Description
Die Erfindung betrifft das im Patentanspruch angegebene Verfahren. Nachstehend werden L-Tryptophan und
5-Hydroxy-L-tryptophan als Tryptophan und Hydroxytryptophan bezeichnet.
Tryptophan ist als Zusatz zu Futtermitteln und Nahrungsmitteln geeignet und sowohl Tryptophan als auch
Hydroxytryptophan werden als Arzneimittel verwendet.
Es ist bekannt, daß Tryptophan oder Hydroxytryptophar. aus Indol oder 5-Hydroxylndol und einer a-Amlnosäure, wie Serin oder Cystein unter der Wirkung eines
Enzyms gebildet wird, das durch einen Mikroorganismus der Genera Proteus, Erwlnia, Escherichla. Pseudomonas
und Aerobacter gebildet wird (Japnalsche Auslegeschrift 46348/1972).
Es wurde nun gefunden, daß das gleiche Enzym die Bildung von Tryptophan aus Indol und /J-Halogenalanin
und jle Bildung von Hydroxytryptophan aus 5-Hydroxyindol und /i-Halogenalanin katalysiert.
Bei dem erfindungsgemäßen Verfahren Ist die Ausbeute an Tryptophan oder Hydroxytryptophan höher als
bei bekannten Verfahren und darüber hinaus ist die Reaktionsrate im Vergleich mit bekannten Verfahren
sehr hoch.
Die zur Bildung dieses Enzyms befähigten Mikroorganismen gehören dem Genus Proteus, Ervlnla, Escherichla, Pseudomonas oder Aerobacter an. Die Mikroorganismen werden In völlig üblichen Medien gezüchtet. Die
Medien enthalten eine Kohlenstoffquelle, Stickstoffquelle, anorganische Ionen und wünschenswert organische Spurennährstoffe. Die Zugabe einer geringen Menge
an Tryptophan oder 5-Hydroxytryptophan fördert die Ausbildung der enzymatlschen Aktivität.
Zu geeigneten Kohlenstoffquellen gehören Kohlenhydrate oder Zuckeralkohole, wie Glucose, Fructose,
Saccharose. Mannose, Maltose, Mannit, Xylose, Galactose. Stärke, Melassen, Sorbit oder Glycerin, organische
Säuren, wie Essigsäure, Zitronensäure, Milchsäure, Fumarsäure oder Maleinsäure und Alkohole, wie Methanol, Äthanol, Propanol oder Butanol. Die verwendete
Stickstoffquelle ist eine übliche Stickstoffquelle, wie
Ammoniumchlorid, Ammoniumsulfat, Ammoniumphosphat, Ammoniumnitrat, Ammoniumcarbonat,
Ammoniumacetat, Harnstoff, Ammoniakwasser oder gasförmiges Ammoniak. Manchmal fördern organische
Spurennährstoffe das Mikrobenwachstum und die Enzymproduktion. Zu diesen organischen Spurennährstoffen gehören Vitamine. Aminosäuren und Rohmaterialien, welche diese organischen Spurennährstoffe enthalten, wie Malsquellwasser, Rindflelchextrakt, Peptone,
Kaseln-Hydrolysat, Sojaproteln-Hydrolysat, Malzextrakt
oder Hefeextrakt. Es werden übliche anorganische Ionen
verwendet, wie Kalium, Natrium, Calcium, Magnesium, Ferrolon, Mangan, Kupfer, Phosphat, Sulfat.
Es ist wünschenswert, daß 0,01 bis 0,5 g/dl Tryptophan oder Hydroxytryptophan dem Medium zugesetzt
werden, um die enzymatische Aktivität zu erhöhen.
Wenn die Mikroorganismen in dem vorstehend angegebenen Medium zwei bis sieben Tage bei pH 4 bis 9 und
bei 27 bis 40° C gezüchtet werden, reichert sich die Aktivität des zur Bildung von Tryptophan und Hydroxytryptophan befähigten Enzyms in den Zellen oder in der
Brühe an.
Die Zellen oder die Brühe werden als solche als Enzym verwendet. Gefriergetrocknete Zellen, Zellhomogenlsat,
ultraschallbehandelte Zellen, Zellautolysate, mit Aceton behandelte Zellen und Enzympräparate warden ebenfalls
als Enzym eingesetzt. In dieser Beschreibung und den Patentansprüchen werden also unter der Bezeichnung
»Zellen« auch Zellhomogenlsate und von ZeKa; abgeleitete Materialien, die die Tryptophanase enthalten, verstanden.
Isolierungs- und Reinigungsverfahren für das Enzym und die enzymologlschen Eigenschaften des Enzyms
wurden von H. Yoshlda el al in FEBS-letters 48(1) 56
(!974) beschrieben.
Das Enzym wird als Tryptophanase bezelchent werden, da es den Abbau von Tryptophan In Indol, Pyruvat und
Ammoniak katalysiert.
Das Reaktionsgemisch enthält das Enzym oder die Ausgangsmaterialien für das Enzym, wie sie vorstehend
gezeigt wurden, /7-Chloralanin und Indol oder 5-Hydroxyindol. Wenn gereinigtes Apoenzym verwendet
wird. Ist Pyridoxal-5-phosphat als Coenzym erforderlich.
Es ist wünschenswert, dem Reaktionsgemisch ein Reduktionsmittel, wie ein Thiosulfat, zuzusetzen, um die
Ausbeute an Tryptophan oder Hydroxytryptophan zu vergrößern.
Das Raktionsgemisch wird bei 5 bis 60° C und einem
pH-Wert von 5 bis 10 gehalten, bis Indol oder 5-Hydroxylndol In Tryptophan oder Hydroxytryptophan übergeführt Ist.
Das in dem Reaktionsgemisch gebildete Tryptophan oder Hydroxytryptophan kann in üblicher Welse Isoliert
und gereinigt werden, wie mit Hilfe von Ionenaustauschchromatographle oder Ausfällung am isoelektrischen
Punkt.
Die in den nachstehenden Beispielen erhaltenen Produkte Tryptophan und Hydroxytryptophan wurden durch
Prüfung Im Gemisch mit authentischen Proben von L-Tryptophan und 5-Hydroxy-L-tryplophan identifiziert.
Proteus morganli IFO 3848 wurde in 60 ml-Antellen
des nachstehend angegebenen Mediums, das in 500 ml-Kolben gehalten wurde, unter Schütteln 20 Stunden bei
30° C gezüchtet.

