Description
- Die vorliegende Erfindung betrifft neue Chinolinoncarbonsäuren, die ausgezeichnete antibakterielle Aktivität gegen grampositive und gramnegative Bakterien aufweisen.
- Die in dem US-Patent Nr. 4,382,892, dem FR-Patent Nr. 2,563,521 und dem US-Patent Nr. 4,528,287 beschriebenen Verbindungen sind als antibakterielle Pyridoncarbonsäuren bekannt. Bei vielen dieser bekannten Produkte treten Probleme auf, so wird zum Beispiel, wenn sie Menschen verabreicht werden, ein ungünstiger Einfluß ausgelöst, wie Krämpfe. Daher ist es das Ziel dieser Erfindung, Chinolinoncarbonsäuren bereitzustellen, die eine starke antibakterielle Aktivität aufweisen und daneben weniger ungünstige Reaktionen auf das ZNS, wie zum Beispiel Krämpfe, auslösen.
- Chinolinoncarbonsäuren mit besonderen Substituenten werden als Verbindungen, die eine gute antibakterielle Aktivität aufweisen, in Chem. Abstracts 108, 109 403 h, Chem. Abstracts 110, 111 571 w, DE-A-3 711 193, EP-A-0 265 230, EP-A-0 172 651 und in EP-A-0 242 789 offenbart.
- Diese Erfindung betrifft Chinolinoncarbonsäuren. Erfindungsgemäße Verbindungen haben an der Position 7 einen Substituenten mit zwei asymmetrischen Kohlenstoffatomen. Der Umfang der Erfindung erstreckt sich auf optisch aktive Verbindungen oder Racemate der allgemeinen Formel (I), wobei cis- und trans- Formen eingeschlossen sind.
- Die vorliegende Erfindung betrifft Chinolinoncarbonsäuren der allgemeinen Formel (I) in der R¹ ein C&sub1;-C&sub4; Alkylrest ist, R² und R³ jeweils identisch oder verschieden ein Wasserstoffatom oder einen C&sub1;-C&sub4;-Alkylrest bedeuten, R&sup4; ein Cyclopropyl-, Phenyl-, Halogenphenyl- oder Thienylrest ist, der gegebenenfalls mit einem C&sub1;-C&sub4;-Alkylrest oder einem Halogenatom substituiert ist und R&sup5; ein Halogenatom ist oder deren pharmazeutisch verträglichen Salze.
- In dieser Erfindung bedeutet ein C&sub1;-C&sub4;-Alkylrest einen gerad- oder verzweigtkettigen C&sub1;-C&sub4;-Alkylrest, einschließlich der Methyl-, Ethyl-, n-Propyl-, Isopropyl-, n-Butyl-, sek.- Butyl-, iso-Butyl-, tert.-Butylgruppe und dergleichen.
- Halogen bedeutet Chlor, Brom, Fluor oder Jod.
- Die erfindungsgemäße Verbindung (I) kann nach den folgenden Verfahren A oder B hergestellt werden. Verfahren A
- wobei R¹, R², R³, R&sup4; und R&sup5; die gleichen Bedeutungen haben wie vorstehend definiert.
- Die Ausgangsmaterialien (II) können durch das in US- Patent Nr. 4,382,892 beschriebene Verfahren hergestellt werden.
- Die erfindungsgemäße Verbindung (I) kann hergestellte werden, indem das Ausgangsmaterial (II) mit den Amin (III) umgesetzt wird. Diese Umsetzung kann in einem Lösungsmittel, wie zum Beispiel Wasser, einem Alkohol, Acetonitril, Dimethylsulfoxid (DMSO) oder Dimethylformamid (DMF) durchgeführt werden. Die Umsetzung wird für eine oder mehrere Stunden bei einer Temperatur zwischen 15 und 200 ºC, vorzugsweise zwischen 80 und 120 ºC oder etwa bei dem Siedepunkt des Lösungsmittel durchgeführt. - wobei R ein Wasserstoffatom oder einen C&sub1;-C&sub4;-Alkylrest bedeutet, R3' eine Aminoschutzgruppe darstellt, und R³, R&sup4; und R&sup5; die gleichen Bedeutungen wie vorstehend definiert haben.
