Abstract
Die Erfindung betrifft ein Verfahren zur Herstellung von Dihydropyridinderivaten, die sich zur Behandlung von Stauungsinsuffizienz des Herzens, Hochdruck oder Angina eignen. Dabei handelt es sich um Verbindungen der Formel, oder von pharmazeutisch vertraeglichen Saeureadditionssalzen davon. Das erfindungsgemaesse Verfahren zeichnet sich dadurch aus, dass man eine Verbindung der Formel I mit einer Verbindung der Formel II und einer Verbindung der Formel III umsetzt. Formeln I, II und III

Die Anwendung der vorliegenden Erfindung erfolgt im Bereich der Arzneimittel, insbesondere auf dem Gebiet der blutdrucksenkenden Arzneimittel.

Es sind bestimmte 4-Phenyl-1,4-dihydropyridinderivate bekannt; vgl. beispielsweise US-A-3 485 847 und 4 044 141.
Ziel der Erfindung
Ziel der Erfindung ist es, ein Verfahren zur Herstellung von Vorläufern von neuen Dihydropyridinderivaten mit wertvollen blutdrucksenkenden Eigenschaften bereitzustellen.

Darlegung des Wesens der Erfindung
. Der Erfindung liegt die Aufgabe zugrunde, ein Verfahren zur Herstellung von Vorläufern von Dihydropyridinderivaten bereitzustellen, die wertvolle blutdrucksenkende Eigenschaften besitzen und sich zur Behandlung von Hochdruck, Stauungsinsuffizienz des Herzens, Angina oder anderen kardiovaskulären Erkrankungen, die einer Behandlung mit Calciumblockern zugänglich sind, wie Migräne oder vasospastische Störungen, eignen.

In der Beschreibung und im Erfindungsanspruch haben, sofern nichts anderes angegeben ist, die einzelnen Ausdrücke die folgenden Bedeutungen:
"^ Der Ausdruck "nieder-Alkyl" bedeutet geradkettige oder verzweigte, einwertige Substituenten, die nur aus Kohlenstoff und Wasserstoff bestehen, keine Mehrfachbindungen aufweisen und 1 bis 4 Kohlenstoffatome besitzen. Beispiele für niedere Alkylreste sind Methyl, Äthyl, n-Propyl, Isopropyl, η-Butyl, Isobutyl, sec.-Butyl und tert.-Butyl.
Der Ausdruck "Halogen" bedeutet Chlor, Brom und Jod.
Der Ausdruck "Schutzgruppe" bedeutet eine Gruppe, die eine vorzeitige Umsetzung von Hydroxyl- oder Carbonylgruppen während der Herstellung der Verbindungen der Erfindung verhindern soll. Beispiele für Schutzgruppen sind Benzyläther, Ketale (einschliesslich acyclische Ketale und cyclische Ketale), wie Acetonide) und Ester. Schutzgruppen lassen sich idealerweise leicht anbringen und unter Bedingungen, die von den bei den Präparationen angewandten Reaktionsbedingungen abweichen, entfernen. Beispielsweise sind Benzyläther unter sauren und basischen Bedingungen, · die bei der Herstellung der. Verbindungen der Formel 1 (siehe unten) angewandt werden, stabil, lassen sich jedoch leicht unter Verwendung von H_ über Pd-C entfernen. Verbindungen mit Mehrfachhydroxygruppen in 1,2- oder 1,3-

