Description
Verfahren zur Herstellung von herbicid wirksamen Anilinderivaten
Die Erfindung betrifft ein Verfahren zur Herstellung von herbicid wirksamen Verbindungen sowie ihre Verwendung.
Es ist z.B. aus den GB-PS 1 164 160 und 1 289 283 bekann,
daß bestimmte Alaninderivate herbicide Wirksamkeit gegenüber wildem Hafer ausüben und darüberhinaus, daß diese
Verbindungen selektiv in Nutzgetreide angewandt werden können, um wilden Hafer, der zusammen mit dem Nutzgetreide
wächst, zu bekämpfen. Es konnte kürzlich gezeigt werden, daß bestimmte andere nahe verwandte Alaninderivate ebenfalls
selektive herbicide Wirksamkeit in Nutzgetreide, besonders gegenüber breitblättrigen Unkräutern besitzen.
Es hat sich ferner gezeigt, daß eines der optisch aktiven Isomeren dieser Alaninderivate sehr viel wirksamer
gegen Unkräuter ist als das andere Isomer,und bisher bestand das einzige Syntheseverfahren in einer Auftrennung
des Gemisches. Erfindungsgemäß wurde nun ein neues stereospezifisches
Verfahren gefunden, bei dem die Notwendigkeit langer und mühsamer Auftrennverfahren entfällt.

Die Erfindung betrifft ein Verfahren zur Herstellung einer Alaninverbindung der folgenden allgemeinen Formel, die an dem
Kohlenstoffatom 2 R-Konfiguration besitzt:

unabhängig ein Wasserstoffatom, eine Alkyl-, Aryl-, Aralkyl-,
Alkaryl-, Alkenylgruppe oder zusammen eine Alkylenbrücke darstellen, die gegebenenfalls durch 1 oder mehrere nicht-Kohlenstoffatome

Molekül austritt und ersetzt wird durch ein Anilin, dem ein Proton fehlt (deprotonated) der Formel III und Z und Y

die oben angegebene Bedeutimg haben) in Gegenwart einer Base und unter erhöhter Temperatur, um die gewünschte Verbindung
der Formel I zu erhalten, in der R ein Wasserstoffatom ist und gegebenenfalls diese Verbindung in das entsprechende
N-Benzoyl- oder N-Thiobenzoylderivat umwandelt.
Die austretende Gruppe X-O- in der Formel II muß, wie oben angegeben, imstande sein, durch ein Anilin der Formel
III, dem ein Proton fehlt, ersetzt zu werden und geeignete Beispiele für derartige Gruppen sind: -0.SO2.Q, -O.CO.Q und
-0.CH(OH)XV,, wobei Q eine Kohlenwasserstoffgruppe, günstigerweise
eine Alkyl- oder Arylgruppe mit bis zu 10 Kohlenstoffatomen und V ein Halogenatom, z.B. ein Chlor-, Fluoroder
Bromatom ist. Vorzugsweise ist die austretende Gruppe X-O- eine -O.SO„.Q-Gruppe, z.B. eine Methansulfonyl- oder
p-*Toluolsulfonylgruppe. Es hat sich gezeigt, daß das Vorhandensein
einer dieser austretenden Gruppen in dem Milchsäurederivat II es ermöglicht, das erwünschte optisch
aktive Isomer der Formel I während der Umsetzung von einer Verbindung der Formel II mit dem Anilin der Formel III zu
erhalten. Durch dieses Verfahren wird nicht nur das gewünschte optisch aktive Isomer erhalten, sondern die Ausbeuten
betragen mehr als 70 % und häufig bis zu 80 % oder darüber.
Die Umsetzung zwischen dem Milchsäurederivat II und dem Anilin III kann bei Temperaturen zwischen 50 und 2000C,
vorzugsweise zwischen 130 und 1700C durchgeführt werden.
Wie bei allen stereospezifischen Verfahren müssen die Reaktionsbedingungen sorgfältig ausgewählt und für jeden
speziellen Fall untersucht werden, um sicherzustellen, daß die.optische Integrität des gewünschten Produktes durch die
Bedingungen nicht nachteilig beeinflußt wird. Eine dieser Bedingungen ist die Art der bei der Reaktion zwischen dem
Milchsäurederivat II und dem Anilin III vorhandenen Base. Die Base kann organisch oder anorganisch sein, wobei die
Anwendung von Alkalicarbonaten oder -bicarbonaten bevorzugt
ist. Besonders gute Ergebnisse werden erhalten mit