- Aminoschutzgruppe bedeutet einen Acylrest, wie zum Beispiel Formyl-, Acetyl- oder Propionylrest.
- (1) Die Verbindung (V) kann hergestellt werden, indem das Ausgangsmaterial (IV) mit dem Amin (III) umgesetzt wird. Diese Umsetzung kann in einem Lösungsmittel, wie zum Beispiel Acetonitril, Dimethylsulfoxid (DMSO), Dimethylfarmamid (DMF), einem Alkohol oder Tetrahydrofuran (THF) durchgeführt werden. Die Umsetzung wird bei einer Temperatur von 0 ºC bis etwa zum Siedepunkt des Lösungsmittel durchgeführt. Um die Umsetzung zu beschleunigen, kann eine anorganische Base, wie zum Beispiel Natriumcarbonat, Kaliumcarbonat, Natriumhydroxid oder Kaliumhydroxid, oder einer organische Base, wie zum Beispiel DBU, Triethylamin und dergleichen, nach herkömmlicher Art und Weise verwendet werden.
- (2) Die Verbindung (VI) kann hergestellt werden, indem die Verbindung (V) mit Ethylorthoformiat umgesetzt wird und die sich ergebende Formylverbindung mit dem Amin R&sup4;NH&sub2; (VIII) kondensiert wird. Die erste Umsetzung wird in Essigsäureanhydrid bei einer Temperatur von 80 ºC bis etwa zum Siedepunkt von Essigsäureanhydrid durchgeführt. Die Kondensation wird in einem Lösungsmittel, wie zum Beispiel Ethanol, Aceton, Dimethylsulfoxid oder Dioxan bei einer Temperatur von 0 bis 50 ºC, vorzugsweise bei etwa Raumtemperatur (15-30 ºC) durchgeführt.
- (3) Die Verbindung (VII) kann durch die Behandlung der Verbindung (VI) mit einer Base in einem Lösungsmittel, wie zum Beispiel DMF, THF, DMSO oder Acetonitril bei einer Temperatur von 0 ºC bis zu etwa dem Siedepunkt des Lösungsmittel hergestellt werden. Als Base können ein Alkalimetallhydrid, wie zum Beispiel Natriumhydrid oder Kaliumhydrid, oder ein organisches Amin, wie zum Beispiel Triethylamin oder Pyridin, nach herkömmlicher Art und Weise verwendet werden.
- (4) Die Verbindung (VII) wird mit Benzylamin umgesetzt, wobei die Benzylaminogruppe an der Position 5 eingeführt wird, anschließend wird die Benzylgruppe durch katalytische Hydrierung entfernt. Die Einführung der Benzylaminogruppe erfolgt in einen Lösungsmittel, wie zum Beispiel Acetonitril, DMF oder DMSO, in einem verschlossenen Rohr bei einer Temperatur von 80 und 150 ºC. Die Hydrierung wird einem Lösungsmittel, wie zum Beispiel Methanol, Ethanol, Essigsäure oder Acetonitril über einem Katalysator, wie zum Beispiel Pd-C, bei einer Temperatur von 0 bis 50 ºC, vorzugsweise bei etwa Raumtemperatur und unter Atmosphärendruck nach herkömmlicher Art und Weise durchgeführt.
- Wenn nötig kann jede Aminoschutzgruppe, wie zum Beispiel Carbobenzoxy-, Trityl- oder Ethoxycarbonylgruppe, an dem Pyrrolidinring oder jede Carboxyschutzgruppe nach herkömmlicher Art und Weise entfernt werden.
- Die Verbindungen der Formel (I) können, wenn es erwünscht ist, nach herkömmlicher Art und Weise in deren Säureadditionssalze überführt werden. Zu den salzbildenden Säuren gehören veranschaulichend anorganische Säuren, wie zum Beispiel Salzsäure, Schwefelsäure oder Phosphorsäure, und organische Säuren, wie zum Beispiel Methansulfonsäure, Milchsäure, Oxalsäure oder Essigsäure.
- Die erfindungsgemäßen Verbindungen können auch in Alkalimetallsalze, wie zum Beispiel Natrium- oder Kaliumsalze überführt werden.

