Description
- Die vorliegende Erfindung betrifft UCN-1028D-Derivate mit einer hemmenden Wirkung auf Proteinkinase C und Antitumorwirkung, die als Antitumormittel verwendbar sind.
- Als Substanz mit einer Perylenchinongrundstruktur, die mit den Verbindungen der vorliegenden Erfindung strukturverwandt ist, ist Phleichrom, das als Pflanzengift aus im Grünland Flecken verursachendem Cladosporium phlei isoliert wird [Agricultural & Biological Chemistry 39, 1683 (1975), J. Jap. Pasture, 28 (4), 426 (1983)], bekannt. Ähnliche Substanzen UCN-1028A und UCN-1028C mit Antitumorwirkung und hemmender Wirkung auf Proteinkinase C werden in der Europäischen Patentveröffentlichung Nr. 0284358 offenbart.
- Die Strukturformeln von UCN-1028A und UCN-1028C sind nachstehend dargestellt.
- Die vorliegende Erfindung stellt UCN-1028D-Derivate [nachstehend als Verbindungen (I) bezeichnet] der allgemeinen Formel (I)
- zur Verfügung, wobei Y - - oder - -O-bedeutet; mit der Maßgabe, daß, wenn Y - - bedeutet, der Perylenchinonring an der Bindung zwischen den 6a- und 6b-Positionen R- Konfiguration annimmt, und daß, wenn Y - -O- bedeutet, der Perylenchinonring an der Bindung zwischen den 6a- und 6b-Positionen 5-Konfiguration annimmt.
- In den Perylenchinonderivaten mit sperrigen Substituenten an den 1- und 12-Positionen und an den 6- und 7-Positionen nimmt die Perylenchinongrundstruktur aufgrund dieser Substituenten keine planare Struktur ein, sondern verdreht sich entlang seiner Achse der Bindung zwischen den 6a- und 6b-Positionen und den 12a- und 12b-Positionen. Es liegen daher Isomere des Perylenchinonringes vor.
- Das heißt, UCN-1028D mit der folgenden Strukturformel
- in der der Perylenchinonring an der Bindung zwischen den 6a- und 6b-Positionen S-Konfiguration einnimmt, wird durch Erhitzen in einer Toluol- oder Xylollösung zu einem Isomeren von UCN-1028D (nachstehend als UCN-1028 iso-D bezeichnet) der folgenden Strukturformel
- isomerisiert, in der der Perylenchinonring an der Bindung zwischen den 6a- und 6b-Positionen R-Konfiguration einnimmt. Diese Verbindungen erreichen das Gleichgewicht bei einem Verhältnis von 1:1.
- UCN-1028D und UCN-1028 iso-D können durch Dünnschichtchromatographie oder Säulenchromatographie unter Verwendung von Silicagel voneinander getrennt werden. UCN-1028 iso-D kann unter den gleichen Bedingungen wie bei der Isomerisierung von UCN-1028D thermisch zu UCN-1028D isomerisiert werden.
- Die vorliegende Erfindung scmießt das UCN-1028D-Derivat und das UCN-1028 iso- D-Derivat und ein Gemisch davon ein.
- Die Verbindungen (I) können durch die folgenden Schritte hergestellt werden.
- UCN-1028D kann durch alkalische Hydrolyse von UCN-1028A oder UCN-1028C erhalten werden.
- Die Hydrolyse von UCN-1028A oder UCN-1028C verläuft zugig durch Behandlung von UCN-1028A oder UCN-1028C mit einer wäßrigen Lösung einer anorganischen Base, wie Natriumhydroxid, Kaliumhydroxid, Bariumhydroxid, Natriumcarbonat oder Kaliumcarbonat, in einem mit Wasser mischbaren Lösungsmittel, wie Methanol, Ethanol, Aceton oder Dimethylsulfoxid bei 0ºC bis 100ºC.
- Die Verbindung (I-1) [Verbindung (I), wobei Y - - bedeutet) und die Verbindung (I-2) [Verbindung (I), wobei Y - -O- bedeutet) können durch die Verfahren 1 beziehungsweise 2 hergestellt werden.
