Description
- Die Erfindung betrifft neue S-Difluormethylhomocysteine und selektive Insektizide, die S-Difluormethylcysteine als Wirkstoff enthalten.
- Methionin ist eine der essentiellen Aminosäuren und sowohl die L-Form als auch die D-Form sind wirksame Pharmazeutika. Methionin kann für die Behandlung von Hepatitis und Toxikosen als lipophiler Nährstoff eingesetzt werden. Bis heute sind noch keine Methionin-Derivate mit insektizider Wirkung in der Literatur beschrieben.
- Blattläuse werden oft als Pflanzenschädlinge in der Landwirtschaft und im Gartenbau diskutiert. Organophosphat-Insektizide, wie Malathion und Pirimicarb, werden als landwirtschaftliche Insektizide gegen solche Blattläuse verwendet. Die Zahl der Blattläuse, die eine Toleranz oder Widerstandsfähigkeit gegenüber diesen Insektiziden besitzen, wächst allerdings. Der Erfindung liegt daher die technische Aufgabe zugrunde neue Verbindungen zur Verfügung zu stellen, die gegenüber Blattläusen wirksam sind, die eine Toleranz besitzen.
- Die Erfindung betrifft daher Verbindungen der allgemeinen Formel:
- in der R und R' jeweils ein Wasserstoffatom oder eine Schutzgruppe bedeuten, oder deren Säureadditions-Salze.
- Erfindungsgemäß bedeutet eine Schutzgruppe eine Amino- oder Carboxy- Schutzgruppe. Amino Schutzgruppen sind Schutzgruppen vom Carbamat- Typ, wie tert.-Butyloxycarbonyl- ( BOC ), p-Biphenylisopropyloxycarbonyl- ( Bpoc ), Diisopropylmethoxycarbonyl- ( Dipmoc ) oder Benzyloxycarbonylgruppen ( Z ), Aralkylreste, wie Trityl- oder Benzylgruppen, und Acylreste, wie Acetyl- oder Trifluoracetylgruppen. Eine Carboxy-Schutzgruppe bedeutet einen Alkylrest, wie eine Methyl-, Ethyl-, Propyl- oder Butylgruppe, einen Aralkylrest, wie eine Benzyl- oder Benzhydrylgruppe, und die Pivaloyloxymethylgruppe.
- Die erfindungsgemäße Verbindung kann nach folgenden Methoden A und B hergestellt werden. Methode A ( Synthese von Racematen ) Schutz von N Schritt Hydrolyse
- wobei M ein Alkalimetallkation bedeutet und R die vorstehende Bedeutung hat.
- L-Homocysteinthiolacton ( IV ) wird in üblicher Weise einer Schutzreaktion für Stickstoffatome unterworfen. Dieses ist ein Verfahren um eine Amino-Schutzgruppe, wie eine BOC, Z, Trityl-, Trifluoracetylgruppe oder einen Acylrest, in die Verbindung ( IV ) einzuführen ( M. Bodanszky und A. Bodanszky, " The Practice of Peptide Synthesis " Springer-Verlag, Tokyo, 1984, und Izumiya, et al., " Peptide Synthesis ", Maruzen, Tokyo, 1985 ). Die Einführung der Schutzgruppen BOC, Trityl, p-Biphenylisopropyloxycarbonyl ( Bpoc ) und Diisopropylmethoxycarbonyl ( Dipmoc ) oder anderer säureempfindlicher Gruppen erfolgt in einem geeigneten Lösemittel bei - 100º C bis 50º C vorzugsweise bei - 50º C bis Raumtemperatur. Ausgangsstoffe zur Einführung von BOC sind beispielsweise di-tert.-Butyldicarbonat, tert.-Butyloxycarbonylchlorid, tert.-Butyloxycarbonylazid, tert.-Butyl-4,6-dimethylpyrimidyl-2-thiolcarbonat und 2-(tert-Butoxycarbonyloxyimino)-2-phenylacetonitril. Tritylchlorid kann zur Einführung der Tritylgruppe verwendet werden. Gegebenenfalls kann eine Base zugesetzt werden. Als Basen können z.B. organische Basen, wie Triethylamin, Pyridin oder ähnliche, und anorganische Basen, wie Natriumhydrogencarbonat, Natriumcarbonat, Natriumhydroxid oder ähnliche verwendet werden. Lösemittel hierzu sind z.B. Alkohole, Ether, Methylenchlorid, Chloroform etc. . Diese Amino-Schutzgruppen bieten einige Vorteile bei der Verhinderung von Racemisierung und der leichten sauren Abspaltung. Wenn die Schutzgruppe die Z-Schutzgruppe ist, wird Verbindung ( IV ) mit einem Ausgangsstoff wie Benzyloxycarbonylchlorid in einem geeigneten Lösemittel in der Gegenwart einer Base bei 0º C bis 100º C, vorzugsweise ungefähr bei Raumtemperatur, umgesetzt. Als Base für die Umsetzung werden z.B. organische Basen wie Triethylamin oder ähnliche und anorganische Basen wie Natriumhydroxid, Natriumhydrogencarbonat oder ähnliche verwendet. Ebenfalls kann das vorstehend genannte Lösemittel eingesetzt werden.