- Die erfindungsgemäßen Verbindungen (I) können Menschen oder Säugetieren oral oder parenteral verabreicht werden. Sie können durch herkömmliche pharmazeutische Praktiken zu Tabletten, Kapseln, Pillen, Granulaten, Injektionen, Suppositorien und zu Sirupen formuliert werden. Zu den pharmazeutisch verträglichen Trägern, Verdünnungsmitteln und Füllstoffen gehören Laktose, Rohrzucker, Weizenstärke, Kartoffelstärke, Magnesiumstearat, Gelatine, Methylzellulose, Agar, Wasser usw. Stabilisatoren, Emulgatoren, Naßfüllstofe, Puffer oder andere Hilfsmittel können, wenn nötig, hinzu gegeben werden. Eine geeignete tägliche Dosierung für einen Erwachsenen beträgt oral 1-500 mg, vorzugsweise 50-100 mg und parenteral 0,1-300 mg in Einzel- oder aufgeteilten Dosen.
- Die folgenden Beispiele, Referenzbeispiele und Formulierungen sind zur Erläuterung der Erfindung aufgeführt.
- Die in den Beispielen und Referenzbeispielen verwendeten Abkürzungen haben folgende Bedeutungen:

- Ein Gemisch aus 298 mg (1,00 mmol) der Verbindung (II- 1), 305 mg (+ )-cis-3-Methylamino-4-methoxy-1-pyrrolidin-Hydrochlorid und 600 ul DBU wird in 3 ml Acetonitril 1 Stunde unter Rückfluß erhitzt. Die sich ergebenden Kristalle werden abfiltriert und mit Acetonitril gewaschen, wobei sich 345 mg der Kristalle ergeben (Schmp. 250-251 ºC, Zers.). Das Rohprodukt wird mit 1 ml 2N-HCl behandelt und aus Methanol/Ethanol umkristallisiert, wobei sich 358 mg des Hydrochlorids (I-1) als gelbe Kristalle ergeben (Schmp. 260-263 ºC, Zers.).

- (1) Ein Gemisch aus 1,8 g (11,4 mmol) cis-3-Acetamino-4- methoxypyrrolidin (III-5), 958 mg NaHCO&sub3; und 3,22 g (11,4 mmol) Ethyl-2,3,4,5,6-pentafluorbenzoylacetat (IV-1) wird in 20 ml Acetonitril gerührt und 1 Stunde unter Rückfluß erhitzt. - (2) Ein Gemisch aus 1,5 g (3,57 mmol) der Verbindung (V-1), 832 ul Ethylorthoformiat und 674 ul Essigsäureanhydrid wird 2 Stunden unter Rückfluß erhitzt und das Reaktionsgemisch wird unter vermindertem Druck konzentriert. Der Rückstand wird mit einer Lösung aus 1 ml Cyclopropylamin in 5 ml Ethanol gemischt und 20 Minuten bei Raumtemperatur gerührt. Das Reaktionsgemisch wird unter vermindertem Druck konzentriert. Das Rohprodukt wird über eine Lobar B -Säule chromatographiert (Toluol:Ethylacetat = 2:1 v/v) und aus Ethanol umkristallisiert, wobei sich 1,10 g (Ausbeute 95%) der Verbindung (VI-1) ergeben, Schmp. 175-176 ºC.

- (3) Zu einer gerührten Lösung aus 630 mg der Verbindung (VI-1) in 3 ml DMF werden bei Raumtemperatur 60 mg 60%-iges NaH gegeben und das Gemisch wird 10 Minuten gerührt. Das Reaktionsgemisch wird mit 7 ml 0,5N-HCl gemischt und die sich ergebenden Kristalle werden abfiltriert, mit Wasser gewaschen und in Methylenchlorid gelöst. Die Lösung wird über MgSO&sub4; getrocknet und konzentriert, der Rückstand wird mit Ethylacetat behandelt. Die sich ergebenden Kristalle werden abfiltriert, wobei sich 594 mg (Ausbeute 97,8%) der Verbindung (VII-1) ergeben. Schmp. 234-235 ºC