- Die Verbindung (I-1) kann durch Umsetzung von UCN-1028D oder UCN-1028 iso- D mit mindestens 2 Äquivalenten eines reaktiven Derivates einer Carbonsäure, zum Beispiel eines Säureanhydrids oder eines Säurehalogenids, wie eines Säurechlorids oder Säurebromids, bei -20 bis 100ºC in einem organischen Lösungsmittel in Gegenwart einer Base hergestellt werden. Als organisches Lösungsmittel können Chloroform, Dichlormethan, Dichlorethan, Pyridin usw. einzeln oder kombiniert verwendet werden. Als Base können Pyridin, 4-Dimethylaminopyridin, 4-Pyrrolidinopyridin, Triethylamin usw. einzeln oder kombiniert verwendet werden. Wenn Pyridin als Base verwendet wird, ist die Verwendung eines zusätzlichen organischen Lösungsmittels unnötig. Die Base wird vorzugsweise in einer aquimolaren oder höheren Menge, basierend auf dem reaktiven Carbonsäurederivat, verwendet.
- Die Verbindung (I-1) kann auch durch Umsetzung von UCN-1028D oder UCN-1028 iso-D mit mindestens 2 Äquivalenten einer Carbonsäure und eines Kondensationsmittels bei -20 bis 100ºC in einem organischen Lösungsmittel in Gegenwart einer Base hergestellt werden. In diesem Verfahren können die vorangehend angeflihrten organischen Lösungsmittel und Basen verwendet werden. Als Kondensationsmittel können N,N'-Dicyclohexylcarbodiimid, N,N'-Diisopropylcarbodiimid, 1-Cyclohexyl-3-(2-morpholinoethyl)carbodiimid, 1-Ethyl-3-(3-dimethylaminopropyl)carbodiimid usw. verwendet werden. Wenn Pyridin als Base verwendet wird, wird das Pyridin vorzugsweise in einer aquimolaren oder höheren Menge, basierend auf der Carbonsäuremenge, verwendet. Falls andere Basen als Pyridin verwendet werden, wird die Verwendung von mindestens 0,05 Äquivalenten einer Base, basierend auf der Carbonsäuremenge, bevorzugt.
- Falls die in den vorangehend beschriebenen zwei Verfahren zur Herstellung der Verbindung (I-1) verwendete Carbonsäure mit einer Aminogruppe, einem Monoalkylaminorest, einer Carboxylgruppe, einer Hydroxygruppe, einer Thiolgruppe oder ähnlichem substituiert ist, werden diese funktionellen Gruppen vor der Umsetzung mit geeigneten Schutzgruppen geschützt, und falls erforderlich werden die Schutzgruppen nach der Umsetzung entfernt. Als Schutzgruppen werden die Gruppen bevorzugt, die unter schwach alkalischen oder sauren Bedingungen entfernt werden können. Einzelheiten zu diesen Schutzgruppen und den Bedingungen zu deren Einführung und Entfernung sind in Green, Protective Groups in Organic Synthesis, John Wiley & Sons Incorporated, 1981, beschrieben.
- Wenn die fünktionelle Gruppe zum Beispiel eine Aminogruppe ist, sind eine p- Methoxybenzyloxycarbonylgruppe, eine t-Butyloxycarbonylgruppe und eine t-Amyloxycarbonylgruppe als Schutzgruppen geeignet. Die Entfernung der Schutzgruppe kann durch Behandlung von Verbindung (I-1) mit mindestens 2 Äquivalenten Trifluoressigsäure bei -20 bis 50ºC in einem organischen Lösungsmittel vom Halogentyp oder in Abwesenheit eines Lösungsmittel bewirkt werden. Beispiele für Lösungsmittel vom Halogentyp sind Chloroform, Dichlormethan und Dichlorethan.