Natriumcarbonat. Wahlweise kann das Amin III selbst als Base angewandt werden.
Das Anilin III kann als Lösungsmittel für die Reaktion angewandt werden, aber es kann auch ein zusätzliches Lösungsmittel
angewandt werden, wenn das die Reaktion unterstützt.
Das als Ausgangssubstanz bei dem erfindungsgemäßen Verfahren
angewandte Milchsäurederivat II kann leicht hergestellt werden durch Umsetzung eines Säurehalogenids der Formel
X-Halogenid oder des entsprechenden Säureanhydrids (X-O-X) oder eines halogenierten Aldehyds mit einem Milchsäurederivat
der Formel IV, das an dem Kohlenstoffatom 2 S-Konfiguration besitzt:

wobei X und V/ die oben angegebene Bedeutung haben. Vorzugsweise
wird die Reaktion in Gegenwart einer Base, z.B. einer organischen Base, wie Pyridin oder Triäthylamin durchgeführt
wird. Im allgemeinen findet die Reaktion bei Raumtemperatur statt, aber es können, wenn das erwünscht ist,
Temperaturen im Bereich von -5 bis +800C angewandt werden.
Das Säurehalogenid oder das entsprechende Säureanhydrid oder halogenierte. Aldehyd kann unter den folgenden Verbindungen
ausgewählt werden
V.S02.Q, V.CO.Q, 0(C0.Q)2 und H.CO.CV, ,
wobei Q und V die oben angegebene Bedeutung haben. Vorzugsweise
wird ein Alkylsulfonyl-oder Arylsulfonylhalogenid angewandt,
z.B. Methansulfonylchlorid oder p-Toluolsulfonylchlorid.
Die Verbindung IV ist eine billige leicht verfügbare Substanz, wenn W ein Wasserstoffatom bedeutet, d.h. sie ist
S-(+)-Milchsäure. Die Ester der Formel IV können bequem aus S-(+)-Milchsäure hergestellt werden durch Veresterung mit
dem entsprechenden Alkohol. Wahlweise kann S-(-)-Äthyllactat,

das ebenfalls eine leicht zugängliche Verbindung ist, durch Umesterung
in das gewünschte Milchsäurederivat IV umgewandelt
werden.
Bei der Herstellung von Verbindungen der allgemeinen Formel I, bei denen ¥ kein Wasserstoffatom ist, kann es, je
nach den Umständen, günstig sein, den Substituenten ¥ in die Ausgangsverbindung IV einzuführen,oder wahlweise kann es
günstiger sein, von der Säure (II; W=H) auszugehen und den
Substituenten ¥ in einer späteren Stufe oder in der letzten Stufe des Verfahrens einzuführen. Gleichermaßen kann es bei
der Herstellung von Verbindungen der Formel I, in denen ¥ ein ¥asserstoffatom ist, günstig sein, von einem Ester auszugehen
und die Ester^in^-Biner Zwischenstufe oder der letzten
Stufe des Verfahren zu entfernen.
Die erfindungsgemäßen Verbindungen der Formel I, bei denen R eine Benzoylgruppe ist, können hergestellt werden
durch Umsetzung einer erfindungsgemäßen Verbindung der Formel I, in der R ein Wasserstoffatom ist mit einem Benzoylhalogenid,
wie Benzoylchlorid. Günstigerweise wird die Reaktion in Gegenwart eines Lösungsmittels , wie Toluol,unter Rückfluß
durchgeführt. Die Verbindungen der Formel I, bei denen
R eine Thiobenzoylgruppe ist, können hergestellt werden durch Umsetzung des entsprechenden Benzoylderivats mit einem
Sulfid, wie P2 S 5·
Vorzugsweise enthalten die Alkyl-, Alkenyl- und Alkylengruppen in der Formel I bis zu 6 Kohlenstoffatome,
die Aryl-, Aralkyl- und Alkarylgruppen bis zu 10 Kohlenstoff atome, das Halogenatom ist, wenn nicht anders angegeben,
ein Chlor- oder Fluoratom und die Heteroatome sind Sauerstoff- und Stickstoffatome.
Nach dem erfindungsgemäßen Verfahren entstehen 2 verschiedene
Klassen von selektiven Herbiciden für Nutzgetreide, und zwar eine erste (Verbindungen der Formel I, bei denen R
ein ¥asserstoffatom ist), die aktiv ist gegen breitblättrige
Unkräuter und eine zweite (Verbindungen der Formel I, bei

