2. Venvendung der nach dem Verfahren gemäss Patentanspruch 1 erhaltenen Polypeptide der Formel I zur Herstellung von Komplexen dieser Verbindungen, dadurch gekennzeichnet, dass man die Polypeptide der Formel I mit komplexbildenden anorganischen Metallverbindungen umsetzt.

Aus Nature 258, 567-8 (1975) ist es bekannt, dass die natürlichen Enkephaline die nachfolgende Struktur besitzen:
H-Tyr-Gly-Gly-Phe-X-OH (X = Leu oder Met-)
Als Säureadditionssalze kommen solche mit organischen Säuren, polymeren Säuren und Salze mit anorganischen Säuren in Frage. Unter den Komplexen sind z. B. anorganische Verbindungen, die sich von Metallen wie Calcium, Magnesium, Aluminium, Cobalt und insbesondere von Zink ableiten lassen, zu nennen.
Die Polypeptide bzw. Polypeptidderivate der obigen Formel können nach für die Synthese von Verbindungen dieser Art allgemein bekannten Methoden hergestellt werden, wobei die Aminosäuren in der festgelegten Reihenfolge einzeln oder nach vorheriger Bildung kleinerer Peptideinheiten miteinander verknüpft werden. Die Verknüpfung der Aminosäuren und/oder Peptideinheiten erfolgt z. B. in der Weise, dass man eine Aminosäure mit geschützter a-Aminogruppe und aktivierter terminaler Carboxylgruppe mit einer Aminosäure oder einem Peptid mit freier a-Aminogruppe und freier oder geschützter terminaler Carboxylgruppe umsetzt oder dass man eine Aminosäure oder ein Peptid mit aktivierter a-Aminogruppe und geschützter terminaler Carboxylgruppe mit einer Aminosäure oder einem Peptid mit freier terminaler Carboxylgruppe und geschützter a-Aminogruppe umsetzt.
Die Carboxylgruppe kann beispielsweise durch Uberführung in ein Säureazid, -anhydrid, -imidazolid, -isoxazolid oder einen aktivierten Ester oder durch Reaktion mittels eines Carbodiimids oder N,N'-Carbonyldiimidazols aktiviert werden.
Vorzugsweise wird als Kondensationsmethode die Carbodiimidmethode, die Azidmethode, die Methode der aktivierten Ester, die Anhydridmethode und die Merrifieldmethode verwendet.
An der Reaktion nicht beteiligte freie, funktionelle Gruppen können beim Aufbau des erfindungsgemässen Peptids durch die von der Synthese langkettiger Peptide her bekannten Schutzgruppen geschützt werden.
Die Umwandlung einer nicht mehr benötigten geschützten Aminogruppe in eine freie Gruppe sowie die Umwandlung einer funktionell abgewandelten Carboxylgruppe in eine freie Carboxylgruppe im Laufe des Verfahrens zur Herstellung der neuen Polypeptide erfolgt nach an sich bekannten Methoden durch Behandlung mit hydrolysierenden bzw. reduzierenden Mitteln.
Die Ausgangsprodukte zur Herstellung der neuen Polypeptide bzw. Polypeptidderivate können, sofern sie bisher nicht bekannt waren, nach den für die Peptidchemie bekannten Methoden erhalten werden, wobei die Aminosäuren einzeln oder nach vorheriger Bildung kleinerer Peptideinheiten miteinander verknüpft werden.
Die Polypeptide bzw. Polypeptidderivate der Formel I und die physiologisch verträglichen Säureadditionssalze dieser Verbindungen weisen im Tierversuch interessante pharmakodynamische Eigenschaften auf. Sie können daher als Heilmittel verwendet werden.
Die Verbindung zeigen eine hohe Affinität zum Opiatrezeptor im Rattenhirn. Die Testierung erfolgt wie beschrieben bei: C. B. Pert and S. H. Snyder, Molecular Pharmacology 10,
868 (1974).
Die Es,,, d. h. die Konzentration bei der 50% des spezi
fisch gebundenen [3Hl-Naloxans verdrängt werden, liegt bei diesen Verbindungen bei 10-5 bis 10" Mol/Liter.
Die neuen Verbindungen können deshalb als Heilmittel, insbesondere zur Linderung von Schmerzzuständen verschiedenster Genese, verwendet werden.
Die zu verwendende Dosis hängt von der verwendeten Verbindung, der Verabreichungsart und der gewünschten Behandlung ab. Im allgemeinen werden befriedigende Resultate mit täglichen Dosen von ungefähr 0,1 bis ungefähr 200 mg/kg Körpergewicht erreicht. Die Administration kann nötigenfalls in mehreren, beispielsweise in 2 bis 4 Anteilen täglich oder auch als Retardform erfolgen. Für grössere Säugetiere liegt die Tagesdosis im Bereich von etwa 10 bis etwa 1000 mg der Substanz; geeignete Dosierungsformen für z. B.
orale Anwendungen enthalten im allgemeinen ungefähr 2,5 bis 1000 mg Wirkstoff.