- Wenn die funktionelle Gruppe eine Carboxylgruppe ist, sind ein Verfahren, in dem eine t-Butylgruppe, eine p-Methoxybenzylgruppe, eine 2,4,6-Trimethylbenzylgruppe, eine Pentamethylbenzylgruppe, eine Benzhydrylgruppe oder eine Methylthiomethylgruppe als Schutzgruppe verwendet und die Schutzgruppe auf ähnliche Art und Weise wie bei der Entfernung der Amino-Schutzgruppe entfernt wird; ein Verfahren, in dem eine t-Butylgruppe, eine p-Methoxybenzylgruppe, eine Tetrahydropyranylgrüppe oder eine Tetrahydrofuranylgruppe als Schutzgruppe verwendet und die Schutzgruppe durch Behandlung mit Essigsäure oder Ameisensäure in wäßrigem Tetrahydrofuran oder hydratisiertem Dioxan entfernt wird; und ähnliche Verfahren anwendbar. Wenn die fünktionelle Gruppe eine Carboxylgruppe ist, kann auch aus der Carboxylgruppe und einer anderen, zur Veresterung verwendbaren, Carboxylgruppe ein intramolekulares Säureanhydrid erzeugt werden.
- Wenn die funktionelle Gruppe eine Hydroxygruppe ist, sind eine Tetrahydropyranylgruppe, eine 4-Methoxytetrahydropyran-4-ylgruppe, eine Tetrahydrofuranylgruppe, eine Triphenylmethylgruppe, eine Methoxymethylgruppe, eine 1-Ethoxyethylgruppe, eine 1-Methyl-1-methoxyethylgruppe, eine t-Butylgruppe, eine Trimethylsilylgruppe, eine t-Butyldimethylsilylgruppe, eine t-Butyldiphenylsilylgruppe, eine Triphenylsilylgruppe usw. als Schutzgruppen geeignet. Die Schutzgruppe kann durch die Behandlung mit einer Säure in Abwesenheit eines Lösungsmittels oder in einem mit Wasser mischbaren organischen Lösungsmittel oder in einem Gemisch eines solchen Lösungsmittels und Wasser entfernt werden. Beispiele für das mit Wasser mischbare organische Losungsmittel schließen Methanol, Ethanol, Tetrahydrofüran und Dioxan ein. Beispiele für die Säure schließen Ameisensäure, Essigsaure, Trifluoressigsaure, p-Toluolsulfonsäure, Pyridinium-p-toluolsulfonat, Ionenaustauscherharz vom Sulfonsäuretyp, Salzsäure und Schwefelsäure ein. Es ist auch möglich, eine Formylgruppe oder Trifluoracetylgruppe als Schutzgruppe zu verwenden. In diesem Fall kann die Schutzgruppe durch die Behandlung mit Ammoniak oder einer anorganischen Base in einem mit Wasser mischbaren organischen Lösungsmittel oder in einem Lösungsmittelgemisch des organischen Lösungsmittels und Wasser entfernt werden. In einem solchen Fall können die vorangehend angeführten mit Wasser mischbaren organischen Lösungsmittel verwendet werden. Als anorganische Base können Kaliumhydrogencarbonat, Natriumhydrogencarbonat, Kaliumcarbonat und Natriumcarbonat angeführt werden. Falls die Hydroxygruppe an den aromatischen Ring des Aryl- oder Aralkylrestes gebunden ist, wird diese phenolische Hydroxygruppe auf eine ähnliche Art und Weise wie vorangehend beschrieben unter Verwendung einer Acetylgruppe als Schutzgruppe geschützt, gefolgt von einer Kondensation mit UCN-1028D oder UCN-1028 iso-D. Die Schutzgruppe kann durch die Behandlung mit Ammoniak oder einer anorganischen Base in einem mit Wasser mischbaren organischen Lösungsmittel oder in einem Gemisch eines solchen Lösungsmittels und Wasser entfernt werden. Als mit Wasser mischbares organisches Lösungsmittel und anorganische Base können die vorangehend angeführten verwendet werden.
- Wenn die fünktionelle Gruppe eine Thiolgruppe ist, können eine p-Methoxybenzylgruppe, eine Diphenylmethylgruppe, eine Triphenylmethylgruppe, eine Acetylgruppe, eine Benzoylgruppe usw. als Schutzgruppen verwendet werden, und die Entfernung der Schutzgruppe kann durch die Behandlung mit Trifluoressigsaure bewirkt werden. Wenn eine Acetyl- oder Benzoylgruppe als Schutzgruppe verwendet wird, kann die Schutzgruppe auch durch die Behandlung mit Ammoniak oder einer anorganischen Base in einem mit Wasser mischbaren organischen Lösungsmittel oder in einem Gemisch eines solchen Lösungsmittels und Wasser entfernt werden. Als mit Wasser mischbares organisches Lösungsmittel und anorganische Base können die vorangehend angeführten verwendet werden.
