Description
Beschreibung
Die vorliegende Erfindung betrifft neue 3-Alkylthio-2-O-carbamoyl-propan-1,2-diol-1-0-phosphocholine
und Verfahren zu ihrer Herstellung.
Die erfindungsgemäßen Verbindungen entsprechen der allgemeinen

und einen geradkettigen oder verzweigten, gesättigten oder ungesättigten Kohlenwasserstoffrest mit 1-20 Kohlenstoffatomen,
Phenyl, durch C,-Co-Alkyl, C,-Cg-Alkoxy, Halogen,
Trif1uormethyl einfach substituiertes Phenyl, Benzyl oder
Wasserstoff bedeuten.

Die erfindungsgemäßen Verbindungen besitzen wertvolle phar-
makologische Eigenschaften und sind daher zur Verwendung 3Ö als Wirkstoffe in Arzneimitteln geeignet.
Von den angeführten, erfindungsgemäßen Verbindungen werden
Substanzen, wie z.B. 3-Hexadecylthio-2-0-methylcarbamoylpropan-l,2-diol-1-0-phosphocholin
oder 2-0-Ethylcarbamoyl-3-
1 octadecylthio-propan-l,2-diol-l-O-phosphocholin besonders
bevorzugt. Die genannten Verbindungen können zur Behandlung des Hochdrucks, verschiedener rheumatischer und einiger
atherosklerotischer Krankheitsbilder sowie zur Tumortherapie am Menschen genutzt werden.
Daneben werden Verbindungen, wie z.B. 3-Hexadecylthio-2-0-hexadecylcarbamoyl-propan-l^-diol-l-O-phosphocholin
bevorzugt, die gegenüber den bisher in der Liposomentechnik verwendeten,
natürlichen oder synthetischen 1,2-Diacyl-glycero-3-phosphocholinen
(z.B. DE-OS 27 12 031) deutliche Vorteile aufweisen, da sie im Gegensatz zu diesen Phospholipiden gegen
Phospholipase Ap beständig sind und damit eine bessere Vermittlung
des eingeschlossenen Wirkstoffes an den Wirkungsort ermöglichen.
Die erfindungsgemäßen Substanzen der Formel I mit RC=H werden
durch Reaktion der Lysoverbindungen II, worin R die in Anspruch 1 genannte Bedeutung hat, mit den entsprechenden Alkyl·
bzw. Aryl isocyanaten der Formel III hergestellt.

Die Reaktion wird zweckmäßig in organischen aprotischen Lösungsmitteln,
wie z.B. Chloroform, Aceton, Dimethylformamid bzw. deren Mischungen, gegebenenfalls unter Anwendung eines
Katalysators, insbesondere einer Lewis-Base, wie z.B. Trie-
thylamin, Pyridin, Dimethylaminopyridin, Dimethylformamid,
bei Temperaturen zwischen 0° und 1000C, vorzugsweise bei
40° - 600C, durchgeführt.
Als Ausgangsverbindungen der Formel II kommen z.B. in Frage:

Die als Ausgangsverbindungen eingesetzten 3-Alkylthiopropan-1,2-diol-l-O-phosphocholine
der Formel II lassen sich nach den üblichen Verfahren, z.B. durch enzymatische Spaltung
(Phospholipase A„) oder milde alkalische Hydrolyse aus den
entsprechenden 2-0-Acyl-3-alkylthio-propan-l,2-diol-l-0-phosphocholinen
synthetisieren.
Aus den genannten Lysoverbindungen II lassen sich durch Umsetzung mit Dialkylcarbamidsäurehalögeniden, insbesondere
mit Chloriden der Formel IV

N,N-Dimethylcarbamidsäurechlorid, N-Ethyl-N-methylcarbarnidsäurechlorid, N,N-Diethyl carbamid saurechlorid, N-Methyl-N-propylcarbamidsäurechlorid, N.N-Dipropylcarbamidsäurechlorid, in indifferenten organischen Lösungsmitteln, wie z.B.
Chloroform, sowie Lösungsmittel gemischen, gegebenenfalls
unter Zusatz von üblichen Basen, wie z.B. tertiären Aminen, Metalloxiden, Metal!carbonaten, die 3-Alkylthio-2-0-dia!kylcarbamoyl-propan-1,2-diol-l-O-phosphocholine der Formel I
herstellen.
Die Herstellung der erfindungsgemäßen Verbindungen wird durch
die folgenden Beispiele näher erläutert. Die Schmelzpunkte wurden mit einem Büchi 510-Schmelzpunktbestimmungsapparat
gemessen und sind nicht korrigiert.
Beispiel 1
3-Hexadecy1thio-2-0-tnethylcarbamoyl-propan-l,2-diol-l-0-phosphocholin.
3-Hexadecy1thio-2-0-tnethylcarbamoyl-propan-l,2-diol-l-0-phosphocholin.
a) ß-Bromethyl-CS-hexadecylthio-2-hydroxy-propyl)-phosphat.
Eine Mischung von 30 g Hexadecy1thio-propan-l,2-diol und
300 ml trockenem Chloroform wird auf O0C abgekühlt und in die Mischung gleichzeitig eine Lösung von 21,6 g
ß-Bromethylphosphorsäuredichlorid in 25 ml Chloroform
und eine Lösung von 7,2 ml Pyridin in 25 ml Chloroform langsam unter Rühren eingetropft. Nach halbstündigem weiteren
Rühren werden nochmals 7,2 ml Pyridin hinzugefügt und die Mischung 12 Stunden bei Raumtemperatur sowie nach
Zugabe von Eiswasser eine weitere Stunde gerührt. Die organische Phase wird abgetrennt, mit Wasser und Natriumchloridlösung
neutral gewaschen, über Natriumsulfat getrocknet und im Vakuum eingeengt. Der Rückstand wird durch