denen R eine Benzoyl- oder Thiobenzoylgruppe ist), die wirksam ist gegen wilden Hafer. Bei beiden Gruppen sind die bevorzugten
Verbindungen solche der Formel I, bei denen Z ein Fluoroder Chloratom, Y ein Wasserstoff-, Chlor- oder Fluoratom und
W eine Alkylgruppe mit bis zu 6 Kohlenstoffatomen, z.B. eine Methyl-, Äthyl- oder Isopropylgruppe ist.
Um die absolute Konfiguration der Produkte, Ausgangssubstanzen und Zwischenprodukte bei dem erfindungsgemäßen Va?-
fahren zu beschreiben, wurde die R- und S-Bezeichnung angewandt, wie sie angegeben ist in Experientia, Band 12, Seite
81-94, 1956.
Das erfindungsgemäße Verfahren wird durch die folgenden Beispiele erläutert, bei denen die Struktur aller synthetisierten
Verbindungen durch das Infrarot- und NMR-Spektrum bestätigt worden ist.
Es wurden die folgenden Verbindungen hergestellt:

Die Verbindungen C, D, H, I, J, K, L und M sind die herbicid wirksamen Verbindungen, die mit Hilfe des erfindungsgemäßen
Verfahrens hergestellt werden und die Verbindungen A, B, E, F und G sind Zwischenprodukte zur Herstellung der
herbicid wirksamen Verbindungen.
Die Verbindungen C, H, J und L sind die rechtsdrehenden selektiven Herbicide gegen breitblättrige Unkräuter in Nutzgetreide
während die Verbindungen D, I, K und M die linksdrehenden selektiven Herbicide gegen wilden Hafer in Nutzgetreide
sind.
Die Verbindungen A bis M wurden nach den Verfahren hergestellt, die in der folgenden Tabelle zusammengefaßt

die Ausgangssubstanz, sind ebenfalls angegeben.

Claims (14)
- Patentansprüchei. Verfahren zur Herstellung von herbicid wirksamen Anilinderivaten "bei denen das Kohlenstoffatom 2 R-Konfiguration hat, der allgemeinen FormelCOOW—Z(Dund in der Y und Z jeweils ein Wasserstoff^ Chlor- oder Fluoratom, W ein Wasserstoffatom, eine Alkyl-, Halogenalkyl-, Alkenyl-, Cycloalkyl-, Aryl-, Alkaryl-, Aralkyl-oder eine Gruppe der Formel -N=CR1Rp bedeuten, wobei R. und Rp jeweils unabhängig ein Wasserstoffatom, eine Alkyl-, Aryl-, Aralkyl-, Alkaryl-, Alkenylgruppe oder zusammen eine Alkylenbrücke darstellen, die gegebenenfalls durch 1 oder mehrere nicht-Kohlenstoffatome unterbrochen ist und R ein Wasserstoffatom oder eine Benzoyl- oder Thiobenzoylgruppe bedeuten und das dadurch gekennzeichnet ist, daß man ein Milchsäurederivat, das an dem Kohlenstoffatom 2 S-Konfiguration besitzt,der FormelCOOWX-O-C" CH(II)umsetzt mit einem Anilin der Formel709820/1013(III)(wobei die Gruppe X-O- eine Gruppe darstellt, die aus dem Molekül austritt und ersetzt wird durch ein Anilin, dem ein Proton fehlt der Formel III,und Z und Ydie oben angegebene Bedeutung haben) in Gegenwart einer Base und unter erhöhter Temperatur, um die gewünschte Verbindung der Formel I zu erhalten, in der R ein Wasserstoffatom ist. und gegebenenfalls diese Verbindung in das entsprechende N-Benzoyl- oder N-Thiobenzoylderivat umwandelt.
- 2. Verfahren nach Anspruch 1, dadurch gekennzeichnet , daß man von einer Verbindung der Formel II ausgeht, bei der die austretende Gruppe X-O-eine Gruppe der Formel -0.SO2.Q, -O.CO.Q oder -0.CH(OH).CV, ist.
- 3. Verfahren nach Anspruch 2, dadurch gekennzeichnet , daß die Gruppe X-O- eine -O.SOp.Q-Gruppe, vorzugsweise eine Methansulfoiiyl- oder p-Toluolsulfonylgruppe ist.
- 4. Verfahren nach Anspruch 1 bis 3 , dadurch gekennzeichnet , daß man die Reaktion zwischen dem Milchsäurederivat der Formel II und dem Anilin der Formel III bei einer Temperatur zwischen 50 und 2000C, vorzugsweise zwischen 130 und 1700C durchführt.
- 5. Verfahren nach Anspruch 1 bis 4, dadurch gekennzeichnet, daß man die Reaktion zwischen dem Milchsäurederivat der Formel II und dem Anilin der Formel III in Gegenwart eines Alkalicarbonats oder -bicarbonats durchführt.
- 6. Verfahren nach Anspruch 5, dadurch gekennzeichnet, daß man als Alkalicarbonat Natriumcarbonat verwendet.../3 709820/1013