Stellung lassen sich durch Kondensation mit einem Keton, z.B. Aceton, unter Bildung von cyclischen Ketalpolyäthern ,schützen. Beispielsweise lassen sich die 1,2-Hydroxygruppen von 1-(2,3-Dihydroxyprop-1-yloxy)-4-(2-hydroxyäthyl)-benzol durch Kondensation mit Aceton unter Bildung von 1-_/""(2,2-Dimethyl-1 ,3-dioxolan-4-yl)-methoxy_7-4-(2-hydroxyäthyl)-benzol schützen, wodurch man eine Umsetzung an der Hydroxyäthylgruppe ohne Beteiligung der Dihydroxypropoxygruppe ermöglicht.
Der Ausdruck "pharmazeutisch verträgliche Säureadditionssalze" bezieht sich auf Salze der vorliegenden Verbindungen, die die gewünschte pharmakologische Aktivität besitzen und die -weder in biologischer Hinsicht noch in anderer Hinsicht unerwünscht sind. Diese Salze werden mit anorganischen Säuren, wie Salzsäure, Bromwasserstoffsäure, Schwefelsäure, Salpetersäure oder Phosphorsäure, oder mit organischen Säuren, wie Essigsäure, Propionsäure, Glykolsäure, Brenztraubensäure, Malonsäure, Bernsteinsäure, Äpfelsäure, Maleinsäure, Fumarsäure, Weinsäure, Citronensäure, Benzoesäure, Zimtsäure, Mandelsäure, Methansulfonsäure, Äthansulfonsäure, p-Toluolsulfonsäure und dergl., gebildet.
Bei der hier verwendeten Nomenklatur handelt es sich im wesentlichen um eine modifizierte Form der IUPAC-Nomen-. klatur. Die erfindungsgemäss hergestellten Verbindungen werden als Derivate von 1,4-Dihydropyridin bezeichnet. In sämtlichen Strukturformeln werden die Positionen der
Verbindungen unter Beginn beim Pyridinstickstoff ,in Uhrzeigerrichtung numeriert. Beispielsweise wird die folgende Verbindung als 2 , 6-Dimethyl-3-methoxycarbonyl-4-(3-nitrophenyl)-5-(2-/~4-(2,3-dihydroxypropoxy)-phenyl_7-äthoxycarbonyl)-1,4-dihydropyridin bezeichnet:

Die Verbindungen der Formel I1 besitzen ein chirales Zentrum am C4 des Dihydropyridinrings und können somit in Form von optischen Isomeren vorliegen. Bestimmte Verbindungen, .bei denen R1 oder Rp asymmetrisch ist, liegen somit als Diastereomere vor. Erfindungsgemäss kommen beliebige Isomere oder Gemische von Isomeren in Frage. Die Isomeren lassen sich nach verschiedenen Methoden, beispielsweise durch selektive Kristallisation oder Säulenchromatographie, auftrennen; vgl. T. Shibanuma et al., Chem. Pharm. Bull.., Bd. 28 (1980), S. 2809 bis 2812. Eine andere Möglichkeit besteht darin, die Verbindungen der Erfindung unter Verwendung von optisch aktiven Reaktanten oder durch eine Kombination von Auftrennung und chiraler Synthese herzustellen. Die Erfindung umfasst die Herstellung sämtlicher optischer Isomeren und beliebiger asymmetrischer Verbindungen der Formel 6 sowie Gemische davon. Optische Isomere der Verbindungen werden mit (+) oder (-) bezeichnet, was die Richtung angibt, in der das chirale Zentrum eine Ebene von polarisiertem Licht dreht. Verbindungen der Erfindung mit mehrfachen chiralen Zentren werden durch Kondensation von optisch aktiven Zwischenprodukten hergestellt und durch Angabe des Drehsymbols der optisch aktiven Zwischenprodukte und durch Angabe der Dihydropyridine Isomeren, aus der die Verbindung hergestellt wird, gekennzeichnet.
Es ist festzuhalten, daß die optische Darstellung des Endprodukts nicht die gleiche Ri'chtung oder Grössenordnung wie die optische Drehung von einem der Zwischenprodukte aufweisen muss.

Diese Arzneimittel' enthalten eine wirksame Menge einer Verbindung der Formel 1 oder.ein pharmazeutisch verträgliches Säureadditionssalz davon und einen pharmazeutisch geeigneten Trägerstoff. ;
Zur Therapie oder Prophylaxe der vorstehend genannten Erkrankungen wird eine wirksame Menge einer Verbindung der Formel 1 verabreicht.
Somit lassen sich die erfindungsgemäss hergestellten Ver-
bindungen der Formel 6 indirekt zur Herstellung von Arzneimitteln zur Behandlung der vorstehend genannten Krankheiten verwenden.

