haben nachgewiesen, daß die Myristinsäure über eine kovalente Bindung an die N-terminale Gruppe von katalytischen Untereinheiten des cyclischen, abhängigen AMP-Kinaseproteins gebunden wird (Proc. Natl. Acad. Sci. USA 79 (1982), 6123-6131).
- Die Anwesenheit dieser endständigen Myristoylgruppe konnte seitdem bei verschiedenen anderen Proteinen, wie Calcineurin B (AITKEN et coll., Febs Letters 150 (1982). Nr. 2. 314-318) oder der Tyrosinproteinkinase (TPK) (BUSS und SEFTON, J. Virol. 53 (1985), 7-12) gezeigt werden.
- Auch im Bereich der Onkogene hat BISHOP nachgewiesen, daß ein transformierendes Protein im Verlaufe der Reifung einer Myristoylierung unterliegt. Es wurde seitdem weiterhin gezeigt, daß diese Stufe der Reifung, die über eine Myristoylierung verläuft, für das Transformationsvermögen dieses Proteins wesentlich ist (KAMPS, BUSS und SEFTON, Proc. Natl. Acad. Sci. USA 82 (1985), 4625- 4628). Dieses Konzept wurde seitdem an einer Vielzahl von anderen transformierenden Proteinen viralen Ursprungs verallgemeinert (RHEE und HUNTER, J. Virol. 61(1987), 1045-1053). Diese Reifung wird durch ein als N-Myristoyltransferase (NMT) bezeichnetes Enzym katalysiert, welches von TOWLER und GLASER in der Hefe nachgewiesen worden ist (Proc. Natl. Acad. Sci. USA 83 (1986), 2812- 2816).
- Darüber hinaus erkennt die NMT praktisch als Cosubstrat nur die Myristinsäure einerseits und andererseits als Substrat die Proteine, die ein Glycin als letzte Aminosäure des N-terminalen Endes aufweisen unter Teilnahme der Peptidsequenz, die an dieses Glycin angrenzt (Teilnahme von 7 Aminosäuren).
- So spielt die Myristoylierung des N-terminalen Glycinrests bestimmter Proteine eine sehr wichtige Rolle bei bestimmten Mechanismen, die bei dertransformation von Zellen und bei der Steuerung ihrer Vermehrung auftreten. Es wurde weiterhin von SHOJI et coll. (japanische Patente JP 63-146851, JP 62-255810 und JP 62-126384)gezeigt, daß Myristoylglycin oder Oligopeptidderivate eine inhibierende Wirkung gegen die Transformation oder Vermehrung von Zellen oder die Vervielfachung von Retroviren ausüben.
- Die Erfindung betrifft insbesondere die Verbindungen der Formel (I)
- in der

- - eine geradkettige oder verzweigte (C&sub1;-C&sub6;)-Alkylgruppe, die nichtsubstituiert oder durch eine oder mehrere Hydroxyl-, Amino-, Carboxy-, Carbamoyl-, Benzylthio-, Methylthio-, Mercapto- oder Phenyl-gruppen (welche nichtsubstituiert oder durch eines oder mehrere Halogenatome oder Hydroxylgruppen, geradkettige oder verzweigte (C&sub1;-C&sub6;)-Alkylgruppen, geradkettige oder verzweigte (C&sub1;-C&sub6;)-Alkoxygruppen oder (CH&sub3;-CH&sub2;-O)&sub2;-PO-CH&sub2;-) substituiert ist),

- - eine (Imidazol-2-yl)-methylgruppe oder eine (Indol-3-yl)-methylgruppe, die nichtsubstituiert oder am Heterocyclus durch eine Benzyl-, Benzhydryl-, Trityl-, Benzyloxymethyl-, Tosyl-, geradkettige oder verzweigte (C&sub1;-C&sub6;)-Alkylgruppe oder Phenylgruppe substituiert ist,
- - eine (1-Azaindolizin-2-yl)-methylgruppe der Formel:

- eine geradkettige oder verzweigte Alkylgruppe mit 6 bis 21 Kohlenstoffatomen, die nichtsubstituiert oder an der endständigen Methylgruppe durch eine Hydroxyl-, Mercapto-, Phenyl- oder Ethinyl-gruppe substituiert ist und worin eine oder mehrere Methylengruppen durch ein Sauerstoffatom, ein Schwefelatom oder eine p-Phenylengruppe ersetzt sein können (mit der Maßgabe, daß, wenn R&sub2; oder R&sub3; mit den Stickstoff- oder Kohlenstoffatomen, an die sie gebunden sind, einen 1,2,3,4-Tetrahydroisochinolin-, Indolin- oder Isoindolin-Ring bilden, R&sub4; von der Gruppe Alk-S-(CH&sub2;)n verschieden ist, worin n einen Wert von 1, 2, 3, 4 oder 5 und Alk eine geradkettige oder verzweigte (C&sub4;-C&sub1;&sub9;)-Alkylgruppe darstellen),
- bedeuten,
- deren Isomere, Diastereoisomere und Epimere sowie derenadditionssalze mit einer pharmazeutisch annehmbaren Säure oder Base.