Die Zellen wurden durch Zentrifugieren aus 1 1 der so
erhaltenen kombinierten Kulturbrühe gewonnen und zu 11 0,l%lger NHXI-NHiOH-Pufferlösung (pH 8,0) gegeben,
die 20 g Indol und 20 g /J-Chlor-L-alanin enthielt.
Das Reaktionsgemisch wurde 20 Stunden unter Rühren bei 37° C gehalten. Dabei wurden 24 g L-Tryptophan
gebildet (bestimmt mit der mikrobiologischen Bestimmungsmethode).
Nach dem Auflösen von ausgefälltem Tryptophan mit NaOH wurde das Reaktionsgemisch zur Entfernung der
Zellen filtriert, der pH-Wert wurde mit HCl auf 4 eingestellt und das Reaktionsgemisch wurde durch eine mit
Aktivkohle gefüllte Kolonne geleitet. L-Tryptophan wurde mit 2n NH1OH eluiert, das Eluat wurde konzentriert
und es wurden 20,2 g L-Tryptophan-Kristalle erhalten.
Zellen von Enterobacter cloacae ATCC 7256 oder Pseudomonas perlurlda ATCC 490, die in der vorstehend
beschriebenen Welse hergestellt wurden, wurden anstelle der Zellen von P/oieus morganli IFO 3848 verwendet.
Sei Verwendung von ATCC 7256 wurden 9,0 g L-Tryptophan
gebildet und bei Verwendung von ATCC 490 wurden 19,1 g L-Tryptophan gebildet.
In einem Vergleichsversuch wurde /J-Chlor-L-alanln
durch 20 g L-Serin ersetzt. In diesem Fall wurden durch die Zellen von Proteus morganli IFO 3848, Enterobacter
cloacae ATCC 7256 bzw. Pseudomonas perlurida ATCC 490 22,3 g 8,0 g bzw. 18,4 g L-Tryptophan gebildet.
Erwinia herblcola ATCC 21433 wurde in gleicher
Welse wie In Beispiel 1 gezüchtet.
Jede so erhaltene Kulturbrühe wuröe mit 1 g /J-Chlor-L-alanin/dl
und 1 g Indol/dl verwtzt und 44 Stunden
unter Rühren bei 370C gehalten. In der Kulturbrühe
wurde 0,70 g Tryptophan/dl gebildet.
Wenn /?-Chlor-L-alanln durch L-Serln In einer Menge
von Ig/dl ersetzt wurde, bildete ATCC 21433 0,62g
Tryptophan/dl.
Zu dem nachstehend angegebenen Grundmedium wurde 0,2 g Tryptophan/dl (Medium A) oder 0,2 g 5-Hydroxy-L-tryptophan/dl
(Medium B) gegeben; 40 ml-Anteile des Mediums wurden In 500 ml-Kolben gegeben
und jeder Anteil wurde mit Proteus morganli IFO 3848 angeimpft. Danach wurden die Kolben während 20 Stunden
unter Schütteln bei 31° C gehalten.