Im vorstehenden Schema haben R, , R.., R., R1- und Y die vorstehend definierten breitest möglichen Bedeutungen; zusätzlich bedeutet * eine Schutzgruppe.
5 .Verbindungen, in denen Y die Bedeutungen -0-, -NH-, -NR2-, -S- oder -S(O)1 5- hat und R1 die Bedeutung A1 oder A3 hat, werden gemäss Schema 1 hergestellt.
Ein zur Bildung des gewünschten R1 geeignetes Alkanol wird ausgewählt. Das Alkanol kann zwei oder mehr -OH-Gruppen aufweisen. Für Verbindungen, in denen R1 die Bedeutung A hat, stellen Zucker zweckmässige Quellen für optisch reine Alkanol-e dar* Die entsprechenden -OH-Gruppen werden mit "üblichen Mitteln geschützt, insbesondere durch BiI-dung von Acetoniden und Benzyläthern. In den Schemata werden geschützte R1 - und Y-Reste mit Sternen gekennzeichnet, z.B. R.*, Y*. Ist ein spezielles optisches Isomer erwünscht, so kann der entsprechende geschützte chirale Alkohol verwendet werden. Beispielsweise sind (+)-Solketale und (-)-Solketale im Handel erhältlich und eignen sich zur Herstellung von Verbindungen, in denen -YR, die Bedeutungen (+)-OCH2CH(OH)CH2OH oder (-)-OCH2CH(OH)CH2OH hat (Stufe 1) .
Das geschützte Alkanol wird sodann mit Tosylchlorid oder einem ähnlichen elektrophilen Reagens unter basischen Bedingungen unter Bildung eines Tosylats oder eines ähnlichen Moleküls mit einer geeigneten austretenden Gruppe umgesetzt (Stufe 2).·
Das Tosylat wird mit einem geeigneten Methyl- 0) -phenylalkylcarboxylat-derivat (2) unter basischer Katalyse, z.B. unter Verwendung von Natriumhydrid in Dimethylformamid (DMF), unter Bildung eines alkylierten CO -Phenylalkylcarboxylat-derivats (3) umgesetzt. Verbindungen der Formel 2, in der Y die Bedeutung -0-, -NH- oder -S- hat, sind bekannt und im Handel erhältlich, z.B. von der Aldrich Chemical Co.. Beispielsweise wird Methyl-(4-hydroxyphenyl)-