- Als pharmazeutisch annehmbare Säuren kann man ohne Einschränkung Chlorwasserstoffsäure, Schwefelsäure, Weinsäure, Maleinsäure, Fumarsäure, Oxalsäure, Methansulfonsäure. Camphersäure etc. nennen.
- Als pharmazeutisch annehmbare Basen kann man ohne Einschränkung Natriumhydroxid, Kaliumhydroxid, tert.-Butylamin... nennen.
- Die Erfindung betrifft weiterhin das Verfahren zur Herstellung der Verbindungen der Formel (I), das dadurch gekennzeichnet ist, daß man:
- 1/ dann, wenn die herzustellenden Derivate der Formel (I) eine Grupve R&sub4; = R4' aufweisen, worin keine Methylengruppe durch ein Schwefelatom oder ein Sauerstoffatom ersetzt ist,
- man eine Verbindung der Formel (II):
- R4' - X' - Z (II)
- in der:
- X' eine Gruppe - -, -SO&sub2;-,
- R&sub4; eine geradkettige oder verzweigte Alkylgruppe mit 6 bis 21 Kohlenstoffatomen, die nichtsubstituiert oder an der endständigen Methylgruppe durch eine Hydroxyl-, Mercapto-, Phenyl- oder Etlilnylgruppe substituiert ist, und worin eine oder mehrere Methylengruppen durch eine p-Phenylengruppe ersetzt sein können,
- Z ein Halogenatom, eine Hydroxylgruppe, eine geradkettige oder verzweigte (C&sub1;-C&sub6;)-Alkoxygruppe, eine Aralkoxygruppe oder eine Gruppe
- bedeuten,
- mit einem gegebenenfalls geschützen (in Form des Racemats oder der Isomeren vorliegenden) Amin der Formel (III)
- in der R&sub2;, R&sub3; und R&sub4; die bezüglich der Formel I angegebenen Bedeutungen besitzen und Y' eine Gruppe der Formeln -CO&sub2;H, -CO(Alkoxy) oder -PO(Alkoxy)&sub2; darstellen, kondensiert,
- so daß man nach der eventuellen Abspaltung der Schutzgruppe eine Verbindung der Formel (I/a) erhält, einem Sonderfall der Verbindungen der Formel (I):
- in der R&sub1;, R&sub2;, R&sub3;, R4', X' und Y' die oben angegebenen Bedeutungen besitzen,
- 2/ dann, wenn die herzustellenden Derivate der Formel (I) eine Grupve R&sub4; = R4" = CH&sub3;-(CH&sub2;)n-, worin mindestens eine der Methylengruppen durch ein Schwefelatom oder ein Sauerstoffatom ersetzt ist, aufweisen,
- man:
- eine Verbindung der Formel (IV)
- A-(CH&sub2;)m-X'-Z (IV)
- in der:
- X' und Z die oben angegebenen Bedeutungen besitzen,
- A ein Halogenatom, eine Mesyloxygruppe oder eine Tosyloxygruppe und
- m kleiner oder gleich n - 1 ist,
- bedeuten,
- und eine der Methylengruppen durch ein Sauerstoffatom, ein Schwefelatom, eine p-Phenylengruppe, eine Gruppe -CH(CH&sub3;)- oder eine Gruppe -C(CH&sub3;)&sub2;- ersetzt sein kann,
- mit einem gegebenenfalls geschützten (in Form des Racemats oder der Isomeren vorliegenden) Amin der Formel (III), wie es oben definiert worden ist, kondensiert zur Bildung einer Verbindung der Formel (V)

- Neben der Tatsache, daß sie neu sind, besitzen die Verbindungen der Formel (I) sehr interessante pharmakologische Eigenschaften.
- Sie stellen starke Inhibitoren der Myristoylierung von Proteinen, wie gag, dar, über das Enzym, das für diese Myristoylierung verantwortlich ist, d. h. die N- Myristoyltransferase (NMT).
- Da NMT sich ln einer Vielzahl von biologischen Quellen findet, kann sie entweder cytosolischen oder mikrosomalen Ursprungs sein, wie es J.A. BOUTIN et coll. gezeigt haben (BIOCHEMICAL Journal (1990), eingereicht). Das mikrosomale Enzym erkennt eine Vielzahl von endogenen Proteinen, Produkte von Onkogenen oder Strukturproteine von Viren. Das cytosolische Enzym seinerseits erkennt die endogenen Proteine schwieriger.
- Die erfindungsgemäßen Verbindungen werden nicht nur von dem mikrosomalen Enzym, sondern auch von dem cytosolischen Enzym erkannt. Sie inhibieren gleichzeitig und in überraschender Weise die mikrosomalen und cytosolischen Wirkungen.