- Das vorstehend erhaltene N-geschützte Homocysteinthiolacton ( III ) wird hydrolysiert, wobei das Dialkalimetallsalz ( II ) entsteht. Diese Umsetzung wird in einem geeigneten Lösemittel einige Minuten bis einige Stunden lang bei 0º C bis 100º C durchgeführt. Als Basen für diese Umsetzung werden z.B. Natriumbicarbonat, Kaliumcarbonat, Kalium-tert.-butoxid und Natriumhydrid verwendet. Als Lösemittel für die Umsetzung werden z.B. Wasser, Alkohole oder wäßrige Alkohole verwendet.
- Das vorstehend erhaltene Alkalimetallsalz ( II ) wird mit Chlordifluormethan in Gegenwart einer Base zur Zielverbindung ( I ) umgesetzt. Difluormethylierungen von Thiolen sind in J. Hine et al., J. Am. Chem. Soc., 79, 5493 ( 1957 ); T. Tsuji et al., J. Antibiotics, XXXVIII, 466 ( 1985 ) M. Hudlicky, " Chemistry of Organic Fluorine Compounds ", 2 nd. Ed., Ellis Horwood Ltd., Chichester, England, p. 444, ( 1972 ) beschrieben. Diese Verfahren sind aber nur auf Verbindungen beschränkt, die einfache funktionelle Gruppen besitzen. Die Verwendung dieser Verfahren für Aminosäuren oder Verbindungen, die eine freie Carboxygruppe besitzen, ist nicht beschrieben. Daher ist hervorzuheben, daß die neuen Difluormethylhomocysteine ( I ) hierdurch einfach und in guter Ausbeute hergestellt werden können. Am Produkt ( I ) kann gegebenenfalls die Schutzgruppe am Stickstoffatom der Aminogruppe entfernt werden und/oder die Carboxygruppe verestert werden. Falls die N-Schutzgruppe eine Benzyloxycarbonylgruppe ist, kann sie durch Hydrieren oder Umsetzung mit HBr/AcOH in bekannter Weise entfernt werden. Falls die Schutzgruppe BOC ist, kann sie durch Umsetzung mit HBr/AcOH, Trifluoressigsäure/organisches Lösemittel ( z.B. Ether, Methylenchlorid, Chloroform, etc. ) oder HCl/organisches Lösemittel ( z.B. Alkohole, Methylenchlorid, Chloroform, etc. ) entfernt werden. Wenn die Schutzgruppe eine Tritylgruppe ist, kann diese leicht durch Behandlung mit Säuren, wie Essigsäure, Trifluoressigsäure, Ameisensäure, Salzsäure, etc., entfernt werden. Ferner wird die Veresterung der Carboxygruppe in einem geeigneten Lösemittel in bekannter Weise, wie mit Säure-Alkohol, einer Diazoverbindung, Thionylchlorid-Alkohol, Säurehalogenid-Base- Alkohol oder ähnlichem durchgeführt. Lösemittel sind z.B. Alkohole, THF ( Tetrahydrofuran ), Ether, Methylenchlorid, etc.. Die Umsetzung wird bei einer Temperatur von - 20º C bis 100º C, vorzugsweise von 0º C bis Raumtemperatur durchgeführt. Die erfindungsgemäß nach diesem Verfahren erhaltene Verbindung ( I ) ist ein vollständiges Racemat. Das mit diesem Verfahren erhältliche Endprodukt ist ein Racemat, auch wenn als Ausgangsprodukt ein optisch aktives L-Homocysteinthiolacton ( IV ) eingesetzt wird. Dies wird durch die Racemisierung im zweiten Schritt verursacht. Daher haben die Erfinder die katalytische Hydrolyse bei Verwendung von Ag-, Hg- und Cu-Salzen als Katalysatoren [ S. Masamune et al., J. Am. Chem. Soc., 97, 3515 ( 1975 ) ] untersucht, um die Racemisierung in Schritt zwei zu unterdrücken. Diese Versuche gelangen nicht. Deshalb wird die Entwicklung einer weiteren Methode zur Herstellung solch einer optisch aktiven Verbindung nachstehend als Verfahren in der folgenden Methode B aufgezeigt. Methode B ( asymmetrische Synthese der L-Form ): Schritt