acetat verwendet, wenn Verbindungen der Formel 1 mit η = 2 und y = para-OR.. erwünscht sind (Stufe 3).
Di"e Carboxylatverbindung 3 wird sodann beispielsweise mit Lithiumaluminiumhydrid (LAH) zu einem Hydroxyalkylbenzolderivat (4), z.B. zu 1-(2-Hydroxyäthyl)-4-(2-benzyloxyäthoxy)-benzol, reduziert (Stufe 4).
Diketen (3-Buteno-ß-lacton, das beispielsweise von der IQ Aldrich Chemical Co. erhältlich ist) wird sodann unter basischen Bedingungen, z.B. unter Verwendung von Triäthylamin in Dimethoxyäthan (DME) unter Bildung eines 6J-Phenylalkylacetoacetat-derivats (5) zum Hydroxyalkylbenzol-derivat 4 gegeben. Beispielsweise wird 1-(2-Hydroxyäthyl)-4- ^g (2-benzyloxyäthoxy)-benzol mit Diketon unter Bildung von 2-J_ 4-(2-Benzyloxyäthoxy)-phenylT-äthylacetoacetat (5) umgesetzt (Stufe 5).
Das CiZ-Phenylalkylacetoacetat-derivat (5) wird sodann mit 2Q einem Alkyl-ß-aminocrotonat (oder Methoxyäthyl-ß-aminocrotonat in Abhängigkeit vom gewünschten Rn.) und einem Benzaldehyd-derivat (in Abhängigkeit vom gewünschten R1.; beispielsweise wird für Verbindungen, in denen R1. meta-NO„ bedeutet, 3-Nitrobenzaldehyd verwendet) unter den Bedinge gungen der Hantzsch-Dihydropyridinsynthese (vgl. beispiels,-weise Fox et al., J. Org. Chem., Bd. 16 (1951), S. 1259) unter Bildung eines 4-Phenyl-2,5-Dimethyl-1,4-dihydropyridindicarboxylat-diesters (β) umgesetzt. sind auch ohne Entfernung der Schutzgruppen aktiv.Wenn die Hydroxyreste durch Ketalbildung geschützt sind, kann eine Entfernung der Schutzgruppen unter Verwendung einer verdünnten starken Säure erreicht werden. Beispielsweise können Acetonide durch Behandlung mit·5-prozentiger HCl in einem protischen Lösungsmittel, vorzugsweise Wasser, im Temperaturbereich von 0 bis 1200C, vorzugsweise etwa 100°C, entfernt werden. Bei Verwendung von Benzyläthern kann die Entfernung der Schutzgruppen durch katalytische Reduktion, z.B. unter Verwendung von Hp und Pd/C bei Drücken von 0,7 bis 14 bar (10 bis 200 ps'i) erreicht werden. Beispielsweise kann 2,6-Dimethyl-3-methoxycarbonyl-4-(3-nitrophenyl)-5-(2-/T-(2-benzyloxyäthoxy)-phenylT-äthoxycarbonyl)-1 ,4-dihydr0Pyr3-di.n'mit H_ über 10-prozentigem Pd/C bei 3,5 bar (50 psi) unter Bildung von 2 ,6-Dimethyl-3-methoxyc ar bon yl-4-(3-nitrophenyl)-5-(2-_/_~~4-(2-hydroxyäthoxy)-pheny_l7-äthoxycarbonyl)-1 ,4-dihydropyridin (1) reduziert werden (Stufe 7).
Sind Verbindungen, in denen Y -NBp- bedeutet, erwünscht, kann eine Verbindung der Formel '6 (Y = -NH-), die beispielsweise gemäss Schema 1 erhalten worden ist, direkt z.B. mit 2-Bromäthanol -unter Bildung einer Dialkylaminoverbindung der Formel 6 alkyliert werden.

Präparation 1 (Herstellung von Ausgangsmaterialien-; HO-R

(A) Bei Verbindungen, in denen R1 die Bedeutung A1 mit Ii
m = 1 und ρ = 1 aufweist, ist die Verwendung von Solketal (2, 2-Dimethyl-4-hydroxymethyl.-1 , 3-dioxolan) , einem Handelsprodukt (z.B. Aldrich Chemical Co.), zweckmässig.
Solketal lässt sich auch gemäss folgendem Verfahren herstellen:
18 g (0,2 Mol) 1,2,3-Glycerin werden zu 20 ml Aceton, 40 ml Benzol und 1 ml HpSO^ gegeben und 2 Stunden unter Verwendung einer Dean-Stark-Falle unter Rückfluss erwärmt. 1^ Das erhaltene Solketal wird zu verdünnter wässriger NaOH gegeben, mit Äther extrahiert und durch Chromatogrpahie an Kieselgel gereinigt.
(B) In entsprechender Weise erhält man gemäss Abschnitt A unter Verwendung von 1,2,4-Butatriol (m = 2, ρ = 1), 1,2,5-Pentatriol (m = 3, ρ = 1) oder 1,2,3,4-Butatetrol (m = 1 , ρ = 2) anstelle von 1 ,2,3-Propyltriol die nachstehend aufgeführten Verbindungen:
2,2-Dimethyl-4-(2-hydroxyäthyl)-r,3-dioxolan; 2,2-Dimethyl-4-(3-hydroxypropyl)-1,3-dioxolan; 2,2-Dimethyl-4,5-di-(hydroxymethyl)-1,3-dioxolan.
(C) Eine andere Möglichkeit zum Schutz der OH-Gruppen
besteht in der Verwendung von Benzyläthern..Beispielsweise 30
werden 8 g (0,2 Mol) 2,2-Dimethyl-4,5-di-(hydroxymethyl)-1,3-dioxolan unter Rückfluss mit 25 g(0,2 Mol) Benzylchlorid und 25 g (0,2 Mol) KpC0~ in 100 ml" Äthanol unter Rückfluss erwärmt. Das Produkt wird zu wässriger NaOH gegeben,
mit Äther extrahiert und durch Chromatographie an Kiesel- ' 35