Das Reaktionsgemisch wird unter vermindertem Druck bis zur Trockne konzentriert und der Rückstand in Methylenchlorid gelöst. Die Lösung wird mit 0,5N-HCl gewaschen, über MgSO&sub4; getrocknet und konzentriert. Der Rückstand wird mit Ethylacetat behandelt und die sich ergebenden Kristalle werden abfiltriert, wobei sich 589 mg (Ausbeute 99,3%) der Verbindung (VII-2) ergeben. Schmp. 191-192 ºC

- (5) Zu einem Gemisch aus 7 ml Essigsäure und 7 ml Ethanol werden 459 mg (0,83 mmol) der Verbindung (VII-2) und 80 mg 5%-iges Pd/C gegeben. Das sich ergebende Gemisch wird bei Raumtemperatur und Atmosphärendruck hydriert und filtriert. Das Filtrat wird unter vermindertem Druck konzentriert und der Rückstand in Methylenchlorid gelöst. Die organische Schicht wird mit Wasser gewaschen, über MgSO&sub4; getrocknet und konzentriert. Der Rückstand wird mit Ethylacetat behandelt und die sich ergebenden Kristalle werden abfiltriert, wobei sich 318 mg (Ausbeute 82,7%) der Verbindung (VII-3) ergeben. Schmp. 199-201 ºC

- (6) 1,5 ml einer 2N-NaOH-Lösung, die 100 mg (0,22 mmol) der Verbindung (VII-3) enthält, werden in einem verschlossenen Rohr bei einer Temperatur von 110 ºC 5 Stunden gerührt. Das Reaktionsgemisch wird mit 1,5 ml 2N-HCl und 1 Tropfen 28%-igem wäßrigen Ammoniak gemischt und unter vermindertem Druck konzentriert. Der Rückstand mit mit Acetonitril extrahiert und die organische Schicht konzentriert. Die sich ergebenden Kristalle werden abfiltriert, wobei sich 54 mg (Ausbeute 63,6%) der Zielverbindung (I-5) ergeben. Schmp. 222-223 ºC

Das Reaktionsgemisch wird unter vermindertem Druck konzentriert, der Rückstand wird mit Wasser gemischt und die sich ergebenden Kristalle werden abfiltriert. Da die Ausgangsverbindung (II-2) in diesem Rohprodukt noch zu etwa einem Drittel enthalten ist, wird eine weitere Umsetzung durchgeführt, indem 102 mg der Verbindung (III-6) und 76 mg DBU zu dem Rohprodukt gegeben werden, das Gemisch in 20 ml DMSO gelöst wird und es im Ölbad 2 Stunden auf 100 ºC erhitzt wird. Das Reaktionsgemisch wird wie vorstehend beschrieben aufgearbeitet. Die sich ergebenden Kristalle werden mit Ether gewaschen und getrocknet, wobei sich 90 mg Rohprodukt ergeben. Dieses Produkt wird in 2 ml einer 36%-igen c-HCl bei Raumtemperatur behandelt. Das Reaktionsgemisch wird unter vermindertem Druck konzentriert und der sich ergebende Rückstand wird zur Kristallisation mit Aceton behandelt. Die sich ergebenden Kristalle werden filtriert, wobei sich 90 mg des Hydrochlorids (I- 6) als gelbe Kristalle ergeben. Schmp. oberhalb 290 ºC, Zers.

- (1) Ein Gemisch aus 500 mg 1-Thienyl-5-benzylamino- 6,7,8-trifluor-1,4-dihydro-4-oxo-3-chinolincarbonsäureethylester (II-3), 443 mg der Verbindung (III-6), 331 mg DBU und 10 ml DMSO wird 3 Stunden in einem Ölbad erhitzt. Das Reaktionsgemisch wird unter vermindertem Druck konzentriert und der Rückstand wird über eine Kieselgelsäule mit Chloroform und Methanol (50 : 1 v/v) als Eluent chromatographiert. Das Eluat wird konzentriert, wobei sich 120 mg (Ausbeute: 19%) der Zielverbindung 1 ergeben.