- Dieser Schritt beschreibt ein Verfahren zur Herstellung des Natriumsalzes von L-Homocystein ( II a ) durch Reduktion von L-Homocystin ( V ) mit Natrium in flüssigem Ammoniak in bekannter Weise ( K. Ramalingam und R. W. Woodard, J. Org. Chem., 49, 1984, 1291 ). Das Natriumsalz von L-Homocystein ( II a ), ein wertvolles Zwischenprodukt der Peptidchemie, kann durch diese Umsetzung ohne Racemisierung erhalten werden. Es wird empfohlen die Isolierung, Reinigung oder Aufbewahrung des Zwischenproduktes ( II a ) wegen seiner Zersetzungsneigung an der Luft ( wegen CO&sub2; oder O&sub2; ) unter Luftausschluß, vorzugsweise unter einer Stickstoffatmosphäre durchzuführen.
- Die im ersten Reaktionsschritt erhaltene Verbindung ( II a ) wird mit Chlordifluormethan in Gegenwart einer Base zur erfindungsgemäßen Verbindung ( I a ) umgesetzt. Es wurde festgestellt, daß optisch aktives L-Difluormethylcystein ( I a ) mittels dieser Umsetzung in einem Schritt ohne Racemisierung erhältlich ist. Daher wird angenommen, daß die Umsetzung eine ausgezeichnete diastereoselektive Synthese für das Produkt ( I a ) ist. Es wird empfohlen, die Umsetzung unter absolutem Luftausschluß durchzuführen.
- Die folgenden Beispiele, Referenz-Beispiele und Formulierungen sollen praktische Ausführungsformen der Erfindung verdeutlichen.
- Eine Lösung von 1,38 g ( 6,35 mmol ) von N-tert.-Butyloxycarbonylhomocysteinthiolacton ( III ) in 8 ml Ethanol wird mit 6,67 ml 2 n Natronlauge gemischt. Das Gemisch wird eine Stunde bei Raumtemperatur gerührt und danach zu einem weißen Pulver eingedampft. Das Pulver wird 2 Tage in einem Trockenapparat getrocknet. Man erhält 279 mg (1 mmol ) Pulver ( II ), das in 2 ml Ethanol gelöst und unter Rühren bei Raumtemperatur mit Chlordifluormethan gemischt wird. Das Gemisch wird 8 Stunden gerührt, mit 1 n HCl unter Eiskühlung neutralisiert und unter vermindertem Druck eingeengt. Der Rückstand wird mit 0,5 n Natronlauge alkalisch gemacht und mit Ether gewaschen. Die Waschlösung wird durch Zugabe von 1 n HCl auf einen pH von 2,0 eingestellt. Das Gemisch wird ausgesalzen und dreimal mit Ethylacetat extrahiert. Die organische Phase wird zweimal mit gesättigter Kochsalzlösung gewaschen. Die Lösung wird über Magnesiumsulfat getrocknet und unter vermindertem Druck eingedampft. Man erhält N-BOC-difluormethylhomocystein ( I-1 ) als Öl.

- Die vorstehend erhaltene Verbindung wird in den Benzhydrylester überführt und mittels einer Silica Gel HPLC-Säule ( Lobar B ) und dem Eluationsgemisch Benzol/Ethylacetat ( 4/1 v/v ) chromatographisch gereinigt. Man erhält 269 mg ( Ausbeute: 59,6 % ) der Esterverbindung.