(E) Chirale Zwischenprodukte der Formel HO-R1* sind im Handel erhältlich oder lassen sich aus handelsüblichen chiralen Aldosen herstellen. Beispielsweise werden 18 g (0,2 Mol) (R)-Glycerinaldehyd (^0 = + 8,7°) zu 20 ml Aceton, 40 ml Benzol und 1 ml HpSO^. gegeben und 2 Stunden unter Verwendung einer Dean-Stark-Falle unter Rückfluss erwärmt. Der erhaltene (R)-2,3-Isopropylidenylglycerinaldehyd wird zu verdünnter wässriger NaOH gegeben, mit Äther extrahiert und getrocknet. Der Glycerinaldehyd wird sodann unter Verwendung von NaBH1,. in Äthanol unter Bildung von (S)-2,2-Dimethyl-4-(2)-hydroxymethyl)-1,3-dioxolan (°<· r> = + 11,5 ) reduziert.
(F) In entsprechender Weise erhält man gemäss Abschnitt
(E) unter Verwendung von (S)-Glycerinaldehyd oder (S)-3,4-Dihydroxybutanal anstelle von (R)-Glycerinaldehyd die folgenden Verbindungen:

(Herstellung von R^*-Tosylaten)
(A) 26 g (0,2 Mol) 2,2-Dimethyl.-4-hydroxymethyl-1 ,3-dioxolan werden zu 38 g (0,2 Mol) p-Toluolsulfonylchlorid in 200 ml Pyridin gegeben und 24 Stunden bei O0C stehengelassen. Sodann wird Wasser zugesetzt und das Gemisch mit • Essigsäureäthylester extrahiert. Der Essigsäureäthylesterextrakt wird mit Wasser und Kochsalzlösung gewaschen, über

(A) 1|8 g Natriumhydrid (50-prozentige Dispersion in Mineralöl, 0,1 Mol) werden zu einer Lösung von 16,6 g (0,1 Mol) Methyl-4-hydroxyphenylacetat (2) (Handelsprodukt, z.B. der Aldrich Chemical Co.) in 150 ml Dimethylformamid gegeben. Das Gemisch wird 15 Minuten gerührt. 28,6 g (0,1 Mol) SoI-

Das als Produkt gebildete Methyl-4-(2,2-dimethyl-1,3-dioxolan-4-yl)-methylsulfinylphenylacetat (3, Y = -S(O)-) wird durch Chromatographie an Kieselgel gereinigt.

dimethyl-1,3-dioxolan-4-yl)-methylsulfonylacetat (3, Y = -S(O)p-) wird durch Chromatographie an Kieselgel gereinigt,

(A) Eine Lösung von 27 g (o,096 Mol) Methyl-4-(2,2-di-
methyl-1,3-dioxolan-4-yl)-methoxyphenylacetat (3) in 200 ml Tetrahydrofuran (THF) wird langsam zu einer gerührten Suspension von 4,0 g Lithiumaluminiumhydrid in 300 ml Tetrahydrofuran gegeben. Das Gemisch wird 2 Stunden bei Raumtemperatur gerührt und sodann nacheinander mit 4 ml Wasser, 30
ml 15-prozentiger NaOH-Lösung und 12 ml Wasser behandelt.
Anschliessend wird das Gemisch filtriert. Das Filtrat wird zur Trockne eingedampft. Der Rückstand wird mit Hexan verrieben. Man erhält 24 g 4-(2,2-Dimethyl-1,3-dioxolan-4-yl)-methoxyphenyläthan-2-ol (4) in Form eines gelbbraunen Feststoffs.