- Die Verwendung der erflndungsgemäßen Verbindungen als NMT-Inhibitoren führt zu einer Inhibierung der Wirkung dieses Enzyms, die deutlich stärker ist als die der in dem Stand der Technik als Inhibitoren der Vermehrung von Krebszellen und Retroviren beschriebenen Verbindungen.
- In der Tat wurde eine umfangreiche Untersuchung der von den erfindungsgemäßen Verbindungen auf die Vermehrung und die Transformation von Zellen ausgeübte Wirkung ausgehend von Krebszellen murinen Ursprungs (L1210) oder menschlichen Ursprungs (HL60) durchgeführt. Nach der Extraktion des Enzyms aus diesem biologischen Medium und der Bestimmung seiner Aktivität scheint es so zu sein, daß die Zugabe der erfindungsgemäßen Verbindungen seine Aktivität in starkem Maße inhlbiert.
- Darüber hinaus entfalten die erfindungsgemäßen Verbindungen eine cytotoxische Wirkung auf in Kultur gezüchtete Krebszellen, wie L1210 (murinen Ursprungs) oder HL60 (menschlichen Ursprüngs). Diese Cytotoxizität hat sich als deutlich stärker erwiesen als diejenige von N-Myristoylglycin auf diese Zellen.
- Andererseits schützen die erfindungsgemäßen Verbindungen über diese Inhibierung der Wirkung von NMT menschliche T-Lymphozytenzellen (CEM) in Kulturen von Infektionen durch den HW-1-Virus.
- Diese Inhibierung der Wirkung ist um so interessanter, als dieses Enzym eine überwiegende Rolle spielt, Insbesondere bei der Reifung entweder von transformierenden Proteinen, die bei bestimmten Krebsen auftreten, oder von Proteinen, die selbst an der Reifung von Viren beteiligt sind.
- Die erfindungsgemäßen Verbindungen können daher potentiell Anwendung finden bei der Behandlung von Krebs und/oder Viruserkrankungen, deren Reifung eine Myristoylierung umfaßt, wie AIDS, Herpes, Hepatitis B, Influenza, Poliomyelitis und Leukämien.
- Die vorliegende Erfindung betrifft weiterhin pharmazeutische Zubereitungen, die als Wirkstoff eine Verbindung der allgemeinen Formel (I) oder eines ihrer Additionssalze mit einer pharmazeutisch annehmbaren Base allein oder ln Kombination mit einem oder mehreren inerten, nichttoxischen Trägermaterialien oder Bindemitteln enthalten.
- Als erfindungsgemäße pharmazeutische Zubereitungen kann man jene nennen, die zur Verabreichung auf oralem, parenteralem oder nasalem Wege geeignet sind, einfache oder dragierte Tabletten, Sublingualtabletten, Sacchets, Päckchen, Gelkapseln, Lutschtabletten, Kompretten, Suppositorien etc.
- Die Dosierung variiert in Abhängigkeit von dem Alter und dem Gewicht des Patienten, der Art und der Schwere der Erkrankung sowie dem Verabreichungsweg. Dieser kann oral, nasal, rektal oder parenteral sein. Ganz allgemein erstreckt sich die Einheitsdosis zwischen 0,1 und 100 mg pro Behandlung bei 1 bis 3 Gaben innerhalb von 24 Stunden.
- Die folgenden Beispiele dienen der weiteren Erläuterung der Erfindung, ohne sie jedoch einzuschränken.
- Die Stellungen der Substituenten von Histidln und Spinacin in den Beispielen werden wie folgt angegeben: Histidin Spinacin
- Die nachfolgend angegebene Herstellung ermöglicht nicht die Herstellung der erfindungsgemäßen Verbindungen, sie führt jedoch zu einem Ausgangsmaterial, das für die Synthese der erfindungsgemäßen Produkte nützlich ist.
- Man beschlckt einen Kolben unter einer Stickstoffatmosphäre mit 200 ml Tetrahydrofuran, 120 mMol Lithlumaluminiumhydrid und gibt dann im Verlaufe von 40 Minuten unter Rühren 120 mMol 2-Carbethoxylmidazo(1,2-a]pyridin, das 30 man nach der von J.G. LOMBARDINO beschriebenen Methode (J. Org. Chem. 30 (1965), 2403-2407) hergestellt hat, in Lösung in 150 ml Tetrahydrofuran zu. Man setzt das Rühren während 20 Stunden bei Raumtemperatur fort. Man hydrolysiert die Mischung mit 50 ml Isopropanol und dann mit 50 ml einer gesättigten Natriumchioridlösung. Nach dem Eindampfen zurtrockne erhält man das erwartete Produkt nach der Reinigung über einer mit Siliciumdioxid be schickten Säule (Elutionslösungsmittel: Dichlormethan/Methanol: 95/5).
- Ausbeute: 31 %.