- Die Verbindung (I-2) [Verbindung (I), wobei Y - -O- bedeutet] kann durch Umsetzung von UCN-1028D oder UCN-1028 iso-D mit mindestens 2 Äquivalenten eines Halogenkohlensäureesters, wie eines Alkylhalogenformiats, eines Aralkylhalogenformiats oder eines Arylhalogenformiats, bei -20 bis 50ºC in einem organischen Lösungsmittel in Gegenwart einer Base hergestellt werden. Als organisches Lösungsmittel und als Base können die vorangehend angeführten einzeln oder kombiniert verwendet werden. Wenn Pyridin als Base verwendet wird, kann die Verwendung eines zusätzlichen organischen Lösungsmittels unnötig sein. Die Base wird vorzugsweise in einer aquimolaren oder höheren Menge, basierend auf dem halogenierten Ameisensäureester, verwendet.
- Die Zwischenprodukte und die erwünschten Produkte in den vorangehend beschriebenen Verfahren können durch herkömmlicherweise in der synthetischen organischen Chemie verwendete Reinigungsverfahren, wie zum Beispiel durch Filtration, Extraktion, Waschen, Trocknen, Einengen, Umkristallisation und verschiedene Arten der Chromatographie, isoliert und gereinigt werden. Die Zwischenprodukte können in folgenden Umsetzungen ohne besondere Reinigung verwendet werden.
- Die Verbindungen (I) kommen manchmal in Form eines Adduktes mit Wasser oder verschiedenen Lösungsmitteln vor. Solche Addukte werden von der vorliegenden Erfindung ebenfalls eingeschlossen.
- Repräsentative Beispiele der Verbindungen (I) werden in Tabelle 1 gezeigt. Tabelle 1
- Anmerkung: Der Perylenchinonring in der vorangehenden Strukturformel der Verbindungen (1) nimmt an der Bindung zwischen den 6a- und 6b-Positionen in Verbindung Nr. 1 S- Konfiguration und in Verbindung Nr.2 R-Konfiguration ein.
- Die hemmende Wirkung auf Proteinkinase C und die Antitumorwirkung der Verbindungen (I) werden nachstehend unter Bezug auf experimentelle Beispiele erläutert.
- Die hemmende Wirkung auf Proteinkinase C wurde mit dem Verfahren von Kikkawa et al. [Journal of Biological Chemistry, 257, 13341 (1982)] gemessen.
- Das heißt, 10 µl einer, ein UCN- 1 028D-Derivat enthaltenden, Testlösung wurden zu 250 µl einer Lösung, die 2,5 µMol Magnesiumacetat, 50 µg Histon Typ II 5 (hergestellt von Sigma Co., Ltd.), 20 µg Phosphatidylserin, 0,8 µg Diolein, 25 rimol CaCl&sub2;, 5 mg rohes Enzym (teilweise gereinigt aus Rattenhirn, nach dem Verfahren von Kikkawa et al.) und 5 µMol Tris-hydrochloridpuffer (pH 7,5) enthielt, zugegeben, gefolgt von einer dreiminütigen Inkubation bei 30ºC. Dann wurde die Phosphorylierung durch Zugabe von 1,25 rimol [γ-³²P]ATP (5 bis 10 x 10³ cpm/nmol) initiiert, gefolgt von einer dreiminütigen Inkubation bei 30ºC. Die Umsetzung wurde durch Zugabe von 25 %iger Trifluoressigsäure (TCA) beendet, und die Reaktionslösung durch eine Celluloseacetatmembran (Porengröße: 0,45 µm) (hergestellt von Toyo Filter Paper Co., Ltd.) filtriert. Nach viermaligem Waschen der Membran mit 5 %iger TCA wurde die auf der Membran verbliebene Radioaktivität gemessen. Als Kontrolle wurde das gleiche Verfahren ohne Zugabe der Testlösung wiederholt und die Radioaktivität ebenso gemessen. Die Konzentration der Testlösung, die im Vergleich zur Kontrolle 50 % Hemmung zeigte, wurde als IC&sub5;&sub0; ausgedrückt.
- Die Ergebnisse werden in Tabelle 2 gezeigt.