(Verbindungen der Formel 4, in der Y.eine Bindung bedeutet: Schema 2)
(A) Eine Lösung von 1,0 g 4-(2-Bromäthyl)-benzoesäure
(Handelsprodukt, z.B. von Trans World Chemicals, Inc., oder nach bekannten Verfahren hergestellt) und 0,055 g Natriumborhydrid in 15 ml Äthanol wird 4 Stunden gerührt. Die Lösung wird zu 20 ml Wasser gegeben, filtriert und destil-1^ liert. Man erhält 1-(2-Bromäthyl)-4-hydroxymethylbenzol.
0,5 g 1-(2-Bromäthyl)-4-hydroxymethylbenzol werden sodann zu einer Lösung von 0,44 g Methylbenzolsulfonat und 0,32 g KpCO-. in 10 ml Methanol gegeben und 4 Stunden umgesetzt. Die Lösung wird sodann mit methanolischer HCl neutralisiert und mit Äther extrahiert. Der Ätherextrakt wird über Na2SO1. getrocknet. Nach Filtration und Entfernung des Lösungsmittels unter vermindertem Druck erhält man 1-(Methoxymethyl)-4-(2-bromäthyl)-benzol (13).
0,32 g Diäthylmalonat werden zu einer Lösung von 0,05 g Natrium in 10 ml Methanol gegeben. Anschliessend werden 0,5 g 1-(Methoxymethyl)-4-(2-bromäthyl)-benzol (13) zugesetzt und 8 Stunden umgesetzt. Die Lösung wird mit metha-
nolischer HCl neutralisiert und mit Äther extrahiert. Der Ätherextrakt wird über Na2SO1. getrocknet und eingedampft. Man erhält 1-_/ 3 , 3-Di- (äthoxycarbonyl )-propy_l7"-4-methoxymethylbenzol (14). Die Methylgruppe wird durch Behandlungmit wässriger HBr in Essigsäure entfernt. Anschliessend
u wird mit wässriger NaOH neutralisiert und mit Äther extrahiert. Der Ätherextrakt wird über Na2SO1, getrocknet und eingedampft. Man erhält 1-_/~3 , 3-Di- (äthoxycarbonyl )-propyl_7-4-hydroxymethylbenzol (4)..

Diäthyl-(3-äthoxycarbonyl)-malonat lässt sich folgendermassen herstellen:
23,9 g Diäthylbrommalonat (Handelsprodukt, z.B. von der Aldrich Chemical Co.) werden mit 2,4 g trockenen Magnesiumspänen in 300 ml trockenem Äther unter Stickstoff bei -10 C umgesetzt. Sodann wird trockenes CO, durch die Lösung geperlt, bis keine Absorption mehr erfolgt. Die Lösung.wird sodann mit 25-prozentiger wässriger HpSG1, neutralisiert, mit Äther extrahiert und über Na2SO1. getrocknet. Der Äther wird unter vermindertem Druck entfernt und der Rückstand mit 1 ml HpSCL und 100 ml Äthanol verestert. Die Lösung wird neutralisiert, mit Äther extrahiert. Nach Trocknen über Na2SO.. und Entfernung des Lösungsmittels unter vermindertem Druck erhält man Diäthyl-~ (3-äthoxycarbonyl)-malonat.