- (2) Zu einer Lösung aus 80 mg der Verbindung 1 in 20 ml Methanol werden 40 mg 10%-iges Pd-C und 2 ml 2N-HCl gegeben und das Gemisch wird bei Raumtemperatur und einem Druck von 5 at 7 Stunden mit Wasserstoff hydriert. Die Lösung wird mit Sellaite filtriert, wobei Pd-C entfernt wird, und das Filtrat wird unter vermindertem Druck konzentriert. Der Rückstand wird in Chloroform gelöst und die Lösung wird mit gesättigter NaHCO&sub3;- Lösung gewaschen. Die Chloroformschicht wird über eine Kieselgelsäule mit Chloroform und Methanol (10 : 1 v/v) als Eluent chromatographiert. Das Eluat wird konzentriert, wobei sich 26 mg (Ausbeute: 39%) der Verbindung 2 als schaumiger Rückstand ergeben.

- (3) Ein Gemisch aus 26 mg der Verbindung 2, 1 ml 2N-HCl, 2 ml Ethanol und 2 ml Wasser wird in einem Ölbad 1 Stunde auf 100 ºC erhitzt. Nach dem Abkühlen wird das Gemisch unter vermindertem Druck konzentriert und der Rückstand mit Methanol behandelt. Nach Entfernung der unlöslichen Bestandteile wird das Filtrat konzentriert. Der sich ergebende Rückstand wird aus Aceton kristallisiert und filtriert, wobei sich 18 mg (Ausbeute: 75%) des Hydrochlorids (I-7) ergeben. Schmp. 280-285 ºC, Zers.

- Zu einer Suspension aus 2,30 g (7,71 mmol) der Verbindung (II-1) in 70 ml trockenem Acetonitril werden 2,842 g der Verbindung (III-7) und 4,61 ml DBU gegeben und das Reaktionsgemisch wird in einem Ölbad bei 100 ºC 1,5 Stunden gerührt. Nach dem Abkühlen des Gemisches werden die sich ergebenden Kristalle abfiltriert, wobei sich 2,971 g (Ausbeute: 91,2%) gelbe Kristalle ergeben. Die Kristalle werden in einem Gemisch aus 7 ml 2N-HCl und 2,3 ml c-HCl aufgelöst und die Lösung wird unter vermindertem Druck konzentriert. Der sich ergebende Rückstand wird mit 50 ml Ethanol gemischt und die Lösung über Nacht bei 5 ºC stehen gelassen. Die sich ergebenden Kristalle werden abfiltriert und aus Wasser-Ethanol-Aceton umkristallisiert, wobei sich 3,102 mg des Hydrochlorids (I-8) als gelbe Kristalle ergeben. Schmp. 278-286 ºC, Zers.

- Zu einer Lösung aus 188 mg (0,6 mmol) 1-Cyclopropyl-5- amino-8-chlor-6,7-difluor-1,4-dihydro-4-oxo-3-chinolincarbonsäure (II-4) in 4 ml Acetonitril werden 231 mg (1,8 mmol) der Verbindung (III-8) und 122 mg (1,2 mmol) Triethylamin gegeben. Das Reaktionsgemisch wird 3 Stunden unter Rückfluß erhitzt. Nach dem Abkühlen werden die sich ergebenden Kristalle abfiltriert. Die Rohkristalle werden aus Methylenchlorid-Methanol-Ethylacetat umkristallisiert, wobei sich 153 mg (Ausbeute: 60,2%) 1-Cyclopropyl-5-amino-8-chlor-6-fluor-7-(cis-3-methylamino-4-methoxy-1- pyrrolidinyl)-1,4-dihydro-4-oxo-3-chinolin-carbonsäure (I-9) ergeben. Schmp. 206-207 ºC.

- Die Verbindung (III-6) wird mit der Verbindung (II-4) in der gleichen Art und Weise umgesetzt wie in Beispiel 9, wobei sich 1-Cyclopropyl-5-amino-8-chlor-6-fluor-7-(cis-3-methylamino- 4-methoxy-1-pyrrolidinyl)-1,4-dihydro-4-oxo-3-chinolincarbonsäure (I-10) (Ausbeute: 48,8%) ergeben. Schmp. 190-192 ºC, Zers.