- Man beschickt einen Kolben mit 40 ml Thionylchlorid und gibt dann portionsweise 34 mMol des in der vorhergehenden Stufe erhaltenen Produkts zu. Man erhitzt das Ganze dann während 20 Minuten zum Sleden am Rückfluß, dampft zur Trockne ein, nimmt den Rückstand mit 100 ml Toluol auf, dampft ein und nimmt erneut mit 50 ml Isopropanol auf. Man filtriert den Niederschlag ab und wäscht ihn mit Dichlormethan.
- Ausbeute: 61 %.
- Man beschickt einen Kolben mit 100 ml Ethanol und dann portionsweise mit 0,056 Grammtom Natrium. Nachdem das Natrium reagiert hat, gibt man 8,05 g Acetamidomalonsäureethylester zu und rührt das Ganze, wobei man die Temperatur auf 20ºC kommen läßt. Dann gibt man im Verlaufe von 10 Minuten portionsweise 19 mMol des in der vorhergehenden Stufe erhaltenen Produkts zu. Man rührt während 18 Stunden bei Raumtemperatur, verdampft das Ethanol und gibt 200 ml 1N Chlorwasserstoffsäure zu. Man wäscht diese wäßrige Phase mit Ethylacetat, stellt mit Natrlumcarbonat alkalisch und extrahiert mit Ethylacetat. Man wäscht die organische Phase dann mit Wasser und dampft ein. Man hydrolysiert das erwartete Produkt, das in Form der Base anfällt, durch Erhitzen während 6 Stunden in 6N Chlorwasserstoffsäure zum Rückfluß zu dem Hydrochlorld, dampft zur Trockne ein, nimmt mit Wasser auf und bindet an ein Harz. Man eluiert mit 10 %-igem Ammoniak, das dann verdampft wird.
- Ausbeute: 74 %
- Man löst 60 mMol N-Hydroxysuccinimld unter Rühren in 200 ml Ethylacetat. Man gibt dann eine Lösung von 60 mMol Myristinsäure in 100 ml Ethylacetat und dann 60 mMol Cyclohexylcarbodiimin zu. Nach 20-stündigem Rühren bei Raumtemperatur werden der gebildete Dicyclohexylharnstoff abfiltriert und das Filtrat eingedampft. Man erhält das erwartete Produkt nach der Umkristallisation aus Ethanol.
- Ausbeute: 75 %

- Unter Anwendung der von Y. LAPIDOT und S. RAPPOPORT (J. of Lipid. Research 8 (1967), 142-145) beschriebenen Methode erhält man ausgehend von dem in der vorhergehenden Stufe beschriebenen Produkt und 3-(S)-Carboxy- (1,2,3,4)-tetrahydrolsochinolin das erwartete Produkt, welches durch Chromatographie über Kieselgel gereinigt wird (Elutionslösungsmittel: CH&sub2;Cl&sub2;/MeOH: 95/5).

- Man züchtet sie in dem Medium RPMI 1640, das mit 50 U/ml Penicillin, 50 uM Streptomycin, 2 mM Glutamin, 10 mM HEPES und 10 % Kalbsfötenserum ergänzt ist und hält ln einer Atmosphäre aus 5 % CO&sub2; und 95 % Luft bei 37ºC.
- Man erntet die Zellen und wäscht sie durch Zentrifugieren bei geringer Geschwindigkeit in PBS. Der letztlich erhaltene Zellen-Zentrifugenrückstand wird erneut in einem HEPES-Puffer 50 mM, pH 7,4, 2 mM EGTA, 1 mM DTT und 1mM PMSF suspendiert. Die Zellen werden durch Ultraschallbehandlung bei 4ºC zerstört und in elner Glas/Glas-Potter-Einrichtung durch Hin- und Herführen homogenisiert. Das Zellenhomogenisat wird anschließend bei geringer Drehzahl (10.000 min&supmin;¹ während 10 Minuten) zentrlfugiert, um die Zellenbruckstücke abzutrennen, wonach man die überstehende Flüssigkeit während 1 Stunde einer Ultrazentrifugation bei 105.000 g unterwirft.
- Anschließend verwendet man die überstehende Flüssigkeit als Quelle für NMT. Die Bestimmung der Aktivität erfolgt nach der von Towler und Glazer beschriebenen Methode (PNAS 83 (1986), 2812) unter Verwendung entweder des vom NH&sub2;-terminalen Ende des Onkogenprodukts pp6osrc: GSSKSKPKDP (DP) oder des vom NH&sub2;-terminalen Ende des Produkts von gag des Strukturproteins des Murinleukämievirus von Moloney: GQTVTTPL (T3) abgeleitete Peptid in einer Endkonzentration von 0,3 mM als Peptidsubstrat.
- Bei diesen Bedingungen zeigen sämtliche erfindungsgemäßen Verbindungen eine deutlich bessere Wirkung als N-Myristoylglycin.