- Die Antitumorwirkung wurde unter Verwendung von mit dem karzinogenen Gen H- ras transformierten BALB 3T3 Zellen (nachstehend als BALB 3T3/H-ras bezeichnet) bestimmt.
- BALB 3T3/H-ras Zellen wurden in einem Medium, umfassend DME-Medium (hergestellt von Nissui Pharmaceutical Co., Ltd.) und 10 % fötales Kälberserum, (nachstehend als Medium A bezeichnet) zu einer Konzentration von 3,0 x 10&sup4; Zellen/ml suspendiert. Die so hergestellte Zellsuspension wurde in einer Menge von 0,1 ml pro Loch in Löcher einer 96-Loch-Mikrotiterplatte gegeben. Nach einer 20-stündigen Kultivierung bei 37ºC in einem CO&sub2;-Inkubator wurden in jedes Loch jeweils 0,1 ml einer entsprechend mit Medium A verdünnten Testlösung gegeben. Die Zellen wurden bei 37ºC weitere 72 Stunden im CO&sub2;-Inkubator kultiviert, und der Kulturüberstand wurde entfernt. Nach einmaligem Waschen mit physiologischer Salzlösung wurde der Rückstand zur Fixierung der Zellen 10 Minuten mit 0,1 ml Methanol behandelt. Danach wurden die Zellen mit 0,1 ml Giemsa-Färbelösung [Giemsa-Stammfärbelösung (Merck Art 9204; hergestellt von Merck Inc.): physiologische Salzlösung 1:10] 5 Minuten angefarbt. Nach dem Entfernen der Färbelösung wurde der Rückstand einmal mit 0,2 ml Wasser gewaschen. Der Farbstoff wurde mit 0,2 ml 0,1 N Salzsäure extrahiert und die Extinktion bei 620 nm mit einem Mikroplate-Meßgerät gemessen. Durch Vergleich der Extinktion unbehandelter Zellen mit der Extinktion mit bekannten Konzentrationen der Testverbindung behandelter Zellen wurde die Konzentration der Testverbindung berechnet, bei der das Zellwachstum um 50 % gehemmt wurde (IC&sub5;&sub0;).

- UCN-1028D-Derivate können einzeln oder zusammen mit pharmazeutisch verträglichen Trägern als Antitumormittel verwendet werden. Ein UCN-1028D-Derivat wird zur Herstellung eines geeigneten Arzneimittels zur Injektion zum Beispiel in einer physiologischen Salzlösung oder in einer wäßrigen Lösung von Glucose, Lactose, Mannitol usw. gelöst. Alternativ wird ein UCN-1028D-Derivat oder ein Salz davon zur Herstellung eines Pulvers zur Injektion gefriergetrocknet oder mit Natriumchlorid vermischt. Das Arzneimittel kann auf dem Fachgebiet der medizinischen Zubereitungen gut bekannte Zusätze, zum Beispiel, falls erforderlich, pharmazeutisch verträgliche Salze, enthalten. Obwohl die Dosierungsmenge der Verbindung in Abhängigkeit des Alters, des Zustandes usw. des Patienten, variiert, ist es angemessen, die Verbindung Säugern, einschließlich Menschen, in einer Menge von 0,01 bis 20 mg/kg/Tag zu verabreichen. Die Verabreichung kann einmal täglich (einzelne Verabreichung der fortlaufende Verabreichung) oder periodisch 1- bis 3mal wöchentlich oder einmal alle 2 bis 3 Wochen intravenös erfolgen. Eine orale Verabreichung in emer annlichen Dosierung und einer ähnlichen Art und Weise ist ebenfalls möglich. Arzneimittel zur oralen Verabreichung schließen Tabletten, Kapseln, Pulver, Granulat, Ampullen usw. ein. Diese Arzneimittel enthalten auf dem Fachgebiet der medizinischen Zubereitungen gut bekannte pharmazeutische Hilfsstoffe. Ferner sind eine intraarterielle Verabreichung, eine intraperitoneale Verabreichung, eine intrathorakale Verabreichung usw. in einer ähnlichen Dosierung und einer ähnlichen Art und Weise ebenfalls möglich.