Ausbeute: 11,9 g mit Schmp. 197-198°C
b) ß-Bromethyl-(2-acetyloxy-3-hexadecylthio-propyl)-phosphat.
14,6 g ß-Bromethyl-(3-hexadecylthio-2-hydroxy-propyl)-phosphat
wenden in 100 ml trockenem Chloroform gelöst und die Lösung mit 7 ml Pyridin und 8,2 ml Essigsäureanhydrid
versetzt. Nach 48-stündigem Rühren bei Raumtemperatur wird wenig Wasser hinzugefügt und nochmals 15 Minuten
gerührt. Die Mischung wird im Vakuum eingeengt und der Rückstand durch Säulenchromatographie (Kieselgel//
3q Chloroform/Methanol) gereinigt.
Ausbeute: 5,1 g mit Schmp. 7 63°C

c) 2-0-Acetyl-S-hexadecylthio-propan-l^-di ο 1-1-0-phosphocho-1

1,8 g ß-Bromethyl-(2-acetyloxy-3-hexadecylthio-propyi)-ü5
phosphat werden in 3 ml Toluol gelöst, die Lösung mit 3 ml einer 20%igen Lösung von Trimethylamin in Toluol
versetzt und 3 Stunden bei 60°C im verschlossenen Gefäß gerührt.

Die Reaktionsmischung wird im Vakuum eingeengt und der Rückstand durch Säulenchromatographie /Kiesel gel//Chloroform/Methanol/Wasser
= 80/20/2, 40/20/2, 20/20/2 (V/V/V)J gereinigt.
Ausbeute: 0,9 g mit Schmp. 246-249°C (Zers.)
d) S-Hexadecylthio-propan-l^-diol-l-O-phosphocholin.
0,8 g 2-0-Acetyl-3-hexadecy1thio-propan-l,2-d1ol-l-0-phosphocholin
werden in 15 ml Ethanol gelöst und 0,2 g gepulvertes Kaliumcarbonat hinzugefügt. Nach 48-stündigem Rühren
werden die Salze abfiltriert und mit Chloroform gewaschen, die vereinigten Filtrate im Vakuum eingeengt
und der Rückstand durch Säulenchromatographie /TCieselgel //
Chloroform/Methanol/Wasser = 80/20/2, 60/20/2, 50/20/2, 40/20/2, 30/20/2, 20/20/2 (V/V/V)7 gereinigt.
Ausbeute: 0,4 g mit Schmp. 252-2540C
e) 3-Hexadecylthio-2-0-methylcarbamoyl-propan-l,2-dio1-1-0-phosphocholin.
0,17 g 3-Hexadecylthio-propan-l^-diol-l-O-phosphocholin
werden in 10 ml trockenem Chloroform und 1 ml Dimethylformamid gelöst und 2 ml Methylisocyanat hinzugefügt.
Nach 48-stündigem Rühren bei 400C wird die Lösung im Vakuum
eingeengt und der Rückstand durch Säulenchromatographie /Kieselge^/Chloroform/Methanol/Wasser = 80/20/2, 60/20/2,
40/20/2, 20/20/2 (V/V/V)7 gereinigt. Ausbeute: 0,14 g mit Schmp. 252-254°C
2-0-Ethylcarbamoyl-3-hexadecylthio-propan-l,2-dio^-l-0-phosphocholin.
0,17 g 3-Hexadecylthio-propan-l,2-diol-l-O-phosphocholin
werden in 10 ml trockenem Chloroform und 1 ml Dimethylformamid gelöst und 1 ml Ethylisocyanat hinzugefügt. Nach 48-stündigem
Rühren bei 4O0C wird die Lösung im Vakuum eingeengt
und 3er Rückstand durch Säulenchromatographie ZXieseIgel/-/

Chloroform/Methanol/Wasser = 80/20/2, 60/20/2, 40/20/2, 20/20/2 (V/V/VJ7 gereinigt.
Ausbeute: 0,15 g mit Schmp. 250-2520C