- Zu einem Gemisch aus 320 mg (1,7 mmol) der Verbindung (II-1) und 376 mg der Verbindung (III-6) werden 10 ml Acetonitril werden und 0,64 ml DBU gegeben und das Reaktionsgemisch wird in einem Wasserbad 1,5 Stunden unter Rückfluß erhitzt. Nach Abkühlung der Lösung werden die sich ergebenden Kristalle abfiltriert, wobei sich 412 mg der Verbindung (I-11) ergeben, die aus Methanol umkristallisiert werden, wobei sich 292 mg der Verbindung (I-11) als gelbe Kristalle ergeben. Schmp. 262-264 ºC, Zers.
- Analyse berechnet (%) für C&sub1;&sub9;H&sub2;&sub2;N&sub4;O&sub4;F&sub2;: C 55,88; H 5,43; N 13-72;
- Gefunden (%): C 55,82; H 5,45; N 13,56;
- (1) Ein racemisches Gemisch aus 10 g der 3,4-cis- Pyrrolidinverbindung 3 wird mit 10 g L(+)-Weinsäure in 50 ml Methanol behandelt und bei Raumtemperatur 3 Stunden stehengelassen. Das sich ergebende L(+)-Tatrat wird abfiltriert und aus Methanol umkristallisiert, wobei sich 6,12 g des L(+)-Tatrats 4 (Schmp. 197-198 ºC, Zers.) ergeben. Das gesamte Filtrat wird zusammengegeben und unter vermindertem Druck verdampft. Nach der Auflösung des Rückstands in gesättigter Kochsalzlösung, wird die Lösung mit K&sub2;CO&sub3; basisch gemacht und mit Ethylacetat extrahiert. Dann wird der Extrakt mit gesättigter Kochsalzlösung gewaschen, über MgSO&sub4; getrocknet und unter vermindertem Druck konzentriert, wobei sich 6,16 g einer öligen Substanz ergeben. Die Substanz wird dann in 40 ml Methanol gelöst. Die Lösung wird mit 6,0 g D(-)-Weinsäure gemischt und bei Raumtemperatur 3 Stunden stehengelassen. Das sich ergebende D(-)-Tatrat wird abfiltriert und aus Methanol umkristallisiert, wobei sich 5,62 g des D(-)- Tatrats 5 (Schmp. 197-198 ºC) ergeben. Die Mutterlauge wird in der gleichen wie vorstehend beschriebenen Art und Weise behandelt, wobei sich L(+)-Tatrat ergibt und nach dem Umkristallisieren 0,57 g L(+)-Tatrat 4 erhalten werden. Die sich ergebende Mutterlauge wird in der gleichen wie vorstehend beschriebenen Art und Weise behandelt und mit D(-)-Weinsäure gemischt, wobei sich 0,37 g D(-)-Tatrat 5 ergeben.

- (2) 6,10 g des des L(+)-Tatrats 4 werden in der gleichen Art und Weise wie in (1) beschrieben behandelt, wobei sich 3,80 g der Verbindung 6 als ölige Substanz ergeben.

- (3) 5,60 g des D(-)-Tatrats 5 werden in der gleichen Art und Weise wie in (1) beschrieben behandelt, wobei sich 3,30 g der Verbindung 7 als ölige Substanz ergeben.

- (4) Eine Lösung von 2,0 g der Verbindung 6 in 3,1N-HCl in 20 ml Methanol wird bei Raumtemperatur eine 1 Stunde gerührt. und unter vermindertem Druck konzentriert. Der sich ergebende Rückstand wird aus Methanol umkristallisiert, wobei sich 1,58 g der Verbindung 8 als hygroskopische Kristalle ergeben. Schmp. 207-208 ºC

- (5) 2,0 g der Verbindung 7 werden in der gleichen Art und Weise umgesetzt wie in (4), wobei sich 1,63 g hygroskopischer Kristalle 9 ergeben. Schmp. 208-209 ºC