- Von den Antitumor-Arzneimittel der vorliegenden Erfindung wird eine Wirkung gegen Leukämie, Magenkrebs, Dickdarmkrebs, Lungenkrebs, Brustkrebs, Gebärmutterkrebs usw. in Säugern, einschließlich Menschen, erwartet.
- Bestimmte spezielle Ausführungsformen der vorliegenden Erfindung werden durch die folgenden Beispiele veranschaulicht.
- UCN-1028D (54,3 mg) wurde in 2,0 ml Dichlormethan und 0,2 ml Pyridin gelöst. Unter Rühren unter Eiskühlung wurden 20 µl Phenylchloroformiat dreimal in 30-Minuten Intervallen zur Lösung zugegeben, und nach weiteren 15 Minuten wurden 10 µl Phenylchloroformiat zugegeben. Die Umsetzung wurde eine weitere Stunde fortgeführt. Dann wurden 30 ml Essigsäureethylester zum Gemisch zugegeben. Die organische Schicht wurde nacheinander mit jeweils 15 ml Wasser (dreimal), 1 M Salzsäure, Wasser, gesättigter wäßriger Natriumhydrogencarbonatlösung und Wasser (zweimal) gewaschen, über wasserfreiem Natriumsulfat getrocknet und unter vermindertem Druck eingeengt. Der Rückstand wurde in einer geringen Menge Dichlormethan gelöst und die Lösung auf eine Silicagel- Dünnschichtchromatographieplatte mit einer Dicke von 0,5 mm (20 cm x 20 cm) aufgetragen und mit Chloroform-Methanol (100:2) entwickelt. Der Teil des Silicagels, auf dem eine Bande des Hauptproduktes entstand, wurde abgekratzt und mit Essigsäureethylester extrahiert. Der Extrakt wurde eingeengt, wobei 68,0 mg (87 %) der Verbindung 1 erhalten wurden.

- In 2,0 ml Dichlormethan und 0,2 ml Pyridin wurden 19,6 mg des im Referenzbeispiel 1 erhaltenen UCN-1028D iso-D gelöst. Unter Rühren unter Eiskühlung wurden 20 µl Benzoylchlorid und nach einer Stunde weitere 20 µl Benzoylchlorid zur Lösung zugegeben. Die Umsetzung wurde weitere 1,5 Stunden fortgeführt. Das Reaktionsgemisch wurde auf ähnliche Art und Weise wie in Beispiel 1 behandelt und der Silicagel-Dünnschichtchromatographie unterworfen, wobei 24,1 mg (89 %) der Verbindung 2 erhalten wurden.

- In einer Argonatmosphäre wurden 1,004 g UCN-1028D in 200 ml Xylol 2 Stunden unter Rückfluß erhitzt. Das Reaktionsgemisch wurde unter vermindertem Druck eingeengt. Der Rückstand wurde auf Silicagel-Dünnschichtchromatographieplatten (3 Platten) mit einer Dicke von 2 mm (20 cm x 20 cm) aufgetragen und mit Chloroform-Methanol (10:1) entwickelt. Der Teil des Silicagels, auf dem eine Bande des Hauptproduktes unter der UCN-1028D-Bande entstand, wurde abgekratzt und mit Essigsäureethylester extrahiert. Der Extrakt wurde eingeengt, wobei 501 mg (49,9 %) der Verbindung UCN-1028 iso-D erhalten wurden.

1. UCN-1028D-Derivat der allgemeinen Formel:
wobei Y- - oder - -O-bedeutet; mit der Maßgabe, daß, wenn
Y- - bedeutet, der Perylenchinonring an der Bindung zwischen den 6a- und 6b-Positionen
R-Konfiguration annimmt, und daß, wenn Y- -O- bedeutet, der
Perylenchinonring an der Bindung zwischen den 6a- und 6b-Positionen S-
Konfiguration annimmt.
2. Arzneimittel, umfassend einen pharmazeutischen Träger und als Wirkstoff eine
wirksame Menge des in Anspruch 1 definierten UCN-1028D-Derivates.
3. Verbindung nach Anspruch 1 zur Verwendung als Medikament.
4. Verwendung einer Verbindung nach Anspruch 1 oder eines Arzneimittels nach
Anspruch 2 zur Herstellung eines Medikamentes mit Antitumorwirkung.