- Insbesondere zeigen die Verbindung des Beispiels 1 einen IC&sub5;&sub0;-Wert von 1,8 10&supmin;&sup7; M, die Verbindung des Beispiels 5 einen IC&sub5;&sub0;-Wert von 6 10&supmin;&sup7; M und die Verbindung des Beispiels 16 einen IC&sub5;&sub0;-Wert von 2 10&supmin;&sup6; M.
- Man züchtet die unsterblichenlymphozyten T (CEM) in der Masse, wie es in Beispiel 57 beschrieben ist, und gewinnt sie dann durch Zentrifugieren und wäscht mit PBS.
- Man behandelt die Zellen mit Ultraschall (3 kurze Perioden von 10 Sekunden) und homogenisiert dann in einer Glas/Glas-Potter-Einrichtung.
- Dieses Homogenisat, in dem sämtliche Zellen zerstört worden sind (Überprüfung unter dem Mikroskop), wird dann während 1 Stunde und 10 Minuten bei 150.000 g zentrifugiert. Man gewinnt unabhängig voneinander den Zentrifugenrückstand (Mikrosomen) und die überstehende Flüssigkeit (Cytosol) und friert die Produkte ein.
- Anschließend mißt man die Wirkung der NMT nach der Verfahrensweise von Towler und Glaser, unterbricht jedoch die Reaktion durch 200 ul Acetonitril, um eine Ausfällung des myristoylierten Peptids zu vermeiden. Dieses Peptid aus Aminosäuren: NH&sub2;GARASVLSGGELDRWEKIRLLCOOH ist von p 18 des HW-1- Virus (Untertyp BRU) abgeleitet.
- Diese Untersuchungen erfolgen mit einer cytosolischen Quelle (40 ul) oder einer mikrosomaien Quelle (20 ul, behandelt mit 4 ul Triton 770, 10 %) von CEM- Zellen in Gegenwart von steigenden Konzentrationen der erfindungsgemäßen Verbindungen (1 bis zur Endkonzentration von 1000 uM). Die während 30 Minuten bei 37ºC inkubierten und mit 200 ul Acetonitril gestoppten Proben werden anschließend mit HPLC analysiert und es wird die Myristoylierung des p18 quantitativ bestimmt.
- Unter diesen Bedingungen zeigt die Verbindung des Beispiels 1 bei der Inkubierung mit der mikrosomalen Fraktion einen IC&sub5;&sub0;-Wert von 10&supmin;&sup4; M und bei der Inkubation in dem Cytosol der Zellen CEM einen IC&sub5;&sub0;-Wert von 8 10&supmin;&sup6; M.
- Die angewandte Methode ist von WEISLOW et coll. beschrieben worden (J. Natl. Cancer Inst. 81(1989), 577).
- Unter diesen Bedingungen zeigt die Verbindung des Beispiels 49 eine Schutzwirkung (EC&sub5;&sub0;) bei einer Konzentration von 10&supmin;&sup5; M und eine Toxizität (IC&sub5;&sub0;) bei einer Konzentration von 2 10&supmin;&sup4; M, so daß der therapeutische Index 20 beträgt.
- Diese Ergebnisse sind in der beigefügten Zeichnung dargestellt.
- Die in einer Kultur vorliegenden Zellen HL60 werden während 6 Tagen mit den Verbindungen behandelt, die in einer minimalen Menge Methanol gelöst sind und in 9 unterschiedlichen Konzentrationen eingesetzt werden. Die ausgewählten Dosierungen liegen um den IC&sub5;&sub0;-Wert herum, der zuvor unter Verwendung der MTT-Analysenmethode ermittelt worden ist (Carmichael et al., Cancer Res. 47 (1986), 936).
- Die Zellen werden gewaschen (450 ul), erneut in dem gleichen vollständigen Medium suspendiert und während 1 Stunde bei 37ºC in Gegenwart von 7 ug/ml für DNA spezifischen Fluorchroms inkubiert.
- Man bewirkt die Messungen mit Hilfe eines Laser-Cytometers, das es ermöglicht, den Prozentsatz der ln der Phase G0 und G1 des Zellzyklus gewachsenen und damit differenzierten Zellen abzuschätzen.
- Bei diesen Bedingungen zeigen die Verbindungen eine deutliche differenzierende Wirkung, die bis zu Dosierungen unterhalb 50 ug/ml reichen können, wie es für die Verbindungen der Beispiele 1, 16 oder 49 zutrifft.