- (1) Zu einer Lösung von 1,5 g (6,93 mmol) der Verbindung 6 in 10 ml Methanol werden 2 ml Di-tert.butyl-dicarbonat gegeben. Das Reaktionsgemisch wird 30 Minuten unter Eiskühlung und weitere 30 Minuten bei Raumtemperatur gerührt. Das Lösungsmittel wird unter vermindertem Druck abdestilliert und der sich ergebenden Rückstand wird über eine Lobar B -Säule chromatographiert (Toluol:Ethylacetat = 4:1 v/v), wobei sich 2,30 g der Verbindung 10 ergeben.
- (2) 1,50 g (6,93 mmol) der Verbindung 7 werden in der gleichen Art und Weise umgesetzt wie in (1), wobei sich 2,30 g der Verbindung 11 ergeben.
- (3) Eine Lösung von 2,2 g (6,90 mmol) der Verbindung 10 in 10 ml DMF wird unter Eiskühlung mit 414 mg 60%-igem NaH gemischt und bei Raumtemperatur 15 Minuten gerührt. Das Reaktionsgemisch wird in Ethylacetat gegossen und die Lösung wird dreimal mit Wasser gewaschen. Der Extrakt wird über MgSO&sub4; getrocknet und unter vermindertem Druck konzentriert. Der Rückstand wird über eine Lobar B -Säule chromatographiert (Toluol:Ethylacetat = 7:1 v/v), wobei sich 2,25 g der Verbindung 12 ergeben.
- (4) 2,30 g (6,90 mmol) der Verbindung 11 werden in der gleichen Art und Weise umgesetzt wie in (3), wobei sich 2,20 g der Verbindung 13 ergeben.
- (5) Zu 2,10 g (6,90 mmol) der Verbindung 12 werden nach und nach 60 ml 3,1N-HCl-MeOH gegeben, das Reaktionsgemisch wird 1,5 Stunden bei Raumtemperatur gerührt und dann unter vermindertem Druck konzentriert. Der Rückstand wird aus Ethanol umkristallisiert, wobei sich 1,25 g der Verbindung 14 als hygroskopische Kristalle ergeben. Schmp. 180-182 ºC
- [α]24D +52,1±1,0º (c=0,973, Methanol)
- berechnete Analyse (%) für C&sub6;H&sub1;&sub6;N&sub2;OCl&sub2;: C 34,80; H 7,90; N 13,52;