Eine Mischung aus 100 mg 2-0-Hexadeclcarbamoyl-l-hexadecylthio-propan-1,2-diol-l-O-phosphocholin,
200 mg Hexadecylisocyanat, 10 ml Chloroform.und 1 ml Dimethylformamid wird bei
600C gerührt (48 Stunden). Das liberschlissige Alkyl i socyanat
wird durch Zugabe von wenig Wasser hydrolysiert und die Mischung im Vakuum weitgehend eingeengt. Der Rückstand wird
durch Säulenchromatographie (Kieselgel//Chloroform/Methanol/
Wasser) gereinigt.
Ausbeute: 93 mg
Analog werden hergestellt:
3-Hexadecylthiο -2-0-oetadecylcarbamoyl-propan-1,2-diol-l-O-phosphochol

2-0-Dimethylcarbamoyl-3-hexadecylthio-propan-1,2-dio1-1-0-phosphocholin,
Eine Mischung aus 100 mg 3-Hexadecylthio-propan-1,2-diol-1-0-phosphocholin,
40 mg Dimethylcarbamidsäurechlorid und
60 mg Silbercarbonat in 10 ml Chloroform wird 24 Stunden bei 500C gerührt, das Lösungsmittel im Vakuum abgezogen und
der Rückstand durch Säulenchromatographie (Kieselgel/ZChloroform/Methanol/Wasser)
gereinigt.
Ausbeute: 83 mg mit Schmp. 239-241°C
l Analog Beispiel 4 werden hergestellt:
2-0-Dimethy!carbamoyl-3-octadecyithio-propan-1,2-dio1-1-0·
phosphocholin,
2-0-Dimethylcarbamoyl~3-eicosylthio-propan-l,2-dio1-1-0-5
phosphocholin.
Claims (7)
- Patentansprüche^/TT)3-Alkylthio-2-0-carbamoyl-propan-l,2-diol-l-0-phosphocholine der allgemeinen Formel IH2C-S-R1

R einen geradkettigen, gesättigten oder ungesät-tigten Kohlenwasserstoffrest mit 10-20 Kohlenstoffatomen3
R , R gleich oder voneinander verschieden sein können und einen geradkettigen oder verzweigten, gesättigten oder ungesättigten Kohlenwasserstoffrest mit 1-10 Kohlenstoffatomen, Phenyl, durch Methyl, Methoxy, Ethoxy, Fluor, Chlor, Brom, Trifluormethyl einfach substituiertes Phenyl, Benzyl oder Wasserstoff
bedeuten. - 3. S-Alkylthio^-O-carbamoyl-propan-l^-diol-l-O-phosphocholine der allgemeinen Formel IworinR einen geradkettigen, gesättigten Kohlenwasserstoffrest mit 16, 18 oder 20 Kohlenstoffatomenρ οR1 R gleich oder voneinander verschieden sein können und einen Methyl-, Ethyl-, Isopropylrest oder Wasserstoff
bedeuten. - 4. S-Alkylthio^-O-carbamoyl-propan-l^-diol-l-O-phosphocholine der allgemeinen Formel IworinR einen geradkettigen, gesättigten oder ungesättigten Kohlenwasserstoffrest mit 10-20 KohlenstoffatomenR^ ein WasserstoffatomR einen geradkettigen, gesättigten oder ungesättig-QQ ten Kohlenwasserstoffrest mit 10-20 Kohlenstoffatomenbedeuten, wobei R* und R3 gleich oder verschieden sein .können.
- 5. 3-Alkylthio-2-0-carbamoyl-propan-l^-diol-l-O-phosphocholine der allgemeinen Formel I

bedeuten, wobei R und R° gleich oder verschieden sein - 6. Verfahren zur Herstellung von Verbindungen der Formel I gemäß den Ansprüchen 1-5, dadurch gekennzeichnet, daß man Lysoverbindungen der allgemeinen Formel II 15H2C-S-R1HC-OH II0
ΐ +H2C-O-P-(CHg)2-N(CH3J30"in der R die in Formel I angegebene Bedeutung besitzt, in einem aprotischen organischen Lösungsmittel, gegebenenfalls unter Zusatz einer Lewis-Base als Katalysator mit einem Isocyanat der Formel IIIR3-N=C=O IIIin der R die in Formel I angegebene Bedeutung hat, umsetzt.1 - 7. Verfahren zur Herstellung von Verbindungen der Formel I gemäß den Ansprüchen 1-3, dadurch gekennzeichnet, daß man Lysoverbindungen der Formel II in einem indifferenten, organischen Lösungsmittel, gegebenenfalls unter Zusatz5 eines Säureakzeptors, mit den entsprechenden Carbamidsäurechloriden der Formel IV umsetzt,R2Cl-CO-N^ _ IVXR3910 in der R und R die in Formel I angegebenen Bedeutungen besitzen.