Claims (8)
1. Verbindungen der allgemeinen Formel (I)
in der
R&sub1;
- ein Wasserstoffatom,
- eine geradkettige oderverzweigte (C&sub1;-C&sub6;)-Alkylgruppe, die nichtsubstituiert
oder durch eine oder mehrere Hydroxyl-, Amino-, Carboxy-, Carbamoyl-,
Benzylthio-, Methylthio-, Mercapto- oder Phenyl-gruppen (welche
nichtsubstituiert oder durch eines oder mehrere Halogenatome oder
Hydroxylgruppen, geradkettige oder verzweigte (C&sub1;-C&sub6;)-Alkylgruppen, geradkettige oder
verzweigte (C&sub1;-C&sub6;)-Alkoxygruppen oder (CH&sub3;-CH&sub2;-O)&sub2;-PO-CH&sub2;-)
substituiert ist),
- eine Phenylgruppe, die nichtsubstituiert oder durch eines oder mehrere
Halogenatome oder Hydroxylgruppen oder geradkettige oder verzweigte (C&sub1;-
C&sub6;)-Alkylgruppen substituiert ist,
- eine (C&sub3;-C&sub7;)-Cycloalkyl-methylgruppe,
- eine (Imidazol-2-yl)-methylgruppe oder eine (Indol-3-yl)-methylgruppe, die
nichtsubstituiert oder am Heterocyclus durch eine Benzyl-, Benzhydryl-,
Trityl-, Benzyloxymethyl-, Tosyl-, geradkettige oder verzweigte
(C&sub1;-C&sub6;)-Alkylgruppe oder Phenylgruppe substituiert ist,
- eine (1-Azaindolizin-2-yl)-methylgruppe der Formel:
R&sub2; ein Wasserstoffatom oder eine geradkettige oder verzweigte
(C&sub1;-C&sub6;)-Alkylgruppe,
R&sub3; ein Wasserstoffatom oder eine geradkettige oder verzweigte
(C&sub1;-C&sub6;)-Alkylgruppe,
oder
wenn R&sub1; ein Wasserstoffatom darstellt,
R&sub2; und R&sub3; gemeinsam mit den Kohlenstoff- und Stickstoffatomen, an die sie
gebunden sind, einen der folgenden Heterocyclen bilden können:
Rx und Rx', die gleichartig oder verschieden sein können, ein Wasserstoffatom,
eine geradkettige oder verzweigte (C&sub1;-C&sub6;)-Alkylgruppe, eine
Hydroxylgruppe, eine geradkettige oder verzweigte (C&sub1;-C&sub6;)-Alkoxygruppe oder ein
Halogenatom,
oder Rx und Rx', wenn sie an zwei benachbarten Kohlenstoffatomen
angeordnet sind, gemeinsam eine Methylendioxy- oder Ethylendioxy-gruppe
bilden,
Ry ein Wasserstoffatom, eine geradkettige oder verzweigte
(C&sub1;-C&sub6;)-Alkylgruppe, eine Arylgruppe, eine Aralkylgruppe, eine Aroylgruppe oder eine
Alylsulfonylgruppe,
X eine der folgenden Gruppen:
Y eine Gruppe -CO-R&sub5; oder
R&sub5; eine Hydroxylgruppe, eine geradkettige oder verzweigte
(C&sub1;-C&sub6;)-Alkoxygruppe, H&sub2;N-CO-CH&sub2;-O-, HO-CH&sub2;-CHOH-CH&sub2;-O-,
R&sub7; und R&sub8;, die gleichartig oder verschieden sein können, ein Wasserstoffatom,
eine geradkettige oder verzweigte (C&sub1;-C&sub6;)-Alkylgruppe oder gemeinsam mit
dem Stickstoffatom, an das sie gebunden sind, einen Pyrrolidin-, Piperidin-,
Morpholin- oder Piperazin-Ring bilden,
R&sub6; und R6', die gleichartig oder verschieden sein können, ein Wasserstoffatom,
eine Hydroxylgruppe oder eine geradkettige oder verzweigte
(C&sub1;-C&sub6;)-Alkoxygruppe,
R&sub4;
eine geradkettige oder verzweigte Alkylgruppe mit 6 bis 21
Kohlenstoffatomen, die nicht substituiert oder an der endständigen Methylgruppe durch
eine Hydroxyl-, Mercapto-, Phenyl- oder Ethinyl-gruppe substituiert ist und
worin mindestens eine der Methvlengruppen durch ein Sauerstoffatom, ein
Schwefelatom oder einen p-Phenylenkern ersetzt ist,
dann, wenn:
R&sub1;
- ein Wasserstoffatom,
- eine geradkettige oder verzweigte (C&sub1;-C&sub6;)-Alkylgruppe, die nicht
substituiert oder durch eine oder mehrere Hydroxyl-, Amino-, Carboxy-,
Carbamoyl-, Benzylthio-, Methylthio-, Mercapto- oder Phenylgruppen
(die nichtsubstituiert oder durch eine Hydroxylgruppe oder eine
geradkettige oder verzweigte (C&sub1;-C&sub6;)-Alkoxygruppe substituiert ist),
- eine nichtsubstituierte Phenylgruppe,
- eine (Imidazol-2-yl)-methylgruppe oder eine (Indol-3-yl)-methylgrup
pe, die nichtsubstituiert oder am Heterocyclus durch eine Methylgruppe
substituiert ist, und
R&sub2;=H, R&sub3;=H,X= - -, Y=COR5'(R5'=OH,Alkoxy)
und in den anderen Fällen:
eine geradkettige oder verzweigte Alkylgruppe mit 6 bis 21
Kohlenstoffatomen, die nichtsubstituiert oder an der endständigen Methylgruppe durch
eine Hydroxyl-, Mercapto-, Phenyl- oder Ethinyl-gruppe substituiert ist und
worin eine oder mehrere Methylengruppen durch ein Sauerstoffatom, ein
Schwefelatom oder eine p-Phenylengruppe ersetzt sein können (mit der
Maßgabe, daß, wenn R&sub2; oder R&sub3; mit den Stickstoff- oder Kohlenstoffatomen, an
die sie gebunden sind, einen 1,2,3,4-Tetrahydroisochinolin-, Indolin- oder
Isoindolin-Ring bilden, R&sub4; von der Gruppe Alk-S-(CH&sub2;)n- verschieden ist,
worin n einen Wert von 1, 2, 3, 4 oder 5 und Alk eine geradkettige oder
verzweigte (C&sub4;-C&sub1;&sub9;)-Alkylgruppe darstellen),
bedeuten,
deren Isomere, Diastereoisomere und Epimere sowie derenadditionssalze mit
einer pharmazeutisch annehmbaren Säure oder Base.