Die erfindungsgemäss hergestellten Verbindungen können als Heilmittel, z. B. in Form pharmazeutischer Präparate, Verwendung finden. Diese enthalten die genannten Verbindungen in Mischung mit einem für parenterale Applikationen geeigneten organischen oder anorganischen Trägermaterial.
Die Verbindungen können auch in Form eines Depotpräparates verabreicht werden.
Es werden folgende Abkürzungen verwendet: Tyr = L-Tyrosyl D-Ala = D-Alanyl Gly = Glycyl Phe = L-Phenylanyl Met = L-Methionyl BOC = tert.-Butyloxycarbonyl OMe = Methoxy DMF = Dimethylformamid Zers.p. = Zersetzungspunkt
In dem folgenden Beispiel, welches die Ausführung des Verfahrens erläutert, den Umfang der Erfindung aber in keiner Weise einschränken soll, erfolgen alle Temperaturangaben in Celsiusgraden. Der Wert c für die optische Drehung beträgt 1.
Beispiel
H-Tyr-(D)-Ala-Gly-Phe-Met-NH2 (HCl)
1,1 g BOC-Tyr-(D)-Ala-Gly-Phe-Met-NH2 werden in 50 ml HCl-haltigem Dioxan gelöst und während einer Stunde bei Raumtemperatur stehen gelassen. Nach Einengen der Lösung im Vakuum wird die Verbindung mit überschüssigem Äther ausgefällt und abfiltriert. Man erhält die Titelverbindung als amorphe Substanz.

Das als Ausgangsmaterial verwendete BOC-Tyr-(D) Ala-Gly-Phe-Met-NH2 wird wie folgt hergestellt: a) BOC-Tyr-(D)-Ala-OH
2.8 g BOC-Tyr-OH und 1,4 ml Triäthylamin werden in 50 ml Tetrahydrofuran gelöst und bei -10" mit 1,0 ml Chlor ameisensäureäthylester versetzt. Nach I 10 Minuten wird eine Lösung, bestehend aus 1,4 g D-Ala-OMe.HCI in 20 ml DMF zugetropft und 3 Stunden bei Raumtemperatur weitergerührt.
Nach Einengen des Lösungsmittels wird der Rückstand in Essigester gelöst und mit HCI l-N und Kaliumbicarbonat l-N gewaschen. Nach Einengen des Lösungsmittels wird der Ester direkt in die Säure überführt, und zwar auf folgende Weise: Man löst den Rückstand in 30 ml Methanol auf und versetzt mit 10 ml 2-N NaOH. Nach l-stündigem Stehen wird das Methanol verdampft und die wässrige Phase wird mit Äther wiederholt gewaschen. Nach Ansäuern mit verdünnter Citronensäure wird die Substanz mit Essigester extrahiert. Die organische Phase wird eingedampft und der Rückstand mit Hilfe von Äther/Petroläther kristallisiert. Man erhält die Titelverbindung vom Smp. 108 (Zers.); ctD20 = -31" (c = 1,0 in DMF).
b) BOC-Tyr-(D)-Ala-Gly-OH
3,7 g BOC-Tyr-(D)-Ala-OH werden in 50 ml Tetrahydrofuran gelöst und mit 1,4 ml Triäthylamin versetzt. Nach Abkühlen auf 10 tropft man 1,0 ml Chlorameisensäureäthylester zu und nach 10 Minuten versetzt man mit einer Lösung von 3,4 g Gly-oBz-Tosylat in 50 ml DMF. Die Aufarbeitung des Esters erfolgt wie unter a angegeben. Der Ester wird wiederum nicht charakterisiert, aber wie folgt in die entsprechende Säure übergeführt: Der Ester wird in 50 ml Methanol gelöst und mit Pd/C katalytisch bis zur konstanten Wasserstoffaufnahme hydriert. Nach Abfiltrieren vom Katalysator und Einengen der Lösung versetzt man mit Äther-Petroläther bis das Produkt als amorphe Verbindung auffällt. Man isoliert die Titelverbindung. Zers.p. 84 ; [celD20 = -14" (c = 1,0 in DMF).
c) BOC-Tyr-(D)-Ala-Gly-Phe-Met-NH2
1,3 g BOC-Tyr-(D)-Ala-Gly-OH und 0,5 ml Triäthylamin werden in 50 ml Tetrahydrofuran gelöst und bei -10 0,3 ml Chlorameisensäureäthylester zugetropft. Nach 10 Minuten versetzt man mit einer Lösung von 1 g HCl.H-Phe-Met-NH2 und 0,5 ml Triäthylamin in 30 ml DMF. Weitere Aufarbeitung wie unter a beschrieben. Man erhält die Titelverbindung vom Zers.p. 72 ; [a1n20= -11" (c = 1,0 in DMF).