Der so erhaltenen Kulturbrühe wurden 2 g ß-CMor-L-alanin/dl
und 2 g Indol/dl sowie 0,2 g Kallumthlosulfat/dl zugesetzt und das Gemisch wurde 40 Stunden
unter Schütteln bei 17° C gehalten.
In der Kulturbrühe aus Medium A wurden 2,4 g L-Tryptophan/dl
festgestellt und 1,8 g Tryptophan/dl wurden In der Kulturbrühe aus Medium B aufgefunden.
Beispiel 4
Proteus morganli IFO 3848 wurde In 100 ml des nachstehend angegebenen Mediums 20 Stunden bei 31,5° C gezüchtet. Die kombinierte Kulturbrühe (2 I) wurde zentrifugiert und die so erhaltenen Zellen wurden in 0,05 m PhosphatpulTer (pH 6,0) suspendiert und mit Hilfe eines Ultraschalloszillators homogenisiert. Die überstehende Flüssigkeit des mit Ultraschall erhaltenen Homogenisats wurde mit Ammoniumsulfat versetzt und die ausgefällten Proteine wurden dialyslert und mit Protamtn behandelt, mit DEAE-Sephadex und danach mit Sephadex G-150 fraktioniert.
Proteus morganli IFO 3848 wurde In 100 ml des nachstehend angegebenen Mediums 20 Stunden bei 31,5° C gezüchtet. Die kombinierte Kulturbrühe (2 I) wurde zentrifugiert und die so erhaltenen Zellen wurden in 0,05 m PhosphatpulTer (pH 6,0) suspendiert und mit Hilfe eines Ultraschalloszillators homogenisiert. Die überstehende Flüssigkeit des mit Ultraschall erhaltenen Homogenisats wurde mit Ammoniumsulfat versetzt und die ausgefällten Proteine wurden dialyslert und mit Protamtn behandelt, mit DEAE-Sephadex und danach mit Sephadex G-150 fraktioniert.

40 mg des so erhaltenen Enzymproteins (40 mg) wurden dem vorstehend angegebenen Reaktionsgemisch
zugesetzt und das Gemisch wurde 20 Stunden bei 37° C gehalten.