Verfahren nach Anspruch 1 bis 6, dadurch gekennzeichnet , daß man ein Milchsäurederi'-vat der Formel II verwendet, das hergestellt worden ist durch Umsetzung eines Säurehalogenids der Formel X-Halogenid oder des entsprechenden Säureanhydrids oder eines halogenierten Aldehyds mit einem Milchsäurederivat, das an dem Kohlenstoffatom 2 S-Konfiguration besitzt,der FormelCOOWHO - C2 - H (IV)CH,wobei X und W die oben angegebene Bedeutung haben.
- 8. Verfahren nach Anspruch 7, dadurch gekennzeichnet, daß man die Reaktion bei einer Temperatur im Bereich von· -5 bis +800C, vorzugsweise bei Raumtemperatur durchführt.
- 9. Verfahren nach Anspruch 7 oder 8, dadurch gekennzeichnet , daß man die Reaktion in Gegenwart einer organischen Base, wie Pyridin oder Triäthylamin, durchführt.
- 10. Verfahren nach Anspruch 7 bis 9, dadurch gekennzeichnet , daß man als Ausgangssubstanz ein Alkylsulfonyl- oder Arylsulfonylhalogenid verwendet.
- 11. '., Verfahren nach Anspruch 10, dadurch gekennzeichnet, daß man als Sulfonylhalogenid Methansulfonylchlorid oder p-Toluolsulfonylchlorid verwendet.0 9 8 2 0/1013
- 12. Verfahren nach Anspruch 1 bis 11, dadurch gekennzeichnet , daß man zur Herstellung einer Verbindung der Formel I, in der R eine Benzoylgruppe ist, eine Verbindung der Formel I, in der R ein Wasserstoffatom ist, mit einem Benzoylhalogenid umsetzt.
- 13. Verfahren nach Anspruch 1 bis 12, dadurch gekennzeichnet , daß bei der Verbindung der Formel I die Alkyl-, Alkenyl- und Alkylengruppen bis zu 6 Kohlenstoffatomen, die Aryl-, Aralkyl- oder Alkarylgruppen bis zu 10 Kohlenstoffatomen enthalten, die Halogenatome Chlor- oder Fluoratome und die Heteroatome Sauerstoff- und/oder Stickstoffatome sind.
- 14. Verfahren nach Anspruch 13, dadurch gekennzeichnet , daß bei den Verbindungen der Formel I Z ein Fluor- oder Chloratom, Y ein Wasserstoff-, Fluor- oder Chloratom und W eine Alkylgruppe mit bis zu 6 Kohlenstoffatomenist.709820/1013