- Die Schutzgruppen der vorstehend erhaltenen Verbindung werden mittels Trifluoressigsäure/Anisol abgespalten. Man erhält die Aminoverbindung als Endverbindung. Es wird mit Dowex- 50W X8 entsalzt und man erhält 77 mg ( Ausbeute: 43 % ) ( D,L )-Difluormethylhomocystein. Schmp. > 195º C ( Zers. )

- In eine Lösung von 2,04 g ( 10,56 mmol) des Natriumsalzes von L- Homocystein in 18 ml Ethanol wird bei Raumtemperatur in kräftigem Strom Chlordifluormethan eingeleitet. Währenddessen werden 2,03 g 2,01 mmol ) Kalium tert.-butoxid in 12ml Ethanol mittels eines Trichters innerhalb 1 bis 2 Stunden tropfenweise zu der Lösung gegeben. Nach Zugabe der Base wird Chlordifluormethan für eine weitere Stunde in die Lösung geleitet. Die Lösung wird dann mit 1 n HCl unter Eiskühlung angesäuert ( pH 4,3) und unter vermindertem Druck eingeengt. Der Rückstand wird zweimal mit Ethanol versetzt, das bis zur völligen Trockne des Rückstandes abdestilliert wird. Danach werden anorganische Salze durch Zugabe von 200 ml Methanol zu dem Rückstand ausgefällt. Die unlöslichen Salze werden abfiltriert und das Filtrat wird eingeengt. Man erhält einen Rückstand von 1,7 g. Der Rückstand wird säulenchromatographisch gereinigt. Die Reinigung erfolgt zunächst mit dem Ionenaustauscherharz Dowex 50W-X8 unter Verwendung von 1 n NH&sub3; als Eluent und anschließend mit HP-20 unter Verwendung von wäßrigem Methanol als Eluent. Man erhält 1,1 g ( Ausbeute: 56 % ) des gewünschten Produktes L-Difluormethylhomocystein.

- Eine vorbestimmte Konzentration der Ausgangslösung wird hergestellt, indem man eine Lösung der Verbindung mit destilliertem Wasser verdünnt, das Tween 20 in einer Konzentration von 100 ppm enthält. Eine Malathion Emulsion und benetzbares Pirimicarb Pulver werden mit destilliertem Wasser verdünnt, um jeweils eine vorbestimmte Konzentration der Testlösung zu erhalten.
- Myzus persicae ( URR-0 Typ ), erhalten von der Chiba Universität, die seit mehreren aufeinanderfolgenden Generationen gezüchtet wurden, wurden als Gruppe empfindlicher Stämme verwendet. Gegen Malathion und Pirimicarb resistente Myzus persicae wurden vom Aburahi Laboratorium von Shionogi & Co., Ltd. bezogen. Diese Blattläuse wurden über mehrere nachfolgende Generationen gezüchtet, wobei solche Tiere, die gegenüber diesen Verbindungen resistent waren, als Gruppe resistenter Stämme verwendet wurden.
- Ein Gefäß aus Polyethylen ( Durchmesser 6 cm, Tiefe 4 cm ) wurde mit 0,3 % Agar Gel gefüllt und ein 3x3 cm großes Stück eines Chinakohlblattes wurde auf den Gel gelegt. Eine flügellose erwachsene Myzus persicae wurde auf das Blatt gesetzt und dort 24 Stunden bei 25º C belassen, so daß sie ihre Larven ablegen konnte. Nach Entfernung der erwachsenen Blattlaus wurden 2 ml der Testlösung mittels einer rotierenden Sprühvorrichtung auf das Blattwerk gegeben. Die Larven der Myzus persicae verblieben für 48 Stunden bei 25º C auf diesem Blattwerk. Anschließend wurde die Mortalität der Larven bestimmt und der LC&sub5;&sub0; - Wert nach der Methode von Billis [ Billis, Ann. Appl. Biol., 22, 134; 307 ( 1935 ) ] berechnet. 1. Verbindungen der allgemeinen Formel:
in der R und R' jeweils ein Wasserstoffatom oder eine Schutzgruppe
bedeuten, oder deren Säureadditions-Salze.
2. Verbindungen nach Anspruch 1 in der optisch aktiven L-Form.
3. Verbindungen nach Anspruch 2 wobei R und R' jeweils ein

4. Verfahren zur Herstellung der Verbindungen nach Anspruch 1,
umfassend die alkalische Hydrolyse einer Verbindung der allgemeinen
Formel:

in der M ein Alkalimetallkation ist und R die in Anspruch 1 angegebene
Bedeutung hat, und Umsetzung dieser Verbindung mit

Schutzgruppe.
5. Verfahren zur Herstellung der Verbindungen nach Anspruch 3,