- (6) 2,10 g (6,40 mmol) der Verbindung 13 werden in der gleichen Art und Weise umgesetzt wie in (5), wobei sich 1,21 g der Verbindung 15 als hygroskopische Kristalle ergeben. Schmp. 178-180 ºC
- [α]24D -53,1±0,9º (c=1,026, Methanol)
- Die antibakterielle Aktivität wurde bestimmt, indem nach dem von der "Japan Society of Chemotherapy" bestimmten Verfahren
- die Mindesthemmkonzentration (MIC) gemessen wurde. Die Ergebnisse sind in Tabelle 1 aufgeführt.
- A, B, C und D in der Tabelle haben folgende Bedeutungen:
- A : Staphylococcus aureus SMITH
- B : Staphylococcus aureus SR 14 (R)
- C : Escherichia coli EC-14
- D : Escherichia coli SR377 (R)
- Die Testmikroorganismen wurden in einer Dichte von 10&sup6; Zellen/ml verwendet. Tabelle 1 Verbindung Nr. *: OFLX : Ofloxacin (Referenzarzneimittel)
- Diese Ergebnisse zeigen, daß die erfindungsgemäßen Verbindungen eine starke antibakterielle Aktivität gegen grampositive und gramnegative Bakterien aufweisen.
Claims (6)
1. Chinolinoncarbonsäuren der allgemeinen Formel I
in der R¹ ein C&sub1;-C&sub4;-Alkylrest ist, R² und R³ jeweils
identisch oder verschieden ein Wasserstoffatom oder
einen C&sub1;-C&sub4;-Alkylrest bedeuten, R&sup4; ein Cyclopropyl-,
Phenyl-, Halo-Phenyl- oder Thienylrest ist, der
gegebenenfalls mit einem C&sub1;-C&sub4;-Alkylrest oder einem
Halogenatom substituiert ist, und R&sup5; ein Halogenatom ist
oder deren pharmazeutisch verträgliche Salze.
2. Verbindung gemäß Anspruch 1, nämlich
(+)-1-Cyclopropyl-5-amino-6,8-difluor-1,4-dihydro-7-
(cis-3-methylamino-4-methoxy-1-pyrrolidinyl)-4-oxo-3-
chinolincarbonsäure,
(-)-1-Cyclopropyl-5-amino-6,8-difluor-1,4-dihydro-7-
(cis-3-methylamino-4-methoxy-1-pyrrolidinyl)-4-oxo-3-
chinolincarbonsäure,
(+)-1-Cyclopropyl-5-amino-6,8-difluor-1,4-dihydro-7-
(cis-3-amino-4-methoxy-1-pyrrolidinyl)-4-oxo-3-chinolincarbonsäure,
(-)-1-Cyclopropyl-5-amino-6,8-difluor-1,4-dihydro-7-
(cis-3-amino-4-methoxy-1-pyrrolidinyl)-4-oxo-3-chinolincarbonsäure,
1-Cyclopropyl-5-amino-6,8-di luor-1,4-dihydro-7-(cis-3-
amino-4-methoxy-1-pyrrolidinyl)-4-oxo-3-chinolincarbonsäure,
1-(2,4-Difluorphenyl)-5-amino-6,8-difluor-1,4-dihydro-7-
(cis-3-methylamino-4-methoxy-1-pyrrolidinyl)-4-oxo-3-
chinolincarbonsäure,
1-Thienyl-6,8-difluor-5-amino-1,4-dihydro-7-(cis-3-methylamino-4-methoxy-1-pyrrolidinyl)-4-oxo-3-chinolincarbonsäure,
1-Cyclopropyl-5-amino-6,8-difluor-1,4-dihydro-7-(cis-3-
methylamino-4-ethoxy-1-pyrrolidinyl)-4-oxo-3-chinolincarbonsäure,
1-Cyclopropyl-5-amino-8-chlor-6-fluor-1,4-dihydro-7-
(cis-3-amino-4-methoxy-1-pyrrolidinyl)-4-oxo-3-chinolincarbonsäure,
1-Cyclopropyl-5-amino-8-chlor-6-fluor-1,4-dihydro-7-
(cis-3-methylamino-4-methoxy-1-pyrrolidinyl)-4-oxo-3-
chinolincarbonsäure, oder
1-Cyclopropyl-5-amino-6,8-difluor-1,4-dihydro-7-(cis-3-
methylamino-4-methoxy-1-pyrrolidinyl)-4-oxo-3-chinolincarbonsäure.
3. Arzneimittel, umfassend eine Verbindung nach Anspruch 1
oder 2 und gegebenenfalls einen pharmazeutisch
verträglichen Träger und/oder Verdünnungsmittel.
4. Arzneimittel nach Anspruch 3, das als antibakterielles
Mittel nützlich ist.
5. Verwendung einer Verbindung nach Anspruch 1 oder 2 für
die Herstellung eines antibakteriellen Mittels.
6. Verfahren zur Herstellung von Chinolinoncarbonsäuren
nach Anspruch 1 oder 2 oder deren pharmazeutisch
verträglichen Salzen, umfassend
(a) die Umsetzung des Ausgangsmaterials (II)
mit dem Amin (III)
oder
(b) die Umsetzung der Verbindung (VII)
wobei R ein Wasserstoffatom oder einen
C&sub1;-C&sub4;-Alkylrest bedeutet, R3' eine Aminoschutzgruppe darstellt
und alle anderen Substituenten wie in Anspruch 1
definiert sind, mit Benzylamin, um die
Benzylaminogruppe
in 5-Stellung einzuführen, und dann
Entfernung der Benzylgruppe durch katalytische Hydrierung
und, falls notwendig, Entfernung jeder
Aminoschutzgruppe oder jeder Carboxylschutzgruppe auf übliche
Weise, und gegebenenfalls Umwandlung der
Verbindungen der Formel (I) in ihre Säureadditionssalze auf
übliche Weise.