2. Verbindung nach Anspruch 1, nämlich
N-Myristoyl-3-carboxy-(1,2,3,4)tetrahydroisochinolin, dessen Enantiomere sowie dessen Additionssalze mit
einer pharmazeutisch annehmbaren Base.
3. Verbindung nach Anspruch 1, nämlich N-Myristoyl-(N'τ-benzyl)-histidin,
dessen Enantiomere sowie dessen Additionssalze mit einer pharmazeutisch
annehmbaren Säure oder Base.
4. Verbindung nach Anspruch 1, nämlich
1-Myristoylamino-2-phenylethanhydrogenphosphonsäureethylester, dessen Enantiomere sowie dessen
Additionssalze mit einer pharmazeutisch annehmbaren Base.
5. Verbindung nach Anspruch 1, nämlich
N-(12-Methoxylauroyl)-phenylalanin, dessen Enantiomere sowie dessen Additionssalze mit einer pharmazeutisch
annehmbaren Base.
6. Verfahren zur Herstellung der Derivate der Formel (I) nach Anspruch 1,
dadurch gekennzeichnet, daß man:
1- dann, wenn die herzustellenden Derivate der Formel (I) eine Gruppe R&sub4; =
R4' aufweisen. worin keine Methylengruupe durch ein Schwefelatom oder ein
Sauerstoffatom ersetzt ist.
man eine Verbindung der Formel (II):
in der:
X' eine Gruppe - -, SO&sub2;,
R4' eine geradkettige oder verzweigte Alkylgruppe mit 6 bis 21
Kohlenstoffatomen, die nichtsubstituiert oder an der endständigen Methylgruppe durch
eine Hydroxyl-, Mercapto-, Phenyl- oder Ethinylgruppe substituiert ist, und
worin eine oder mehrere Methylengruppen durch eine p-Phenylengruppe
ersetzt sein können,
Z ein Halogenatom, eine Hydroxylgruppe, eine geradkettige oder verzweigte
(C&sub1;-C&sub6;)-Alkoxygruppe, eine Aralkoxygruppe oder eine Gruppe
bedeuten,
mit einem gegebenenfalls geschützen (in Form des Racemats oder der Isomeren
vorliegenden) Amin der Formel (III)
in der R&sub2;, R&sub3; und R&sub4; die bezüglich der Formel I angegebenen Bedeutungen
besitzen und Y' eine Gruppe der Formeln -CO&sub2;H, -CO(Alkoxy) oder -PO(Alkoxy)&sub2;
darstellen, kondensiert,
so daß man nach der eventuellen Abspaltung der Schutzgruppe eine Verbindung
der Formel (I/a) erhält, einem Sonderfall der Verbindungen der Formel (I):
in der R&sub1;, R&sub2;, R&sub3;, R&sub4;', X' und Y' die oben angegebenen Bedeutungen besitzen,
2- dann, wenn die herzustellenden Derivate der Formel (I) eine Gruppe R&sub4; =
R4' = CH&sub3;-(CH&sub2;)n-, worin mindestens eine der Methvlengruppen durch ein
Schwefelatom oder ein Sauerstoffatom ersetzt ist, aufweisen,
man:
eine Verbindung der Formel (IV)
A - (CH&sub2;)m - X' - Z (IV)
in der:
X' und Z die oben angegebenen Bedeutungen besitzen,
A ein Halogenatom, eine Mesyloxygruppe oder eine Tosyloxygruppe und
m kleiner oder gleich n - 1 ist,
bedeuten,
und eine der Methylengruppen durch ein Sauerstoffatom, ein Schwefelatom, eine
p-Phenylengruppe, eine Gruppe -CH(CH&sub3;)- oder eine Gruppe -C(CH&sub3;)&sub2;- ersetzt
sein kann,
mit einem gegebenenfalls geschützten (in Form des Racemats oder der Isomeren
vorliegenden) Amin der Formel (III), wie es oben definiert worden ist,
kondensiert zur Bildung einer Verbindung der Formel (V)
in der A, m, X', R&sub1;, R&sub2;, R&sub3; und Y' die oben angegebenen Bedeutungen besitzen,
welche man mit einem Derivat der Formel (VI):
R&sub9; - B - M (VI)
in der:
R&sub9; eine Gruppe der Formel CH&sub3;-(CH&sub2;)p- die nichtsubstituiert oder an der
endständigen Methylgruppe durch eine Hydroxyl-, Mercapto-, Phenyl- oder
Ethinylgruppe substituiert ist und bei der eine der Methylengruppen durch
ein Sauerstoffatom, ein Schwefelatom, eine p-Phenylengruppe, eine Gruppe
-CH(CH&sub3;)- oder eine Gruppe -C(CH&sub3;)&sub2;- ersetzt sein kann,
B ein Sauerstoffatom oder ein Schwefelatom,
M ein aus Natrium, Kalium oder Caesium ausgewähltes Metall und
p einen solchen Wert besitzt, daß m + p kleiner oder gleich n - 1 ist,
bedeuten,
umsetzt, so daß man nach der eventuellen Abspaltung der Schutzgruppe eine
Verbindung der Formel (I/b) erhält, einem Sonderfall der Verbindungen der
Formel (I):
in der R&sub9;, B, m, X', R&sub1;, R&sub2;, R&sub3;und Y' die oben angegebenen Bedeutungen
besitzen,
welche Derivate der Formeln (I/a) und (I/b) man auch vereinfacht wie folgt
schreiben kann:

a dann, wenn X' eine Gruppe der Formel
darstellt, durch katalytische Hydrierung ln die Verbindungen der Formel
(I/c) umgewandelt werden können, einem Sonderfall der Verbindungen der
Formel (I):
in der R&sub1;, R&sub2;, R&sub3;, R&sub4; und Y' die oben angegebenen Bedeutungen besitzen,
b dann, wenn Y' eine Gruppe -CO(Alkoxy) oder -PO(Alkoxy)&sub2; darstellt,
vollständig oder teilweise verselft werden können, so daß man die Verbindungen
der Formeln (I/d), (I/e) bzw. (I/f) erhält, Sonderfälle der Verbindungen der
Formel (I):
worin R&sub1;, R&sub2;, R&sub3;, R&sub4; und X' die oben angegebenen Bedeutungen besitzen,
c dann, wenn Y' eine Gruppe -CO&sub2;H darstellt, ln Gegenwart von
Caesiumcarbonat mit Hilfe von Chloracetamid in die Verbindung der Formel (I/g)
umgewandelt
werden können, einem Sonderfall der Verbindungen der Formel (I):
in der R&sub1;, R&sub2;, R&sub3;, R&sub4; und X' die oben angegebenen Bedeutungen besitzen,
d dann, wenn Y' eine Gruppe -CO&sub2;H darstellt,
mit Hilfe von Isopropylidenglycerol zu einer Verbindung der Formel (I/h)
verestert werden können, einem Sonderfall der Verbindungen der Formel (I):
in der R&sub1;, R&sub2;, R&sub3; und R&sub4; die oben angegebenen Bedeutungen besitzen,
welches Derivat der Formel (I/h) in saurem Medium hydrolysiert werden
kann zur Bildung der Verbindung der Formel (I/i), einem Sonderfall der
Verbindungen der Formel (I):
in der R&sub1;, R&sub2;, R&sub3; und R&sub4; die oben angegebenen Bedeutungen besitzen,
welche Verbindungen der Formeln (I/a) bis (I/i) gegebenenfalls mit Hilfe einer
klassischen Reinigungsmethode gereinigt werden können, dessen Isomeren mit
Hilfe einer klassischen Trennmethode getrennt werden können und welche man
mit einer pharmazeutisch annehmbaren Säure oder Base in ihre
Säureadditionssalze umwandeln kann.
7. Pharmazeutische Zubereitungen enthaltend als Wirkstoff mindestens eine
Verbindung nach einem der Ansprüche 1 bis 5 allein oder in Kombination mit
einem oder mehreren inerten, nichttoxischen, pharmazeutisch annehmbaren
Trägermaterialien.
8. Pharmazeutische Zubereitungen nach Anspruch 7 für die Behandlung von
Krebs und/oder Virenerkrankungen, deren Ausbrechen eine Myristoylierung
impliziert, wie AIDS, Herpes, Hepatitis B, Influenza, Poliomyelitis oder Leukämie.