ρ O ist, gemäss folgendem Verfahren herzustellen: Eine. Lösung von 29,6 g (0,096 Mol) 1 -_/~3,3-Di-(äthoxycarbonyl)-propy_l7-1 -methoxymethylbenzol (14, ρ ·= 0) in 200 ml Tetrahydrofuran wird langsam zu einer gerührten
Suspension von 4,0 g-Lithiumaluminiumhydrid in 300 ml ο
Tetrahydrofuran gegeben·.' Das Gemisch wird 2 Stunden bei Raumtemperatur gerührt und sodann nacheinander mit 4 ml Wasser, 4 ml 15-prozentiger NaOH-Lösung und 12 ml Wasser behandelt.' Das Gemisch wird filtriert. Das Filtrat wird zur Trockne eingedampft. Der Rückstand wird mit Hexan verrieben. Man erhält W~3 , 3- (Hydroxymethyl )-propyl7-1 methoxymethyl-benzol. ~
Das Produkt' wird sodann mit 17,5 g PBr-. in 100 ml Äther
bei 00C 4 Stunden umgesetzt. Die Lösung wird filtriert un 15
mit Wasser gewaschen. Die Atherphase wird über Na2SOj4 getrocknet. Nach Entfernen des Äthers unter vermindertem Druck erhält man 1 -Γ~3 , 3-Di- (brommethyl )-propy_l7-1 -methoxymethylbenzol .
Der Rückstand wird sodann mit 2,4 g trockenen Magnesiumspänen in 300 ml trockenem Äther unter Stickstoff bei -100C umgesetzt. Sodann wird trockenes C0„ durch die Lö'su geperlt, bis keine Absorption mehr erfolgt. Anschliessend
wird die Lösung mit 25-prozentiger wässriger H„S0k neutralisiert und mit Äther extrahiert. Der nach Trocknen über Na2SO1, und Entfernen des Äthers unter vermindertem Druck erhaltene Rückstand wird mit 1 ml HpSO1. und 100 ml Äthanol verestert. Die Lösung wird neutralisiert und mit Äther extrahiert. Nach Trocknen über Na^SO1, und Entfernen des Lösungsmittels unter vermindertem Druck erhält man 1 -]_ 3,3-Di-(carboxymethyl)-propyl7-1-methoxymethylbenzol (p = 1).
(Herstellung von Acetoacetat-derivaten 5)
(A) Eine Lösung von 24 g (0,95 Mol) 4-(2,2-Dimethyl-1,3-dioxolan-4-yl)-methoxyphenyläthan-2-ol) (4) in 50 ml

Dimethoxyäthan und 1 ml Triäthylamin wird unter Rückfluss erwärmt und tropfenweise mit 8,0 ml Diketen versetzt. Nach beendeter Zugabe wird die Lösung 2 Stunden unter Rückfluss erwärmt. Die Lösungsmittel werden unter vermindertem Druck entfernt. Der Rückstand wird mit Wasser und Diäthyläther ausgeschüttelt. Die Ätherphase wird über Na-SOj, getrocknet und eingedampft. Der Rückstand wird durch Mitteldruck-Chromatographie an Kieselgel (50% Äther-Hexan) gereinigt. Man erhält 25 g 4-(2 , 2-Dimethyl-i., 3-dioxolan-4-yl)-methoxyphenyläthylacetoacetat (5) in Form eines farblosen Öls.
(B) In entsprechender Weise erhält man gemäss Abschnitt (A) unter Verwendung der gemäss den Präparationen 5 und 6 her-

Der nach Entfernen des Lösungsmittels unter vermindertem Druck erhaltene Rückstand wird durch Mitteldruck-Chromatographie an Kieselgel'(80% Äther-Hexan) gereinigt..Man erhält 21 g 2,6-Dimethyl-3-methoxycarbonyl-4-(3-nitrophenyl)-5-(2-/~4-(2,2-dimethyl-1,3- dioxolan-4-yl)-methoxyphenyl7-äthoxycarbonyl)-1,4-dihydropyridin (6) in Form eines orangefarbenen Öls.