In dem erhaltenen Reaktionsgemisch wurden pro 100 ml 2 g Tryptophan aufgefunden.
Wenn anstelle von /5-ChIor-L-alanlr. 2,0 g Serln/dl verwendet
wurden, wurden 1,85 g L-TrypiOyhnn/dl angerelchert.
In dem Medium gemäß Beispiel 1 wurde Tryptophan durch 2 g 5-Hydroxy-L-tryptophan ersetzt und Proteus
morganii IFO 3848 wurde in gleicher Weise wie In Beispiel
1 in dem Medium gezüchtet.
Die Zellen wurden durch Zentrifugleren gewonnen
und in 1 I O,l%lger NH4CI-NH4OH-Pufferlösung (pH 8,0)
suspendiert, die 20 g 5-Hydroxyindol und 20 g /J-Chlorvi
L-alanln enthielt. Das Gemisch wurdo 20 Stunden unter
Rühren bei 37° C gehalten. Das Reaktionsgemisch enthielt
14,1 g 5-Hydroxy-L-Tryptophan.
In gleicher Welse wie in Beispiel 1 wurden aus dem Reaktionsgemisch 7,5 g kristallines /?-Hydroxy-L-3
> tryptophan gewonnen.
Wenn /7-Chlor-L-alanin durch 20 g L-Serln ersetzt
wurde, wurden In dem Reaktionsgemisch 13,6 g 5-Hydroxy-L-tryptophan
aufgefunden.
Eine Kulturbrühe von Proteus morganli IFO 3848 wurde In gleicher Welse wie In Beispiel 1 hergestellt. Zu
100 ml der Kulturbrühe wurden Ig 5-Hydroxylndol,
0,2 g Na2SO1, 0,5 g NH4CI und Ig /3-Chlor-L-alanin
b5 gegeben, der pH-Wert wurde mit Ammoniakwasser auf
8,0 eingestellt und das Gemisch wurde 20 Stunden bei Raumtemperatur gehalten. Die Kulturbrühe enthielt
dann 0,88 g 5-Hydroxy-L-tryptophan/dl.

In gleicher Weise wie in Beispiel 5 wurde Proteus morgani!
durch Erwinia herbicola ATCC 21433 oder Enterobacter cloacae ATCC 7256 ersetzt. ATCC 21433 bildete
0,71 g 5-Hydroxy-L-tryptophan/dt während ATCC 7256 0,41 g 5-Hydroxy-L-tryptophaii/dl bildete.

Enzymprotein von Proteus morganii IFO 3848 wurde in gleicher Weise wie in Beispiel 4 gebildet und 60 mg
des Enzymproteins wurden zu dem nachstehend angegebenen Reaktionsgemisch zugemischt.

Reaktionsgemisch:

Das Reaktionsgemisch wurde 20 Stunden bei 37° C gehalten. Danach enthielt das Reaktionsgemisch 0,87 g
5-Hydroxy-L-tryptophan/dI.
Zellen von Proteus morganii IFO 3848, die in gleicher
Weise wie in Beispiel 1 gebildet woFden warer., wurden
in 1 I O,l%lger NH4C1-NH4OH-Pufferlösung (ph 8,0) suspendiert,
die 20 g Indol und 20 g /J-Brom-L-alanin enthielt.
Der Pffer wurde 20 Stunden unter Rühren bei 37C C gehalten und enthielt dann 19,5 g 5-Hydroxy-L-tryptophan.

Das in Beispiel 5 verwendete ß-Chlor-L-alanin wurde
durch /?-Brom-L-alanin ersetzt und 5,7 g 5-Hydroxy-L-tryptophan
wurden in gleicher Weise wie In Beispiel 5 hergestellt.

Claims (1)
- Patentanspruch:Verfahren zur Herstellung von L-Tryptophan oder 5-Hydroxy-L-tryptophan durch Umsetzung eines Indol oder 5-HydroxyIndoI und eine Aminosäure enthaltenden wäßrigen Gemisches in Gegenwart von Tryptophanase und Gewinnung des erhaltenen Produkts aus dem wäßrigen Gemisch, dadurch gekennzeichnet, daß man ein Tryptophanase, Indol oder 5-Hydroxyindol sowie /i-Halogenalanin enthaltendes wäßriges Gemisch bei 5 bis 60° C hält, bis die Bildung von L-Tryptophan oder 5-Hydroxy-L-tryptophan erfolgt ist.