(Herstellung einer Verbindung der Formel 1 nach Schema 1 )
Eine Lösung von 21 g 2,ö-Dimethyl-S-methoxycarbonyl-4-(3-nitrophenyl)-5-(2-_/~4-(2,2-dimethyl-1 ,3-dioxolan-4-yl)-methoxyphenyl7-äthoxycarbonyl)-1,4-dihydropyridin (6) i-n 150 ml Aceton und 50 ml Wasser wird mit 10 ml Salzsäure behandelt. Das Gemisch wird 6 Stunden unter Rückfluss erwärmt. Nach Zugabe von 500 ml Wasser wird das Gemisch mit Äther extrahiert. Die Stherphase wird über NapSOj. getrocknet und eingedampft. Das erhaltene Öl wird durch Mitteldruck-Chromatographie an Kieselgel (90% Essigsäureäthylester-Hexan) gereinigt. Man erhält 12 g 2,6-Dimethyl-3-methoxycarbonyl-4-(3-n-itrophenyl)-5.-(2-_/~4-(2 , 3-dihydroxypropoxy)-pheny_l7-äthoxycarbonyl )-1 ,4-dihydropyridin (1 ) in Form eines dicken Öls, das nach Verreiben mit Essigsäureäthylester kristallisiert; F. 117 bis 1180C. Berechnet für C27H30N2O9: C-6I,59, H-5,74, N-5,32. Gefunden: C-61,42, H-5,76, N-5,31.

Claims (1)
- Erfindunqsanspruch1. Verfahren zur Herstellung einer Verbindung der Formel . ·(CH2)(6)oder von pharmazeutisch verträglichen Säureadditionssalzen davon, worin η eine ganze Zahl von 0 bis 8 ist; Y -0-, -NH-, -NR2-, -S-, -S(O)-, -S(O)2- oder eine Bindung bedeutet;und R2 jeweils unabhängig voneinander A. A2, Ao oder..A1. bedeuten, wobeiA1 -(CHg)111(CHOG1) CH2OG2 bedeutet;A2 -(CH2)qCH(3_r)r"(CH2)s0G1_7r bedeutet;A0 -(CH-) CH,, v/~(CHo) COOR- 7 bedeutet; und 3 2 q (3-Γ)— 2 p 3— rA1. -(CH2) COOR, bedeutet; wobei m eine ganze Zahl von 1 bis 8 ist; ρ eine ganze Zahl von 0 bis 4 ist; q eine ganze Zahl von 0 bis 8 ist; r 2 oder 3 ist;s eine ganze Zahl von 1 bis 4 ist;R, H oder Alkyl mit 1 bis 18 Kohlenstoffatomen bedeutet;G- und Gp jeweils -H oder -CHpC^H,- bedeuten oder G1 und ein benachbartes G1 zusammen einen Rest der Formel-C(R )(R, )- oder G1
a D ιund G~ zusammen einen Rest der Formel-C(R )(R. )- bedeuten, wobei R und R, unabhängig voneinau a Dander nieder-Alkyl bedeuten, wobei mindestens einer der Reste G1 oder G0 nicht -H bedeutet;-NO2, -CF, oder Halogen bedeutet;2811.86- 390434- 33 -R5 nieder-Alkyl oder -CH2CH2OCH, bedeutet, gekennzeichnet dadurch, dass man eine Verbindung der Formelin der Y -0-, -NH-, -NR2-, -S-, -S(O)-, -S(O)2- oder eine Bindung bedeutet; R.J A.., Ap, A-, oder A1, bedeutet, wobei A1 -(CH2)m(CHOH) CH2OH bedeutet;A2- -(CH2)'CH (3_r ^"(CH2 )s0H_7r bedeutet;A -(CH2) CH (3_r )jf"(CH2) C00R3_7r bedeutet; und -(CH2)mCOOR3 bedeutet; wobeim eine ganze Zahl von 1 bis 8 ist; ρ eine ganze Zahl von 0 bis 4 ist; q eine ganze Zahl von 0 bis 8 ist; r 2 oder 3 ist;s eine ganze Zahl von 1 bis U ist; und R H oder Alkyl mit 1 bis 18 Kohlenstoffatomen bedeutet; und* eine Schutzgruppe bedeutet, mit einer Verbindung der FormelCHO 30in der R1. -NO2, -CF, oder Halogen bedeutet; und einer Verbindung der Formelin der R5 nieder-Alkyl oder -CH2CH2OCH3 bedeutet, umsetzt9R11RG- 390434
